Investigation of VEGF and PDGF signals in vascular formation by 3D culture models using mouse ES cells

Vascular formation in vivo involves several processes and signal cascades subsequently occurring in the embryo. Several models by ES cells have been reported for analysis in vitro. We show here a 3D culture system using collagen gel (AteloCell) as a simple and useful system for investigating vascular formations and analyzing the roles of factors in vivo. Although VEGF and PDGF are growth factors with multi-potentials for vascular formation, their sequential roles have not been elucidated. We investigated the effects of VEGF and PDGF B signals for vascular formation by a 3D culture system that embedded embryoid bodies (EBs) from ES cells into a collagen gel. After embedding EBs in the collagen gel with a medium containing VEGF, EBs gave off CD105 immunopositive vessels as the initial step of vasculogenesis. When the factor in the culture medium for EBs was switched from VEGF to PDGF B after 5 days of culture, the morphological features of vessels varied, suggesting the occurrence of vascular-type differentiation. After 11 days of 3D culture, vessels in both groups cultured with VEGF alone and switching to VEGF B at day 5 showed Flk-1 immunoreactivity. Some blood vessels cultured with PDGF B after day 5 expressed either EphrinB2 (arteriole marker) or Flt-4 (lymphatic marker) immunoreactivity, but vessels cultured with VEGF alone exhibited neither of them. Vessels cultured with these two factors could not differentiate into a venous type. The present study indicates that VEGF is the initial signal for vasculogenesis, and that PDGF B is probably involved in vascular diversification.

Exploring factors that motivate nursing students to engage in skills practice in a laboratory setting:A descriptive qualitative design

Objectives:The aim of this study was to explore factors that motivate students to engage in skills practice in a laboratory setting,and to identify their motivation types and the regulatory ***:Semi-structured interviews were conducted with 23 nursing students from three universities between November 2017 and January 2018.A thematic analysis was used to identify factors associated with students’motivation to engage in skills practice in a *** types and the regulatory styles of student motivation were identified based on the self-determination ***:Seven motivating factors were *** factors included the students’desire“to acquire the skills necessary to work as a nurse”,the“desire to improve skills in preparation for clinical practicum”,and their felt“obligations to patients as a nurse”.Moreover,“the impetus to study arising from the objective evaluation of oneself and others”and“wanting to pass the skills examination”motivated the students to engage in skills practice.A“learning environment that facilitates students’learning”and the“supportive involvement of educators”facilitated their *** on the self-determination theory,the students were found to embrace extrinsic motivation with four regulatory styles of motivation,namely integrated,identified,introjected,and external ***:Nurse educators should understand the motivating factors of students,and help students embrace a more internally controlled motivation by helping them envision their future careers as nurses,and by fostering their ethical duty to care for patients.

Improvement of regional cerebral blood flow after oral intake of ranched-chain amino acids in patients with cirrhosis

AIM： To evaluate the effect of oral intake of branchedchain amino acids （BCAA） on brain perfusion in patients with liver cirrhosis. METHODS： Single photon emission computed tomography scans were performed in 43 patients with cirrhosis and in 15 age-matched healthy subjects. Twenty-nine out of forty-three patients were randomly treated with either BCAA granules or placebo, and single photon emission computed tomography was performed before and after the treatment. We measured the regional cerebral blood flow values using a threedimensional stereotaxic region of interest template. RESULTS： Cirrhotic patients had regions of significant hypoperfusion in the bilateral central （right P = 0.039, P〈0.05; left P = 0.006 P〈0.01）, parietal （right P = 0.028, P〈0.05; left P = 0.009, P〈0.01）, angular （right P = 0.039, P〈0.05; left P = 0.008, P〈0.01）, and left pericallosal segments （P = 0.038 P〈0.05） as compared with healthy subjects. A significant increase in cerebral perfusion was observed 70 min after the oral intake of BCAA in the angular （right P = 0.012, P〈0.05; left P = 0.049, P〈0.05）, temporal （right P = 0.012, P〈0.05; left P=0.038, P〈0.05）, pericallosal segments （right P = 0.025, P〈0.05; left P = 0.049, P〈0.05） and left precentral （P = 0.044, P〈0.05）, parietal （P = 0.040, P〈0.05） and thalamus （P = 0.033, P〈0.05）. No significant change in perfusion was observed in the placebo group. CONCLUSION： Administration of BCAA rapidly improves cerebral perfusion.

Loss of Masticatory Function Affects Growth and Development of the Mandibular Condyle in Rats

The effects of childhood masticatory function loss and soft foods on the mandibular condyle have been the subject of much research. However, the corresponding bone turnover is not fully understood. The purpose of the present study was to clarify the effects of a lack of teeth and a soft food diet during the growth period on bone turnover in the mandibular condyle. We divided 3-week-old Wistar rats into the following three groups: 1) Extraction group: The maxillary molars were extracted at the age of 4 weeks, and animals were fed powdered standard feed. 2) Powder group: Animals were fed powdered standard feed without tooth extraction. 3) Control group: Animals were fed solid standard feed without tooth extraction. Non-decalcified thin-slice specimens of sagittal sections of the mandibular condyle were obtained at the age of 20 weeks for histological analysis. We used micro-CT analysis and bone histomorphometry to measure bone volume (BV), bone mineral content (BMC), bone mineral density (BMD), bone microstructure, bone resorption, and osteogenesis in the mandibular condyle, and we compared the results among groups. In the extraction and the powder groups, we found deformation and disruption of the arrangement of chondrocytes, coagulation of chondrocytes, and duplication of the tidemark in the cartilage. We also found an increase in multinuclear osteoclasts in the cancellous bone. We found a reduction in BV, BMC, and BMD in the extraction and powder groups compared to the control group, as well as a reduction of bone volume, a lowering of osteogenesis parameters, and an increase in bone resorption parameters in the secondary cancellous bone. These results suggest that a lack of teeth and a soft food diet during the growth period cause a decline in bone microstructure, a decrease in osteogenesis, and an increase in bone resorption.

AIM To evaluate the efficacy and safety of a regimen containing sofosbuvir(SOF) and ledipasvir(LDV) in Japanese patients aged ≥ 75 years with hepatitis C genotype *** This multicenter, retrospective study consisted of 246 Japanese patients with HCV genotype 1 at nine centers in Miyazaki prefecture in Japan. Demographic, clinical, virological, and adverse effects(AE)-related data obtained during and after SOF/LDV therapy were collected from medical records. These patients were divided into two groups, younger(aged < 75 years) and elderly(aged ≥ 75 years). Virological data and AEs were analyzed by age *** The sustained virological response(SVR) rates at 12 wk after treatment were 99.2%, 99.4%, and 98.7% in the overall population and in patients aged < 75 and ≥ 75 years, respectively. Common AEs during therapy were headache, pruritus, constipation, and insomnia. These occurred in fewer than 10% of patients, and their incidence was not significantly different between the younger and elderly groups. Two patients discontinued treatment, one due to a skin eruption and the other due to cerebral bleeding. CONCLUSION Compared with younger patients, elderly patients had a similar virological response and tolerance to SOF/LDV therapy.

Relation of stathmin expression with tumor invasion in human tongue cancer

Background: Stathmin is a cytosolic phosphoprotein expressed at high levels in most human cancer cells。Stathmin expression and phosphorylation are linked to the control of cell differentiation and proliferation。However, few studies of stathmin expression in human oral squamous cell carcinoma have been reported。We performed immunohistochemical staining of stathmin and analyzed the relation with the mode of invasion in human tongue cancer。Methods: Oral biopsy specimens of 42 cases of tongue cancer were used。Paraffin sections were deparaffinized and primary antibody (anti-stathrnin monoclonal antibody) was added。The mode of invasion was divided into five types from grades 1 to 4d according to yamamoto (Cancer51;2175-80,1983)。Of these 42cases,29were grades 2+3 and 13 were grades 4c+4d。Results: No obvious immunostaining of stathmin was observed in normal tongue epithelium。Positive expression of stathmin in tumor cell was detected in all 42 cases of tongue cancer。However, the expression pattern was clearly different depending on mode of invasion。In cases of grades 2+3, stathmin expression observed in cancer cells located inside of the cancer nests。In cases of grade 4c+4d, strong positive staining for stathmin was detected throughout the small cancer nests。Extensive staining was observed in tumor cells invading diffusely into the deepest portion of the tissue。Conclusions: Expression of stathmin in human tongue cancer cells correlates with the mode of invasion。Thus, stathmin may play an important role in the invasion of tongue cancer。
