Efficient energy storage devices with suitable electrode materials,that integrate high power and high energy,are the crucial requisites of the renewable power source,which have unwrapped new possibilities in the sustainable development of energy and the ***,a facile collagen microstructure modulation strategy is proposed to construct a nitrogen/oxygen dual-doped hierarchically porous carbon fiber with ultrahigh specific surface area(2.88 m^(2.g^(-1))and large pore volume(4.56 cm^(3)g^(-1))via local microfibrous breakage/disassembly of natural structured *** operando spectroscopy and density functional theory unveil that the dual-heteroatom doping could effectively regulate the electronic structure of carbon atom framework with enhanced electric conductivity and electronegativity as well as decreased diffusion resistance in favor of rapid pseudocapacitive-dominated Li^(+)-storage(353 mAh g^(-1)at 10 A g^(-1)).Theoretical calculations reveal that the tailored micro-/mesoporous structures favor the rapid charge transfer and ion storage,synergistically realizing high capacity and superior rate performance for NPCF-H cathode(75.0 mAh g^(-1)at 30 A g^(-1)).The assembled device with NPCF-H as both anode and cathode achieves extremely high energy density(2.0 Wh kg^(-1))with maximum power density(42.00 W kg^(-1))and ultralong lifespan(80%capacity retention over 10000 cycles).

In this work,a new methodology is presented to mainly solve the fluid–solid interaction(FSI)*** methodology combines the advantages of the Newmark precise integral method(NPIM)and the dual neural network(DNN)*** NPIM is employed to modify the exponential matrix and loading vector based on the DNN integral *** involves incorporating the basic assumption of the Newmark-βmethod into the dynamic equation and eliminating the acceleration term from the dynamic equilibrium *** a result,the equation is reduced to a first-order linear equation ***,the PIM is applied to integrate the system step by step within the *** DNN method is adopted to solve the inhomogeneous term through fitting the integrand and the original function with a pair of neural networks,and the integral term is solved using the Newton–Leibniz *** examples demonstrate that the proposed methodology significantly improves computing efficiency and provides sufficient precision compared to the DNN *** is particularly evident when analyzing large-scale structures under blast loading conditions.

Soybean mosaic virus(SMV),an RNA virus,is the most common and destructive pathogenic virus in soybean *** newly developed CRISPR/Cas immune system has provided a novel strategy for improving plant resistance to viruses;hence,this study aimed to engineer SMV resistance in soybean using this ***,multiple sgRNAs were designed to target positive-and/or negative-sense strands of the SMV HC-Pro ***,the corresponding CRISPR/CasRx vectors were constructed and transformed into *** inoculation with SMV,39.02.,35.77%,and 18.70%of T_(1)plants were confirmed to be highly resistant(HR),resistant(R),and mildly resistant(MR)to SMV,respectively,whereas only 6.50%were identified as susceptible(S).Additionally,qRT-PCR and DAS-ELISA showed that,both at 15 and 30 d post-inoculation(dpi),SMV accumulation significantly decreased or was even undetectable in HR and R plants,followed by MR and S ***,the expression level of the CasRx gene varied in almost all T_(1)plants with different resistance level,both at 15 and 30 ***,when SMV resistance was evaluated in the T_(2.generation,the results were similar to those recorded for the T_(1)*** findings provide new insights into the application of the CRISPR/CasRx system for soybean improvement and offer a promising alternative strategy for breeding for resistance to biotic stress that will contribute to the development of SMV-immune soybean germplasm to accelerate progress towards greater soybean crop productivity.

The flower of Syringa pubescens Turcz. （SP）， was used as both medicine and food in china， exhibited a variety of biological activities. However， it remains unknown whether SP has an effect on ameliorative nonalcoholic fatty liver disease （NASH）. In this study， the improvement of SP ethyl acetate extract （SPE） on NASH was evaluated and the potential mechanisms were explored. The glycosides of SPE were determined by an HPLC method. HepG2.cell lines were treated with free fatty acid to clarify the improvement effect and mechanisms of SPE on NASH in vitro. C57BL/6J mice were treated by 60 kcal% high-fat diet （HFD） combined with carbon tetrachloride (CCl4) to establish the NASH model in vivo. The results showed SPE could inhibited the hepatic injury and lipid steatosis by decreasing the levels of ALT， AST， TG and TC in vitro and vivo. The SPE suppressed the oxidative stress and inflammation by restraining the generation of ROS， MDA， IL-1β， IL-6 and TNF-α and enhancing the activity of antioxidant enzymes including SOD， CAT and GSH-Px in NASH model. Moreover， SPE declined the level of fibrosis markers including HYP， Col Ⅰ and α-SMA in HFD combined CCl4（HC）-induced mice. Mechanically， SPE inhibited hepatocellular Keap1 expression and promoted Nrf2.nuclear translocation， which suppressing the phosphorylation of IκBα/NF-κB. In addition， SPE down-regulated the level of TGF-β1 and phosphorylation of Smad2.to mitigate fibrosis. In brief， SPE could significantly alleviate lipid accumulation， oxidative stress， inflammation and fibrosis via regulating Nrf2.HO-1， IκBα/NFκB and TGF-β1/Smad2.pathways in the progression of NASH， which indicated that SPE had the potential to be a novel and effective drug or food supplements for the improvement of NASH.

BACKGROUND In this study,we retrospectively analysed macrophage infiltration and podocyte injury in three patients with diffuse proliferative lupus nephritis(LN)who un-derwent repeated renal *** SUMMARY Clinical data of three diffuse proliferative LN patients with different pathological characteristics(case 1 was LN IV-G(A),case 2.was LN IV-G(A)+V,and case 3 was LN IV-G(A)+thrombotic microangiopathy)were *** patients underwent repeated renal biopsies 6 mo later,and renal biopsy specimens were *** infiltration was assessed by CD68 expression detected by immunohistochemical staining,and an immunofluorescence assay was used to detect podocin expression to assess podocyte *** treatment,Case 1 changed to LN III-(A),Case 2.remained as type V LN lesions,and Case 3,which changed to LN IV-S(A),had the worst *** observed reduced macro-phage infiltration after ***,two of the patients with active lesions after treatment still showed macrophage infiltration in the renal *** treatment,the three patients showed discontinuous expression of ***,the integrity of podocin was restored after treatment in Case *** It may be possible to reverse podocyte damage and decrease the infiltrating ma-crophages in LN patients through effective treatment.

The evolution of ablative Rayleigh–Taylor instability(ARTI)induced by single-mode stationary and time-varying perturbations in heat flux is studied numerically in two *** with the stationary case,time-varying heat-flux perturbation mitigates ARTI growth because of the enhanced thermal smoothing induced by the wave-like traveling heat flux.A resonance is found to form when the phase velocity of the heat-flux perturbation matches the average sound speed in the ablation *** the resonant regime,the coherent density and temperature fluctuations enhance the electron thermal conduction in the ablation region and lead to larger ablation pressure and effective acceleration,which consequently yield higher linear growth rate and saturated bubble *** enhanced effective acceleration offers increased implosion velocity but can also compromise the integrity of inertial confinement fusion shells by causing faster ARTI growth.

CRISPR-Cas12. genome engineering systems have been widely used in plant research and crop breeding. To date, the performance and use of anti-CRISPR-Cas12. systems have not been fully established in plants. Here, we conduct in silico analysis to identify putative anti-CRISPR systems for Cas12.. These putative anti-CRISPR proteins, along with known anti-CRISPR proteins, are assessed for their ability to inhibit Cas12. cleavage activity in vivo and in planta. Among all anti-CRISPR proteins tested, AcrVA1 shows robust inhibition of Mb2.as12. and LbCas12. in E. coli. Further tests show that AcrVA1 inhibits LbCas12. mediated genome editing in rice protoplasts and stable transgenic lines. Impressively, co-expression of AcrVA1 mitigates off-target effects by CRISPR-LbCas12., as revealed by whole genome sequencing. In addition, transgenic plants expressing AcrVA1 exhibit different levels of inhibition to LbCas12. mediated genome editing, representing a novel way of fine-tuning genome editing efficiency. By controlling temporal and spatial expression of AcrVA1, we show that inducible and tissue specific genome editing can be achieved in plants. Furthermore, we demonstrate that AcrVA1 also inhibits LbCas12.-based CRISPR activation(CRISPRa) and based on this principle we build logic gates to turn on and off target genes in plant cells. Together, we have established an efficient anti-CRISPR-Cas12. system in plants and demonstrate its versatile applications in mitigating off-target effects, finetuning genome editing efficiency, achieving spatial-temporal control of genome editing, and generating synthetic logic gates for controlling target gene expression in plant cells.
