Mutation breeding of high 4-androstene-3,17-dione-producing Mycobacterium neoaurum ZADF-4 by atmospheric and room temperature plasma treatment

Steroid medication is used extensively in clinical applications and comprises a large and vital part of the pharmaceutical industry. However, the difficulty of separating 4-androstene-3,17-dione （AD） from 1,4-androstadiene- 3,17-dione （ADD） restricts the application of the microbial transformation of phytosterols in the industry. A novel at- mospheric and room temperature plasma （ARTP） treatment, which employs helium as the working gas, was used to generate Mycobacterium neoaurum mutants producing large amounts of AD. After treatment of cultures with ARTP, four mutants were selected using a novel screening method with a color assay. Among the mutants, M. neoaurum ZADF-4 was considered the best candidate for industrial application. When the fermentation medium contained 15 g/L phytosterols and was cultivated on a rotary shaker at 160 r/min at 30 ~C for 7 d, （6.28＋0.11） g/L of AD and （0.82＋0.05） g/L of ADD were produced by the ZADF-4 mutant, compared with （4.83＋0.13） g/L of AD and （2.34＋0.06） g/L of ADD by the original strain, Iv~. neoaururn ZAD. Compared with ZAD, the molar yield of AD increased from 48.3% to 60.3% in the ZADF-4 mutant. This result indicates that ZADF-4 may have potential for industrial production of AD.

Phospholipase D(PLD)is an essential biocatalyst for the biological production of phosphatidylserine and phospholipid ***,the efficient heterologous expression of PLD is limited by its cell *** this study,a PLD was secretory expressed efficiently in Bacillus subtilis with an activity around 100 U/mL.A secretory expression system containing the signal peptide SPEstA and the dual-promoter PHpaII-SrfA was estab-lished,and the extracellular PLD activity further reached 119.22 U/mL through scale-up fermentation,191.30-fold higher than that of the *** optimum reaction conditions,a 61.61%conversion ratio and 21.07 g/L of phosphatidylserine production were ***,the synthesis system of PL derivates was established,which could efficiently synthesis novel PL *** results highlight that the secretory expression system constructed in this study provides a promising PLD producing strain in industrial application,and laid the foundation for the biosynthesis of phosphatidylserine and other PL *** far as we know,this work re-ports the highest level of extracellular PLD expression to date and the enzymatic production of several PL der-ivates for the first time.

The biological treatment of wastewater with high concentrations of ammonia nitrogen has become a hot research issue,but there are limited reports on the mechanism of ammonia nitrogen utilization by *** this paper,a transcriptomic approach was used to investigate the differences in gene expression at 500.0 mg/L(Amo 500)and 100.0 mg/L(Amo 100)ammonium concentrations to reveal the mechanism of ammonia nitrogen removal from water by Pseudomonas stutzeri *** transcriptome data showed 1015(459 up-regulated and 556 down-regulated)differentially expressed genes with functional gene annotation related to nitrogen source metabolism,glycolysis,tricarboxylic acid cycle,extracellular polysaccharide synthesis,energy conversion and transmembrane transport,revealing the metabolic process of ammonium nitrogen conversion to biological ni-trogen in *** F2 through *** verify the effect of ammonium transporter protein(AmtB)of cell membrane on assimilation,a *** F2-ΔamtB mutant strain was obtained by constructing a knockout plasmid(pK18mobsacB-ΔamtB),and it was found that the growth characteristics and ammonium removal rate of the mutant strain were significantly reduced at high ammonium *** carbon source components and dissolved oxygen conditions were optimized after analyzing the transcriptome data,and the ammonium removal rate was increased from 41.23%to 94.92%with 500.0 mg/L ammonium *** study of *** F2 transcript level reveals the mechanism of ammonia nitrogen influence on microbial assimilation process and improvement strategy,which provides a new strategy for the treatment of ammonia nitrogen wastewater.

An Immune-Inspired Approach with Interval Allocation in Solving Multimodal Multi-Objective Optimization Problems with Local Pareto Sets

In practical engineering,multi-objective optimization often encounters situations where multiple Pareto sets(PS)in the decision space correspond to the same Pareto front(PF)in the objective space,known as Multi-Modal Multi-Objective Optimization Problems(MMOP).Locating multiple equivalent global PSs poses a significant challenge in real-world applications,especially considering the existence of local *** identifying and locating both global and local PSs is a major *** tackle this issue,we introduce an immune-inspired reproduction strategy designed to produce more offspring in less crowded,promising regions and regulate the number of offspring in areas that have been thoroughly *** approach achieves a balanced trade-off between exploration and ***,we present an interval allocation strategy that adaptively assigns fitness levels to each *** strategy ensures a broader survival margin for solutions in their initial stages and progressively amplifies the differences in individual fitness values as the population matures,thus fostering better population ***,we incorporate a multi-population mechanism that precisely manages each subpopulation through the interval allocation strategy,ensuring the preservation of both global and local *** results on 21 test problems,encompassing both global and local PSs,are compared with eight state-of-the-art multimodal multi-objective optimization *** results demonstrate the effectiveness of our proposed algorithm in simultaneously identifying global Pareto sets and locally high-quality PSs.

Although pseudocapacitive manganese dioxide(MnO_(2))integrates the high-power merit of carbonaceous materials with the high-energy merit of battery-type materials,it still has a long way to go in achieving a more satisfactory balance of higher energy and power density,and in decoupling the relationship of structural characteristics with energy storage *** realize such goals,a bottom-up[WO_(6)]-perturbed[MnO_(6)]assembly strategy has been developed here due to their similar structure,yet mismatched lattice *** facile protocol is capable of finely controlling the morphology and crystal structure of MnO_(2)by adjusting its internal[WO_(6)]***,the as-prepared W_xMnO_(2)is treated as an ideal platform to scrutinize the correlations of the structure with the energy storage *** operando Raman spectra and finite element analysis have fully demonstrated the superiority of the locally ordered defects-enriched structure of W_(0.02)-MnO_(2),which could reach a favorable balance between the ion diffusion equilibrium time and the number of active *** a result,the W_(0.02)-MnO_(2)is able to deliver a high capacitance of 292 F·g^(-1)at a current density of 1 A·g^(-1)and a remarkable rate performance with a 60%capacity retention at a current density of 50 A·g^(-1).The further unveiled structure-performance relationship provides a guideline for the design of better pseudocapacitive energy storage devices.

The denoising of microseismic signals is a prerequisite for subsequent analysis and *** this research,a new microseismic signal denoising algorithm called the Black Widow Optimization Algorithm(BWOA)optimized VariationalMode Decomposition(VMD)jointWavelet Threshold Denoising(WTD)algorithm(BVW)is *** BVW algorithm integrates VMD and WTD,both of which are optimized by ***,this algorithm utilizes VMD to decompose the microseismic signal to be denoised into several Band-Limited IntrinsicMode Functions(BLIMFs).Subsequently,these BLIMFs whose correlation coefficients with the microseismic signal to be denoised are higher than a threshold are selected as the effective mode functions,and the effective mode functions are denoised using WTD to filter out the residual low-and intermediate-frequency ***,the denoised microseismic signal is obtained through *** ideal values of VMD parameters and WTD parameters are acquired by searching with BWOA to achieve the best VMD decomposition performance and solve the problem of relying on experience and requiring a large workload in the application of the WTD *** outcomes of simulated experiments indicate that this algorithm is capable of achieving good denoising performance under noise of different intensities,and the denoising performance is significantly better than the commonly used VMD and Empirical Mode Decomposition(EMD)*** BVW algorithm is more efficient in filtering noise,the waveform after denoising is smoother,the amplitude of the waveform is the closest to the original signal,and the signal-to-noise ratio(SNR)and the root mean square error after denoising are more *** case based on Fankou Lead-Zinc Mine shows that for microseismic signals with different intensities of noise monitored on-site,compared with VMD and EMD,the BVW algorithm ismore efficient in filtering noise,and the SNR after denoising is higher.

This study presents a probabilistic seismic risk model for the Beijing–Tianjin–Hebei region in China. The model comprises a township-level residential building exposure model, a vulnerability model derived from the Chinese building taxonomy,and a regional probabilistic seismic hazard model. The three components are integrated by a stochastic event-based method of the OpenQuake engine to assess the regional seismic risk in terms of average annual loss and exceedance probability curve at the city, province, and regional levels. The novelty and uniqueness of this study are that a probabilistic seismic risk model for the Beijing–Tianjin–Hebei region in China is developed by considering the impact of site conditions and epistemic uncertainty from the seismic hazard model.

Objective Ferritin,initially acting as an iron-storage protein,was found to be associated with metabolic *** study was designed to investigate the association between serum ferritin and metabolic-associated fatty liver disease(MAFLD)using data from the National Health and Nutrition Examination Survey(NHANES)of the United State of *** A cross-sectional study was conducted,enrolling a total of 2145 participants from the NHANES in the 2017–2018 *** steatosis and liver fibrosis were assessed by ultrasound images and several non-invasive *** regression analysis was conducted to determine the associations between serum ferritin concentration and MAFLD and liver *** The analysis revealed that participants with higher serum ferritin levels(Q3 and Q4 groups)had a higher prevalence of MAFLD than those with the lowest serum ferritin levels[Q3 vs.Q1:OR=2.17(1.33,3.53),P<0.05 in fatty liver index(FLI);Q4 vs.Q1:OR=3.13(1.91,5.13),P<0.05 in FLI].Additionally,participants with the highest serum ferritin levels(Q4 group)displayed a higher prevalence of liver fibrosis[Q4 vs.Q1:OR=2.59(1.19,5.62),P<0.05 in liver stiffness measurement;OR=5.06(1.12,22.94),P<0.05 in fibrosis-4 index],with significantly increased risk observed in participants with concomitant diabetes[OR=7.45(1.55,35.72),P=0.012].Conclusion Our study revealed that elevated serum ferritin levels are associated with a higher prevalence of MAFLD and advanced liver fibrosis in *** serum ferritin levels combined with diabetes are important risk factors for liver fibrosis.
