Reading Time and DNA Sequence Preference of TET3 CXXC Domain Revealed by Single-Molecule Profiling

Recognition of CpG dinucleotide DNA in epigenetic information flow plays a pivotal role for cellular differentiation and development.The TET3 CXXC domain binds to CpG DNA,serving a basic epigenetic information reading mechanism.During the selective recognition of a CpG motif by a CXXC domain from crowded binding sites in a gene sequence,the protein-DNA interactions are beyond CpG dinu-cleotide.However,the selective binding dynamics of CpG within a long DNA context by epigenetic enzymes have been rarely exploit-ed,which is hard for ensemble methods to probe.Here,we used single-molecule magnetic tweezers to quantitatively examine the dynamics of TET3's CXXC domain on a Hoxa9 promoter DNA.Our single-molecule binding profile revealed that CXXC-DNA interactions involve both CpG motifs and their flanking sequences.The residence time of TET3 CXXC differs by about 1000 times in five distin-guished CpG clusters in the context of a CpG island.Moreover,we performed multi-state hidden Markov modeling analysis on the zip-ping/unzipping dynamics of a CpG hairpin,discovering TET3 CXXC's preference on CpG motifs regarding the-2 to+2 flanking bases.Our results shed light on the selective binding dynamics of a CXXC on a gene sequence,facilitating studies on epigenetic information reading mechanisms.

The development of a multifunctional oil adsorbing material which could effectively and quickly separate oily wastewater is one of the focuses in water environment restoration.In this study,bamboo charcoal(BC)was used as an improver to modify polyurethane(PU)foam.The results of scanning electron microscope(SEM)and Fourier-transform infrared spectroscopy(FTIR)revealed that the addition of BC could effectively improve the mechanical properties of PU.The adsorption data exhibited that the BC-loaded PU(BC/PU)foam composites effectively removed seven organic solvents(OSs,including octane,petroleum ether,soybean oil,chlorobenzene,1,2-dichloroethane,n-hexane,cyclohexane),and the maximum adsorption capacity of BC/PU was 23.6 g g^(−1)when BC content was 5%.The order of pseudo-second-order kinetic constants and maximum adsorption capacity of seven OSs was octane

Background:Gallbladder cancer(GBC)is the most common malignant tumor of biliary tract.Isoliquiritigenin(ISL)is a natural compound with chalcone structure extracted from the roots of licorice and other plants.Relevant studies have shown that ISL has a strong anti-tumor ability in various types of tumors.However,the research of ISL against GBC has not been reported,which needs to be further investigated.Methods:The effects of ISL against GBC cells in vitro and in vivo were characterized by cytotoxicity test,RNA-sequencing,quantitative real-time polymerase chain reaction,reactive oxygen species(ROS)detection,lipid peroxidation detection,ferrous ion detection,glutathione disulphide/glutathione(GSSG/GSH)detection,lentivirus transfection,nude mice tumorigenesis experiment and immunohistochemistry.Results:ISL significantly inhibited the proliferation of GBC cells in vitro.The results of transcriptome sequencing and bioinformatics analysis showed that ferroptosis was the main pathway of ISL inhibiting the proliferation of GBC,and HMOX1 and GPX4 were the key molecules of ISL-induced ferroptosis.Knockdown of HMOX1 or overexpression of GPX4 can reduce the sensitivity of GBC cells to ISL-induced ferroptosis and significantly restore the viability of GBC cells.Moreover,ISL significantly reversed the iron content,ROS level,lipid peroxidation level and GSSG/GSH ratio of GBC cells.Finally,ISL significantly inhibited the growth of GBC in vivo and regulated the ferroptosis of GBC by mediating HMOX1 and GPX4.Conclusion:ISL induced ferroptosis in GBC mainly by activating p62-Keap1-Nrf2-HMOX1 signaling pathway and downregulating GPX4 in vitro and in vivo.This evidence may provide a new direction for the treatment of GBC.

With over a million cases detected each year,skin disease is a global public health problem that diminishes the quality of life due to its difficulty to eradicate,propensity for recurrence,and potential for post-treatment scarring.Photodynamic therapy(PDT)is a treatment with minimal invasiveness or scarring and few side effects,making it well tolerated by patients.However,this treatment requires further research and development to improve its effective clinical use.Here,a piezoelectric-driven microneedle(PDMN)platform that achieves high efficiency,safety,and non-invasiveness for enhanced PDT is proposed.This platform induces deep tissue cavitation,increasing the level of protoporphyrin IX and significantly enhancing drug penetration.A clinical trial involving 25 patients with skin disease was conducted to investigate the timeliness and efficacy of PDMN-assisted PDT(PDMN-PDT).Our findings suggested that PDMN-PDT boosted treatment effectiveness and reduced the required incubation time and drug concentration by 25%and 50%,respectively,without any anesthesia compared to traditional PDT.These findings suggest that PDMN-PDT is a safe and minimally invasive approach for skin disease treatment,which may improve the therapeutic efficacy of topical medications and enable translation for future clinical applications.

Ultrafast Carbonized Wood of Electrode-Scaled Aligned-Porous Structure for High-Performance Lithium Batteries

The use of carbonized wood in various functional devices is attracting considerable attention due to its low cost,vertical channels,and high electrical conduction.However,the conventional carbonization method requires a long processing time and an inert atmosphere.Here,a microwave-assisted ultrafast carbonization technique was developed that carbonizes natural wood in seconds without the need for an inert atmosphere,and the obtained aligned-porous carbonized wood provided an excellent electrochemical performance as an anode material for lithium-ion batteries.This ultrafast carbonization technique simultaneously produced ZnO nanoparticles during the carbonization process that were uniformly distributed on the alignedporous carbon.The hierarchical structure of carbonized wood functionalized with ZnO nanoparticles was used as a host for achieving high-performance lithium-sulfur batteries:the highly conductive carbonized wood framework with vertical channels provided good electron transport pathways,and the homogeneously dispersed ZnO nanoparticles effectively adsorbed lithium polysulfide and catalyzed its conversion reactions.In summary,a new method was developed to realize the ultrafast carbonization of biomass materials with decorated metal oxide nanoparticles.

The adaptive transition of glioblastoma stem cells and its implications on treatments

Glioblastoma is the most malignant tumor occurring in the human central nervous system with overall median survival time<14.6 months.Current treatments such as chemotherapy and radiotherapy cannot reach an optimal remission since tumor resistance to therapy remains a challenge.Glioblastoma stem cells are considered to be responsible for tumor resistance in treating glioblastoma.Previous studies reported two subtypes,proneural and mesenchymal,of glioblastoma stem cells manifesting different sensitivity to radiotherapy or chemotherapy.Mesenchymal glioblastoma stem cells,as well as tumor cells generate from which,showed resistance to radiochemotherapies.Besides,two metabolic patterns,glutamine or glucose dependent,of mesenchymal glioblastoma stem cells also manifested different sensitivity to radiochemotherapies.Glutamine dependent mesenchymal glioblastoma stem cells are more sensitive to radiotherapy than glucose-dependent ones.Therefore,the transition between proneural and mesenchymal subtypes,or between glutamine-dependent and glucose-dependent,might lead to tumor resistance to radiochemotherapies.Moreover,neural stem cells were also hypothesized to participate in glioblastoma stem cells mediated tumor resistance to radiochemotherapies.In this review,we summarized the basic characteristics,adaptive transition and implications of glioblastoma stem cells in glioblastoma therapy.

自适应光学(AO)技术是一种能够补偿大气湍流的有效手段,但由于存在系统固有时延,变形镜(DM)上的补偿波前滞后于实测畸变波前,导致AO技术对高时间频率大气湍流的校正效果明显下降。因此,开展大气湍流前向预测研究对于抵消AO系统固有时延、提升系统校正带宽具有重要的研究意义和应用价值。本文提出了一种基于注意力机制的AO波前时空预测网络,该网络同时考虑了大气湍流的时间与空间特征,可通过连续6帧先验波前斜率信息预测未来第2帧的波前斜率。在具有两帧延迟的AO系统仿真中,所提预测网络使得波前校正残差均方根(RMS)下降了约40%,并且在不同的大气湍流强度下均表现出稳定的预测精度,预测残差RMS仅为真实畸变波前RMS的5.00%。最后使用1 km激光大气传输系统采集的实验数据进行了测试,验证了开环斜率预测网络的有效性。

为提高振动压路机作业过程的压实质量,研究了振动压路机的能量传递模型与作业参数优化。首先基于U-K方程建立“碾轮-被压实材料”的系统振动动力学模型,结合能量守恒提出振动压路机能量传递模型;然后以振动频率和压实速度作为寻优工作参数,构建路面压实质量的优化模型;最后通过案例验证证明了所提方法的可行性与有效性。结果表明:工作频率与被压实材料的固有频率比值保持在√2~2范围内,可避免共振带来的影响;初始阶段低速碾压,材料性能稳定后提高碾压速度可保证高效压实,得出振动频率在21.8~27 Hz、压实速度在2.36~2.91 km/s范围内,可达到最优的压实效果。所提的能量传递模型、作业参数优化模型为保障振动压路机压实质量奠定了基础,为提高振动压路机压实质量和效率提供了参考。

Understanding the scale-up of fermentation processes from the viewpoint of the flow field in bioreactors and the physiological response of strains

The production capability of a fermentation process is predominately determined by individual strains,which ultimately affected ultimately by interactions between the scale-dependent flow field developed within bioreactors and the physiological response of these strains.Interpreting these complicated interactions is key for better understanding the scale-up of the fermentation process.We review these two aspects and address progress in strategies for scaling up fermentation processes.A perspective on how to incorporate the multiomics big data into the scale-up strategy is presented to improve the design and operation of industrial fermentation processes.
