Experimental study of solid-liquid origami composite structures with improved impact resistance

In this paper,a liquid-solid origami composite design is proposed for the improvement of impact resistance.Employing this design strategy,Kresling origami composite structures with different fillings were designed and fabricated,namely air,water,and shear thickening fluid(STF).Quasi-static compression and drop-weight impact experiments were carried out to compare and reveal the static and dynamic mechanical behavior of these structures.The results from drop-weight impact experiments demonstrated that the solid-liquid Kresling origami composite structures exhibited superior yield strength and reduced peak force when compared to their empty counterparts.Notably,the Kresling origami structures filled with STF exhibited significantly heightened yield strength and reduced peak force.For example,at an impact velocity of 3 m/s,the yield strength of single-layer STF-filled Kresling origami structures increased by 772.7%and the peak force decreased by 68.6%.This liquid-solid origami composite design holds the potential to advance the application of origami structures in critical areas such as aerospace,intelligent protection and other important fields.The demonstrated improvements in impact resistance underscore the practical viability of this approach in enhancing structural performance for a range of applications.

A modified single edge V-notched beam method for evaluating surface fracture toughness of thermal barrier coatings

The surface fracture toughness is an important mechanical parameter for studying the failure behavior of air plasma sprayed(APS)thermal barrier coatings(TBCs).As APS TBCs are typical multilayer porous ceramic materials,the direct applications of the traditional single edge notched beam(SENB)method that ignores those typical structural characters may cause errors.To measure the surface fracture toughness more accurately,the effects of multilayer and porous characters on the fracture toughness of APS TBCs should be considered.In this paper,a modified single edge V-notched beam(MSEVNB)method with typical structural characters is developed.According to the finite element analysis(FEA),the geometry factor of the multilayer structure is recalculated.Owing to the narrower V-notches,a more accurate critical fracture stress is obtained.Based on the Griffith energy balance,the reduction of the crack surface caused by micro-defects is corrected.The MSEVNB method can measure the surface fracture toughness more accurately than the SENB method.

Background:Although immune checkpoint inhibitors(ICIs)against programmed cell death protein 1(PD-1)and its ligand PD-L1 have demonstrated potency towards treating patients with non-small cell lung carcinoma(NSCLC),the potential association between Kirsten rat sarcoma viral oncogene homolog(KRAS)oncogene substitutions and the efficacy of ICIs remains unclear.In this study,we aimed to find point mutations in the KRAS gene resistant to ICIs and elucidate resistance mechanism.Methods:The association between KRAS variant status and the efficacy of ICIs was explored with a clinical cohort(n=74),and confirmed with a mouse model.In addition,the tumor immune microenvironment(TIME)of KRASmutant NSCLC,such as CD8+tumor-infiltrating lymphocytes(TILs)and PD-L1 level,was investigated.Cell lines expressing classic KRAS substitutions were used to explore signaling pathway activation involved in the formation of TIME.Furthermore,interventions that improved TIME were developed to increase responsiveness to ICIs.Results:We observed the inferior efficacy of ICIs in KRAS-G12D-mutant NSCLC.Based upon transcriptome data and immunostaining results from KRAS-mutant NSCLC,KRAS-G12D point mutation negatively correlated with PD-L1 level and secretion of chemokines CXCL10/CXCL11 that led to a decrease in CD8+TILs,which in turn yielded an immunosuppressive TIME.The analysis of cell lines overexpressing classic KRAS substitutions further revealed that KRAS-G12D mutation suppressed PD-L1 level via the P70S6K/PI3K/AKT axis and reduced CXCL10/CXCL11 levels by down-regulating high mobility group protein A2(HMGA2)level.Notably,paclitaxel,a chemotherapeutic agent,upregulated HMGA2 level,and in turn,stimulated the secretion of CXCL10/CXCL11.Moreover,PD-L1 blockade combined with paclitaxel significantly suppressed tumor growth compared with PD-L1 inhibitor monotherapy in a mouse model with KRAS-G12D-mutant lung adenocarcinoma.Further analyses revealed that the combined treatment significantly enhanced the recruitment of CD8+TILs via the up-regulation

Dear Editor,Lung cancer still remains the leading cause of cancer death worldwide,80-90%of which are non-small cell lung cancer(NSCLC).In 2-3%of NSCLC patients,mutations in human epidermal growth factor receptor 2(HER2)have been identified as oncogenic drivers.However,HER2 has been poorly reported as a therapeutic target for advanced NSCLC despite of the outstanding improvements in breast cancer receiving HER2-targeting regimens.

Metabolic reprogramming in triple-negative breast cancer

Since triple-negative breast cancer(TNBC)was first defined over a decade ago,increasing studies have focused on its genetic and molecular characteristics.Patients diagnosed with TNBC,compared to those diagnosed with other breast cancer subtypes,have relatively poor outcomes due to high tumor aggressiveness and lack of targeted treatment.Metabolic reprogramming,an emerging hallmark of cancer,is hijacked by TNBC to fulfill bioenergetic and biosynthetic demands;maintain the redox balance;and further promote oncogenic signaling,cell proliferation,and metastasis.Understanding the mechanisms of metabolic remodeling may guide the design of metabolic strategies for the effective intervention of TNBC.Here,we review the metabolic reprogramming of glycolysis,oxidative phosphorylation,amino acid metabolism,lipid metabolism,and other branched pathways in TNBC and explore opportunities for new biomarkers,imaging modalities,and metabolically targeted therapies.

Dear Editor,In recent years,the rapid development of immunotherapy has brought survival benefits for lung squamous cell carcinoma(LUSC)patients who have rare treatment options.Nevertheless,there are still some patients with immunotherapy resistance,which is the root cause of the low benefit rate and disease progression of the overall population,and it is an urgent problem to be solved.Except for tumor mutation burden,the tumor immune microenvironment(TIME)as characterized by infiltration of CD8+tumor-infiltrating lymphocytes(TILs)and PD-L1 expression has been associated with the efficacy of immune checkpoint inhibitors(ICIs).In our previous studies,we have demonstrated that psoriasin,also known as S100 calcium-binding protein A7(S100A7),not only accelerates tumor progression via the activation of the p-Erk pathway in LUSC and the p-Erk and p-FAK pathways in esophageal squamous cell carcinomas(ESCC)but also remodels the tumor microenvironment by promoting angiogenesis and M2 macrophage infiltration in ESCC.1,2 However,the role of S100A7 in the TIME and immunotherapy efficacy in LUSC remains to be elucidated.

The integrity of lysosomes is of vital importance to survival of tumor cells.We demonstrated that LW-218,a synthetic flavonoid,induced rapid lysosomal enlargement accompanied with lysosomal membrane permeabilization in hematological malignancy.LW-218-induced lysosomal damage and lysosome-dependent cell death were mediated by cathepsin D,as the lysosomal damage and cell apoptosis could be suppressed by depletion of cathepsin D or lysosome alkalization agents,which can alter the activity of cathepsins.Lysophagy,was initiated for cell self-rescue after LW-218 treatment and correlated with calcium release and nuclei translocation of transcription factor EB.LW-218 treatment enhanced the expression of autophagy-related genes which could be inhibited by intracellular calcium chelator.Sustained exposure to LW-218 exhausted the lysosomal capacity so as to repress the normal autophagy.LW-218-induced enlargement and damage of lysosomes were triggered by abnormal cholesterol deposition on lysosome membrane which caused by interaction between LW-218 and NPC intracellular cholesterol transporter 1.Moreover,LW-218 inhibited the leukemia cell growth in vivo.Thus,the necessary impact of integral lysosomal function in cell rescue and death were illustrated.

预测干部容错:效率考量、合法性压力与领导特征——基于机器学习方法的预测建模

国家对于干部容错机制的诉求愈发强烈,但是近年来该政策在国内的扩散速度逐步放缓。要解决上述问题,关键在于研究干部容错机制的扩散逻辑。在政策创新扩散逻辑相关研究中,既有研究方法大多遵循“样本内解释”的思路。这使得相关研究结论的科学性,尤其是泛化能力,难以得到保证。为突破这一局限,本文采用“样本外预测”的思路来探索干部容错机制的扩散逻辑。具体来看,本文基于政策创新扩散理论构建了分析框架,使用机器学习方法训练了地方政府采纳干部容错机制的预测模型,并在确保模型预测性能的前提下呈现干部容错机制的扩散逻辑。模型解读结果表明,干部容错机制的扩散主要由行动者逻辑主导,其次是效率逻辑,最后是合法性逻辑;效率逻辑、合法性逻辑及行动者逻辑三个维度中,最具影响力的特征分别为治理规模、同级采纳和主官任期。地方政府采纳干部容错机制的概率与治理规模具有负向关系,与同级采纳具有正向关系,与主官任期具有倒U形关系。为推进干部容错机制有序发展,本文建议地方政府强化机制扩散中的领导驱动效应、科学规划干部容错机制扩散路径。
