Objective:Celastrol is a pentacyclic triterpenoid extracted from the traditional Chinese medicinal herb,Tripterygium *** study aims to provide a scientific basis for the rational development and use of celastrol in breast ***:A quantitative chemical biology approach was used to investigate the protein targets and molecular mechanisms of celastrol in breast cancer ***:Low-concentration celastrol exerted an anti-tumor effect by directly binding to hydroxysteroid dehydrogenase-like 2(HSDL2)and inhibiting its ***,the expression of the pro-apoptotic protein,Bcl-2-associated X(BaX),increased,the level of the anti-apoptotic protein,B-cell lymphoma-2(Bcl-2),decreased,and the rate of apoptosis *** the transfection of cells with si-HSDL2,the apoptosis rate was similar to that observed after the administration of ***,apoptosis was reversed by the overexpression of ***,our mass spectrometry(MS)data indicated a relationship between HSDL2 and the mitogen-activated protein kinase(MAPK)signaling *** also found that the expression of HSDL2 was directly related to the degree of extracellular signal-regulated kinase(ERK)***:Celastrol may promote apoptosis by suppressing the HSDL2/MAPK/ERK signaling pathway.

目的探讨终末期肾脏病(ESRD)患者成纤维细胞生长因子-23(FGF23)血清浓度与心力衰竭及全因死亡的相关性。方法采用前瞻性队列研究,纳入医院肾脏内科收治的无心衰症状的ESRD患者,采用基线问卷和体格检查、超声心动图检查、实验室检查收集患者数据,采用酶联免疫吸附法(ELISA)检测患者血清FGF23浓度。随访时间2年,以随访发生新发的心力衰竭(ACC/AHA stage C-D)和全因死亡为复合终点结局事件,采用Cox比例风险模型分析患者发生结局事件的危险因素,通过亚组分析和交互作用分析,进一步探讨FGF23与结局事件的关联在不同亚组中是否存在异质性。结果该研究最终纳入ESRD患者107例,平均年龄(52.00±12.51)岁,男性39(36.45%)例,中位随访时间为23个月(21,25个月),出现结局事件32(29.9%)例,其中新发心衰22(20.6%)例,全因死亡10(9.3%)例。该研究结果显示结局事件组患者血清FGF23浓度显著高于非事件组[(4.40±1.16)pmol/ml vs(3.85±0.82)pmol/ml,P<0.05]。Cox比例风险模型结果显示升高的FGF23可以增加ESRD患者发生结局事件的风险(HR=1.730,95%CI:1.164~2.570,P=0.007)。亚组分析显示FGF23水平与性别对于结局事件发生风险存在交互作用,尤其在男性ESRD患者中升高的FGF23风险更高(P_(-交互作用)<0.05)。结论升高的血清FGF23是ESRD患者发生心衰和全因死亡的独立危险因素,尤其在男性患者中风险更高。

目的利用欧洲五维健康评定量表(EQ-5D-5L)调查腹膜透析(PD)患者健康相关生活质量(HRQOL)并分析其影响因素,为改善患者生活质量提供建议。方法纳入医院肾脏内科住院治疗并规律随访的PD患者,收集患者一般人口学资料、实验室检查指标。采用6分钟步行实验(6-MWT)评估患者运动耐力,抑郁筛查评价量表(PHQ-9)评价患者抑郁状态,EQ-5D-5L量表进行HRQOL评分,多因素线性回归分析EQ-5D-5L健康效用值的影响因素。结果研究纳入90例PD患者,年龄18~70(49.44±11.41)岁,EQ-5D-5L的健康效用值最高为1.00分,最低为-0.01分,平均(0.92±0.15)分。多因素线性回归分析显示胆红素升高(β=-0.009,P=0.018)、C-反应蛋白升高(β=-0.005,P<0.001)、PHQ-9评分升高(β=-0.008,P=0.014)与PD患者EQ-5D-5L健康效用值呈负向关联,6分钟步行距离升高(β=0.005,P=0.018)与PD患者EQ-5D-5L健康效用值呈正向关联。结论胆红素和CRP水平、抑郁状态、运动耐量是影响PD患者HRQOL的主要影响因素。

On the Optimal Initial Inner-Core Size for Tropical Cyclone Intensification: An Idealized Numerical Study

Recent observational and numerical studies have revealed the dependence of the intensification rate on the inner-core size of tropical cyclones (TCs). In this study, with the initial inner-core size (i.e., the radius of maximum wind-RMW) varied from 20-180 km in idealized simulations using two different numerical models, we found a nonmonotonic dependence of the lifetime maximum intensification rate (LMIR) on the inner-core size. Namely, there is an optimal inner-core size for the LMIR of a TC. Tangential wind budget analysis shows that, compared to large TCs, small TCs have large inward flux of absolute vorticity due to large absolute vorticity inside the RMW. However, small TCs also suffer from strong lateral diffusion across the eyewall, which partly offsets the positive contribution from large inward flux of absolute vorticity. These two competing processes ultimately lead to the TC with an intermediate initial inner-core size having the largest LMIR. Results from sensitivity experiments show that the optimal size varies in the range of 40-120 km and increases with higher sea surface temperature, lower latitude, larger horizontal mixing length, and weaker initial TC intensity. The 40-120 km RMW corresponds to the inner-core size most commonly found for intensifying TCs in observations, suggesting the natural selection of initial TC size for intensification. This study highlights the importance of accurate representation of TC inner-core size to TC intensity forecasts by numerical weather prediction models.

针对引黄灌区灌溉用水效率的综合评价问题,利用灌溉用水流向跟踪法,构建了由19个确定性指标、3个子系统组成的开放型评价指标体系,按照多重赋权理念,确定了在多层次模糊评价体系中的各个层采用多种赋权方法。在指标体系的第1层(输入层),通过计算指标数据所携带的熵值对其赋予数学权重;在第2层(准则层),采用熵值权重法、等权重法和经验赋权法等对3个子系统进行多重赋权,获得和比较不同权重对评价结果的影响。选择三义寨引黄灌区为研究实例,对2005—2014年共计10 a的灌溉用水效率进行综合评价,经过赋权计算,第1种熵值权向量为(0.307,0.348,0.345),第2种等权向量为(0.333,0.333,0.334),第3种主观权向量为(0.400,0.200,0.400)3种不同权重的评价结果呈现出整体趋势和走向基本一致,权重对于评价结果的影响非常有限。根据多重赋权的评价结果,2013年均为10 a中灌溉用水效率的最优年份,评价等级为优,综合评价平均值为0.993。此评价结果与三义寨引黄灌区的实际情况相符,2013年在降雨量和地下水相对较少的情况下,灌溉实际引水量和实际灌溉面积都保持了较高水平,保证了灌溉的效果,也确保了灌区作物的丰收。根据第二层的评价值可以清晰地看到工程因素和管理因素两个子系统的动态变化过程,2007年之前都处于较低发展水平,2009年之后进入稳步发展阶段,从而查找到灌溉用水过程中存在的问题和薄弱的环节,为下一步引黄灌区的科学规划和可持续发展提供数据基础和技术支撑。

Since 1980,both the intensity and duration of summer heatwaves in the middle and high latitudes of the Northern Hemisphere have significantly increased,leading this region to become a critical area for a significant increase in the frequency of intense and long-lived extreme *** found that stronger and more persistent high-pressure systems and lower soil moisture before the events were the main drivers of intense and long-lived extreme heatwaves in western Europe and the middle and high latitudes of North ***,in eastern Europe and Siberia,lower cloud cover before events is also a main driver of this type of extreme heatwave,in addition to the above *** factors are coupled with each other and can change heatwave intensity and duration by influencing surface radiation processes during *** the self-organizing map classification method,we found that 6 weather patterns with increased frequency,intensity,and duration were the main dynamic reasons leading to the increase in intense and long-lived extreme heatwaves after *** addition,the decrease in summer average soil moisture in most areas of the mid-high latitudes of the Northern Hemisphere and the decrease in average cloud cover in eastern Europe and Siberia are found to be the main thermodynamic reasons leading to the increase in these extreme heatwaves.

In this study,we investigated how empathic neural responses unfold over time in different empathy networks when viewing same-race and other-race individuals in dynamic painful *** recorded magnetoencephalography signals from Chinese adults when viewing video clips showing a dynamic painful(or non-painful)stimulation to Asian and White models’faces to trigger painful(or neutral)*** found that perceived dynamic pain in Asian models modulated neural activities in the visual cortex at 100 ms–200 ms,in the orbitofrontal and subgenual anterior cingulate cortices at 150 ms–200 ms,in the anterior cingulate cortex around 250 ms–350 ms,and in the temporoparietal junction and middle temporal gyrus around 600 ms after video *** dynamic pain in White models modulated activities in the visual,anterior cingulate,and primary sensory cortices after 500 *** findings unraveled earlier dynamic activities in multiple neural circuits in response to same-race(vs other-race)individuals in dynamic painful situations.

The composition of serum is extremely complex,which complicates the discovery of new pharmacodynamic biomarkers via serum proteome for disease prediction and ***,nanoparticles have been reported to efficiently reduce the proportion of high-abundance proteins and enrich lowabundance proteins in ***,we synthesized a silica-coated iron oxide nanoparticle and developed a highly efficient and reproducible protein corona(PC)-based proteomic analysis strategy to improve the range of serum proteomic *** identified 1,070 proteins with a median coefficient of variation of 12.56%using PC-based proteomic analysis,which was twice the number of proteins identified by direct *** were also more biological processes enriched with these *** applied this strategy to identify more pharmacodynamic biomarkers on collagen-induced arthritis(CIA)rat model treated with methotrexate(MTX).The bioinformatic results indicated that 485 differentially expressed proteins(DEPs)were found in CIA rats,of which 323 DEPs recovered to near normal levels after treatment with *** strategy can not only help enhance our understanding of the mechanisms of disease and drug action through serum proteomics studies,but also provide more pharmacodynamic biomarkers for disease prediction,diagnosis,and treatment.

Cardioprotective effects of combination of notoginseng total saponins and safflower total flavonoids against myocardial infarction in rats

In this study, the cardioprotective mechanism of the combination of notoginseng total saponins and safflower total flavonoids（CNS） was investigated due to its excellently efficacy against myocardial infarction（MI） in rats. After the left anterior descending coronary artery（LADCA） ligation, rats were orally administered with CNS for 7 consecutive days. CNS prevented MI-induced pathophysiological changes and significantly decreased plasma levels of myocardial enzymes, including creatine kinase MB isoenzyme（CK-MB）, lactate dehydrogenase（LDH） and aspartate aminotransferase（AST）. Further investigation revealed that CNS attenuated the production of inflammatory factors in plasma, including tumor necrosis factor-alpha（TNF-α）, interleukin-6（IL-6） and interleukin-1β（IL-1β）. Moreover, CNS treatment decreased the expression of caspase-3 at the mR NA level in infarct tissue. Our findings demonstrated that the anti-inflammatory and anti-apoptotic properties of CNS might confer its cardioprotection against MI in rats.
