Applying nanotechnology to boost cancer immunotherapy by promoting immunogenic cell death

Tumor immunotherapy,especially immune checkpoint blockade(ICB),has revolutionized the cancer field.However,the limited response of tumors to immunotherapy is a major obstacle.Tumor immunogenic cell death(ICD)is a death mode of tumor cells that can promote tumor immunity.ICD can induce strong an-titumor immune responses through the ectopic exposure of calreticulin on the plasma membrane surface and the release of the non-histone nuclear protein high-mobility group box 1(HMGB1),ATP,and in-terferon(IFN),thus activating an adaptive immune response against dead cell-associated antigens and enhancing the therapeutic effect of tumor immunotherapy.Chemotherapy,radiotherapy,photothermal therapy,magneto-thermodynamics therapy,nanopulse stimulation,and oncolytic virus therapy can all induce a strong antitumor immune response by ICD.In addition,the application of nanotechnology can precisely target drug delivery and improve the efficacy of immunotherapy.Here we introduce the basic concepts and molecular mechanisms underlying the induction of ICD.Then,we summarize and discuss the progress in the application of nanotechnology in immunotherapy to promote ICD.Finally,we attempt to define the challenges and future directions in this area to extend the benefits of ICD to a broader patient population.

Spatial-temporal variations of natural suitability of human settlement environment in the Three Gorges Reservoir Area A case study in Fengjie County,China

With rapid environmental degeneration and socio-economie development,the human settlement environment (HSE)has experienced dramatic changes and attracted attention from different communities.Consequently,the spatial-temporal evaluation of natural suitability of the human settlement environment (NSHSE)has become essential for understanding the patterns and dynamics of HSE,and for coordinating sustainable development among regional populations,resources,and environments.This study aims to explore the spatialtemporal evolution of NSHSE patterns in 1997,2005,and 2009 in Fengjie County near the Three Gorges Reservoir Area (TGRA).A spatially weighted NSHSE model was established by integrating multi-source data (e.g.,census data,meteorological data,remote sensing images,DEM data,and GIS data)into one framework,where the Ordinary Least Squares (OLS)linear regression model was applied to calculate the weights of indices in the NSHSE model.Results show that the trend of natural suitability has been first downward and then upward, which is evidenced by the disparity of NSHSE existing in the south,north,and central areas of Fengjie County. Results also reveal clustered NSHSE patterns for all 30 townships.Meanwhile,NSHSE has significant influence on population distribution,and 71.49% of the total population is living in moderate and high suitable districts.

Effect of ball milling time on the microstructure and compressive properties of the Fe–Mn–Al porous steel

In the present work,Fe–Mn–Al–C powder mixtures were manufactured by elemental powders with different ball milling time,and the porous high-Mn and high-Al steel was fabricated by powder sintering.The results indicated that the powder size significantly decreased,and the morphology of the Fe powder tended to be increasingly flat as the milling time increased.However,the prolonged milling duration had limited impact on the phase transition of the powder mixture.The main phases of all the samples sintered at 640℃ were α-Fe,α-Mn and Al,and a small amount of Fe2Al5 and Al8Mn5.When the sintering temperature increased to 1200℃,the phase composition was mainly comprised of γ-Fe and α-Fe.The weight loss fraction of the sintered sample decreased with milling time,i.e.,8.3wt% after 20 h milling compared to15.3wt% for 10 h.The Mn depletion region(MDR) for the 10,15,and 20 h milled samples was about 780,600,and 370 μm,respectively.The total porosity of samples sintered at 640℃ decreased from ~46.6vol% for the 10 h milled powder to ~44.2vol% for 20 h milled powder.After sintering at 1200℃,the total porosity of sintered samples prepared by 10 and 20 h milled powder was ~58.3vol% and ~51.3vol%,respectively.The compressive strength and ductility of the 1200℃ sintered porous steel increased as the milling time increased.

Effects of Mechanical Milling and Sintering Temperature on the Densification, Microstructure and Tensile Properties of the Fe–Mn–Si Powder Compacts

This work reported on the effects of mechanical milling and sintering temperature on the densification,microstructure and mechanical properties of the Fe–28Mn–3Si（wt%） alloy. Elemental Fe, Mn and Si powders were used as the starting materials, and two batches of powder mixture were prepared： one was blended elemental（BE） powder mixture; the other was mechanically milled（MM） powder mixture milled for5 h using planetary ball milling. Both powder mixtures were pressed under a uniaxial pressure of 400 MPa,and subsequently sintered in a high vacuum furnace for 3 h at 1000, 1100, 1200 and 1300 °C. It was found that Mn depletion region（MDR） was formed on the surface of all the sintered samples. The sintered BE compacts had a low density（〈68.2%） at all temperatures, while the density of the sintered MM compacts increased drastically from ~65% at 1000 °C to ~91% at 1300 °C. All the sintered MM compacts were composed of a predominant γ-austenite and minor ε-martensite. In comparison, additional（Fe, Mn）3Si phase was observed in the BE alloys sintered at 1000 °C, and a single α-Fe phase was identified in the BE compact sintered at 1300 °C. The tensile properties of the sintered MM compacts increased significantly with the temperature and were significantly higher than those of their BE counterparts.

The water snail Pomacea canaliculata retracts the discoidal and multi-layered operculum to protect the soft body from being attacked by predators,and releases it when threats lifted.However,the duration of the operculum retraction is usually less than that of the operculum protraction.In this paper,we elucidate the biological compliant mechanism of the operculum.By using confocal laser scanning microscopy,we find that the operculum has compliant sandwiched layers between hard layers.The layered structure results in a compliant mechanism with a bidirectional stiffness for the locking and unlocking processes of the operculum.A mathematical model is derived to rationalize the bidirectional stiffness mechanism of the operculum.In addition,we carry out the experiments on the locking and unlocking processes.The experimental results show that the locking tension is about two-fifths of the unlocking tension of the operculum.Moreover,based on the mechanical properties of the operculum with the layered structure,we designed an operculum-inspired structure,which may have a variety of potential applications in combined driving patterns.

Difference analysis of Doppler ultrasound blood flow of Cunkou(radial artery)pulse,Renying(carotid artery)pulse,and Fuyang(anterior tibial artery)pulse

OBJECTIVE:To analyze the Doppler ultrasound blood flow of Renying(carotid artery)pulse,Cunkou(radial artery)pulse,and Fuyang(anterior tibial artery)pulse in the normal group and the functional constipation with gastrointestinal heat(FCGH)group,and to compare and explore the differences of Renying,Cunkou and Fuyang pulses.METHODS:Sixty normal subjects and 60 patients with gastrointestinal heat constipation were collected in the department of ultrasound,Beijing Anzhen Hospital,Capital Medical University.Doppler ultrasound was used to observe the blood flow indexes including maximum systolic velocity(Vp),maximum diastolic velocity(Vd),mean velocity(Vm),pulse index(PI),resistance index(RI),vascular diameter(D),and circulation blood flow cycle(ET)of Renying pulse,Cunkou pulse and Fuyang pulse in the normal group and patients with gastrointestinal heat constipation.The differences of these three pulses were compared between the normal group and the FCGH group.RESULTS:The PI,Vp,Vm,RI,and D of the three pulses in the normal group were statistically significant different(P<0.01).In the comparison of Doppler flow diagram of three pulses in the FCGH group,there were significant differences in PI,Vd,Vp,Vm,RI,and D(P<0.01).CONCLUSION:Under normal conditions,the indexes of Renying pulse Doppler flow diagram,Cunkou pulse Doppler flow diagram,and Fuyang pulse Doppler flow diagram are significantly different.In the FCGH group,most of the indexes of Renying pulse Doppler flow diagram,Cunkou pulse Doppler flow diagram,and Fuyang pulse Doppler flow diagram are different.This result verified the necessity of simultaneous examinations of all Renying,Cunkou,and Fuyang pulses in modern Chinese medicine.

Analysis of mesoscopic mechanical dynamic characteristics of ballast bed with under sleeper pads

The meso-dynamical behaviour of a high-speed rail ballast bed with under sleeper pads(USPs)was studied.The geometrically irregular refined discrete element model of the ballast particles was constructed using 3D scanning techniques,and the 3D dynamic model of the rail-sleeper-ballast bed was constructed using the coupled discrete element method-multiflexible-body dynamics(DEM-MFBD)approach.We analyse the meso-mechanical dynamics of the ballast bed with USPs under dynamic load on a train and verify the correctness of the model in laboratory tests.It is shown that the deformation of the USPs increases the contact area between the sleeper and the ballast particles,and subsequently the number of contacts between them.As the depth of the granular ballast bed increases,the contact area becomes larger,and the contact force between the ballast particles gradually decreases.Under the action of the elastic USPs,the contact forces between ballast particles are reduced and the overall vibration level of the ballast bed can be reduced.The settlement of the granular ballast bed occurs mainly at the shallow position of the sleeper bottom,and the installation of the elastic USPs can be effective in reducing the stress on the ballast particles and the settlement of the ballast bed.

Single color image super-resolution using sparse representation and color constraint

Color image super-resolution reconstruction based on the sparse representation model usually adopts the regularization norm(e.g.,L1 or L2).These methods have limited ability to keep image texture detail to some extent and are easy to cause the problem of blurring details and color artifacts in color reconstructed images.This paper presents a color super-resolution reconstruction method combining the L2/3 sparse regularization model with color channel constraints.The method converts the low-resolution color image from RGB to YCbCr.The L2/3 sparse regularization model is designed to reconstruct the brightness channel of the input low-resolution color image.Then the color channel-constraint method is adopted to remove artifacts of the reconstructed highresolution image.The method not only ensures the reconstruction quality of the color image details,but also improves the removal ability of color artifacts.The experimental results on natural images validate that our method has improved both subjective and objective evaluation.

Impacts of communication delay on vehicle platoon string stability and its compensation strategy:A review

Based on the four-element model,this paper reviewed the important research progress in vehicle platoon,compared the advantages and disadvantages of different models in each element longitudinally,and summarized the linkage between each element horizontally.The stability criteria are briefly reviewed from three dimensions:local stability,string stability,and traffic flow stability.The impact of communication delay on vehicle platoon is quantitatively summarized from two aspects:the variation of controller gains and the variation of headway time values.Aiming at the inevitable communication delay in vehicle platoon,the compensation strategies are analyzed from five levels.(1)Optimizing the communication network structure.(2)Reconstructing acceleration information.(3)Tuning controller gains.(4)Constructing a multi-branch selection structure.(5)Improving the controller.The results show that,although these compensation strategies can alleviate the negative impact of communication delay to a certain extent,they also have some defects such as difficulty in adapting to complex and various real road conditions,poor accuracy and real-time performance,insufficient security,and limited application scenarios.It is necessary to further improve the accuracy and real-time performance of the device,design an encrypted and scalable network architecture to ensure communication security and adaptability,and conduct further real vehicle testing.

Can whole steps of grain production be outsourced? Empirical analysis based on the three provinces of Jiangsu,Jilin,and Sichuan in China

Certain outsourcing services for agricultural management in China,such as pest control in grain production,have experienced prolonged sluggishness,contrasting with the relatively high level of outsourcing services observed in harvesting,land preparation,and sowing.This study examines the feasibility of implementing whole-step outsourcing in grain production by conducting a case study of rice and maize production in Jiangsu,Jilin,and Sichuan provinces in China.The provision of outsourcing services hinges on two essential conditions:technological advancements fostering specialized production and economies of scale,coupled with a market size sufficient to realize the aforementioned potential economies of scale.The results showed that outsourcing pest control or harvesting services had varying economies of scale.The outsourcing services in pest control were less common than in harvesting services,and their marginal growth space of the economies of scale with technological change was also smaller.Determined by the operational characteristics of pest control itself,the market scale of its professional services is small.Therefore,achieving the whole-step outsourcing of grain production necessitates not only technological innovation but also effective policy interventions to overcome the constraints of market scale.Such interventions include(1)optimizing crop layouts between planning regions and reducing land fragmentation and(2)supplying timely and effective inter-regional agricultural information for service providers aided by information technology.
