OBJECTIVE:To evaluate the effects of the wenshen Jianpi recipe(温肾健脾方,WJR)on immune reconstruction and natural killer(NK)cells in immunological non-responders(INRs)of people living with human immunodeficiency virus(HIV)(PLWH)and propose new therapeutic strategies for HIV.METHODS:Based on Traditional Chinese Medicine treatment principle“invigorating Qi and warming Yang in the spleen and kidneys”,WJR combined with antiretroviral therapy(ART)therapy was performed in a randomized,double-blind,placebo-controlled study of 60 patients with non-responders.The randomized process was executed by the Clinical Evaluation Center of China Academy of Chinese Medical Sciences.Sixty patients who met the inclusion criteria obtained random numbers(that is the drug number)was randomly divided into a treatment group and a placebo control group according to a 1∶1 ratio.CD4+T cell counts and natural killer(NK)cells counts were evaluated at baseline and 12-week,24-week follow-ups.RESULTS:Four participants received random numbers and did not enter the group due to the patient’s own reasons.A total of 56 patients were enrolled,including 28 in the treatment group and 28 in the control group.CD4+T cell counts in the treatment group were significantly increased at week 24(P=0.01<0.05),which were significantly higher than those in the control group(P=0.01<0.05).Although no significant differences were observed between two groups,the CD56bri CD16-NK cell counts in the treatment group were significantly increased after duration.and CD56dim CD16+NK cell counts in the treatment group were significantly higher than those in the control group after 24 weeks of treatment(P=0.025<0.05).As compared with the control group,the treatment group had significantly lower CD56neg CD16+NK cell counts after 24 weeks of treatment(P=0.023<0.05).CONCLUSIONS:WJR promotes the immune reconstruction of INRs and redistribution of NK cell subsets,notably decreasing CD56neg CD16+NK cell counts in INRs.However,the redistribution of NK cell subsets is not beneficial for imm

Life-cycle cost(LCC)theory can be effectively applied to improve the efficiency and quality of power plant equipment and asset management.However,specific aspects of the LCC calculation and evaluation model require further research for practical application.This paper proposes an LCC assessment model for the management of electric power plant equipment during its service life.A membership function method based on fuzzy logic is used to improve the allocation of modernization and overhaul projects to multiple equipment assets.An LCC assessment model and evaluation system for power equipment are proposed and successfully applied to the equipment and project management of a Guangzhou power plant in the China Southern Power Grid,providing a decision-making mechanism that facilitates efficient operation and optimal utilization of power plant equipment and assets.

Responsiveness and minimal clinically important difference of the Chinese version of the Low Vision Quality of Life Questionnaire after cataract surgery

AIM: To investigate the Chinese version of the Low Vision Quality of Life Questionnaire(CLVQOL) as an instrument for obtaining clinically important changes after cataract surgery.METHODS: Patients underwent cataract surgery in Shanghai General Hospital, Shanghai Jiao Tong University, who fit the inclusion criteria were recruited. Two CLVQOLs were administered, including a preoperative CLVQOL and a CLVQOL at the end of the 3 mo follow-up period, and were completed using face-to-face interviews or phone interviews conducted by trained investigators. The minimal clinically important difference(MCID) was calculated using an anchor-based method and a distribution method. In addition, the responsiveness of the questionnaire was measured.RESULTS: A total of 155 residents were enrolled. The average visual acuity(VA) preoperatively was 0.08(SD=0.05), and it increased to 0.47(SD=0.28) at the end of followup. Statistically significant positive changes in the CLVQOL scores indicated significant improvement of vision related quality of life after cataract surgery. With the larger value between the two results as the final value, the MCID values of the CLVQOL(scores of the four scales as well as the total score) were 8.94, 2.61, 4.34, 3.10 and 17.63, respectively. The CLVQOL has both good internal and external responsiveness.CONCLUSION: CLVQOL scores are appropriate instruments for obtaining clinically important changes after cataract surgery. This study is an effective exploration for establishingcataract surgery efficacy standards, which helps clinical and scientific research workers in ophthalmology to gain a more in-depth understanding when using CLVQOL.

Resin composition: An indicator of oil maturity as revealed by fourier transform ion cyclotron resonance mass spectrometry

A pyrolysis experiment was carried out on a Dongying Depression kerogen sample to separate the resin from the oil. Fourier transform ion cyclotron resonance mass spectrometry (FT-ICR MS) with a positive-ion detector was used to detect the relative proportional changes in the compounds of the resin. During the whole pyrolysis experiment, the relative ratio of resin exceed 10% of the soluble component at each temperature point. Five compounds were detected from the resin: N1, N1O1, N1O_(2), O1, and O_(2). To research the changes in the proportions of the compounds during pyrolysis clearly, these five compounds were divided into three classes: N1, N1Ox, and Ox. The N1 class has the largest proportion in resin at the beginning of the pyrolysis, while Ox class has the least proportion. And the relationship between the number and the molecular mass of three classes compound was researched. With increasing maturity, the proportion of N1 and the N1Ox class decreased rapidly, while the Ox class increased slowly. Through researching these resin compounds, it was found that an inversion in the proportions of above three compounds appeared at the end of the oil window. At the same time, we found that the DBE and carbon number of resin compounds have changed obviously during the pyrolysis: the DBE increased, while the carbon number decreased significantly. And the details of the change of each compound have been researched. This research extends our knowledge of judging the maturity of crude oil during the pyrolysis through the characteristics of compounds in resin and provides the new index based on resin for the evaluation of thermal evolution stage and hydrocarbon generation capacity of source rocks.

Simulation of Horizontal-Two-Dimension Focused Waves Using A Two-Layer Boussinesq-Type Model

Accurate simulation of the horizontal-two-dimension(H2D)focused wave group in deep water requires high accuracy of a numerical model.The two-layer Boussinesq-type model(Liu and Fang,2016;Liu et al.,2018)with the highest spatial derivative of 2 has high accuracy in both linear and nonlinear properties.Based on the further development of the velocity equations(Liu et al.,2023),the H2D numerical model for water waves is established with the prediction-correction-iteration model in the finite difference method,and a composite fourth-order Adams-Bashforth-Moulton scheme is used for time integration.The wave generation method proposed by Hsiao et al.(2005)is applied and calibrated in this H2D model.The numerical calculations lead to the following three main conclusions:First,compared with the analytical solution of Stokes linear waves,the calculated velocity profiles show higher accuracy by using the improved velocity formulas.Second,the simulations of the focused multidirectional wave group are carried out,and good agreements are found,demonstrating that the present H2D numerical model shows high accuracy in simulating focused multidirectional wave groups,and the effectiveness of the improved velocity formulas is also validated.Furthermore,the velocity profiles throughout the computational domain at the time of maximum wave crest are given.Finally,the FFT method is used to obtain the amplitude with different frequencies for several locations,and the changes of the wavelet energy spectrum at different locations are presented for several cases.

The long non-coding RNA PILNCR2 increases low phosphate tolerance in maize by interfering with miRNA399-guided cleavage of ZmPHT1s

Theopen reading regions of ZmPHT1s(inorganic phosphate[Pij transporters)inmaize possess target sites of microRNA399(miR399).However,the relationship between miR399 and ZmPHT1s and its functional importance in response to Pi deficiency remain to be explored.We show here that ZmPHT1;1,ZmPHT1;3,and ZmPHT1;13 are the targets of ZmmiRNA399.We found that a long non-coding RNA,PILNCR2(Pi-deficiency-induced IncRNA 2),is transcribed from the opposing DNA strand of ZmPHT1;1 and predominantly localized in the cytoplasm.A ribonuclease protection assay and an RNA-RNA binding assay showed that PILNCR2 and ZmPHT1s could form the RNA/RNA duplexes in vivo and in vitro.A co-expression assay in N.benthamiana revealed that the PILNCR2/ZmPHT1 RNA/RNA duplexes interfere with miR399-guided cleavage of ZmPHT1 mRNAs.Overexpression of PILNCR2 increased low-Pi tolerance in maize,whereas its knockout and knockdown decreased low-Pi tolerance in maize.Consistently,ZmPHT1;3 and ZmPHT1;13 mRNA abundance was increased in transgenic plants overexpressing PILNCR2 but reduced in its knock-out mutants,suggesting that PILNCR2 positively regulates the mRNA abundance of ZmPHT1;3 and ZmPHT1;13 in maize.Collectively,these results indicate that PILNCR2 plays an important role in maize Pihomeostasisby interfering with miRNA399-guided cleavageof ZmPHT1mRNAs.

Experimental Investigation on the Vibration Attenuation of Tensegrity Prisms Integrated with Particle Dampers

Tensegrities made of tensile strings and compressed struts possess large strength-to-mass ratio values but tend to experience non-negligible vibration in dynamic environments because of poor structural damping.Here,we introduce particle dampers into a tensegrity prism to attenuate vibration,with the goal of establishing a lightweight and efficient approach of vibration suppression.To integrate the particle dampers and the tensegrity,a novel strut structure formed by assembling a solid strut and a hollow strut is devised,in which granular materials are inserted to develop a particle damper.The vibration attenuation performance of the tensegrity prism is investigated through exciter tests.According to the experimental parametric study,the influences of system parameters including excitation magnitude,the filling height of particles,particle size and the configuration of tensegrity prism on the vibration attenuation performance is analyzed.In addition,the experimental results regarding the dependence of the vibration attenuation on the system parameters are interpreted by the mechanism of collisions and frictions between particles and between particles and struts.The maximal vibration attenuation ratio of 76%can be achieved in the experiments.Thus,this research can provide insights into the design of lightweight tensegrity structures where vibration suppression is important,particularly in some dynamic environments.

BACKGROUND Increasing data indicated that long noncoding RNAs(lncRNAs)were directly or indirectly involved in the occurrence and development of tumors,including hepatocellular carcinoma(HCC).Recent studies had found that the expression of lncRNA HAND2-AS1 was downregulated in HCC tissues,but its role in HCC progression is unclear.Ultrasound targeted microbubble destruction mediated gene transfection is a new method to overexpress genes.AIM To study the role of ultrasound microbubbles(UTMBs)mediated HAND2-AS1 in the progression of HCC,in order to provide a new reference for the treatment of HCC.METHODS In vitro,we transfected HAND2-AS1 siRNA into HepG2 cells by UTMBs,and detected cell proliferation,apoptosis,invasion and epithelial-mesenchymal transition(EMT)by cell counting kit-8 assay,flow cytometry,Transwell invasion assay and Western blotting,respectively.In addition,we transfected miR-837-5p mimic into UTMBs treated cells and observed the changes of cell behavior.Next,the UTMBs treated HepG2 cells were transfected together with miR-837-5p mimic and tissue inhibitor of matrix metalloproteinase-2(TIMP2)overexpression vector,and we detected cell proliferation,apoptosis,invasion and EMT.In vivo,we established a mouse model of subcutaneous transplantation of HepG2 cells and observed the effect of HAND2-AS1 silencing on tumor formation ability.RESULTS We found that UTMBs carrying HAND2-AS1 restricted cell proliferation,invasion,and EMT,encouraged apoptosis,and HAND2-AS1 silencing eliminated the effect of UTMBs.Additionally,miR-873-5p targets the gene HAND2-AS1,which also targets the 3’UTR of TIMP2.And miR-873-5p mimic counteracted the impact of HAND2-AS1.Further,miR-873-5p mimic solely or in combination with pcDNA-TIMP2 had been transformed into HepG2 cells exposed to UTMBs.We discovered that TIMP2 reversed the effect of miR-873-5p mimic caused by the blocked signalling cascade for matrix metalloproteinase(MMP)2/MMP9.In vivo results showed that HAND2-AS1 silencing significantly inhibited tumor formation in mice.CON

Enhanced electrochemical performance of garnet-based solid-state lithium metal battery with modified anodic and cathodic interfaces

Due to high ionic conductivity and wide electrochemical window,the garnet solid electrolyte is considered as the most promising candidate electrolyte for solid-state lithium metal batteries.However,the high contact impedance between metallic lithium and the garnet solid electrolyte surface seriously hampers its further application.In this work,a Li-(ZnO)_(x)anode is prepared by the reaction of zinc oxide with metallic lithium and in situ coated on the surface of Li_(6.8)La_(3)Zr_(1.8)Ta_(0.2)O_(12)(LLZTO).The anode can be perfectly bound to the surface of LLZTO solid electrolyte,and the anode/electrolyte interfacial resistance was reduced from 2319 to 33.75Ω·cm^(2).The Li-(ZnO)_(0.15)|LLZTO|Li-(ZnO)_(0.15) symmetric battery exhibits a stable Li striping/plating process during charge-discharging at a constant current density of0.1 m A·cm;for 100 h at room temperature.Moreover,a Li-(ZnO)_(0.15)|LLZTO-SPE|LFP full battery,comprised of a polyethylene oxide-based solid polymer electrolyte(SPE)film as an interlayer between LiFePO4(LFP)cathode and LLZTO solid electrolyte,presents an excellent performance at 60℃.The discharge capacity of the full battery reaches 140 mA·h·g^(-1)at 0.1 C and the capacity attenuation is less than3%after 50 cycles.
