家庭养老能力评价指标体系构建与实证分析

家庭养老能力事关社会养老服务需求与发展,亟需构建科学有效的家庭养老能力评价指标体系。文章利用CHARLS2018年调查数据,构建出由自我养老能力、配偶养老能力和子女养老能力组成的3个准则指标,共有9个子准则指标和17个观测指标构成的家庭养老能力综合评价指标体系。采用熵权法和复相关系数法确定指标权重,并运用U检验、H检验和空间相关性分析家庭养老能力及其分维度的分布特征。结果显示:子女养老能力、自我养老能力和配偶养老能力权重系数分别为0.555、0.288和0.157。家庭养老能力的得分为0.433±0.091,子女养老能力、自我养老能力和配偶养老能力分维度得分分别为0.121±0.056、0.066±0.019和0.245±0.069,均呈现右偏分布,子女养老能力维度失分最多,自我养老能力维度差异性最大。女性、农村和西部地区老人的家庭养老能力显著弱于男性、城市和东部地区老人。低龄老人自我养老能力和配偶养老能力显著较高,但子女养老能力也显著低于高龄老人。中国家庭养老能力居前三位的是上海市、北京市和新疆维吾尔自治区,云南省、甘肃省和安徽省居后三位。研究发现中国家庭养老能力总体得分不高,自我养老能力与子女养老能力之间呈现“互补”态势。家庭养老能力分布呈现性别、城乡和区域差异性。养老服务体系需基于当前家庭养老能力及其分维度的地区差异性,因地制宜发展。

Pelvic recurrence of urothelial carcinoma after radical cystectomy is considered infrequent but the prognosis is poor. The incidence of pelvic recurrence after radical cystectomy was 4%-19%, depending on the clinical stage of the bladder malignancy]＇2 Pelvic recurrences usually occur within the first 2-3 years, and most of the patients die within 15 months after diagnosis of the recurrence.l＇3＇4 The treatment for pelvic recurrence was combined systemic chemotherapy, with a median survival period of 4-7monthsY Here we report a long-term survival case who received intra-arterial chemotherapy as a mono-therapy for pelvic recurrence, which developed 9 months after radical cystectomy.

Unisexual reproduction is generally relevant to polyploidy, and unisexual vertebrates are often considered an evolutionary “dead end” due to the accumulation of deleterious mutations and absence of genetic diversity. However, some unisexual polyploids have developed strategies to avoid genomic decay, and thus provide ideal models to unveil unexplored evolutionary mechanisms, from the reproductive success to clonal diversity creation. This article reviews the evolutionary mechanisms for overcoming meiotic barrier and generating genetic diversity in unisexual vertebrates, and summarizes recent research advancements in the polyploid Carassius complex. Gynogenetic gibel carp(Carassius gibelio) is a unique amphitriploid that has undergone a recurrent autotriploidy and has overcome the bottleneck of triploid sterility via gynogenesis. Recently, an efficient strategy in which ploidy changes, including from amphitriploid to amphitetraploid, then from amphitetraploid to novel amphitriploid, drive unisexual-sexual-unisexual reproduction transition and clonal diversity has been *** on this new discovery, multigenomic reconstruction biotechnology has been used to breed a novel strain with superior growth and stronger disease resistance. Moreover, a unique reproduction mode that combines both abilities of ameiotic oogenesis and sperm-egg fusion,termed as ameio-fusiongensis, has been discovered, and it provides an efficient approach to synthesize sterile allopolyploids. In order to avoid ecological risks upon escape and protect the sustainable property rights of the aquaculture seed industry, a controllable fertility biotechnology approach for precise breeding is being developed by integrating sterile allopolyploid synthesis and gene-editing *** review provides novel insights into the origin and evolution of unisexual vertebrates and into the attempts being made to exploit new breeding biotechnologies in aquaculture.

目的筛选抗低钙应答V抗原(LcrV)的人源单克隆抗体,分析抗体基因特点,检测抗体与抗原结合的特异性、亲和力及功能。方法使用PCR方法从鼠疫疫苗临床试验志愿者外周血细胞cDNA扩增抗体轻、重链可变区基因片段,构建ScFv噬菌体抗体文库。用LcrV对文库进行富集筛选,将获得的抗体基因表达成人IgG1后,测定抗体与LcrV抗原结合的特异性、亲和力、免疫调节功能以及保护能力。结果成功构建了鼠疫耶尔森菌人源ScFv抗体文库,库容量为7.54×10^(8)。抗体文库经LcrV筛选后获得了3株抗LcrV抗体,命名为RV-B4、RV-D1和RV-E8。3株抗体重链为VH1-46和VH3-30,轻链分别为VL1-51、VK3-20和VK1-39。3株抗体经ELISA及Western blot验证均与LcrV特异性结合,其与V抗原结合的解离常数(KD)分别为2.1 nmol/L、1.24 nmol/L和42 nmol/L。RV-D1体外降低人THP-1分泌TNF-α,动物攻毒试验中未发现抗LcrV抗体的保护效果。结论从鼠疫疫苗免疫人群获得了靶向低钙应答V抗原的人源抗体,以结合抗体为主,不能有效阻断病菌感染。所获得的单抗为鼠疫免疫基础研究、鼠疫诊断应用提供了候选材料。

Objective In China, 24 cases of human infection with highly pathogenic avian influenza(HPAI) H5 N6 virus have been confirmed since the first confirmed case in 2014. Therefore, we developed and assessed two H5 N6 candidate vaccine viruses(CVVs).Methods In accordance with the World Health Organization(WHO) recommendations, we constructed two reassortant viruses using reverse genetics(RG) technology to match the two different epidemic H5 N6 viruses. We performed complete genome sequencing to determine the genetic stability. We assessed the growth ability of the studied viruses in MDCK cells and conducted a hemagglutination inhibition assay to analyze their antigenicity. Pathogenicity attenuation was also evaluated in vitro and in *** The results showed that no mutations occurred in hemagglutinin or neuraminidase, and both CVVs retained their original antigenicity. The replication capacity of the two CVVs reached a level similar to that of A/Puerto Rico/8/34 in MDCK cells. 目的探讨血液透析患者血压变异性对移植物动静脉内瘘(arteriovenous graft,AVG)功能的影响。方法回顾性收集2020年12月用AVG为血管通路的患者作为研究对象,分析其连续4周血液透析时血压变异性,根据血压变异性中位数分为对照组(血压变异性>17.3,n=40)和观察组(血压变异性≤17.3,n=40),观察其血栓发生率及辅助初级通畅率,探讨其相关性。结果共纳入80例患者,观察1年内临床数据。对照组和观察组AVG血栓形成率分别为77.5%(31/40)和22.5%(9/40),2组比较差异有统计学意义(χ^(2)=24.200,P<0.001);对照组12个月辅助初级通畅率低于观察组(Log-rank χ^(2)=63.870,P<0.001)。结论血压变异性越高,血流动力学变化越大,移植物动静脉内瘘血栓形成的风险越高,建议临床上定期评估患者透析中血压变异性,并采取针对性的干预措施,降低血栓形成,提高辅助初级通畅率。
