Genetic mutations associated with sensitivity to neoadjuvant chemotherapy in metastatic colon cancer:A case report and review of literature

BACKGROUND Colorectal liver metastases(CLM)occur in 15%-30%of patients with colorectal cancer(CRC).Advancements in next generation sequencing(NGS)can provide more precise prognoses for cancer patients and help guide clinical ***,the genetic variants that predict high sensitivity to neoadjuvant chemotherapy remain unclear,especially in patients with *** aim of this study was to identify the relevant genetic variants in a single CLM patient and to summarize the current evidence on mutations and single nucleotide polymorphisms(SNPs)that objectively predict sensitivity to neoadjuvant *** SUMMARY A 76-year-old male patient,who was diagnosed as stage IV colon cancer with liver metastases,was found to have APC/TP53/KRAS *** showed a good therapeutic response to 12 courses of oxaliplatin regimens combined with *** analysis of the patient identified 5 genes with 7 detected SNPs that may be related to a better response to chemotherapy *** addition,a critical literature review was performed based on a standardized appraisal form after selecting the ***,21 eligible studies were appraised to assess the association between gene mutations and good *** in KRAS,TP53,SMAD4,and APC were identified as being associated with a poor response to chemotherapy drugs,whereas mutations of CREBBP and POLD1 were associated with longer overall *** NGS can identify precise predictors of response to neoadjuvant chemotherapy,leading to improved outcomes for CRC patients.

BACKGROUND There is growing evidence proving that many human carcinomas, including colon cancer, can overexpress immunoglobulin(Ig); the non B cancer cell-derived Ig usually displayed unique V(D)J rearrangement pattern that are distinct from B cell-derived Ig. Especially, the cancer-derived Ig plays important roles in cancer initiation, progression, and metastasis. However, it still remains unclear if the colon cancer-derived Ig can display unique V(D)J pattern and sequencing, which can be used as novel target for colon cancer *** To investigate the Ig repertoire features expressed in human colon cancer *** Seven cancerous tissue samples of colon adenocarcinoma and corresponding noncancerous tissue samples were sorted by fluorescence-activated cell sorting using epithelial cell adhesion molecule as a marker for epithelial cells. Ig repertoire sequencing was used to analyze the expression profiles of all 5 classes of Ig heavy chains(IgH) and the Ig repertoire in colon cancer cells and corresponding normal epithelial *** We found that all 5 IgH classes can be expressed in both colon cancer cells and normal epithelial cells. Surprisingly, unlike the normal colonic epithelial cells that expressed 5 Ig classes, our results suggested that cancer cells most prominently express IgG. Next, we found that the usage of Ig in cancer cells caused the expression of some unique Ig repertoires compared to normal cells. Some VH segments, such as VH3-7, have been used in cancer cells, and VH3-74 was frequently present in normal epithelial cells. Moreover, compared to the normal cell-derived Ig, most cancer cell-derived Ig showed unique VHDJH ***, even if the same VHDJH pattern was seen in cancer cells and normal cells, cancer cell-derived IgH always displayed distinct hypermutation hot *** We found that colon cancer cells could frequently express IgG and unique IgH repertoires, which may be involved in carcinogenesis of colon

A Simplified Method for the Stress Analysis of Underground Transfer Structures Crossing Multiple Subway Tunnels

According to the design specifications,the construction of extended piles involves traversing the tunnel’s upper region and extending to the underlying rock *** address this challenge,a subterranean transfer structure spanning multiple subway tunnels was *** on the function of piles in the transfer structure as springs with axial and bending stiffness,and taking into account the force balance and deformation coordination conditions of beams and plates within the transfer structure,we established a simplified mechanical model that incorporates soil stratification by combining it with the Winkler elastic foundation beam *** resolved established simplifiedmechanicalmodel employed finite difference technology and the Newton-Simpsonmethod,elucidating the mechanical mechanism of the transfer *** research findings suggest that the load carried by the upper structural columns can be transferred to the pile foundation beneath the beams through the transfer structure,subsequently reaching the deep soil layer and ensuring minimal impact on adjacent *** established simplified analysis method can be used for stress analysis of the transfer structure,concurrently considering soil stratification,pile foundation behavior,and plate *** pile length,pile section size,and beam section size within the transfer structure should account for the characteristics of the upper load,ensuring an even distribution of the beam bending moment.

FPGA implementation of 500-MHz high-count-rate high-time-resolution real-time digital neutron-gamma discrimination for fast liquid detectors

Fast neutron flux measurements with high count rates and high time resolution have important applications in equipment such as *** this study,real-time neutron and gamma discrimination was implemented on a self-developed 500-Msps,12-bit digitizer,and the neutron and gamma spectra were calculated directly on an FPGA.A fast neutron flux measurement system with BC-501A and EJ-309 liquid scintillator detectors was developed and a fast neutron measurement experiment was successfully performed on the HL-2 M tokamak at the Southwestern Institute of Physics,*** experimental results demonstrated that the system obtained the neutron and gamma spectra with a time accuracy of 1 *** count rates of up to 1 Mcps,the figure of merit was greater than 1.05 for energies between 50 keV and 2.8 MeV.

Novel wood-plastic composite fabricated via modified steel slag:Preparation,mechanical and flammability properties

A novel method was developed to enhance the utilization rate of steel slag(SS).Through treatment of SS with phosphoric acid and aminopropyl triethoxysilane(KH550),we obtained modified SS(MSS),which was used to prepare MSS/wood-plastic composites(MSS/WPCs)by replacing talcum powder(TP).The composites were fabricated through melting blending and hot *** mechanical and combustion properties,which comprise heat release,smoke release,and thermal stability,were systematically *** can improve the mechanical strength of the composites through grafting reactions between wood powder and ***,MSS/WPC#50(16wt%MSS)with an MSS-to-TP mass ratio of 1:1 exhibited optimal comprehensive *** with those of WPC#0 without MSS,the tensile,flexural,and impact strengths of MSS/WPC#50 were increased by 18.5%,12.8%,and 18.0%,***,the MSS/WPC#50 sample achieved the highest limited oxygen index of 22.5%,the highest vertical burning rating at the V-1 level,and the lowest horizontal burning rate at 44.2 mm/*** formation of a dense and stable char layer led to improved thermal stability and a considerable reduction in heat and smoke releases of MSS/WPC#***,the partial replacement of TP with MSS slightly compromised the mechanical and flame-retardant properties,possibly due to the weak grafting caused by SS powder *** findings suggest the suitability of MSS/WPCs for high-value-added applications as decorative panels indoors or outdoors.

Controllable Condensation of Aromatics and Its Mechanisms in Carbonization

In order to obtain liquefied products with higher yields of aromatic molecules to produce mesophase pitch,a good understanding of the relevant reaction mechanisms is *** molecular dynamics simulations were used to study the thermal reactions of pyrene,1-methylpyrene,7,8,9,10-tetrahydrobenzopyrene,and mixtures of pyrene with 1-octene,cyclohexene,or *** reactant conversion rates,reaction rates,and product distributions were calculated and compared,and the mechanisms were analyzed and *** results demonstrated that methyl and naphthenic structures in aromatics might improve the conversion rates of reactants in hydrogen transfer processes,but their steric hindrances prohibited the generation of high *** naphthenic structures could generate more free radicals and presented a more obvious inhibition effect on the condensation of polymers compared with the methyl side *** was discovered that when different olefins were mixed with pyrene,1-octene primarily underwent pyrolysis reactions,whereas cyclohexene mainly underwent hydrogen transfer reactions with pyrene and styrene,mostly producing superconjugated biradicals through condensation reactions with *** the mixture systems,the olefins scattered aromatic molecules,hindering the formation of pyrene trimers and higher *** to the reactive molecular dynamics simulations,styrene may enhance the yield of dimer and enable the controlled polycondensation of pyrene.

Engineering surface framework TiO6 single sites for unprecedented deep oxidative desulfurization

Catalytic oxidative desulfurization（ODS） using titanium silicate catalysts has emerged as an efficient technique for the complete removal of organosulfur compounds from automotive fuels. However, the precise control of highly accessible and stable-framework Ti active sites remains highly challenging. Here we reveal for the first time by using density functional theory calculations that framework hexa-coordinated Ti（TiO6） species of mesoporous titanium silicates are the most active sites for ODS and lead to a lower-energy pathway of ODS. A novel method to achieve highly accessible and homogeneously distributed framework TiO6active single sites at the mesoporous surface has been developed. Such surface framework TiO6species exhibit an exceptional ODS performance. A removal of 920 ppm of benzothiophene is achieved at60°C in 60 min, which is 1.67 times that of the best catalyst reported so far. For bulky molecules such as4,6-dimethyldibenzothiophene（DMDBT）, it takes only 3 min to remove 500 ppm of DMDBT at 60°C with our catalyst, which is five times faster than that with the current best catalyst. Such a catalyst can be easily upscaled and could be used for concrete industrial application in the ODS of bulky organosulfur compounds with minimized energy consumption and high reaction efficiency.

The Effect of Digital Transformation on Corporate Green innovation

The digital transformation that induces the disruption of traditional production models is as crucial as the green development emphasized during the construction of an ecological ***,there is ongoing debate about whether,and under what contextual conditions,digital transformation promotes corporate green *** data from A-share listed enterprises on the Shanghai and shenzhen stock exchanges from 2012 to 2019,this study employs a fixed effect model to analyze the effect of digital transformation on green innovation and its specific *** are four findings.(1)Digital transformation significantly improves green innovation in *** conclusion is supported by quantile analysis and endogenous treatment based on a multi-period difference-in-differences model.(2)Digital transformation stimulates green innovation by enhancing absorptive capacity and internal control.(3)Industry heterogeneity analysis indicates that,compared with non-state-owned enterprises,digital transformation has a greater positive effect on green innovation in state-owned *** positive effect of digital transformation on green innovation is significant in technology-intensive enterprises but not in non-technology-intensive enterprises.(4)External context tests show that in regions with stringent environmental regulations,digital transformation significantly promotes green innovation,whereas in regions with weak environmental regulations,its effect is not ***,the positive correlation between digital transformation and green innovation remains largely unchanged as the level of green finance increases.
