Comparison of SO_2 column retrievals from BRD and DOAS algorithms

Atmospheric SO_2 has a significant impact on the urban environment and global climate. Band Residual Difference Algorithm(BRD) and Differential Optical Absorption Spectroscopy(DOAS) were used respectively by NASA and ESA science team to derive SO_2 columns from satellite observations, but there are few studies on the comparison and validation of BRD and DOAS SO_2 retrievals under the same observation conditions. In this study, the radiative transfer model SCIATRAN was firstly used to validate the accuracies of BRD and DOAS SO_2 retrievals, and analyse the uncertainty of SO_2 retrieval caused by band selection,O_3 absorption, aerosol, surface reflectance, solar and viewing zenith angle. Finally, BRD and DOAS algorithms were applied to the same radiances from satellite observations, and comparisons of BRD and DOAS SO_2 retrievals were conducted over volcanic eruption and North China. Results show that, for the case with low SO_2 columns, BRD SO_2 retrievals have higher retrieval accuracy than DOAS, but typical seasonal variation with high SO_2 column in winter and low in summer can be more clearly discernible in DOAS SO_2 retrievals than BRD from satellite observations. For the case with high SO_2 columns, the differences between BRD(310.8–314.4 nm) and DOAS(315–327 nm) retrievals are large, and the value and accuracy of BRD(310.8–314.4 nm) SO_2 retrievals are lower than those of DOAS(315–327 nm) retrievals. Compared with the SO_2 inputs in forward model, both BRD(310.8–314.4 nm) and DOAS(315–327 nm) SO_2 retrievals are underestimated for the case with high SO_2 columns. The selection of wavelength range can significantly affect the accuracy of SO_2 retrieval. The error of BRD SO_2 retrieval from 310.8–314.4 nm is lower than other bands in the ultraviolet spectral region(306–327 nm). The increase of wavelength in the ultraviolet spectral region306–330 nm can reduce the underestimation of DOAS SO_2 retrievals in the case of high SO_2 column, but slight overestimation o

Fast CO_(2) Retrieval Using a Semi-Physical Statistical Model for the High-Resolution Spectrometer on the Fengyun-3D Satellite

China’s Fengyun-3D meteorological satellite launched in December 2016 carries the high-resolution greenhouse-gases absorption spectrometer(GAS)aimed at providing global observations of carbon dioxide(CO_(2)).To date,GAS is one of the few instruments measuring CO_(2) from the near-infrared *** orbit,the oxygen(O_(2))A band suffers a disturbance,and the signal-to-noise ratio(SNR)is significantly lower than the nominal *** leads to difficulties in the retrieval of surface pressure and hence a degradation of the retrieval of the column-averaged CO_(2) dry air mole fraction(XCO_(2))if a full physics retrieval algorithm is ***,a fast CO_(2) inverse method,named semi-physical statistical algorithm,was developed to overcome this *** instrument characteristics,the semi-physical statistical algorithm,and the results of comparison with ground-based measurements over land were introduced in this ***_(2) can be obtained from three bands,namely,the O_(2) A,weak CO_(2),and strong CO_(2) bands,with compensation from the Medium Resolution Spectral Imager-2(MERSI-2)products,ECMWF Reanaly-sis v5(ERA-5)data,and Total Carbon Column Observing Network(TCCON)*** eigenvectors of covariance matrices and the least square fits were used to derive retrieval coefficients and yield cloud-free *** addition to the GAS radiance,some key factors necessary for the accurate estimations of XCO_(2) were also taken as input information(e.g.,air mass,surface pressure,and a priori XCO_(2)).The global GAS XCO_(2) restricted over land was compared against the simultaneously collocated observations from *** retrieval algorithm can mitigate the issue caused by the low SNR of the O_(2) A band to a certain ***,through site-by-site comparisons,GAS XCO_(2) agreed well with the average precision(1σ)of 1.52 ppm and bias of−0.007 *** seasonal variation trends of GAS XCO_(2) can be clearly seen at TCCON sites on the 1-yr timescale.

CRISPR-Cas9 mediated LAG-3 disruption in CAR-T cells

T cells engineered with chimeric antigen receptor （CAR） have been successfully applied to treat advanced refractory B cell malignancy. However, many challenges remain in extending its application toward the treatment of solid tumors. The immunosuppressive nature of tumor microenvironment is considered one of the key factors limiting CART efficacy. One negative regulator of T cell activity is lymphocyte activation gene-3 （LAG-3）. We successfully generated LAG-3 knockout T and CAR-T cells with high efficiency using CRISPR-Cas9 mediated gene editing and found that the viability and immune phenotype were not dramatically changed during in vitro culture. LAG-3 knockout CART cells displayed robust antigen-specific antitumor activity in cell culture and in murine xenograft model, which is comparable to standard CAR-T cells. Our study demonstrates an efficient approach to silence immune checkpoint in CAR-T cells via gene editing.

Retrieval experiment was made for global total column ozone using the first year measurements of Total Ozone Unit (TOU) on board the second generation polar orbiting meteorological satellite of China, FY-3/A. The retrieval results were analyzed and validated by comparison with AURA/OMI, Meteop/GOME-2 global ozone products and ground-based ozone measurement data. The qualititative comparisons over the globe especially over Antarctica and the Tibetan Plateau show that the spatial and temporal distribution characteristics are consistent with OMI and GOME-2 products. The quantitative comparisons with ground-based measurements and AURA/OMI ozone product were made over 74 stations, the TOU total ozone retrieval has a 3% rms relative error compared with AURA/OMI ozone product and 4.2% rms relative error with ground-based measurements. The maximum difference between satellite retrieval and ground-based measurements was found in the Antarctica ozone hole. The TOU global ozone product is operational and distributed to all users.

为了有效评价和利用核桃种质资源,以甘肃省85份农家品种核桃坚果为研究对象,对20个数量性状进行测定,分析各性状的变异系数,并对这20个数量性状进行K-S检验和分级。结果表明:甘肃省实生核桃居群坚果性状变异系数为8.56%~27.32%,平均为15.53%,说明甘肃省核桃坚果主要数量性状的变异较大,遗传多样性丰富。油酸含量、仁重、锌含量、单果重、壳厚等指标的变异系数较高,分别为27.32%、26.35%、24.96%、23.50%、22.80%,品种选育中宜将这几个数量性状作为主要参考指标。K-S检验结果表明,1-5级出现的频率分别为10%、20%、40%、20%、10%,符合正态分布。核仁不饱和脂肪酸含量稳定,多不饱和脂肪酸含量高于大豆油、玉米胚芽油和芝麻油。

Changes in PM2.5 sensitivity to NOx and NH3 emissions due to a large decrease in SO2 emissions from 2013 to 2018

The authors evaluated and compared the behavior of PM2.5 with respect to NOx and NH3 emission changes in high(the year 2013)and low(the year 2018)SO2 emission *** groups of simulations were conducted based on anthropogenic emissions from China in 2013 and 2018,*** each group of simulations,a respective 25%reduction in NOx and NH3 emissions were assumed.A sensitivity factor(β)was defined as the relative change in PM2.5 concentration due to 1%change in NOx or NH3 *** the high SO2 emissions case,PM2.5 was more sensitive to NH3(0.31)emissions change than NOx(0.21).Due to the significant decrease in SO2 emissions from the high to low SO2 emissions case,the sensitivity of PM2.5 to NOx increased to 0.33,while its sensitivity to NH3 decreased to *** result implies that now and in the future,PM2.5 is/will be less sensitive to NH3 emissions change,while NOx emissions control is more effective in reducing the surface PM2.5 ***,in the low SO2 emissions case,the sensitivities of PM2.5 to NOx and NH3 in winter were higher than those in summer,indicating that to dealwith severewinter hazemore attention should be paid to the emissions control of inorganic PM2.5 precursors,especially NOx.

Combining LPJ-GUESS and HASM to simulate the spatial distribution of forest vegetation carbon stock in China

It is very important in accurately estimating the forests＇ carbon stock and spatial distribution in the regional scale because they possess a great rate in the carbon stock of the terrestrial ecosystem. Yet the current estimation of forest carbon stock in the regional scale mainly depends on the forest inventory data, and the whole process consumes too much labor, money and time. And meanwhile it has many negative influences on the forest carbon storage updating. In order to figure out these problems, this paper, based on High Accuracy Surface Modeling （HASM）, proposes a forest vegetation carbon storage simulation method. This new method employs the output of LPJ-GUESS model as initial values of HASM and uses the inventory data as sample points of HASM to simulate the distribution of forest carbon storage in China. This study also adopts the seventh forest resources statistics of China as the data source to generate sample points, and it also works as the simulation accuracy test. The HASM simulation shows that the total forest carbon storage of China is 9.2405 Pg, while the calculated value based on forest resources statistics are 7.8115 Pg. The forest resources statistics is taken based on a forest canopy closure, and the result of HASM is much more suitable to the real forest carbon storage. The simulation result also indicates that the southwestern mountain region and the northeastern forests are the important forest carbon reservoirs in China, and they account for 39.82% and 20.46% of the country＇s total forest vegetation carbon stock respectively. Compared with the former value （1975-1995）, it mani- fests that the carbon storage of the two regions do increase clearly. The results of this re- search show that the large-scale reforestation in the last decades in China attains a signifi- cant carbon sink.

Programmed cell death protein-1(PD-1)-mediated immunosuppression has been proposed to contribute to the limited clinical efficacy of chimeric antigen receptor T(CAR-T)cells in solid *** generated PD-1 and T cell receptor(TCR)deficient mesothelin-specific CAR-T(MPTK-CAR-T)cells using CRISPR-Cas9 technology and evaluated them in a dose-escalation study.A total of 15 patients received one or more infusions of MPTK-CAR-T cells without prior *** dose-limiting toxicity or unexpected adverse events were observed in any of the 15 *** best overall response was stable disease(2/15 patients).Circulating MPTK-CAR-T cells peaked at days 7–14 and became undetectable beyond 1 ***-positive CAR-T cells rather than TCR-negative CAR-T cells were predominantly detected in effusion or peripheral blood from three patients after *** further confirmed the reduced persistence of TCR-deficient CAR-T cells in animal *** results establish the preliminary feasibility and safety of CRISPR-engineered CAR-T cells with PD-1 disruption and suggest that the natural TCR plays an important role in the persistence of CAR-T cells when treating solid tumors.

Excellent Electrochemical Performances of Intrinsic Polyaniline Nanofibers Fabricated by Electrochemical Deposition

A simple route to synthesize the polyaniline(PANI) nanofibers with diameter about 150 nm was reported. In this strategy, the PANI nanofibers were fabricated by electrochemical deposition by using two-electrode configuration in 0.01 M aniline and 0.01 M H_2SO_4 electrolytes. The as-prepared materials were characterized by scanning electron microscopy(SEM), infrared spectroscopy(FTIR), Raman spectroscopy and thermogravimetric analysis(TGA). The electrochemical properties of the PANI nanofibers electrode as supercapacitor materials were investigated. The PANI nanofibers electrode showed high capacitance of 485 F·g^(-1)at 0.1 A·g^(-1), and the decrease in the specific capacitance is about 3.5% in 1 000 cycles. The results indicate that the PANI nanofibers electrode shows high stability and retains its electrochemical capacitance property over 1 000 cycles, suggesting PANI nanofibers have promising applications in high-performance supercapacitors.
