Instant formation of nanopores on flexible polymer membranes using intense pulsed light and nanoparticle templates

The development of simple and high-throughput approaches to yield solid-state nanopores on large surface membranes may facilitate the prevalence of nanopore analysis technology and in-vitro diagnosis using portable devices.However,solidstate nanopores are typically realized by complex and highend nanofabrication equipments.Here,we present a method to achieve nanopores on polymer membranes using,silver nanoparticles(AgNPs)as templates and intense pulsed light(IPL)as a heating source.The density and size of nanopores are controllable by adjusting the spin coating rate,the concentration of nanoparticle suspension,and the size of nanoparticles(NPs).The temperature of the AgNPs can rapidly reach 1132 K under instant heating of photothermal effect through light irradiation in 2 ms,resulting in localized melting and decomposition of an underneath polycarbonate(PC)membrane to yield nanopores with sizes ranging from 10 to 270 nm.After removing the nanoparticle residues,the flexible membrane with nanopores can be integrated into a flow cell to achieve a nanopore sensor that has been used to measure the translocation behaviors of bovine serum albumin(BSA).The results have demonstrated the capability of the sensor in protein denaturation identification.This low-cost and highthroughput technique to fabricate solid-state nanopores on flexible polymeric membranes may facilitate the development of more nanopore-based flexible sensors that can be integrated with other flexible components for wearable diagnosis.

跨省界流域能否有效治理是关系我国生态环境健康发展的重要命题,也是困扰环境保护的痛点所在。跨省界流域在行政区边界治理成效不彰根源于科层治理的边界约束,即科层治理的范围、对象与治理能力的边界共同形构了有限的治理空间。面对跨省界流域治理的困境,地方政府努力拓展治理边界,探索出外部强制权力介入、建立区域管理机构、签订多边府际协议和构建多元自组织系统等四种路径。虽然取得了一定治理成效,但其尝试仍立足于内嵌的科层体制,无法克服科层治理的固有局限。因此,有必要利用现代信息技术,延伸科层治理范围,扩展科层治理对象,提高科层治理能力,用现代信息治理技术为科层体制赋能,促使科层制超越治理边界,才能在现有制度框架内实现流域边界良治善治。

Fragaria vesca,commonly known as wild or woodland strawberry,is the most widely distributed diploid Fragaria species and is native to Europe and Asia.Because of its small plant size,low heterozygosity,and relative ease of genetic transformation,F.vesca has been a model plant for fruit research since the publication of its Illumina-based genome in 2011.However,its genomic contribution to octoploid cultivated strawberry remains a long-standing question.Here,we de novo assembled and annotated a telomere-to-telomere,gap-free genome of F.vesca‘Hawaii 4’,with all seven chromosomes assembled into single contigs,providing the highest completeness and assembly quality to date.The gap-free genome is 220785082 bp in length and encodes 36173 protein-coding gene models,including 1153 newly annotated genes.All 14 telomeres and seven centromeres were annotated within the seven chromosomes.Among the three previously recognized wild diploid strawberry ancestors,F.vesca,F.iinumae,and F.viridis,phylogenomic analysis showed that F.vesca and F.viridis are the ancestors of the cultivated octoploid strawberry F.×ananassa,and F.vesca is its closest relative.Three subgenomes of F.×ananassa belong to the F.vesca group,and one is sister to F.viridis.We anticipate that this high-quality,telomere-to-telomere,gap-free F.vesca genome,combined with our phylogenomic inference of the origin of cultivated strawberry,will provide insight into the genomic evolution of Fragaria and facilitate strawberry genetics and molecular breeding.

锚筋桩在传统的边坡治理、地基加固等领域已经广泛使用,较于抗滑桩或抗滑墙等施工时具有设备轻便、桩位布置灵活、施工速度快、对场地的适应性强等特点,取得了较好的工程效果。但在一些地质条件复杂、岩层风化程度较高的破碎体,锚筋桩造孔出现了难以避免的塌孔和无法成孔的现象,若采用常规泥浆护壁和固结等方式将导致施工工序复杂、工期较长,且施工成本较高,而采用微型钢管桩的钻孔技术,可有效解决此类问题。通过对杨房沟水电站采用微型钢管桩技术的案例研究,成功解决大型崩坡积体处理中复杂地质条件下易塌孔、无法成孔的情况。

Genome-wide association analysis identified molecular markers associated with important tea flavor-related metabolites

The characteristic secondary metabolites in tea(theanine,caffeine,and catechins)are important factors contributing to unique tea flavors.However,there has been relatively little research on molecular markers related to these metabolites.Thus,we conducted a genome-wide association analysis of the levels of these tea flavor-related metabolites in three seasons.The theanine,caffeine,and catechin levels in Population 1 comprising 191 tea plant germplasms were examined,which revealed that their heritability exceeded 0.5 in the analyzed seasons,with the following rank order(highest to lowest heritabilities):(+)-catechin>(−)-gallocatechin gallate>caffeine=(−)-epicatechin>(−)-epigallocatechin-3-gallate>theanine>(−)-epigallocatechin>(−)-epicatechin-3-gallate>catechin gallate>(+)-gallocatechin.The SNPs detected by amplified-fragment SNP and methylation sequencing divided Population 1 into three groups and seven subgroups.An association analysis yielded 307 SNP markers related to theanine,caffeine,and catechins that were common to all three seasons.Some of the markers were pleiotropic.The functional annotation of 180 key genes at the SNP loci revealed that FLS,UGT,MYB,and WD40 domain-containing proteins,as well as ATP-binding cassette transporters,may be important for catechin synthesis.KEGG and GO analyses indicated that these genes are associated with metabolic pathways and secondary metabolite biosynthesis.Moreover,in Population 2(98 tea plant germplasm resources),30 candidate SNPs were verified,including 17 SNPs that were significantly or extremely significantly associated with specific metabolite levels.These results will provide a foundation for future research on important flavor-related metabolites and may help accelerate the breeding of new tea varieties.

A full-length c DNA library from leaf and root tissues of cassava(Manihot esculenta) Arg7 and one accession of its wild ancestor W14(M. esculenta ssp.flabellifolia) has been constructed. The library is comprised of four sub-libraries, containing 32640 recombinant clones, 6028 c DNA clones from their 5′ ends, and 128 clones from the 3′ ends were sequenced. In total, 5013high-quality expressed sequence tags(ESTs) and 1259 unigenes were obtained. Of these, 746 unigenes were identified by their sequence homologies to ESTs from model plants, and 323 unigenes were mapped onto 114 different KEGG pathways. From these, 24 differentially expressed genes involved in starch metabolism and photosynthesis were identified and five of them were selected to compare their expression level between Arg7 and W14. Notably, Arg7 has a higher net photosynthesis rate in leaves, higher ribulose-1,5-bisphosphate carboxylase oxygenase activities in leaves, and higher AGPase activity in roots. This resource is the first EST collection from wild cassava and should be of value for gene discovery, genome annotation and studies of Manihot evolution.

采用薄膜固相微萃取与气质联用技术对市场占有率较高的4种牛油火锅底料品牌的B端和C端产品的挥发性风味物质进行了比较分析,并构建主成分评分模型对其风味进行了差异性评价。结果表明,B端产品共检测有154种风味物质,C端产品则有158种,而8个样品检测出25种相同的风味物质,相同品牌的风味物质中,C端产品的烃类物质占比明显较低。利用主成分分析对火锅底料中的风味物质组成进行分析,结果表明,7个主成分因子累积方差贡献率达到100%,能有效表征火锅底料的风味物质信息。最后构建主成分分析模型,计算综合评价指数,对其风味进行评价排序,Pearson相关性分析发现主成分综合得分与感官评价结果相关性为极显著(P<0.01),相关系数为0.908,说明所构建的模型能够有效地对火锅底料进行风味评价。

Dengue virus(DENV) and Zika virus(ZIKV) have spread throughout many countries in the developing world and infect millions of people every year, causing severe harm to human health and the economy. Unfortunately, there are few effective vaccines and therapies available against these viruses. Therefore, the discovery of new antiviral agents is critical.Herein, a scorpion venom peptide(Smp76) characterized from Scorpio maurus palmatus was successfully expressed and purified in Escherichia coli BL21(DE3). The recombinant Smp76(rSmp76) was found to effectively inhibit DENV and ZIKV infections in a dose-dependent manner in both cultured cell lines and primary mouse macrophages. Interestingly,rSmp76 did not inactivate the viral particles directly but suppressed the established viral infection, similar to the effect of interferon(IFN)-b. Mechanistically, rSmp76 was revealed to upregulate the expression of IFN-b by activating interferon regulatory transcription factor 3(IRF3) phosphorylation, enhancing the type-Ⅰ IFN response and inhibiting viral infection.This mechanism is significantly different from traditional virucidal antimicrobial peptides(AMPs). Overall, the scorpion venom peptide Smp76 is a potential new antiviral agent with a unique mechanism involving type-Ⅰ IFN responses,demonstrating that natural AMPs can enhance immunity by functioning as immunomodulators.

Macadamia from the family Proteaceae is a plant native to Australia and has long been favoured by people for its crispy and high nutritional and medicinal value.Here,the genome of GUIRE 1(GR1),a highly heterozygous superior cultivar of macadamia nut,was sequenced and assembled using nanopore sequencing,and a 807-Mb genome(contig N50,1.9 Mb;scaffold N50,54.70 Mb)and 14 chromosomes were obtained.A total of 453 Mb(about 55.95%)repetitive sequences and 37,657 protein-coding genes were obtained by gene annotation and homologous protein comparison.Proteaceae diverged from Nelumbonaceae nearly 115.37 million years ago and from Rubiaceae about 140 million years ago.A genome-wide duplication(WGD)event occurred in macadamia 41 million years ago based on the WGD analysis.The functional enrichment analysis of M.integrifolia-specific gene families revealed their roles in signal transduction,protein phosphorylation,protein binding,and defense response.Here,a highly heterozygous genome of M.integrifolia was unlocked to provide a database for breeding and molecular mechanism research.
