CmMYB3-like negatively regulates anthocyanin biosynthesis and flower color formation during the post-flowering stage in Chrysanthemum morifolium

Color fading caused by a decrease in anthocyanin accumulation during the post-flowering stage significantly affects postharvest quality of chrysanthemum.However,the underlying mechanism by which anthocyanin accumulation decreases during the post-flowering stage still unclear,which greatly restricts design of molecular breeding in chrysanthemum.Here,a chrysanthemum SG7 R2R3 MYB transcription factor(TF),CmMYB3-like,was identified to have a function in regulating anthocyanin biosynthesis during the post-flowering stage.Quantitative real time PCR(qRT-PCR)assays showed that the expression of CmMYB3-like was gradually downregulated when anthocyanin content increased during the flowering stage and was significantly upregulated during the post-flowering stage.Genetic transformation of chrysanthemum and dual-luciferase assays in N.benthamiana leaves showed that CmMYB3-like suppressed anthocyanin accumulation by inhibiting the transcription of CmCHS and CmANS directly and that of CmF3H indirectly.However,overexpression or suppression of CmMYB3-like did not affect the biosynthesis of flavones or flavonols.Genetic transformation of chrysanthemum revealed that the overexpression of CmMYB3-like inhibited anthocyanin accumulation,but its suppression prevented the decrease in anthocyanin accumulation during the post-flowering stage.Our results revealed a crucial role of CmMYB3-like in regulating the color of petals during the post-flowering stage and provided a target gene for molecular design breeding to improve the postharvest quality of chrysanthemum.

Antibody-drug conjugates(ADCs),which combine the advantages of monoclonal antibodies with precise targeting and payloads with efficient killing,show great clinical therapeutic value.The ADCs’payloads play a key role in determining the efficacy of ADC drugs and thus have attracted great attention in the field.An ideal ADC payload should possess sufficient toxicity,low immunogenicity,high stability,and modifiable functional groups.Common ADC payloads include tubulin inhibitors and DNA damaging agents,with tubulin inhibitors accounting for more than half of the ADC drugs in clinical development.However,due to clinical limitations of traditional ADC payloads,such as inadequate efficacy and the development of acquired drug resistance,novel highly efficient payloads with diverse targets and reduced side effects are being developed.This perspective summarizes the recent research advances of traditional and novel ADC payloads with main focuses on the structure-activity relationship studies,co-crystal structures,and designing strategies,and further discusses the future research directions of ADC payloads.This review also aims to provide valuable references and future directions for the development of novel ADC payloads that will have high efficacy,low toxicity,adequate stability,and abilities to overcome drug resistance.

Time-lagged and cumulative effects of drought and anthropogenic activities on China's vegetation greening from 1990 to 2018

Previous studies have confirmed the time-lagged and cumulative effects of drought and anthropogenic activities on vegetation growth,but these studies focus on the time-lagged effect of drought and are poorly known how vegetation productivity responds to anthropogenic activities.Here,based on the reconstructed Normalized Difference Vegetation Index,the Standardized Precipitation Evapotranspiration Index and land use degree comprehensive index,we diagnosed the spatiotemporal pattern of vegetation and drought,investigated time-lagged and cumulative effects of drought and anthropogenic activities over China through the month where the maximum correlation coefficient occurred.It revealed that the browning trend of 32.21%of vegetated lands was covered by overall greening,especially northwestern China.Drought intensified with a rate of 0.0014/year.in 66.41%and 54.57%of the vegetated lands had time-lagged and cumulative response to drought,with a shorter timescales of 1–4 months,indicating the higher sensitivity of vegetation growth to drought.There was a U-shaped relationship between moisture conditions and vegetation response time.49.9%of China’s vegetation showed time-lagged effects to anthropogenic activities,with a longer timescales of 6–10 years,demonstrating that anthropogenic activities triggered ecological changes but vegetation ecosystems cannot keep pace.The accumulated and time-lagged years declined with increased land use intensity.

Influences of different modifiers on the disintegration of improved granite residual soil under wet and dry cycles

The disintegration of granite residual soil is especially affected by variations in physical and chemical properties. Serious geologic hazards or engineering problems are closely related to the disintegration of granite residual soil in certain areas. Research on the mechanical properties and controlling mechanisms of disintegration has become a hot issue in practical engineering. In this paper, the disintegration characteristics of improved granite residual soil are studied by using a wet and dry cycle disintegration instrument, and the improvement mechanism is analyzed. The results show that the disintegration amounts and disintegration ratios of soil samples treated with different curing agents are obviously different. The disintegration process of improved granite residual soil can be roughly divided into 5 stages:the forcible water intrusion stage, microcrack and fissure development stage, curing and strengthening stage, stable stage, and sudden disintegration stage. The disintegration of granite residual soil is caused by the weakening of the cementation between soil particles under the action of water. When the disintegration force is greater than the anti-disintegration force of soil, the soil will disintegrate. Cement and lime mainly rely on ion exchange agglomeration, the inclusion effect of curing agents on soil particles, the hard coagulation reaction and carbonation to strengthen granite residual soil. Kaolinite mainly depends on the reversibility of its own cementation to improve and strengthen granite residual soil. The reversibility of kaolinite cementation is verified by investigating pure kaolinite with a tensile, soaking, drying and tensile test cycle. Research on the disintegration characteristics and disintegration mechanism of improved granite residual soil is of certain reference value for soil modification.

Domain adaptation(DA)for semantic segmentation aims to reduce the annotation burden for the dense pixellevel prediction task.It focuses on tackling the domain gap problem and manages to transfer knowledge learned from abundant source data to new target scenes.Although recent works have achieved rapid progress in this field,they still underperform fully supervised models with a large margin due to the absence of any available hints in the target domain.Considering that few-shot labels are cheap to obtain in practical applications,wc attempt to leverage them to mitigate the performance gap between DA and fully supervised methods.The key to this problem is to leverage the few-shot labels to learn robust domain-invariant predictions effectively.To this end,we first design a data perturbation strategy to enhance the robustness of the representations.Furthermore,a transferable prototype module is proposed to bridge the domain gap based on the source data and few-shot targets.By means of these proposed methods,our approach can perform on par with the fully supervised models to some extent.We conduct extensive experiments to demonstrate the effectiveness of the proposed methods and report the state-of-the-art performance on two popular DA tasks,i.e.,from GTA5 to Cityscapes and SYNTHIA to Cityscapes.

The role of phot1 in triggering hypocotyl phototropism and optimizing growth orientation has been wellcharacterized in Arabidopsis, whereas the role of Zmphot1 in maize remains largely unclear. Here, we show that Zmphot1 is involved in blue light-induced phototropism. Compared with Atphot1, Zmphot1exhibited a weaker phototropic response to very low-fluence rates of blue light( 10 μmol m-2s-1) than Atphot1. Notably, blue light exposure induced Zmphot1-green fluorescent protein(GFP), but not Atphot1-GFP, to form the aggregates in the cytoplasm of Nicotiana benthamiana cells. Furthermore, by generating the chimeric phot1 proteins, we found that the serine-threonine kinase(STK) domain at the C-terminus is responsible for a more volatile membrane association of Zmphot1. Consistently, the chimeric phot1 protein fusing the STK domain of Zmphot1 with other domains of Atphot1 responded similarly as Zmphot1 to both low and high fluence rates of blue light. Interestingly, although both Zmphot1 and Atphot1 interact with AtNPH3, Zmphot1 induced weaker dephosphorylation of NONPHOTOTROPIC HYPOCOTYL 3(NPH3) than Atphot1. Together, our findings indicate that Zmphot1 and Atphot1 exhibit different photosensory function during phototropic response and that the STK domain may play a key role in determining their properties.

Dear Editor,Human monkeypox(MPX),an endemic disease in equatorial Africa,1 has been causing outbreaks in non-endemic countries since early May 2022.Interestingly,many confirmed cases have no history of travel to Africa.This is the first time many MPX cases have been reported concurrently in both non-endemic and endemic countries in disparate geographical areas.2 In response,we have analyzed 27 genes or sequences from 643 full-length human monkeypox virus(MPXV)genomes collected after January 1 and submitted by August 7,2022(Supplementary Table 1).We divided these 643 genomes into 24 clusters with a 95%identity threshold(Supplementary Table 2).
