The heavy metal cadmium(Cd) is detrimental to crop growth and threatens human health through the food chain. To cope with Cd toxicity,plants employ multiple strategies to decrease Cd uptake and its root-to-shoot translocation. However, genes that participate in the Cd-induced transcriptional regulatory network, including those encoding transcription factors, remain largely unidentified. In this study, we demonstrate that ENDO-BETA-MANNASE 7(MAN7) is necessary for the response of Arabidopsis thaliana to toxic Cd levels. We show that MAN7 is responsible for mannase activity and modulates mannose content in the cell wall, which plays a role in Cd compartmentalization in the cell wall under Cd toxicity conditions. Additionally,the repression of root growth by Cd was partially reversed via exogenous application of mannose,suggesting that MAN7-mediated cell wall Cd redistribution depends on the mannose ***, we identified a basic leucine zipper(bZIP) transcription factor, bZIP44, that acts upstream of MAN7 in response to Cd *** dual-luciferase assays indicated that bZIP44 directly binds to the MAN7 promoter region and activates its transcription. Loss of bZIP44 function was associated with greater sensitivity to Cd treatment and higher accumulation of the heavy metal in roots and ***, MAN7 overexpression relieved the inhibition of root elongation seen in the bzip44mutant under Cd toxicity conditions. This study thus reveals a pathway showing that MAN7-associated Cd tolerance in Arabidopsis is controlled by bZIP44 upon Cd exposure.

1-氨基环丙烷-1-羧酸氧化酶(1-aminocyclopropane-1-carboxylate oxidase, ACO)是乙烯合成过程中的关键限速酶之一。本研究以毛竹(Phyllostachys edulis)为材料，对PeACO基因家族进行全基因组鉴定及表达分析，以探究PeACOs在毛竹茎秆木质化进程中的影响。本研究通过与水稻(Oryza sativa) ACO基因序列比对，在毛竹基因组中鉴定得到11条具有完整2OG-FeII＿Oxy结构域的PeACOs序列；理化分析显示，PeACOs基因编码304～445个氨基酸，蛋白分子量为34 105.83～49 417.78 D；进化关系和基因结构分析显示，同一分支的基因对在基因结构和蛋白水平上保持高度的保守性；共线性分析得出ACO基因家族在毛竹中共有6对共线性基因，说明基因组复制事件对PeACOs基因数目扩张有重要影响。基因表达分析显示，PeACOs在毛竹各组织均有表达，尤其在快速生长期的笋中表达丰度最高；剥除处于快速生长期的毛竹箨鞘导致毛竹茎秆木质化程度加深，大部分PeACOs基因的表达量显著上调，且具有相似结构的基因对表达趋势相似，提示PeACO基因家族可能在木质化进程中发挥重要作用。本研究为深入探讨毛竹ACO基因家族功能提供了参考依据。

背景:针灸是缓解腰椎间盘突出症腰痛的有效方法,但其机制目前尚未明确。JAK2/STAT3信号通路相关因子可调节机体炎症反应,参与神经病理性疼痛的过程。目的:基于JAK2/STAT3信号通路研究针刺对腰椎间盘突出症模型大鼠的作用机制。方法:采用随机数字表法将40只SD大鼠分为假手术组、模型组、针刺组与针刺+激动剂组,每组10只。模型组、针刺组及针刺+激动剂组采用自体髓核移植法构建L5腰椎间盘突出症模型,造模3 d后,针刺组开始进行针刺治疗(作用于阳陵泉、肾俞、环跳、大肠俞等穴位),针刺+激动剂组造模后第6,12,18天针刺治疗前向L4/L5椎间隙鞘内注射JAK2激动剂香豆霉素A1,针刺治疗1次/d,20 min/次,连续治疗15 d。造模前及造模后3,6,9,12,15,18 d,检测大鼠机械缩足阈值;造模后18 d,检测血清炎症因子水平,苏木精-伊红染色观察L5-L6组织形态,RT-PCR检测L5-L6组织JAK2、STAT3 mRNA表达,Western Blot检测L5-L6组织JAK2、STAT3、p-JAK2及p-STAT3蛋白表达。结果与结论:①模型组大鼠造模后不同时间点的机械缩足阈值均低于假手术组(P<0.05),针刺组大鼠造模后9,12,15,18 d的机械缩足阈值均高于模型组(P<0.05),针刺+激动剂组大鼠造模后9,12,15,18 d的机械缩足阈值均低于针刺组(P<0.05);②与假手术组比较,模型组白细胞介素6、肿瘤坏死因子α、神经递质P物质、脑部神经肽Y水平均升高(P<0.05);与模型组比较,针刺组4种炎症因子水平均降低(P<0.05);与针刺组比较,针刺+激动剂组4种炎症因子水平均升高(P<0.05);③苏木精-伊红染色显示模型组大鼠腰椎退行性变化明显,针刺组与针刺+激动剂组大鼠腰椎退行性变化减轻,针刺组减轻更明显;④与假手术组比较,模型组JAK2与STAT3 mRNA表达、p-JAK2与p-STAT3蛋白表达均升高(P<0.05);与模型组比较,针刺组JAK2与STAT3 mRNA表达、p-JAK2与p-STAT3蛋白表达均降低(P<0.05);与针刺组比较,针刺+激动剂组JAK2与STAT3 mRNA表达、p-JAK2与p-STAT3蛋白表达均升高(P<0.05);⑤结果表明,针刺干预可通过降低腰椎间盘突出症模型大鼠的炎症反应来缓解疼痛,其作用机制可能与抑制JAK2/STAT3信号通路有关。

A micro-chip exploding foil initiator based on printed circuit board technology

Conventional exploding foil initiator (EFI) in ignition or detonation applications hosts many performance advantages, but was hindered by the bulky, inaccurate, inefficient and expensive shortcomings. We highlight a novel micro-chip exploding foil initiator (McEFI) using printed circuit board (PCB) technology. The structural parameters were determined based on energy coupling relationship at the component interfaces. Next, the prototype McEFI has been batch-fabricated using PCB technology, with a monolithic structure of 7.0 mm (l) × 4.5 mm (w) × 4.0 mm (δ). As expected, this PCB-McEFI illustrated the successful firing validations for explosives pellets. This paper has addressed the cost problem in both military munitions and civil markets wherever reliable, insensitive and timing-dependent ignition or detonation are involved.

Pairwise testing is an effective test generation technique that requires all pairs of parameter values to be by at least one test case. It has been proven that generating minimum test suite is an NP-complete problem covered Genetic algorithms have been used for pairwise test suite generation by researchers. However, it is always a time-consuming process, which leads to significant limitations and obstacles for practical use of genetic algorithms towards large-scale test problems. Parallelism will be an effective way to not only enhance the computation performance but also improve the quality of the solutions. In this paper, we use Spark, a fast and general parallel computing platform, to parallelize the genetic algorithm to tackle the problem. We propose a two-phase parallelization algorithm including fitness evaluation parallelization and genetic operation parallelization. Experimental results show that our algorithm outperforms the sequential genetic algorithm and competes with other approaches in both test suite size and computational performance. As a result, our algorithm is a promising improvement of the genetic algorithm for pairwise test suite generation.
