Background Infantile epileptic spasms syndrome(IESS)is a serious disease in infants,and it usually evolves to other epilepsy types or syndromes,especially refractory or super-refractory focal *** adrenocorticotropic hormone(ACTH)is one of the first-line and effective treatment plans for IESS,it has serious side effects and is not sufficiently *** A retrospective study of the clinical outcomes of ACTH combined with magnesium sulfate(MgSO4)therapy for IESS in two hospital centers was *** major outcome of the single and combined treatment was evaluated by changes in seizure frequency and improvements in hypsarrhythmia electroencephalography(EEG).To reduce the confounding bias between the two groups,we used SPSS for the propensity score matching(PSM)*** We initially recruited 1205 IESS patients from two Chinese hospitals and treated them with ACTH combined with MgSO4 and ACTH *** 1005 patients were enrolled in the treatment(ACTH combined with MgSO4:744,ACTH:261),and both treatment plans had a more than 55%response ***,compared to patients treated with ACTH alone,those patients treated with ACTH combined with MgSO4 had better performance in terms of the seizure frequency and hypsarrhythmia *** PSM,the two groups also showed significant differences in responder rate[70.8%(95%confidence interval,CI)=66.7%–74.8%)vs.53.8%(95%CI=47.4%–60.2%),Phe lead time to treatment and the number of antiseizure medications taken before treatment were two factors that may affect the clinical *** with less than 3 months of lead time responded to the treatment much better than those with>3 months(Phe overall incidence of adverse reactions in the ACTH combined with MgSO4 group was much lower than that in the ACTH group(31.4%vs.63.1%,Phe treatment,only

The cooling storage ring (CSR) external-target experiment (CEE) is a spectrometer used in construction to study the properties of nuclear matter in high-baryon density regions at the heavy-Ion Research Facility in Lanzhou (HIRFL). This study presents the design, simulation, manufacturing, and testing of a half-size prototype of a multi-wire drift chamber (MWDC) for the CEE. First, the performance of the MWDC connected to homemade electronics was simulated. The results demonstrated that an energy resolution of 18.5% % for 5.9-keV X-rays and a position resolution of 194 mu m for protons can be achieved by the current design. Because the size of the largest MWDC reached 176 cm x 314 cm, a set of 98 cm x 98 cm prototypes was built using the new techniques. The positioning accuracy of the anode wires in this prototype is better than 20 mu m. After optimization, using commercially available electronics, the prototype can achieved an energy resolution of 19.7% % for a Fe-55 X-ray source. The CEE-MWDC detector and electronics were simultaneously tested. An energy resolution of 22% % was achieved for the Fe-55 source; the track residuals were approximately 330 mu m for the cosmic rays. The results demonstrate that the current design and techniques meet the requirements of the CEE-MWDC array.

Fluoropyrimidine-based combination chemotherapy plus targeted therapy is the standard initial treatment for unresectable metastatic colorectal cancer(mCRC),but the prognosis remains *** phase 3 trial(***:NCT03950154)assessed the efficacy and adverse events(AEs)of the combination of PD-1 blockade-activated DC-CIK(PD1-T)cells with XELOX plus bevacizumab as a first-line therapy in patients with mCRC.A total of 202 participants were enrolled and randomly assigned in a 1:1 ratio to receive either first-line XELOX plus bevacizumab(the control group,n=102)or the same regimen plus autologous PD1-T cell immunotherapy(the immunotherapy group,n=100)every 21 days for up to 6 cycles,followed by maintenance treatment with capecitabine and *** main endpoint of the trial was progression-free survival(PFS).The median follow-up was 19.5 *** PFS was 14.8 months(95%CI,11.6-18.0)for the immunotherapy group compared with 9.9 months(8.0-11.8)for the control group(hazard ratio[HR],0.60[95%CI,0.40-0.88];p=0.009).Median overall survival(OS)was not reached for the immunotherapy group and 25.6 months(95%CI,18.3-32.8)for the control group(HR,0.57[95%CI,0.33-0.98];p=0.043).Grade 3 or higher AEs occurred in 20.0%of patients in the immunotherapy group and 23.5%in the control groups,with no toxicity-associated deaths *** addition of PD1-T cells to first-line XELOX plus bevacizumab demonstrates significant clinical improvement of PFS and OS with well tolerability in patients with previously untreated mCRC.

Plants that grow in extreme environments represent unique sources of stress-resistance genes and *** mongolicus(Leguminosae)is a xerophytic evergreen broadleaf shrub native to semi-arid and desert regions;however,its drought-tolerance mechanisms remain poorly ***,we report the assembly of a reference-grade genome for ***,describe its evolutionary history within the legume family,and examine its drought-tolerance *** assembled genome is 843.07 Mb in length,with 98.7%of the sequences successfully anchored to the nine chromosomes of *** genome is predicted to contain 47611 protein-coding genes,and 70.71%of the genome is composed of repetitive sequences;these are dominated by transposable elements,particularly longterminal-repeat *** analyses revealed two whole-genome duplication(WGD)events at 130 and 58 million years ago(mya)that are shared by the genus Ammopiptanthus and other legumes,but no species-specific WGDs were found within this *** genome reconstruction revealed that the *** genome has undergone fewer rearrangements than other genomes in the legume family,confirming its status as a"relict plant".Transcriptomic analyses demonstrated that genes involved in cuticular wax biosynthesis and transport are highly expressed,both under normal conditions and in response to polyethylene glycol-induced *** induction of genes related to ethylene biosynthesis and signaling was also observed in leaves under dehydration stress,suggesting that enhanced ethylene response and formation of thick waxy cuticles are two major mechanisms of drought tolerance in *** expression of AmERF2,an ethylene response factor unique to ***,can markedly increase the drought tolerance of transgenic Arabidopsis thaliana plants,demonstrating the potential for application of *** genes in crop improvement.
