MicroRNA-370-5p inhibits pigmentation and cell proliferation by downregulating mitogen-activated protein kinase kinase kinase 8 expression in sheep melanocytes

In mammals,microRNAs(miRNAs)play key roles in multiple biological processes by regulating the expression of target genes.Studies have found that the levels of miR-370-5p expression differ significantly in the skins of sheep with different hair colors;however,its function remains unclear.In this study,we investigated the roles of miR-370-5p in sheep melanocytes and found that the overexpression of miR-370-5p significantly inhibited cell proliferation(P<0.01),tyrosinase activity(P=0.001)and significantly reduced(P<0.001)melanin production.Functional prediction revealed that the 3′-untranslated region(UTR)of MAP3K8 has a putative miR-370-5p binding site,and the interaction between these two molecules was confirmed using luciferase reporter assays.In situ hybridization assays revealed that MAP3K8 is expressed in the cytoplasm of melanocytes.The results of quantitative RT-PCR and Western blotting analyses revealed that overexpression of miR-370-5p in melanocytes significantly inhibits(P<0.01)MAP3K8 expression via direct targeting of its 3′UTR.Inhibition of MAP3K8 expression by siRNA-MAP3K8 transfection induced a significant inhibition(P<0.01)of melanocyte proliferation and significant reduction(P<0.001)in melanin production,which is consistent with our observations for miR-370-5p.Target gene rescue experiments indicated that the expression of MAP3K8 in melanocytes co-transfected with miR-370-5p and MAP3K8-cDNA(containing sites for the targeted binding to miR-370-5p)was significantly rescued(P≤0.001),which subsequently promoted significant increases in cell proliferation(P<0.001)and melanin production(P<0.01).Collectively,these findings indicate that miR-370-5p plays a functional role in inhibiting sheep melanocyte proliferation and melanogenesis by downregulating the expression of MAP3K8.

miR-99a-5p inhibits target gene FZD5 expression and steroid hormone secretion from goat ovarian granulosa cells

MicroRNA(miRNA) has vital regulatory effects on the proliferation, differentiation and secretion of ovarian granulosa cells, but the role of miR-99a-5p in goat ovarian granulosa cells(GCs) is unclear. Both miR-99a-5p and Frizzled-5(FZD5) were found to be expressed in GCs in goat ovaries via fluorescence in situ hybridization and immunohistochemistry, respectively, and FZD5 was verified(P<0.001) as a target gene of miR-99a-5p by double luciferase reporter gene experiments. Furthermore, FZD5 mRNA and protein expression were both found to be regulated(P<0.05) by miR-99a-5p in GCs. Moreover, the overexpression of miR-99a-5p or knockdown of FZD5 suppressed(P<0.05) estradiol and progesterone secretion from the GCs, as determined by ELISA. In summary, miR-99a-5p inhibits target gene FZD5 expression and estradiol and progesterone synthesis in GCs. Our study thus provides seminal data and new insights into the regulatory mechanisms of follicular development in the goat and other animals.

Carbon and Nitrogen Storage in Inner Mongolian Grasslands: Relationships with Climate and Soil Texture

Understanding the spatial variability of soil carbon(C) storage and its relationship with climate and soil texture is critical for developing regional C models and for predicting the potential impact of climate change on soil C storage. On the basis of soil data from a transect across the Inner Mongolian grasslands, we determined the quantitative relationships of C and nitrogen(N) in bulk soil and particle-size fractions(sand, silt, and clay) with climate and soil texture to evaluate the major factors controlling soil C and N storage and to predict the effect of climate changes on soil C and N storage. The contents of C and N in the bulk soil and the different fractions in the 0–20 and 20–40 cm soil layers were positively correlated with the mean annual precipitation(MAP) and negatively correlated with the mean annual temperature(MAT). The responses of C storage in the soil and particle-size fractions to MAP and MAT were more sensitive in the 0–20 cm than in the 20–40 cm soil layer. Although MAP and MAT were both important factors influencing soil C storage, the models that include only MAP could well explain the variation in soil C storage in the Inner Mongolian grasslands. Because of the high correlation between MAP and MAT in the region, the models including MAT did not significantly enhance the model precision. Moreover, the contribution of the fine fraction(silt and clay) to the variation in soil C storage was rather small because of the very low fine fraction content in the Inner Mongolian grasslands.

Immunization of Male Mice with a New Recombinant GnRH Fusion Protein Reduces the Testicular Function

The objective of the present study was to evaluate the effect of an immunogenic maltose-binding protein-gonadotropin releasing hormone （GnRH-6-MBP） using genetic engineering. The synthetic mammalian tandem repeated GnRH hexamer gene was inserted into the expression plasmid pMAL-c2x. Recombinant GnRH-6-MBP protein was over- expressed in E.coli strain BL21. Amylose resin with affinity chromatograph was used to purify target protein. The reactiongenicity of fusion protein was identified by indirect enzyme linked immunosorbent assay （ELISA）, and the antigenicity and biological effects of GnRH-6-MBP were tested in mice. In the experiment, 20 male Kunming white mice of 20 d old were randomly divided into treatment and control group. Ten mice were immunized with 100 μg GnRH-6-MBP administered subcutaneously （s.c.） thrice at 2-week intervals with GnRH-6-MBP. Mice were sacrificed after 3 weeks following the booster injection, the testis was removed, weighed and measured, and the histological structure was observed. The reactiongenicity of fusion protein to GnRH antibody was much higher than the control. Active immunization against GnRH-6-MBP reduced remarkably （P 〈 0.01） the length and weight of the testis, and shortened the girth and width of the testis （P 〈 0.05）, and suppressed testicular spermatogenesis compared to the control mice. These results indicate that the recombinant GnRH-6-MBP acted as a strong immunogen and caused atrophy of the testis.

Switches in transcriptome functions during seven skeletal muscle development stages from fetus to kid in Capra hircus

Skeletal muscle accounts for about 40% of mammalian body weight, the development of which is a dynamic, complex and precisely regulated process that is critical for meat production. We here described the transcriptome expression profile in 21 goat samples collected at 7 growth stages from fetus to kid, including fetal 45(F45), 65(F65), 90(F90), 120(F120), and 135(F135) days, and birth 1(B1) day and 90(B90) days kids. Paraffin sections combined with RNA-seq data of the 7 stages divided the transcriptomic functions of skeletal muscle into 4 states: before F90, F120, F135 and B1, and B90. And the dynamic expression of all 4 793 differentially expressed genes(DEGs) was identified. Furthermore, DEGs were clustered by weighted gene correlation network analysis into 4 modules(turquoise, grey, blue and brown) that corresponded to these 4 states. Functional and pathway analysis indicated that the active genes in the stages before F90(turquoise) were closely related to skeletal muscle proliferation. The DEGs in the F120-related module(grey) were found to participate in the regulation of skeletal muscle structure and skeletal muscle development by regulating t RNA. The brown module(F135 and B1) regulated fatty acid biological processes to maintain the normal development of muscle cells. The DEGs of B90 high correlation module(blue) were involved the strengthening and power of skeletal muscle through the regulation of actin filaments and tropomyosin. Our current data thus revealed the internal functional conversion of the goat skeletal muscle in the growth from fetus to kid. The results provided a theoretical basis for analyzing the involvement of m RNA in skeletal muscle development.

苯并[a]芘(Bap)是一种由5个苯环组成的分布广泛、广受关注、“三致”毒性极强的多环芳烃,也是主要的环境污染物监测对象。Bap在土壤等环境中长期积累,可通过食物链的累积和放大效应威胁人体健康。Bap污染土壤的高效修复迫在眉睫,本文综述了Bap的产生、污染现状与毒性效应、不同修复技术的优缺点。物理/化学方法具有修复周期短、范围广,但成本高且易产生二次污染等特点;生物修复成本低、操作简单、环境效益好,但修复周期长、效果受多因素影响。在实际治理过程中应综合考虑成本和效果,重点关注微生物修复及相关联合修复技术的应用,将为今后开展Bap类有机污染土壤的绿色修复提供科学思路。

Theoretical study on the effect of H_(2)O on the formation mechanism of NO_(x) precursors during indole pyrolysis

The incineration technology of kitchen waste is one of the effective technologies to achieve the resource utilization of municipal solid waste.Pyrolysis is an important stage of incineration.Indole is a rich initial product in the pyrolysis process of kitchen waste,and the presence of H_(2)O has a significant impact on the decomposition of indole to form NO_(x) precursors.Therefore,this study uses density functional theory method to study the effect of H_(2)O on the thermal decomposition of indole to produce NH_(3),HNCO,and HCN.When H_(2)O participates in the reaction,it can provide oxidative groups to generate a new product HNCO,which is different from the previous findings by indole pyrolysis without the presence of H_(2)O.Meanwhile,this study theoretically proves that NH_(3) is easier to form than HCN.This is consistent with the phenomenon that NH_(3) release is higher than HCN release in pyrolysis experiment.In addition,compared with the individual pyrolysis of indole,the participation of H_(2)O reduces the energy barriers for the formation of NH_(3) and HCN during indole pyrolysis,thereby promoting the formation of NH_(3) and HCN.

Genetically predicted fatty liver disease and risk of psychiatric disorders: A mendelian randomization study

BACKGROUND Non-alcoholic fatty liver disease(NAFLD)and alcohol-related liver disease(Ar-LD)constitute the primary forms of chronic liver disease,and their incidence is progressively increasing with changes in lifestyle habits.Earlier studies have do-cumented a correlation between the occurrence and development of prevalent mental disorders and fatty liver.AIM To investigate the correlation between fatty liver and mental disorders,thus ne-cessitating the implementation of a mendelian randomization(MR)study to elu-cidate this association.METHODS Data on NAFLD and ArLD were retrieved from the genome-wide association studies catalog,while information on mental disorders,including Alzheimer's disease,schizophrenia,anxiety disorder,attention deficit hyperactivity disorder(ADHD),bipolar disorder,major depressive disorder,multiple personality dis-order,obsessive-compulsive disorder(OCD),post-traumatic stress disorder(PTSD),and schizophrenia was acquired from the psychiatric genomics consor-tium.A two-sample MR method was applied to investigate mediators in signifi-cant associations.RESULTS After excluding weak instrumental variables,a causal relationship was identified between fatty liver disease and the occurrence and development of some psychia-tric disorders.Specifically,the findings indicated that ArLD was associated with a significantly elevated risk of developing ADHD(OR:5.81,95%CI:5.59-6.03,P<0.01),bipolar disorder(OR:5.73,95%CI:5.42-6.05,P=0.03),OCD(OR:6.42,95%CI:5.60-7.36,P<0.01),and PTSD(OR:5.66,95%CI:5.33-6.01,P<0.01).Meanwhile,NAFLD significantly increased the risk of developing bipolar disorder(OR:55.08,95%CI:3.59-845.51,P<0.01),OCD(OR:61.50,95%CI:6.69-565.45,P<0.01),and PTSD(OR:52.09,95%CI:4.24-639.32,P<0.01).CONCLUSION Associations were found between genetic predisposition to fatty liver disease and an increased risk of a broad range of psychiatric disorders,namely bipolar disorder,OCD,and PTSD,highlighting the significance of preven-tive measures against psychiatric disorders in patients with fatty liver

Chaiqin Chengqi Decoction as an adjuvant treatment for mild/moderately severe hypertriglyceridemic acute pancreatitis: A retrospective study

Hypertriglyceridemia is the third leading cause of acute pancreatitis(AP),and its incidence is increasing.Due to its relatively insidious etiology,it is easy to be ignored in the early stages.In China,Chaiqin Chengqi Decoction(CQCQD)has long been employed for treating AP.AIM To evaluate the effectiveness of CQCQD in patients diagnosed with mild/moderately severe hypertriglyceridemic AP(HTG-AP).METHODS In this study,the clinical data of 39 patients with HTG-AP admitted from January 2019 to November 2022 were collected.The changes of blood lipids,gastrointestinal symptoms,and abdominal pain before and after treatment were analyzed and compared between the two groups.RESULTS Twenty patients were treated with the conventional HTG-AP regimen,and 19 patients were additionally treated with CQCQD.After receiving treatment,the triglycerides(TG)level of the CQCQD group was lower than that of the CQCQD group(3.14±0.25 mmol/L vs 4.96±0.47 mmol/L,P<0.01).After 3 d of treatment,the patients in the CQCQD group had more bowel movements than the control group(2.51±0.25 times vs 1.00±0.17 times,P=0.01).The gastrointestinal function of most patients returned to normal,and the acute gastrointestinal injury score was significantly lower than that of the control group(0.11±0.07 vs 0.42±0.11,P<0.01).CONCLUSION In patients with HTG-AP,CQCQD can significantly reduce the TG level,shorten the recovery time of defecation,significantly improve the gastrointestinal function.
