Meeting the damage tolerance requirements for engineering-grade titanium alloys pose a significant challenge in achieving high fracture toughness in direct energy deposition(DED)titanium alloys.This work primarily investigated the relationship between the microstructure and the fracture toughness of DED new Ti-6Al-4V-1Mo alloy.Two types of microstructures were designed via two process strategies:high-line energy density(he)and low-line energy density(LE).Relative to LE samples,he samples possess larger-sized microstructural characteristics(coarser grain boundaryα(α_(GB)),largerαcolonies,and coarserαlaths).Lessα/βphase boundaries were formed by coarserαlaths in the he samples,increasing the movement of dislocations,resulting in tensile strength decreasing from 1007.1 MPa(LE)to 930.8 MPa(he)and elongation increasing from 10.8%(LE)to 15.7%(he).Also,he samples exhibited an excellent fracture toughness of 114.0 MPa m^(1/2),significantly higher than that of LE samples(76.8 MPa m^(1/2)).An analysis of crack propagation paths was conducted to investigate the factors contributing to toughening.The primary factor enhancing toughness is the frequent obstruction of cracks by coarseαGB and largeαcolonies in he samples.Particularly,the pretty large-angle deflections induced by the superposition effect of coarseαGB and largeαcolonies play a vital of significant role.These factors induced the long and tortuous high-energy pathways,which resulted in ultimately improved fracture toughness.The discovered microstructural toughening mechanisms can serve as a reference for future studies involving titanium alloys,offering insights on how to enhance fracture toughness by achieving similar characteristics.

Background The reproductive performance of chickens mainly depends on the development of follicles.Abnor-mal follicle development can lead to decreased reproductive performance and even ovarian disease among chick-ens.Chicken is the only non-human animal with a high incidence of spontaneous ovarian cancer.In recent years,the involvement of circRNAs in follicle development and atresia regulation has been confirmed.Results In the present study,we used healthy and atretic chicken follicles for circRNA RNC-seq.The results showed differential expression of circRALGPS2.It was then confirmed that circRALGPS2 can translate into a protein,named cir-cRALGPS2-212aa,which has IRES activity.Next,we found that circRALGPS2-212aa promotes apoptosis and autophagy in chicken granulosa cells by forming a complex with PARP1 and HMGB1.Conclusions Our results revealed that circRALGPS2 can regulate chicken granulosa cell apoptosis and autophagy through the circRALGPS2-212aa/PARP1/HMGB1 axis.

Plant morphogenesis relies on precise gene expression programs at the proper time and position which is orchestrated by transcription factors(TFs)in intricate regulatory networks in a cell-type specific manner.here we introduced a comprehensive single-cell transcriptomic atlas of Arabidopsis seedlings.This atlas is the result of meticulous integration of 63 previously published scRNA-seq datasets,addressing batch effects and conserving biological variance.This integration spans a broad spectrum of tissues,including both below-and above-ground parts.Utilizing a rigorous approach for cell type annotation,we identified 47 distinct cell types or states,largely expanding our current view of plant cell compositions.We systematically constructed cell-type specific gene regulatory networks and uncovered key regulators that act in a coordinated manner to control cell-type specific gene expression.Taken together,our study not only offers extensive plant cell atlas exploration that serves as a valuable resource,but also provides molecular insights into gene-regulatory programs that varies from different cell types.

PLATZ is a novel zinc finger DNA-binding protein that plays an important role in regulating plant growth and development and resisting abiotic stress.However,there has been very little research on the function of this family gene in tomatoes,which limits its application in germplasm resource improvement.Therefore,the PLATZ gene family was identified and analyzed in tomato,and its roles were predicted and verified to provide a basis for in-depth research on SlPLATZ gene function.In this study,the PLATZ family members of tomato were identified in the whole genome,and 19 SlPLATZ genes were obtained.Functional prediction was conducted based on gene and promoter structure analysis and RNA-seq-based expression pattern analysis.SlPLATZ genes that responded significantly under different abiotic stresses or were significantly differentially expressed among multiple tissues were screened as functional gene resources.SlPLATZ17 was selected for functional verification by experiment-based analysis.The results showed that the downregulation of SlPLATZ17 gene expression reduced the drought and salt tolerance of tomato plants.Tomato plants overexpressing SlPLATZ17 had larger flower sizes and long,thin petals,adjacent petals were not connected at the base,and the stamen circumference was smaller.This study contributes to understanding the functions of the SlPLATZ family in tomato and provides a reference for functional gene screening.

At present,China is facing new strategic opportunities as well as risks and challenges in its development.Faced with the challenges brought by current global major risks,China urgently needs to build a group of world-class enterprises as ballast stones.Accelerating the development of a number of world-class enterprises is an important support for China to enhance its comprehensive strength and international competitiveness,and is crucial for the realization of the strategic blueprint for socialist modernization.After more than forty years of reform and opening up,Chinese enterprises have rapidly developed and grown stronger,with their position rising and their influence in the global market increasing continuously.However,Chinese enterprises are“large in size but not yet strong enough”.In this context,this paper systematically analyzes the practical significance and theoretical connotation of building world-class enterprises and the status quo of building world-class enterprises in China and takes CNOOC as an example to deeply analyze the practices of large central enterprises to build world-class enterprises at the levels of group and subsidiaries,providing a reference for Chinese enterprises to accelerate their progress towards world-class enterprises.

MXenes have aroused intensive enthusiasm because of their exotic properties and promising applications.However,to date,they are usually synthesized by etching technologies.Developing synthetic technologies provides more opportunities for innovation and may extend unexplored applications.here,we report a bottom-up gas-phase synthesis of Cl-terminated MXene(Ti_(2)CCl_(2)).The gas-phase synthesis endows Ti_(2)CCl_(2) with unique surface chemistry,high phase purity,and excellent metallic conductivity,which can be used to accelerate polysulfide conversion kinetics and dramatically prolong the cyclability of Li-S batteries.In-depth mechanistic analysis deciphers the origin of the formation of Ti_(2)CCl_(2) and offers a paradigm for tuning MXene chemical vapor deposition.In brief,the gas-phase synthesis transforms the synthesis of MXenes and unlocks the hardly achieved potentials of MXenes.

The process of domestication has led to dramatic differences in behavioral traits between domestic dogs and gray wolves. Whole-genome research found that a class of putative positively selected genes were related to various aspects of learning and memory, such as long-term potentiation and long-term depression. In this study, we constructed a single-nucleus transcriptomic atlas of the dog hippocampus to illustrate its cell types, cell lineage and molecular features. Using the transcriptomes of 105 057 nuclei from the hippocampus of a Beagle dog, we identified 26 cell clusters and a putative trajectory of oligodendrocyte development.Comparative analysis revealed a significant convergence between dog differentially expressed genes(DEGs) and putative positively selected genes(PSGs). Forty putative PSGs were DEGs in glutamatergic neurons, especially in Cluster 14, which is related to the regulation of nervous system development. In summary, this study provides a blueprint to understand the cellular mechanism of dog domestication.

目的探讨钛表面构筑的有序微纳米分级结构复合羟基磷灰石涂层对骨髓间充质干细胞(BMMSC)黏附、增殖及成骨分化的影响,以期构筑出具有良好成骨活性的钛试样。方法在纯钛试样(对照组)表面制备有序微纳米分级结构(MN组),通过交替循环浸泡法在MN组钛试样表面进一步沉积羟基磷灰石(HA组),扫描电镜观察3组钛试样的表面形貌。将BMMSC接种至3组钛试样表面,4′,6-二脒基-2-苯基吲哚染色法检测0.5、1.0、2.0 h后细胞黏附情况,扫描电镜观察2 d后细胞黏附形态,细胞计数试剂盒法检测1、3、7 d时细胞增殖情况,茜素红染色法和实时荧光定量PCR分别检测14 d后细胞外基质矿化和成骨相关基因表达情况,每组每项实验各3个试样。结果HA组钛试样保留了MN组钛试样原有的20μm孔径的微米凹坑阵列,且原有的70 nm管径的二氧化钛纳米管上出现均匀的花瓣样羟基磷灰石沉积。与对照组相比,MN组和HA组BMMSC顺着微米结构充分铺展和交汇,并伸出较多伪足和伪板,HA组试样表面这种趋势表现得更明显。HA组各时间点早期细胞黏附数量均显著大于对照组和MN组(P0.05)。HA组细胞外基质矿化检测A值(0.607±0.011)显著大于对照组和MN组(分别为0.268±0.025和0.522±0.022)(t=-0.25,P<0.001;t=-0.34,P<0.001)。HA组成骨相关基因表达水平均显著高于对照组和MN组(P<0.05)。结论钛表面构筑的有序微纳米分级结构复合羟基磷灰石涂层可促进BMMSC的细胞黏附和成骨分化,并支持BMMSC细胞增殖,具有良好的成骨活性。

目的系统评价直肠癌患者术后低位前切除综合征发生风险预测模型。方法计算机检索PubMed、Web of Science、Embase、Cochrane Library、Scopus、CINHAL、CNKI、CBM、WanFang Data和VIP数据库,搜集与研究目的相关的研究,检索时限均为建库至2023年6月13日。由2名研究者独立筛选文献,按照预测模型系统评价的关键评估和数据提取清单(CHARMS)提取数据,应用预测模型研究偏倚风险评估工具(PROBAST)评价研究质量。结果共纳入14项研究。所有研究都报告了模型区分度,其中10项研究报告了校准。8项研究对模型进行了内部验证,5项研究进行了外部验证。纳入模型最常见的预测因子为肿瘤距肛缘的距离、新辅助治疗、吻合口瘘、BMI。仅5项研究的整体适用性较好,所有研究均有很高的偏倚风险,偏倚风险主要来源于研究对象、结果、分析领域。结论直肠癌患者术后低位前切除综合征发生风险预测模型尚存在诸多不足,未来研究可考虑对现有模型进行外部验证和重新校准,或参照方法学指南构建新的预测模型并进行验证。
