无可信第三方的数据匿名化收集协议

针对半诚信的数据收集者对包含敏感属性(SA)数据收集和使用过程中可能造成隐私泄露问题,该文在传统模型中增加实时的数据领导者,并基于改进模型提出一个隐私保护的数据收集协议,确保无可信第三方假设前提下,数据收集者最大化数据效用只能建立在K匿名处理过的数据基础上。数据拥有者分布协作的方式参与协议流程,实现了准标识(QI)匿名化后SA的传输,降低了数据收集者通过QI关联准确SA值的概率,减弱内部标识揭露造成隐私泄露风险;通过树形编码结构将SA的编码值分为随机锚点和补偿距离两份份额,由K匿名形成的等价类成员选举获取两个数据领导者,分别对两份份额进行聚集和转发,解除唯一性的网络标识和SA值的关联,有效防止外部标识揭露造成的隐私泄露;建立符合该协议特性的形式化规则并对协议进行安全分析,证明了协议满足隐私保护需求。

高斯核密度估计方法检测健康数据异常值

针对智能穿戴设备普及背景下,利用运动手环采集的活动数据存在未知异常数据的问题,提出一种基于高斯核密度估计的健康数据异常值检测方法。首先采用t-分布邻域嵌入算法对数据集进行特征提取,增强数据局部结构能力;接着利用高斯核局部密度代替局部异常因子算法中的局部可达密度,提出基于高斯核密度估计离群因子(GKDELOF)算法,推导分析了该算法判断阈值的稳定性;最后在UCI标准数据集上进行仿真实验,验证算法的准确性,并在选取的真实运动手环所采集的活动数据集上进行实验分析。实验结果表明,该方法能够解决由活动复杂多样性造成的健康数据稀疏问题,准确检测出异常值。

Background Studying the composition and developmental mechanisms in mammary gland is crucial for healthy growth of newborns. The mammary gland is inherently heterogeneous, and its physiological function dependents on the gene expression of multiple cell types. Most studies focused on epithelial cells, disregarding the role of neighboring *** Here, we constructed the largest transcriptomic dataset of porcine mammary gland cells thus far. The dataset captured 126,829 high-quality nuclei from physiological mammary glands across five developmental stages(d 90 of gestation, G90;d 0 after lactation, L0;d 20 after lactation, L20;2 d post natural involution, PI2;7 d post natural involution, PI7). Seven cell types were identified, including epithelial cells, adipocytes, endothelial cells, fibroblasts cells, immune cells, myoepithelial cells and precursor cells. Our data indicate that mammary glands at different developmental stages have distinct phenotypic and transcriptional signatures. During late gestation(G90), the differentiation and proliferation of adipocytes were inhibited. Meanwhile, partly epithelial cells were completely differentiated. Pseudo-time analysis showed that epithelial cells undergo three stages to achieve lactation, including cellular differentiation, hormone sensing, and metabolic activation. During lactation(L0 and L20), adipocytes area accounts for less than 0.5% of mammary glands. To maintain their own survival, the adipocyte exhibited a poorly differentiated state and a proliferative capacity. Epithelial cells initiate lactation upon hormonal stimulation. After fulfilling lactation mission, their undergo physiological death under high intensity lactation. Interestingly, the physiological dead cells seem to be actively cleared by immune cells via CCL21-ACKR4 pathway. 针对大多数现有谱聚类算法处理大规模数据集时面临聚类精度低、大规模相似度矩阵存储开销大的问题,提出一种结合度量融合和地标表示的自编码谱聚类算法。引入相对质量概念进行节点评估,选取最具代表性的点作为地标点,通过稀疏表示近似获得图相似度矩阵,以降低存储开销。同时考虑到近邻样本的几何分布和拓扑分布的信息,融合欧氏距离与Kendall Tau距离来度量地标点与其他样本之间的相似度,提高聚类精度;以栈式自编码器取代拉普拉斯矩阵特征分解,将所获得的相似度矩阵作为自编码器的输入,通过联合学习嵌入表示和聚类来进一步提高聚类精度。在5个大规模数据集上的实验验证了本文算法的有效性。

Immunogenic dying tumor cells hold promising prospects as cancer vaccines to activate systemic immunity against both primary and metastatic ***,X-ray-induced dying tumor cells are rich in highly immunogenic tumor-associated antigens and self-generated dsDNA as potent ***,we found that the X-ray induction process can result in the excessive exposure of phosphatidylserine in cancer vaccines,which can specifically bind with the MerTK receptor on macrophages,acting as a“checkpoint”to facilitate immune silence in the tumor ***,we developed a novel strategy combining X-ray-induced cancer vaccines with UNC2250,a macrophage MerTK“checkpoint inhibitor,”for treating peritoneal carcinomatosis in colon *** incorporating UNC2250 into the treatment regimen,immunosuppressive efferocytosis of macrophages,which relies on MerTK-directed recognition of phosphatidylserine on vaccines,was effectively ***,the immune analysis revealed that this combination strategy promoted the maturation of dendritic cells and M1-like repolarization of macrophages,thereby simultaneously eliciting robust adaptive and innate *** innovative approach utilizing X-ray-induced vaccines combined with a checkpoint inhibitor may provide valuable insights for developing effective cancer vaccines and immunotherapies targeting colon cancer.
