Association analysis of an anti-obesity mechanism and key ripened Pu-erh tea bioactive components by mimicking human general tea drinking

Pu-erh tea， a traditional chinese beverage， performs an anti-obesity function， but the correlation between its components and efficacy remains unknown. Here， we screened two Pu-erh teas with significant anti-obesity efficacies from 1. teas. In vitro experiments revealed that lipid accumulation in L02.cells and lipid synthesis in 3T3-L1.cells were significantly better inhibited by Tea-B than Tea-A. Further in vivo experiments using model mice revealed that the differences in chemical components generated two pathways in the anti-obesity efficacy and mechanism of Pu-erh teas. Tea-A changes the histomorphology of brown adipose tissue (BAT) and increases the abundance of Coriobacteriaceae＿UCG＿002.and cyclic AMP in guts through high chemical contents of cyclopentasiloxane， decamethyl， tridecane and 1.2.3-trimethoxybenzene， eventually increasing BAT activation and fat browning gene expression; the high content of hexadecane and 1.2.dimethoxy-benzene in Tea-B reduces white adipose tissue (WAT) accumulation and the process of fatty liver， increases the abundance of Odoribacter and sphinganine 1.phosphate， inhibits the expression of lipid synthesis and transport genes. These mechanistic findings on the association of the representative bioactive components in Pu-erh teas with the anti-obesity phenotypes， gut microbes， gut metabolite structure and anti-obesity pathways， which were obtained for the first time， provide foundations for developing functional Pu-erh tea.

Stutzerimonas have been extensively studied due to their remarkable metabolic and physiological ***,research on its phages is currently *** this study,we isolated a novel double-stranded DNA(dsDNA)phage,vB_SstM-PG1.from the marine environment that infects Stutzerimonas stutzeri *** dsDNA genome is 372.4 bp long with a G/C content of 64.1.%and encodes 54 open reading *** phage possesses a tail packaging structure that is different from known Stutzerimonas stutzeri phages and exhibits structural protein characteristics similar to those of temperate *** addition,two genes of toxin-antitoxin system,including YdaS_antitoxin and HEPN_SAV_61.7,were found in the vB_SstM-PG1.genome and play important roles in regulating host growth and *** phylogenetic tree and comparative genomic analysis,it has been determined that vB_SstM-PG1.is not closely related to any phages previously identified in the GenBank ***,it has a connection with enigmatic,uncultured ***,the vB_SstM-PG1.virus exhibits an average nucleotide identity of over 70%with six uncultivated viruses identified in the IMG/VR v4 *** significant finding has resulted in the identification of a novel viral genus known as Metabovirus.

超导磁体的场强与超导材料的载流能力、磁体口径和低温环境有密切关系.为了在中高温区域实现高磁场强度的超导磁体,采用国产第二代高温超导带材,成功绕制出内直径为1.0 mm的高温超导线圈.该超导线圈在77,65,55和46 K不同温区下进行了性能测试,其最大运行电流分别为65,1.7,2.7和338 A,对应的中心磁场强度分别为0.78,1.77,3.1.4.08 T.所研制的超导线圈的中平面上磁场基本一致.

Integrase strand transfer inhibitors (INSTIs) have emerged as the first-line choice for treating human immunodeficiency virus (HIV) infection due to their superior efficacy and safety. However, the impact of INSTIs on the development of neuropsychiatric conditions in people living with HIV (PLWH) is not fully understood due to limited data. In this study, we conducted a cross-sectional examination of PLWH receiving antiretroviral therapy, with a specific focus on HIV-positive men who have sex with men (MSM) on INSTI-based regimens (n = 61. and efavirenz (EFV)-based regimens (n = 2.). Participants underwent comprehensive neuropsychiatric evaluations and multimodal magnetic resonance imaging (MRI) scans, including T1.weighted images and resting-state functional MRI. Compared to the EFV group, the INSTI group exhibited primarily reduced gray matter volume (GMV) in the right superior parietal gyrus, higher regional homogeneity (ReHo) in the left post-central gyrus, lower ReHo in the right orbital part of the inferior frontal gyrus, and increased voxel-wise functional connectivity for the seed region in the left inferior temporal gyrus with clusters in the right cuneus. Furthermore, the analysis revealed a main effect of antiretroviral drugs on GMV changes, but no main effect of neuropsychiatric disorders or their interaction. The repeated analysis of participants who did not switch regimens confirmed the GMV changes in the INSTI group, validating the initial findings. Our study demonstrated gray matter atrophy and functional brain changes in PLWH on INSTI-based regimens compared to those on EFV-based regimens. These neuroimaging results provide valuable insights into the characteristics of brain network modifications in PLWH receiving INSTI-based regimens.

Aqueous Zn-iodine(Zn-I_(2.)batteries have attracted extensive research interest as an emerging redox conversion energy storage system due to the low cost and high ***,the shuttling effects of polyiodides arising from incomplete redox conversion and inhomogeneous Zn plating on the Zn anode surface always hinder the commercial application of Zn-I_(2.*** this work,a two-birds-with-one-stone strategy is reported for long-life Zn-I_(2.*** on the strategy,the sulfonate-functionalized carbon fiber not only acts as the excellent iodine limiter to inhibit iodine species shuttling,but also as the uniform Zn plating guidance layer on the Zn anode surface to prevent the inhomogeneous deposition of Zn^(2.).Consequently,a superior cycling stability(a capacity of 1.4 mAh g^(-1.after 1.,000 cycles at 5 A g^(-1.)is *** calculations illustrate that sulfonate groups successfully induce charge redistribution on the carbon substrate,thereby strengthening the electronic interactions of the iodine species with the carbon *** charge-enriched sulfonate groups can guide the uniform deposition of Zn^(2.)through a strong Coulombic effect with Zn^(2.).This work gives a new perspective on the integrated design of cathodes and anodes for rechargeable batteries.

Differentiation potential of bone marrow mesenchymal stem cells into retina in normal and laser-injured rat eye

Bone marrow mesenchymal stem cells (MSCs) can develop into hematopoietic and mesenchymal lineages but have not been known to participate in the production of retina. Here we report that bone marrow mesenchymal stem cells, after being subretinally transplanted into normal or Nd: YAG laser-injured rat eye, can integrate into RPE layer, photoreceptor layer, bi-polar cell layer and ganglion layer. DAPI-labeling detection was used to trace the origin of the repopulating cells. DAPI fluorescence was used to identify retina cells of bone marrow origin 1., 2., 35 and 50 days after transplantation. No formation of rosettes was found but some random cells were found at the end of the observation. MSCs-originated cells spread more widely in the injured retinas than in the normal ones. Immunohistochemical detection showed that though the cells could express neuronal nuclei (NeuN), neuron specific enolase (NSE), glial fibrillary acidic protein (GFAP) and cytokeratin (CK), the proteins expression in the injured transplantation group was abnormal in some region compared with that in the normal transplantation group. Elec-troretinogram (ERG) showed that ERG-b wave of the injured transplantation group is significantly higher than that of the two laser-injured control groups. These results suggest that a proportion of MSCs can differentiate into retina-like structure in vivo and the differentiation differs in normal and laser-injured retinas.
