Curcumin Interferes with TGF-β1-Induced Fibrosis in NRK-49F Cells by Reversing ADAMTS18 Gene Methylation

Objective: To explore the molecular mechanism by which curcumin affects renal interstitial fibrosis（RIF） progression by regulating ADAM metallopeptidase with thrombospondin type 1 motif 18（ADAMTS18） methylation. Methods: NRK-49F cells RIF model were induced with transforming growth factor β1（TGF-β1）. Effects of different concentrations of curcumin（0, 10, 20, and 30 μmol/L） on cell proliferation, cell cycle, cell apoptosis as well as cyclin D1 expression were analyzed by cell counting kit-8, flow cytometry and Western blot, respectively. ADAMTS18 methylation levels were determined by methylation-specific polymerase chain reaction. ADAMTS18, fibronectin（FN）, type Ⅰ collagen（Col-Ⅰ） and alpha-smooth muscle actin（α-SMA） mRNA and protein expressions were analyzed by real-time PCR（RT-PCR） and Western blot, respectively. Meanwhile, cells were treated with 50 mmol/L 5-aza-2'-deoxycytidine（5-aza-dC, demethylation agent） for 72 h. Effect of curcumin on extracellular matrix（ECM） deposition was evaluated by immunochemical staining and Western blot. NRK-49F cells were transfected with ADAMTS18 small interfering RNA and grouped into a normal control, ADAMTS18-knock-out（KO）, and ADAMTS18-KO+ 30 μmol/L curcumin groups, and whether curcumin can reverse the effect of ADAMTS18 knockdown on RIF was evaluated. Results: Compared with the control group, TGF-β1 significantly inhibited the proliferation of NRK-49F cells, blocked the G1/G0phase, promoted cell apoptosis and inhibited cyclin D1 expression（Pent concentrations of curcumin, 30 μmol/L curcumin significantly reversed these processes（P<0.01）. Immunochemical staining and Western blot results showed that curcumin significantly inhibited the deposition of FN, Col-Ⅰ and α-SMA（P<0.01）. Curcumin and 5-zaz-dC had synergistic effects, promoting ADAMTS18 expression, removing ADAMTS18 methylation, and reducing ECM deposition. ADAMTS18 knockdown promoted ECM accumulation, and curcumin reversed this process（P<0.01）. Conclusion: TGF-β1 induced fibrosis in NRK-49F cells.

Curcumin Inhibits Proliferation of Renal Cell Carcinoma in vitro and in vivo by Regulating miR-148/ADAMTS18 through Suppressing Autophagy

Objective: To explore the effect of curcumin on the proliferation of renal cell carcinoma and analyze its regulation mechanism. Methods: In RCC cell lines of A498 and 786-O, the effects of curcumin(2.5, 5, 10 μmo/L) on the proliferation were analyzed by Annexin V+PI staining. Besides, A498 was inoculated into nude mice to establish tumorigenic models, and the model mice were treated with different concentrations of curcumin(100, 200, and 400 mg/kg), once daily for 30 days. Then the tumor diameter was measured, the tumor cells were observed by hematoxylin-eosin staining, and the protein expressions of miR-148 and ADAMTS18were detected by immunohistochemistry. In vitro, after transfection of miR-148 mimics, miR-148 inhibitor or si-ADAMTS18 in cell lines, the expression of ADAMTS18 was examined by Western blotting and the cell survival rate was analyzed using MTT. Subsequently, Western blot analysis was again used to examine the autophagy phenomenon by measuring the relative expression level of LC3-Ⅱ/LC3-Ⅰ;autophagy-associated genes,including those of Beclin-1 and ATG5, were also examined when miR-148 was silenced in both cell lines with curcumin treatment. Results: Curcumin could inhibit the proliferation of RCC in cell lines and nude mice. The expressions of miR-148 and ADAMTS18 were upregulated after curcumin treatment both in vitro and in vivo(Pent or miR-148 inhibitor reversed these results, that is, both of them promoted the cell survival rate. Conclusion: Curcumin can inhibit the proliferation of renal cell carcinoma by regulating the miR-148/ADAMTS18 axis through the suppression of autophagy in vitro and in vivo. There may exist a positive feedback loop between mi R-148 and ADAMTS18 gene in RCC.

Conservation of Traditional Rice Varieties in a Globally Important Agricultural Heritage System (GIAHS):Rice-Fish Co-Culture

The traditional rice-fish farming system is selected as a ＂globally important agricultural heritage system＂（GIAHS） by the Food and Agriculture Organization （FAO）,United Nations Development Programme （UNDP）,and Global environment Facility （GEF）,etc.In Zhejiang Province of China,where the pilot site for this GIAHS farming system is located,we compared the use of traditional rice varieties in rice-fish co-culture and rice monoculture.Further,we determined how traditional rice varieties were performed in this rice-fish system.Only 19% of the farmers who practiced rice monoculture planted traditional varieties while 52% of farmers who practiced rice-fish co-culture planted traditional varieties.Traditional varieties represented 13% of the total land cultivated under rice in the rice-fish system but only 2% in the rice monoculture system.In the rice-fish system,yield was lower for traditional rice varieties than hybrid varieties but application of fertilizers and pesticides was also lower.In a field experiment in the rice-fish system without pesticides,rice planthopper numbers and sheath blight incidence were lower from three traditional varieties than one hybrid variety;yields were 8 to 32% lower from the traditional varieties than the hybrid.Our results showed that traditional rice varieties can be preserved through conserving GIAHS rice-fish co-culture.Our study also indicated that traditional rice varieties can survive in the rice-fish system because these varieties are helpful to the whole system and beneficial to the farmers.

论文研究了复杂构造区初始层速度约束反演问题。首先得出初始层速度模型的精度对偏移速度建模的质量和迭代稳定尤为重要,常规方法在构造复杂区使初始速度模型产生的误差易导致最终偏移速度模型地质意义失真、真实构造特征掩盖等问题。提升初始速度模型与地下地质特征的匹配和精度成为关键。基于此,论文提出了基于自适应速度趋势函数的约束层速度反演方法。在常规方法的基础上,利用复相关法在时间域剖面上获得地质构造的连续性与变化,并将其作为构造响应算子形成针对性的影响域和权重融入速度趋势函数中,使该函数自适应地质构造特征,提升初始速度模型的精度。再通过对基准速度和速度趋势对深度的梯度进行样条拟合,构建趋势函数的全局高频异常约束,有效预防在构造突变区产生异常高频,进一步增强算法的稳健和可靠性。理论模型和实际数据的测试,验证了本文方法的有效性,反演得到的层速度模型更符合地质规律,并能更好地解决构造复杂区层速度模型的连续性问题,有利于提高复杂构造叠前深度偏移成像精度。

为了比较福寿螺(Pomacea canaliculata)和部分本地螺种耐旱能力的差异,探究福寿螺成功入侵的机制,测定了不同干旱时长下成年以及幼年的福寿螺、中国圆田螺(Cipangopaludina chinesis)和梨形环棱螺(Sinotaia quadrata)的存活率,并分析了干旱胁迫对3种螺体内自由水、结合水和抗氧化酶的影响。结果表明,福寿螺和中国圆田螺的存活率远高于梨形环棱螺;中国圆田螺的自由水含量最高,而福寿螺管理自身水分能力最强。福寿螺的CAT(过氧化氢酶)活性、SOD(超氧化物歧化酶)活性和T-AOC(总抗氧化能力)水平在干旱胁迫下都有不同程度的增加,并高于2种本地螺。3种螺耐旱性表现为福寿螺>中国圆田螺>梨形环棱螺。

Factors influencing willingness to participate in ophthalmic clinical trials and strategies for effective recruitment

AIM:To explore the factors influencing individuals’willingness to participate in ophthalmic clinical trials.METHODS:A questionnaire survey was conducted from January to April 2021 among patients and their family members at Zhongshan Ophthalmic Center,Sun Yat-sen University,in Guangzhou,China.The survey gathered data on respondents’willingness,demographic and socioeconomic profiles,as well as their reasons and concerns regarding engagement in clinical trials.RESULTS:Of the 1078 residents surveyed(mean age 31.2±13.1y;65.8%females)in Guangzhou,749(69.5%)expressed a willingness to participate in future ophthalmic clinical trials.Specific characteristics associated with greater willingness included a younger age,lower annual income,higher education,prior participation experience,previous ophthalmic treatment,and a better understanding of clinical trials.With the exception of age,these characteristics were significantly linked to a higher willingness.The primary barrier to participation,expressed by 64.8%of those willing and 54.4%of those unwilling,was“Uncertain efficacy”.In terms of motivations,the willing group ranked“Better therapeutic benefits”(35.0%),“Professional monitoring”(34.3%),and“Trust in healthcare professionals”(33.1%)as their top three reasons,whereas the unwilling participants indicated“Full comprehension of the protocol”(46.2%)as the key facilitator.CONCLUSION:This study reveals a substantial willingness to participate in ophthalmic clinical trials and demonstrates the predictive role of demographic and socioeconomic factors.Variations in motivators and concerns between willing and unwilling participants highlight the significance of tailored recruitment strategies.Importantly,the need for and trust in healthcare professionals stand out as powerful motivations,underscoring the importance of enhancing physician-patient relationships,adopting patient-centered communication approaches,and addressing individualized needs to improve accrual rates.

Design Strategies for Aqueous Zinc Metal Batteries with High Zinc Utilization: From Metal Anodes to Anode-Free Structures

Aqueous zinc metal batteries(AZMBs)are promising candidates for next-generation energy storage due to the excellent safety, environmental friendliness, natural abundance, high theoretical specific capacity, and low redox potential of zinc(Zn) metal. However,several issues such as dendrite formation, hydrogen evolution, corrosion, and passivation of Zn metal anodes cause irreversible loss of the active materials. To solve these issues, researchers often use large amounts of excess Zn to ensure a continuous supply of active materials for Zn anodes. This leads to the ultralow utilization of Zn anodes and squanders the high energy density of AZMBs. Herein, the design strategies for AZMBs with high Zn utilization are discussed in depth, from utilizing thinner Zn foils to constructing anode-free structures with theoretical Zn utilization of 100%, which provides comprehensive guidelines for further research. Representative methods for calculating the depth of discharge of Zn anodes with different structures are first summarized. The reasonable modification strategies of Zn foil anodes, current collectors with pre-deposited Zn, and anode-free aqueous Zn metal batteries(AF-AZMBs) to improve Zn utilization are then detailed. In particular, the working mechanism of AF-AZMBs is systematically introduced. Finally, the challenges and perspectives for constructing high-utilization Zn anodes are presented.

Background:Hepatocellular carcinoma(HCC)has a poor long-term prognosis.The competition of circular RNAs(circRNAs)with endogenous RNA is a novel tool for predicting HCC prognosis.Based on the alterations of circRNA regulatory networks,the analysis of gene modules related to HCC is feasible.Methods:Multiple expression datasets and RNA element targeting prediction tools were used to construct a circRNA-microRNA-mRNA network in HCC.Gene function,pathway,and protein interaction analyses were performed for the differentially expressed genes(DEGs)in this regulatory network.In the proteinprotein interaction network,hub genes were identified and subjected to regression analysis,producing an optimized four-gene signature for prognostic risk stratification in HCC patients.Anti-HCC drugs were excavated by assessing the DEGs between the low-and high-risk groups.A circRNA-microRNA-hub gene subnetwork was constructed,in which three hallmark genes,KIF4A,CCNA2,and PBK,were subjected to functional enrichment analysis.Results:A four-gene signature(KIF4A,CCNA2,PBK,and ZWINT)that effectively estimated the overall survival and aided in prognostic risk assessment in the The Cancer Genome Atlas(TCGA)cohort and International Cancer Genome Consortium(ICGC)cohort was developed.CDK inhibitors,PI3K inhibitors,HDAC inhibitors,and EGFR inhibitors were predicted as four potential mechanisms of drug action(MOA)in high-risk HCC patients.Subsequent analysis has revealed that PBK,CCNA2,and KIF4A play a crucial role in regulating the tumor microenvironment by promoting immune cell invasion,regulating microsatellite instability(MSI),and exerting an impact on HCC progression.Conclusions:The present study highlights the role of the circRNA-related regulatory network,identifies a four-gene prognostic signature and biomarkers,and further identifies novel therapy for HCC.
