The concept and structure of serial-connection multi-chamber (SCMC) micropumps with cantilever valves is introduced. The SCMC micropump, which can be manufactured using conventional production techniques and materials, has a multi-layer circular planar structure. The border-upon piezoelectric actuators of a SCMC micropump work in anti-phase, as a result the pumping performance is similar to that of several single-chamber pumps running in series. The theoretical analysis shows that the pumping performance of a SCMC micropump depends not only on the characteristic and geometrical parameters of the piezoelectric actuators, but also on the number of pump chambers. Both flowrate and pressure of a SCMC pump can be enhanced to a certain extent. Four piezoelectric micropumps with different chambers were fabricated and tested. The testing results show that the enhancing extents of the flowrate and pressure of a SCMC piezoelectric micropump are different. The maximum flowrate and pressure of the four-chamber pump achieved are 2.5 times and 3.6 times those of the single-chamber pump achieved.

The persistence infection of low-risk type （type 6 or type 11） of human papillomavirus （HPV） is the main cause of genital warts. Given the high rate of recurrence after treatment, the use of a new molecular agent is certain to be of value. The aim of this study was to achieve targeted inactivation of viral E7 gene in keratinocytes using the reprogrammed clustered regularly interspaced short palindromic repeats （CRISPR）/CRISPR-associated （Cas） 9 system. To accomplish this, a universal CRISPR-Cas9 system for targeting both HPV6/11 E7 genes was constructed by using a dual guide RNA vector. After transfection of the vector into E7-transfromed keratinocytes, the expression level of E7 protein was measured using western-blot analysis and the sequence of the E7 gene was determined using Sanger sequencing. Cell proliferation was analyzed by CCK-8 assay, and cell apoptosis was evaluated by Hoechst 33258 staining, flow cytometry analysis and ELISA assay. The results indicated that both HPV6/11 E7 genes can be inactivated by the single CRISPR-Cas9 system. Furthermore, silencing of E7 led to inhibition of cell proliferation and induction of apoptosis in E7-transfromed keratinocytes but not in normal keratinocytes. Our data suggested that the reprogrammed CRISPR-Cas9 system has the potential for the development of an adjuvant therapy for genital warts.

Target tracking is one of the most important applications of wireless sensor networks. Optimized computation and energy dissipation are critical requirements to save the limited resource of sensor nodes. A new robust and energy-efficient collaborative target tracking framework is proposed in this article. After a target is detected, only one active cluster is responsible for the tracking task at each time step. The tracking algorithm is distributed by passing the sensing and computation operations from one cluster to another. An event-driven cluster reforming scheme is also proposed for balancing energy consumption among nodes. Observations from three cluster members are chosen and a new class of particle filter termed cost-reference particle filter （CRPF） is introduced to estimate the target motion at the cluster head. This CRPF method is quite robust for wireless sensor network tracking applications because it drops the strong assumptions of knowing the probability distributions of the system process and observation noises. In simulation experiments, the performance of the proposed collaborative target tracking algorithm is evaluated by the metrics of tracking precision and network energy consumption.

Effect of Climate Variability and Human Activities on Runoff in the Jinghe River Basin, Northwest China

Much attention has recently been focused on the effects of climate variability and human activities on the runoff. In this study, we analyzed 56-yr(1957–2012) runoff change and patterns in the Jinghe River Basin(JRB) in the arid region of northwest China. The nonparametric Mann–Kendall test and the precipitation-runoff double cumulative curve(PRDCC) were used to identify change trend and abrupt change points in the annual runoff. It was found that the runoff in the JRB has periodically fluctuated in the past 56 yr. Abrupt change point in annual runoff was identified in the JRB, which occurred in the years around 1964 and 1996 dividing the long-term hydrologic series into a natural period(1957 – 1964) and a climate and man-induced period(1965 – 1996 and 1997 – 2012). In the 1965 – 1996 period, human activities were the main factor that decreased runoff with contribution of 88.9%, while climate variability only accounted for 11.1%. However,the impact of climate variability has been increased from 11.1% to 47.5% during 1997 – 2012, showing that runoff in JRB is more sensitive to climate variability during global warming. This study distinguishes theeffect of climate variability from human activities on runoff, which can do duty for a reference for regional water resources assessment and management.
