Two new C-22 steroidal lactone glycosides, named dracaenoside A and B were isolated from the methanol extract of the fresh stem of Dracaena cochinchinensis. Their structures were established as (20S)3b,14a,16b-trihydroxy pregn-5-ene-22-carboxylic acid (22,16)-lactone 3-O-a- L-rhamnopyranosyl (1→2)[ a-L-rhamnopyranosyl (1→4)]-b-D-glucopyranoside and (20S)3b, 14a, 16b-trihydroxy pregn-5-ene-22-carboxylic acid (22,16)-lactone 3-O-a-L-rhamnopyranosyl (1→2) [b-D-glucopyranosyl(1→3)]-b-D-glucopyranoside by means of 2D NMR spectral and chemical methods. It is the first time that steroidal lactone glycosides were isolated from the genus Dracaena.

The lack of axonal regeneration is the major cause of vision loss after optic nerve injury in adult mammals. Activating the PI3K/AKT/mTOR signaling pathway has been shown to enhance the intrinsic growth capacity of neurons and to facilitate axonal regeneration in the central nervous system after injury. The deletion of the mTOR negative regulator phosphatase and tensin homolog （PTEN） enhances regeneration of adult corticospinal neurons and ganglion cells. In the present study, we used a tyrosine-mutated （Y444F） AAV2 vector to efficiently express a short hairpin RNA （shRNA） for silencing PTEN expression in retinal ganglion cells. We evaluated cell survival and axonal regeneration in a rat model of optic nerve axotomy. The rats received an intravitreal injection of wildtype AAV2 or Y444F mutant AAV2 （both carrying shRNA to PTEN） 4 weeks before optic nerve axotomy. Compared with the wildtype AAV2 vector, the Y444F mutant AAV2 vector enhanced retinal ganglia cell survival and stimulated axonal regeneration to a greater extent 6 weeks after axotomy. Moreover,post-axotomy injection of the Y444F AAV2 vector expressing the shRNA to PTEN rescued ～19% of retinal ganglion cells and induced axons to regenerate near to the optic chiasm. Taken together, our results demonstrate that PTEN knockdown with the Y444F AAV2 vector promotes retinal ganglion cell survival and stimulates long-distance axonal regeneration after optic nerve axotomy. Therefore, the Y444F AAV2 vector might be a promising gene therapy tool for treating optic nerve injury.

AIM: To investigate the biological impacts of 'hot-spot'mutations on genotype B and C HBV X proteins (HBx).METHODS: Five types of'hot-spot' mutations of genotype B or C HBV X genes, which sequentially lead to the amino acid substitutions of HBx as I127T, F132Y, K130M+V131I,I127T+K130M+V131I, or K130M+V131I+F132Y, respectively,were generated by means of site-directed *** evaluate the anti-proliferative effects, HBx or related mutants' expression vectors were transfected separately to the Chang cells by lipofectamine, and the cells were cultured in hygromycin selective medium for 14 d, drug-resistant colonies were fixed with cold methanol, stained with Giemsa dyes and scored (increase of the colonies indicated the reduction of the anti-proliferation activity,and vice versa). Different types of HBx expression vectors were co-transfected separately with the reporter plasmid pCMVβ to Chang cells, which were lysed 48 h post-transfection and the intra-cellular β-galactosidase activities were monitored (increase of the β-galactosidase activities indicated the reduction of the transactivation activity, and vice versa). All data obtained were calculated by paired-samples ***: As compared to standard genotype B HBx,mutants of I127T and I127T+K130M+V131I showed higher transactivation and anti-proliferative activities, while the mutants of F132Y, K130M+V131I, and K130M+V131I+F132Y showed lower activities. As compared to standard genotype C HBx, I127T mutant showed higher transactivation activity, while the other four types of mutants showed no differences. The artificial endocrine system (AES) is a new branch of natural computing which uses ideas and takes inspiration from the information processing mechanisms contained in the mammalian endocrine system. It is a fast growing research field in which a variety of new theoretical models and technical methods have been studied for dealing with complex and significant problems. An overview of some recent advances in AES modeling and its applications is provided in this paper, based on the major and latest works. This review covers theoretical modeling, combinations of algorithms, and typical application fields. A number of challenges that can be undertaken to help move the field forward are discussed according to the current state of the AES approach.

A nano-laminated Y_(3)Si_(2)C_(2) ceramic material was successfully synthesized via an in situ reaction between YH_(2)and SiC using spark plasma sintering technology.A MAX phase-like ternary layered structure of Y_(3)Si_(2)C_(2) was observed at the atomic-scale by high resolution transmission electron *** lattice parameters calculated from both X-ray diffraction and selected area electron diffraction patterns are in good agreement with the reported theoretical *** nano-laminated fracture of kink boundaries,delamination,and slipping were observed at the tip of the Vickers *** elastic modulus and Vickers hardness of Y_(3)Si_(2)C_(2) ceramics(with 5.5 wt%Y_(2)O_(3)) sintered at 1500℃were 156 and 6.4 GPa,*** corresponding values of thermal and electrical conductivity were 13.7 W·m^(-1)·K^(-1) and 6.3×10^(5)S·m^(-1),respectively.
