Genes important for survival or reproduction in Varroa destructor identified by RNAi

The Varroa mite,(Varroa destructor),is the worst threat to honey bee health *** explore the possibility of using RNA interference to control this pest, we determined the effects of knocking down various genes on Varroa mite survival and ***-stranded RNA (dsRNA)of six candidate genes (Da,Pros26S,RpL8, RpL11,RpPO and RpS13)were synthesized and each injected into Varroa mites,then mite survival and reproduction were *** of dsRNA for Da (Daughterless)and Pros26S (Gene for proteasome 26S subunit adenosine triphosphatase)caused a significant reduction in mite survival,with 3.57%±1.94% and 30.03%±11.43% mites surviving at 72 h post-inj ection (hpi),*** mites injected with green fluorescent protein (GFP)-dsRNA showed survival rates of 81.95%±5.03% and 82.36 ±2.81%,respectively. Injections of dsRNA for four other genes (RpL8,RpL11,RpPO and RpS13)did not affect survival significantly,enabling us to assess their effect on Varroa mite *** number of female offspring per mite was significantly reduced for mites injected with dsRNA of each of these four genes compared to their GFP-dsRNA *** of the target genes was verified by real-time polymerase chain reaction for two genes important for reproduction (RpL8,RpL11)and one gene important for survival (Pros26S). In conclusion,through RNA interference,we have discovered two genes important for mite survival and four genes important for mite *** genes could be explored as possible targets for the control of Varroa destructor in the future.

Methoprene does not affect juvenile hormone titers in honey bee （Apis mellifera） workers

Methoprene, a juvenile hormone （JH） analog, is a widely used insecticide that also accelerates behavioral development in honey bees （Apis mellifera）. JH regulates the transition from nursing to foraging in adult worker bees, and treatment with JH or methoprene have both been shown to induce precocious foraging. To determine how methoprene changes honey bee behavior, we compared JH titers ofmethoprene-treated and untreated bees. Behavioral observations confirmed that methoprene treatment significantly increased the number of precocious foragers in 3 out of 4 colonies. In only 1 out of 4 colonies, however, was there a significant difference in JH titers between the methoprene- treated and control bees. Further, in all 4 colonies, there was no significant differences in JH titers between precocious and normal-aged foragers. These results suggest that methoprene did not directly affect the endogenous JH secreted by corpora allata. Because methoprene caused early foraging without changing workers＇ JH titers, we conclude that methoprene most likely acts directly on the JH receptors as a substitute for JH.

Ions in the bulk of solvent-free ionic liquids bind into ion pairs and *** competition between the propensity of ions to stay in a bound state,and the reduction of the energy when unbinding in electric field,determines the portion of free ions in the electrical double *** present the simplest possible mean-field theory to study this effect."Cracking"of ion pairs into free ions in electric field is accompanied by the change of the dielectric response of the ionic *** predictions from the theory are verified and further explored by molecular dynamics simulations.A particular finding of the theory is that the differential capacitance vs potential curve displays a bell shape,despite the low concentration of free charge carriers,because the dielectric response reduces the threshold concentration for the bell-to camelshape *** presented theory does not take into account overscreening and oscillating charge distributions in the electrical double *** in spite of the simplicity of the model,its findings demonstrate a clear physical effect:a preference to be a charged monopole rather than a dipole(or higher order multipole)in strong electric field.

The contemporary role of metastasectomy in the management of metastatic RCC

Metastasectomy was initially described in the 1970s as a therapeutic strategy for patients with metastatic renal cell *** that time,systemic therapy options have grown exponentially,most recently with the introduction of *** aimed to review the contemporary literature regarding the role of metastasectomy in the era of targeted therapy and ***,metastasectomy has benefited patients with small volume,single-organ metastases,with favorable outcomes amongst younger,healthier patients with metastases to specific *** interplay between the employment of metastasectomy and systemic therapy has been limited to small,retrospective series with significant patient selection *** recently,investigators have conducted randomized controlled trials exploring the use of targeted therapies in the adjuvant setting after *** randomized data suggested no benefit in using sorafenib in this setting,and a subsequent study demonstrated possible harm in using pazopanib after ***,the role of other novel systemic therapies,including immunotherapy,nor the timing of use,have been meaningfully *** appears to be a valuable therapeutic option in the properly selected patient,requiring a multi-disciplinary management strategy and,pending future trials,a multimodal treatment approach.

In this paper,we present the performance of a recently developed in-situ spin-exchange optically pumped^(3)He-neutron spin filter system at the China Spallation Neutron Source(CSNS).The system achieved a^(3)He polarization of over 74%at the beamline *** of neutron transmission experiment results reveals a neutron polarization of>90%and an average transmission of 27%for 2.2Åneutrons,which were maintained for a duration of 120 h of beam *** the best of our knowledge,this is the first in-situ hyperpolarized^(3)He system incorporated on a neutron beamline in *** technology is expected to provide stable,wide-angle,and highly polarized neutrons at the CSNS for materials research and fundamental neutron physics.

Polarized 3He neutron spin filters(NSFs)can be used as a vital tool for neutron polarization production and *** China Spallation Neutron Source(CSNS),as one of the major neutron facilities in China,has committed resources to the development of a polarized ^(3)He NSF program to support its growing polarized neutron research.A spin-exchange optical pumping(SEOP)-based polarized ^(3)He system and other necessary hardware for NSF transport has been recently *** performance of the system is benchmarked using an in-house developed cell named“Trident”.Neutron beam measurements yield a ^(3)He polarization of 77%with over 200 h of on-beam relaxation *** this newly developed SEOP system with the recently reported cell fabrication station,CSNS is now capable of the fully self-sustained production of ^(3)He NSFs that shall support its future neutron polarization research.

Genome-wide Analysis of Transcriptional Variability in a Large Maize-Teosinte Population

Gene expression regulation plays an important role in controlling plant phenotypes and adaptation. Here, we report a comprehensive assessment of gene expression variation through the transcriptome analyses of a large maize-teosinte experimental population. Genome-wide mapping identified 25 660 expression quantitative trait loci （eQTL） for 17 311 genes, capturing an unprecedented range of expression variation. We found that local eQTL were more frequently mapped to adjacent genes, displaying a mode of expression piggybacking, which consequently created co-regulated gene clusters. Genes within the co-regulated gene clusters tend to have relevant functions and shared chromatin modifications. Distant eQTL formed 125 significant distant eQTL hotspots with their targets significantly enriched in specific functional cate- gories. By integrating different sources of information, we identified putative trans- regulators for a variety of metabolic pathways. We demonstrated that the bHLH transcription factor R1 and hexokinase HEX9 might act as crucial regulators for flavonoid biosynthesis and glycolysis, respectively. Moreover, we showed that domestication or improvement has significantly affected global gene expression, with many genes targeted by selection. Of particular interest, the Bx genes for benzoxazinoid biosynthesis may have undergone coordinated cis-regulatory divergence between maize and teosinte, and a transposon insertion that inactivates Bx12 was under strong selection as maize spread into temperate environments with a distinct herbivore community.
