P-glycoprotein regulating biphasic insulin secretion in rat pancreatic beta cells

Background A 65-kD mdrl （multi-drug resistance protein 1, P-glycoprotein）-Iike protein has been suggested to be the regulatory protein to the chloride channel protein 3 （CIC-3） mediating insulin granules acidification and release in mouse pancreatic beta cells. But the protein has not been deeply investigated. In this study, we identified existence of the 65-kda protein in rat islets and preliminarily explored its biological functions. Methods Total RNAs of rat kidneys served as positive controls, and pancreas, islets and INS-1 cells were extracted for reverse-transcript PCR （RT-PCR）, respectively. The cDNAs were run with specific primers selected from the mRNA of abcblb encoding P-glycoprotein. All PCR products were visualized in agarose gel electrophoresis and sequenced. Homogenates of rat islets and INS-1 cells were applied to SDS-PAGE. P-glycoprotein was detected by a specific monoclonal antibody, C219. Biphasic insulin release was measured in static incubations of rat islets with radioimmunology assay. Results Compared with positive control, expression of the P-glycoprotein mRNA segments were detected in the islets, INS-1 cells and pancreas. Sequence analysis confirmed that the PCR products were matched with mRNA of P-glycoprotein. A 65-kda protein was recognized by the antibody in the islets homogenate but not in that of INS-1 cells in Western-blotting. Instead, the homogenate of INS-1 cells contained a 160-kda protein recognized by the antibody. Insulin secretion of rat islets were stimulated by high glucose （16.7mmol/L）, and showed biphasic curve during 60-minute incubation. After co-incubation with cyclosporine A （CsA）, specific inhibitor to P-glycoprotein, the second phase of insulin secretion was reduced significantly while the first phase was not influenced. Conclusions The 65-kda protein expressed in rat islets is most likely a mini-P-glycoprotein. It may play a key role regulating biphasic insulin release.

Application potential of high performance steels for weight reduction and efficiency increase in commercial vehicles

The fast-growing economy and the gradually established highway system have boosted the road trans- portation for both passenger and cargo over the last decade in China. From 2000 to 2010 Chinese GDP increased by around 10.15% annually and the sales of medium and heavy trucks by around 18.87% （sales increased from 0.2 million in 2000 to 1.3 million in 2010） according to the National Bureau of Statistics of People＇s Republic of China. Today commercial vehicles consume almost the same amount of fuel as pas- senger cars in China although the number of commercial vehicles is only about one fourth of passenger cars. It is estimated that around 50% of imported fuel to China each year will be consumed by vehicle transportation. This si- tuation will worsen fuel shortage problems in the long run and at the same time it is partially responsible for the ever- worsening air pollution in China. Due to the widespread overloading in China, lightweight development in commer- cial vehicles has fallen far behind that of passenger cars with the consequences that Chinese commercial vehicles consume in average about 20% more fuel, especially the heavy trucks, compared to European models. Under these circumstances it is essential to reduce the vehicle fuel consumption and in- crease the transport efficiency. The key solution thereby is to implement lightweight design in commercial vehicles as it has been successfully practiced over the last decade in the passenger cars. This paper summarizes highlights given in presentations during the ＂International seminar on the ap- plication of high strength steels in light weight commercial vehicles＂ with the focus on the development and application of Nb alloyed high performance steels made for lightweight commercial vehicles.

Two-dimensional bimetallic selenium-containing metal-organic frameworks and their calcinated derivatives as electrocatalysts for overall water splitting

The use of two-dimensional(2D)layered metal-organic frameworks(MOFs)as self-sacrificial templates has been proven to be a successful method to create high-efficiency Selenium(Se)-containing electrocatalysts for overall water ***,two strategies are then utilized to introduce Se element into the Co–Fe MOF,one being the etching of as-prepared MOF by SeO_(2) solution,and the other,the replacing of SCN−with SeCN−as the construction *** electrochemical activity of the pristine 2D MOF and their calcinated derivatives for catalyzing the hydrogen evolution reaction(HER)and oxygen evolution reaction(OER)is evaluated and further *** is found that the effect of introducing Se on improving electrochemical catalytic activity is significant for the HER ***,the calcinated derivative in the replacing method exhibits an overpotential of 235 mV for HER and 270 mV for OER at a current density of 10 mA/*** comparing the two methods of introducing Se element into MOF,similar electrocatalytic activity can be achieved on the their calcinated *** high electrochemical performance of 2D CoFe-MOF derivatives may be resulted from the unique 2D hierarchical porous structure and strong synergistic effect between different components in the material.

Probabilistic seismic inversion based on physics-guided deep mixture density network

Deterministic inversion based on deep learning has been widely utilized in model parameters *** by logging data,seismic data,wavelet and modeling operator,deterministic inversion based on deep learning can establish nonlinear relationships between seismic data and model ***,seismic data lacks low-frequency and contains noise,which increases the non-uniqueness of the *** conventional inversion method based on deep learning can only establish the deterministic relationship between seismic data and parameters,and cannot quantify the uncertainty of *** order to quickly quantify the uncertainty,a physics-guided deep mixture density network(PG-DMDN)is established by combining the mixture density network(MDN)with the deep neural network(DNN).Compared with Bayesian neural network(BNN)and network dropout,PG-DMDN has lower computing cost and shorter training time.A low-frequency model is introduced in the training process of the network to help the network learn the nonlinear relationship between narrowband seismic data and low-frequency *** addition,the block constraints are added to the PG-DMDN framework to improve the horizontal continuity of the inversion *** illustrate the benefits of proposed method,the PG-DMDN is compared with existing semi-supervised inversion *** synthetic data examples of Marmousi II model are utilized to quantify the influence of forward modeling part,low-frequency model,noise and the pseudo-wells number on inversion results,and prove the feasibility and stability of the proposed *** addition,the robustness and generality of the proposed method are verified by the field seismic data.

Closed-Form Solution for a Circular Nanohole with Surface Effects Under Uniform Heat Flux

This paper investigates the steady-state thermoelastic problem of a circular nanohole embedded in an infinitely large elastic plane subjected to a uniform far-field heat flux.A lowly conductive surface model is used to account for the effects of surface phonon scattering,while the complete Gurtin–Murdoch model is utilized to characterize the effects of surface tension and surface *** closed-form solution to the temperature and stress field surrounding the hole is derived in the context of complex variable *** numerical examples are presented to analyze the influence of surface effects on thermal stress *** is shown that surface effects induce notable increases in normal and shear stresses around the ***,all three stress components(hoop,normal,and shear)in the vicinity of the hole exhibit substantial augmentation with increasing surface tension and surface *** particular,it is found that the presence of surface effects amplifies the variation in stress gradients and intensifies stress concentration around the hole.

A flat-foldable equiangular spiral folding pattern inspired by sunflowers for deployable structures

Flasher origami pattern has been widely utilized to improve the stowage efficiency of deployable ***,flasher origami cannot be folded fully flat,and they still have great potential for optimization in terms of storage volume and folding *** this paper,a flat foldable equiangular spiral folding pattern inspired by the sunflower disk is ***,a parametric design method for this equiangular spiral crease diagram is introduced in ***,a kinematic model of the equiangular spiral folding pattern is established based on the kinematic equivalence between rigid origami and spherical linkages.A simulation of the developed model demonstrates that the equiangular spiral folding pattern can be folded *** the folded ratio as an evaluation index,the calculated results and experiments show that the equiangular spiral crease pattern can yield fewer creases and improve stowage efficiency in comparison to flasher origami *** spiral folding pattern can save a considerable amount of space and provide a new approach to spatially deployable structures.
