The coronavirus disease 2.19(COVID-19)pandemic has devastated global *** key host factors essential for SARS-CoV-2.RNA replication is expected to unravel cellular targets for the development of broad-spectrum antiviral drugs which have been quested for the preparedness of future viral ***,we have identified host proteins that associate with nonstructural protein 12.nsp12.,the RNA-dependent RNA polymerase(RdRp)of SARS-CoV-2.using a mass spectrometry(MS)-based proteomic *** the candidate factors,CDK2.Cyclin-dependent kinase 2.,a member of cyclin-dependent kinases,interacts with nsp12.and causes its phosphorylation at T2.,thus facilitating the assembly of the RdRp complex consisting of nsp12.nsp7 and nsp8 and promoting efficient synthesis of viral *** crucial role of CDK2.in viral RdRp function is further supported by our observation that CDK2.inhibitors potently impair viral RNA synthesis and SARS-CoV-2.*** together,we have discovered CDK2.as a key host factor of SARS-CoV-2.RdRp complex,thus serving a promising target for the development of SARS-CoV-2.RdRp inhibitors.

Background: Severe fever with thrombocytopenia (SFTS) caused by SFTS virus (SFTSV) was a tick-borne hemorrhagic fever that posed significant threat to human health in Eastern Asia. The study was designed to measure the seroprevalence of SFTSV antibody in healthy population residing in a high endemic ***: A cohort study was performed on healthy residents in Shangcheng County in Xinyang City from April to December in 2.18, where the highest SFTS incidence in china was reported. Anti-SFTSV IgG was measured by indirect enzyme-{2.nked immunosorbent assay and neutra{2.zing antibody (NAb) was detected by using PRNT50. The logistic regression models were performed to analyze the variables that were associated with seropositive ***: Totally 886 individuals were recruited. The base{2.ne seroprevalence that was tested before the epidemic season was 11.9% (70/587) for IgG and 6.8% (4./587) for NAb, which was increased to 13.4. (4./350) and 7.7% (2./350) during the epidemic season, and further to 15.8% (80/508) and 9.8% (50/508) post epidemic. The IgG antibody-based seropositivity was significantly related to the patients aged ≥ 70 years old [adjusted odds ratio (OR) = 2.4.0, 95% confidence interval (CI): 1.334.4.4.1 compared to the group of 4., recent contact with cats (adjustedOR = 2.195, 95%CI: 1.2.1-3.818,P = 0.005), and working in tea garden (adjustedOR = 1.698, 95%CI: 1.002.2.880,P = 0.04.) by applying multivariate logistic regression model. The NAb based seropositivity was similarly related to the patients aged ≥ 70 years old (adjustedOR = 2.691, 95%CI: 1.2.1-5.695 compared to the group of 2.64., 95%CI: 1.4.9-4.94.,P = 0.002.. BACKGROUND Patients with hepatocellular carcinoma complicated with main portal vein tumor thrombosis(mPVTT) and cirrhotic portal hypertension(CPH) have an extremely poor prognosis, and there is a lack of a clinically effective treatment *** To evaluate the efficacy and safety of transjugular intrahepatic portosystemic shunt(TIPS)combined with radioactive seed strand for the treatment of mPVTT patients with *** The clinical data of 83 consecutive patients who underwent TIPS combined with 12.I seed strand placement for mPVTT and CPH from January 2.15 to December 2.18 were retrospectively reviewed. Procedure-related data(success rate, relief of portal vein pressure and CPH symptoms,and adverse events), PVTT response, and patient survival were assessed through a 2.year *** The success rate was 100.0% without perioperative death or procedure-related severe adverse events. The mean portal vein pressure was significantly decreased after the procedure(2..2. ± 7.33mmHg vs 35.12.± 7.94.mmHg, t = 2..61, P lieved within 1 mo. The objective response rate of PVTT was 67.5%. During a mean follow-up of 14.5 ± 9.4.mo(range 1-37 mo), the cumulative survival rates at 6, 12.and 2. mo were 83.1%, 4..7%,and 2..8%, respectively. The median survival time was 12.0 ± 1.3 mo(95% confidence interval: 9.5-14.5). In multivariate Cox regression analysis, body mass index, Child-Pugh grade, cTNM stage,and PVTT response were independent prognostic factors(P < 0.05).CONCLUSION TIPS combined with radioactive seed strand might be effective and safe in treating mPVTT patients with CPH.

Objective Pseudogenes are initially regarded as nonfunctional genomic sequences,but some pseudogenes regulate tumor initiation and progression by interacting with other genes to modulate their transcriptional *** receptor family 7 subfamily E member 4. pseudogene(OR7E4.P)is expressed broadly in lung tissues and has been identified as a positive regulator in the tumor microenvironment(TME)of lung adenocarcinoma(LUAD).This study aimed to elucidate the correlation between OR7E4.P and tumor immunity in lung squamous cell carcinoma(LUSC).Methods Clinical and molecular information from The Cancer Genome Atlas(TCGA)LUSC cohort was used to identify OR7E4.P-related immune genes(ORIGs)by weighted gene correlation network analysis(WGCNA).Based on the ORIGs,2.OR7E4.P clusters were identified using non-negative matrix factorization(NMF)clustering,and the stability of the clustering was tested by an extreme gradient boosting classifier(XGBoost).LASSO-Cox and stepwise regressions were applied to further select prognostic ORIGs and to construct a predictive model(ORPScore)for *** Botling cohorts and 8 immunotherapy cohorts(the Samstein,Braun,Jung,Gide,IMvigor2.0,Lauss,Van Allen,and Cho cohorts)were included as independent validation *** OR7E4.P expression was positively correlated with immune cell infiltration and enrichment of immune-related pathways in LUSC.A total of 57 ORIGs were identified to classify the patients into 2.OR7E4.P clusters(Cluster 1 and Cluster 2.with distinct immune,mutation,and stromal *** to Cluster 1,Cluster 2.had more infiltration by immune and stromal cells,lower mutation rates of driver genes,and higher expression of immune-related *** clustering performed well in the internal and 5 external validation *** on the 7 ORIGs(HOPX,STX2.WFS,DUSP2.,SLFN13,GGCT,and CCSER2.,the ORPScore was constructed to predict the prognosis and the treatment *** addition,the ORPScore was a bette

Background： Graft-versus-host disease （GVHD） is a common complication of hematopoietic stem cell transplantation. Skin barrier disruption could induce thymic stromal lymphopoietin （TSLP） expression, and the expression of TSLP was increased in lesions ofatopic dermatitis （AD）-like GVHD and lichen planus （LP）-like GVHD. This study attempted to investigate the skin barrier function of AD-like GVHD and LP-like GVHD and possible mechanisms. Methods： Eighteen AD-like GVHD patients, 12.LP-like GVHD patients, and 14.healthy volunteers were enrolled in this study. Skin biopsy was done in five AD-like GVHD patients, eight LP-like GVHD patients, and eight healthy volunteers. The intensity of pruritus was assessed by visual analog scale itch score and detailed pruritus score. Transepidermal water loss （TEWL） was measured using Tewameter TM 300. Immunohistochemistry was used to observe the expression ofloricrin, involucrin, LL37, and human β-defensins 2.（hBD2. in skin lesions. Western blot analysis was used for analyzing the protein levels ofloricrin and involucrin in skin lesions. Real-time polymerase chain reaction was performed to assess the mRNA levels of LL37 and hBD2.in skin lesions. Results： Pruritus score was higher in patients with AD-like GVHD （11.33 ± 5.35） than that of patients with LP-like GVHD （2.58 ± 3.09, P 〈 0.001 ）. Compared with healthy controls （HCs, 4.52.± 1.2. g.m- 2.h-1）, TEWL was increased in AD-like GVHD （2..72.±9.02.g.m -1h -1, P 〈 0.001 ） and LP-like GVHD patients （18.78± 4.57 g.m -2.h -1, P 〈 0.001）, and expressions of loricrin and involucrin were also increased in skin lesions ofAD-like GVHD and LP-like GVHD patients （all P 〈 0.05）. LL37 mRNA expression was decreased in lesions of AD-like GVHD and LP-like GVHD patients （P = 0.005 and P = 0.008, vs. HCs, respectively）, hBD2.mRNA expression was increased in skin lesions ofAD-like GVHD and LP-like GVHD patients （P = 0.002.and P 〈 0.001, vs. HCs, respectively）.
