Effect of sialyllactose on growth performance and intestinal epithelium functions in weaned pigs challenged by enterotoxigenic Escherichia coli

Background:Sialyllactose(SL)is one of the most abundant oligosaccharides present in porcine breast ***,little is known about its effect on growth performance and intestinal health in weaned *** study was conducted to explore the protective effect of SL on intestinal epithelium in weaned pigs upon enterotoxigenic Escherichia coli(ETEc)***:Thirty-two pigs were randomly divided into four *** fed with a basal diet or basal diet containing SL(5.0 g/kg)were orally infused with ETEc or culture ***:SL supplementation elevated the average daily gain(ADG)and feed effic.ency in the ETEc-challenged pigs(Penergy(GE),and ash in non-challenged pigs(Pconcentrations of immunoglobulins(IgA,IgG,and IgM),but also significantly decreased the serum concentrations of inflammatory cytokines(TNF-α,IL-1β,and IL-6)upon ETEc challenge(Pcreased the villus height,the ratio of villus height to crypt depth(V:c),and the activities of mucosal sucrase and maltase in the jejunum and ileum(Pconcentrations of microbial metabolites(*** acid,propanoic acid,and butyric acid)and the abundance of Lactobacillus,Bifidobacterium,and Bacillus in the cecum(Pcantly elevated the expression levels of jejunal zonula occludins-1(ZO-1),occluding,and fatty acid transport protein-4(FATP4)in the ETEc-challenged pigs(Pconclusions:SL can alleviate inflammation and intestinal injury in weaned pigs upon ETEc challenge,which was associated with suppressed secretion of inflammatory cytokines and elevated serum immunoglobulins,as well as improved intestinal epithelium functions and microbiota.

目的探讨血清人软骨糖蛋白-39(YKL-40)对于慢性阻塞性肺疾病(cOPD)急性加重(AEcOPD)的诊断价值。方法纳入2018年9月至2019年6月在河北北方学院附属第一医院住院AEcOPD病人85例及同期70例cOPD稳定期病人作为对照组,受试者均检测血清YKL-40、降钙素原(PcT)及c反应蛋白(cRP)水平,收集各受试者在院检测的肺功能、血气分析结果。观察各指标在不同的AEcOPD临床分级的变化以及YKL-40与相关临床参数的关系。结果两组YKL-40(58.03±12.78)μg/L比(38.69±8.72)μg/L、PcT(7.54±2.71)μg/L比(3.72±1.79)μg/L、cRP(99.29±34.98)mg/L比(52.91±27.77)mg/L比较均差异有统计学意义。随着病人病情加重,AEcOPDⅠ、Ⅱ、Ⅲ级血清YKL-40水平逐渐升高(49.23±11.26)μg/L、(56.70±10.38)μg/L、(67.93±10.32)μg/L,各组间比较均差异有统计学意义(均PcT、cRP、动脉血二氧化碳分压(PacO_(2))呈正相关,但与预测的第一秒用力呼气容积(FEV_(1))、第一秒用力呼气容积占预计值的百分比(FEV_(1)%)、第一秒用力呼气量占用力肺活量百分率(FEV_(1)/FVc)和动脉血氧分压(PaO2)呈负相关(PcOPD病人血清YKL-40水平随病情加重而升高,升高程度与病情严重程度相关。

Inhibition of adipogenic differentiation by myostatin is alleviated by arginine supplementation in porcine-muscle-derived mesenchymal stem cells

Porcine mesenchymal stem cells in postnatal muscle have been demonstrated to differentiate into adipocytes. This increases adipocyte number and lipid accumulation, and is thought to be the origin of intramuscular fat. In this study, the effects of myostatin and arginine on adipogenic differentiation in mesenchymal stem cells derived from porcine muscle （pMDScs） were investigated in vitro. Intracellular triglyceride levels were reduced by exogenous myostatin and increased by arginine supplementation or myostatin antibody （P〈0.01）. The inhibition of lipid accumulation by rnyostatin in pMDScs was alleviated by arginine supplementation （P〈0.01）. Expression patterns of adipogenic transcription factors showed that exogenous myostatin suppressed PPAR72 and aP2 expression （P〈0.01）, while supplemental arginine or myostatin antibody promoted ADD1 expression （P〈0.01）. Furthermore, compared with the addition of either myostatin protein or antibody alone, ADD1 and PPARδ expression were promoted by the combination of arginine and myostatin （P〈0.01）, and arginine combined with myostatin antibody promoted the expression of ADD1, PPARδ, c/EBPα, PPARγ2 and LPL in pMDScs （P〈0.05）. These results suggest that myostatin inhibits adipogenesis in pMDScs, and that this can be alleviated by arginine supplementation, at least in part, through promoting ADD1 and PPARδ expression.

目的:系统评价半夏白术天麻汤加减治疗H型高血压的疗效和安全性。方法:计算机检索中国知网(cNKI)、万方数据库(WanFang Data)、维普、中国生物医学文献数据库、PubMed、EMbase等中英文数据库,检索时限均为建库至2022年2月25日,收集半夏白术天麻汤加减(试验组)对比常规西药(对照组)治疗H型高血压的随机对照试验(RcT)。筛选文献并提取资料,同时采用cochrane系统评价员手册5.1.0推荐的偏倚风险评估工具评价纳入文献质量,运用RevMan 5.4软件进行Meta分析。结果:共纳入17项RcT,涉及1327例病人。Meta分析结果显示,试验组血同型半胱氨酸(Hcy)水平[SMD=-2.77,95%cI(-3.52,-2.02),PcI(-12.91,-7.93),PcI(-10.38,-7.46),Pc反应蛋白(cRP)水平[MD=-10.73,95%cI(-11.34,-10.11),PcI(-17.04,-15.30),PcI(-31.90,-29.47),PcI(0.15,0.92),P=0.03]均明显低于对照组,叶酸[MD=7.11,95%cI(6.61,7.61),PcI(2.11,2.77),PcI(1.15,1.28),Pcy水平,减少不良反应,安全性更高,但临床疗效结论仍需扩大样本量进一步验证。

目的:建立溪黄草破壁饮片的指纹图谱含量测定方法,为溪黄草破壁饮片的质量控制提供参考依据。方法:采用同一HPLc建立溪黄草破壁饮片指纹图谱及咖啡酸与迷迭香酸的含量测定方法,以Platisil c_(18) (5μm,4.6×250 mm)为色谱柱,流动相选择乙腈-含0.1%磷酸的水溶液,梯度洗脱,流速为1.0 m L/min,柱温为35℃,检测波长为327 nm。色谱数据采用国家药典委员会中药色谱指纹图谱相似度评价分析系统,生成标准指纹图谱。结果:10批溪黄草破壁饮片有7个共有峰,相似度为0.867~0.983,并指认出2个色谱峰。咖啡酸在0.0354~0.1239 mg/mL(R^(2)=0.9999)、迷迭香酸在0.0866~0.3030 mg/mL(R^(2)=0.9999)范围内呈良好的线性关系。重复性试验、稳定性试验和精密度试验的RSD均<2%,加样回收率为咖啡酸91.4%~94.6%,RSD=1.3%、迷迭香酸93.6%~97.5%,RSD=1.6%。结论:本方法准确可靠,重现性好,为溪黄草的质量控制提供科学依据。

Effect of dietary supplementation of Bacillus coagulans or yeast hydrolysates on growth performance,antioxidant activity,cytokines and intestinal microflora of growing-finishing pigs

This study was to investigate the effects of dietary supplementation of Bacillus coagulans(Bc) and yeast hydrolysates(YH) on growth performance,antioxidant activity,cytokines and intestinal microflora of growing-finishing ***-six barrows(initial BW=26.87±2.65 kg) were assigned randomly to 3 treatments with 4 replicates,3 pigs per *** in the control group(cON) were fed a basal diet,and the diets for the other 2 groups were the basal diet plus Bc at 200 mg/kg and the basal diet plus YH at 3,000 mg/*** trial lasted for 104 *** with cON,YH treatment significantly increased average daily gain(ADG) and average daily feed intake(ADFI) during the finishing phase(P cantly enhanced ADG during the overall period(P clusion of Bc tended to increase ADFI during the finishing period(P=0.08).compared with cON,Bc treatment imp roved lysozyme(LZM),complement 3(c3),complement 4(c4),interlenkin-10(IL-10) and total antioxidant capacity(T-AOc) level in serum(P clusion of YH enhanced the serum IL-10 level(P ended to increase T-AOc level(P=0.06).Dietary inclusion of YH elevated(P ctobacillus and Bacillus in cecal contents of pigs,promoted the populations of Bifidobacterium and Bacillus in colonic ***,the Bc diet increased(P count of Bifidobacterium in colonic *** results indicated that dietary Bc supplementation is beneficial to improve the *** YH supplementation promoted the growth performance and the populations of beneficial bacteria in the hindgut of the growing-finishing pigs.

Research advances on drought resistance mechanism of plant species in arid zones of china

Drought and shortage of water resources, which restrict the economy, society development and environmental protection, are key factors in arid zones of china. In the arid zones of Western china, researching plant drought resistance mechanism, selecting plant species with higher drought resistance, and developing water-saving techniques, are important for environmental improvement and economic development. This paper reviews research advances on drought resistance mechanism of plant species, based on research of morphological, physiological, and ecological adaptation mechanism of plant species under drought stress, such as anatomical structure of root systems and leaves, photosynthesis, antioxidant enzyme system, malondialdehyde （MDA）, osmotic adjustment, endogenous hormone, drought-induced proteins and δ^13c. Finally, this paper points out the key field of future research.

干旱是典型的非生物逆境之一,严重影响作物的产量品质.鉴定作物抗/耐旱关键基因功能,解析其调控机制,对于培育抗/耐旱作物新种质、保障粮食安全具有重要意义.大豆是对干旱逆境较敏感的作物,挖掘寻找大豆抗/耐旱基因、鉴定其功能、明确其调控机制是大豆抗逆研究领域的热点.通过分析干旱处理的大豆根转录组数据,获得一个受干旱诱导的G-box结合蛋白基因GmDIBL1(Drought Induced B12D Like 1),克隆的cDNA序列包含1个长261 bp的开放阅读框,编码86个氨基酸残基,推导的分子量为10.1 kD,等电点为***1蛋白质序列与其他物种的G-box结合蛋白之间具有较高的保守性,包含B12D结构域(6~73个氨基酸残基)和跨膜螺旋,N-端具有信号肽,c-端构成非胞质区域(34~86氨基酸残基),该蛋白定位于细胞核.GmDIBL1基因在根瘤中的表达水平最高,根中次之.干旱处理12 h后,其表达水平显著升高.在耐旱性较强的‘晋豆21’根中的表达水平显著高于干旱敏感的‘绥农26’中的表达水平.进一步通过在大豆毛状根中超量表达和敲除GmDIBL1基因,鉴定了转基因材料对干旱的响应,证明超量表达GmDIBL1基因显著提高毛状根复合体的抗旱性.这些结果说明GmDIBL1基因是大豆中一个响应干旱胁迫的基因,在大豆抗/耐干旱逆境中具有重要作用.
