Background:Muscle is the complex and heterogeneous tissue,which comprises the primary edible part of the trunk of fish and *** studies have shown that dietary isoleucine(Ile)exerts beneficial effects on growth in aquatic ***,there were limited studies regarding the benefits of Ile on fish muscle and their effects on flesh quality and muscle ***,this study was conducted to explore whether dietary Ile had affected flesh quality and muscle growth in hybrid bagrid catfish(Pelteobagrus vachelli♀×Leiocassis longirostris♂).Methods:A total of 630 hybrid fish,with an initial average body weight of 33.11±0.09 g,were randomly allotted into seven experimental groups with three replicates each,and respectively fed seven diets with 5.0,7.5,10.0,12.5,15.0,17.5,and 20.0 g Ile/kg diets for 8 ***:In the present study,we demonstrated that Ile significantly:(1)increased muscle protein and lipid contents and the frequency distribution of myofibers with≤20μm and≥50μm of diameter;(2)improved pH value,shear force,cathepsin B and L activities,hydroxyproline content,resilience,cohesiveness,and decreased cooking loss,lactate content,hardness,springiness,gumminess,and chewiness;(3)decreased reactive oxygen species(ROS),malondialdehyde(MDA),and protein carbonyl(PC)contents,GCLC and Keap1 mRNA levels,and up-regulated CuZnSOD,CAT,GPX1a,GST,and Nrf2 mRNA levels;(4)up-regulated the insulin-like growth factor 1,2(IGF-1,IGF-2),insulin-like growth factor 1 receptor(IGF-1R),proliferating cell nuclear antigen(PCNA),Myf5,Myod,Myog,Mrf4,and MyHC mRNA levels,and decreased MSTN mRNAlevel;(5)increased muscle protein deposition by activating AKT-TOR-S6K1 and AKT-FOXO3a signaling ***:These results revealed that dietary Ile improved flesh quality,which might be due to increasing nutritional content,physicochemical,texture parameters,and antioxidant ability;promoting muscle growth by affecting myocytes hyperplasia and hypertrophy,and muscle protein depos

Hens are raised apart from roosters in modern poultry production, a substantial change from their natural social structure. We compared productivity, injuries, behavior, physiology, microbiome and transcriptome of hens housed with(R+) or without(R–)roosters to quantify the effects of this change in social structure. Hens were raised free-range from 70 to 280 days when 30 birds per treatment were assigned to battery cages until Day 315(R+C vs. R–C), while 30 birds per treatment remained in free-range pens(R+F vs. R–F). Response to a novel environment and object, behavioral time budgets, cecum microbiome, blood composition and transcriptomic sequencing of thigh muscle and spleen were analyzed. Hens housed without roosters showed better survival, consumed less food, produced more eggs and had better feed conversion. R+F hens clustered around the rooster and were less mobile in the novel environment and object tests. R+F hens displayed the richest microbiome, and the presence of roosters resulted in differentially expressed genes related to muscle development, cellular processes, environmental information processing and immune function. Removing roosters from housed hens intensified desirable characteristics favored by domestication probably operating by deprivation of mating behavior and reduced fear, along with altered microbial and genetic function.

Assessment of structural brain changes in patients with type 2 diabetes mellitus using the MRI-based brain atrophy and lesion index

Patients with type 2 diabetes mellitus(T2 DM) often have cognitive impairment and structural brain *** magnetic resonance imaging(MRI)-based brain atrophy and lesion index can be used to evaluate common brain changes and their correlation with cognitive function,and can therefore also be used to reflect whole-brain structural changes related to T2 DM.A total of 136 participants(64 men and 72 women,aged 55–86 years) were recruited for our study between January 2014 and December *** participants underwent MRI and Mini-Mental State Examination assessment(including 42 healthy control,38 T2 DM without cognitive impairment,26 with cognitive impairment but without T2 DM,and 30 T2 DM with cognitive impairment participants).The total and sub-category brain atrophy and lesion index scores in patients with T2 DM with cognitive impairment were higher than those in healthy *** in the brain atrophy and lesion index of gray matter lesions and subcortical dilated perivascular spaces were found between non-T2 DM patients with cognitive impairment and patients with T2 DM and cognitive *** adjusting for age,the brain atrophy and lesion index retained its capacity to identify patients with T2 DM with cognitive *** findings suggest that the brain atrophy and lesion index,based on T1-weighted and T2-weighted imaging,is of clinical value for identifying patients with T2 DM and cognitive *** matter lesions and subcortical dilated perivascular spaces may be potential diagnostic markers of T2 DM that is complicated by cognitive *** study was approved by the Medical Ethics Committee of University of South China(approval ***20131109003) on November 9,2013,and was retrospectively registered with the Chinese Clinical Trial Registry(registration *** CTR1900024150) on June 27,2019.

目的研究健脾降脂方针刺对脾虚湿盛所致肥胖病患者的临床疗效及对患者空腹血脂的影响。方法采用随机对照原则,分为治疗组62例、对照组64例。治疗组采用健脾降脂方(内庭、公孙、足三里、中脘、丰隆)加减针刺治疗,对照组采用"脐周八穴"(双侧滑肉门、天枢、外陵及水分、阴交)针刺治疗。10 d为1个疗程,治疗3个疗程。观察其疗效及治疗前后患者体重、体重指数(BMI)、腰围、臀围、空腹血脂的变化情况。结果①2组自身比较治疗前后患者的体重、BMI、腰围、臀围均具有统计学差异(P<0.05);治疗组治疗前后血清总胆固醇(TC)、甘油三酯(TG)具有统计学差异(P<0.05)。②2组临床总有效率具有统计学差异(P<0.05)。③组间比较治疗后2组的体重、臀围具有统计学差异(P<0.05),BMI具有显著统计学差异(P<0.01);TC、TG具有统计学差异(P<0.05)。结论健脾降脂方针刺治疗脾虚湿盛所致肥胖较"脐周八穴"具有更好的疗效,主要体现在减少体重、BMI、臀围,以及TC、TG的降低方面。

AIM： To explore effects of huoxiangzhengqi liquid （HXZQ） on enteric mucosal immune responses in mice with Bacillus dysenteriae and Salmonella typhimurium induced diarrhea （BSD）. METHODS： BSD was induced in Balb/c mice by oral administration with Bacillus dysenteriae and Salmonella typhimurium. HXZQ was administrated from the day of diarrhea induction at dosages of 5.21 g/kg and 0.52 g/kg, respectively. The onset of diarrhea and lasting time were recorded. Peyer＇s patches and peripheral lymphocytes were prepared for flow cytometry, and levels of TNF-α in peripheral blood and enteric tissue homogenates were determined with ELISA. Student＇s t test was employed for statistics. RESULTS： Mice in BSD group started showing continuous diarrhea on the day of induction until the fourth day when they were sacrificed. Diarrhea in the mice of HXZQ high and low dose groups lasted for 36 and 54 h, respectively. There were more CD4^＋ and CD8^＋ cells in peripheral blood, fewer CD4^＋ cells in Peyer＇s patches in BSD mice compared to normal mice. Fewer CD4^＋ and CD8^＋ cells was shown in the mice in HXZQ high group compared to BSD mice. In Peyer＇s patch, there were more CD8^＋ cells in mice in HXZQ high and low dose groups and more CD4^＋ in mice in HXZQ high group. Higher levels of TNF-α in peripheral blood and intestinal tissue homogenates in BSD group were observed. Mice in HXZQ high group showed decreased levels of TNF-α in peripheral blood and enteric tissue homogenates. CONCLUSION： The immune regulation of CD4^＋ and CD8^＋ cells in Peyer＇s patch and suppression of TNF-α levels in enteric homogenates may partially explain the effect of HXZQ on improvement of BSD.

Low-molecular-weight photoresponsive supramulecular hydrogel based on a dicationic azobenzene-bridged pyridinium hydrogelator

Low-molecular-weight supramolecular hydrogels are of significant attractive soft materials as particular functions can be facilely introduced by the straightforward fabrication of such self-assembled systems. In this study, an azobenzene-bridged dicationic pyridinium salt was synthesized, from which photoresponsive supramolecular hydrogel could be fabricated through the π-π stacking and hydrophobic interactions in the aqueous solution. By taking advantages of the UV-vis light induced E/Z photoisomerization behaviors of the incorporated azobenzene photochromophore, reversible gel-sol transformation of such supramolecular hydrogel could be achieved under the alternating UV-vis irradiation conditions. We believed that this photoresponsive supramolecular hydrogel will be a good supplementary in the creation of intelligent soft material.

Influence of Interspecies interactions on bacterial community assembly in the active and permafrost layers on the Qinghai-Tibet Plateau

Bacterial communities play a crucial role in permafrost biogeochemical cycling and ecosystem function maintenance. Bacterial interaction is one of the main factors in shaping soil bacterial communities. However， how would bacterial interaction influence the bacterial communities in permafrost of the Qinghai–Tibet Plateau (QTP) remains largely unknown. Here we collected paired soil samples from both the active and permafrost layers of two typical QTP permafrost regions in October 2020 for Tuotuohe River (TTH) and May 2022 for Aerjin (ARJ)， and investigated the bacterial communities and the role of interactions in structuring the bacterial community and its assembly process through amplicon sequencing of the 16S rRNA gene. Our study revealed distinct bacterial communities， with significant differences in the relative abundances of Proteobacteria (P<0.05)， Acidobacteriota (P<0.001)， Bacteroidota (P<0.05)， and Planctomycetota (P<0.001) between the active layer and the permafrost layer. More importantly， we found that interspecies interactions， including both positive and negative associations， were strongly correlated with bacterial alpha-diversity and played a significant role in community variation and assembly process. Our findings also showed that the community assembly in both the active and permafrost layers was primarily driven by homogeneous selection of deterministic processes， with interspecies interactions accounting for more than 58% and 63% of all assembly mechanisms， respectively. This is the first study to quantify the contribution of bacterial interactions in shaping the bacterial community and its assembly process in permafrost of QTP， highlighting the importance of considering interspecies interactions in future modeling efforts. Our work also emphasizes the necessity of including interspecies interactions in microbial process projections to reduce uncertainty.

Background and Objectives The relationship between left atrial(LA)size and congestive heart failure(CHF)is well recognized;however,there is little information on the association of pulmonary vein(PV)diameter and *** purpose of this study was to investigate the changes of PV and LA sizes in CHF patients by multislice computed tomography(MSCT)angiography using a new 64-slice *** and Results We assessed diameters of PVs ostium and LA by 64-slice MSCT with three-dimensional reconstruction in 25 CHF patients and in 26 age-and sex-matched non-CHF *** with controls,CHF patients showed significant greater diameters of left superior pulmonary vein(LSPV)and right inferior pulmonary vein(RIPV)in both anteriorposterior(AP)and superior-inferior(SI)directions(P<0.01),significant dilation of right superior pulmonary vein(RSPV)in AP direction(P<0.05),as well as significant increase of LA transverse,AP,and SI diameters(P<0.01).Conclusion Significant dilation of PVs with simultaneous LA enlargement was demonstrated in CHF *** anatomic and geometric changes may participate in the perpetuation of AF.
