The various advantages of organic polymer monoliths, including relatively simple preparation processes,abundant monomer availability, and a wide application range of pH, have attracted the attention of chromatographers. Organic polymer monoliths prepared by traditional methods only have macropores and mesopores, and micropores of less than 50 nm are not commonly available. These typical monoliths are suitable for the separation of biological macromolecules such as proteins and nucleic acids, but their ability to separate small molecular compounds is poor. In recent years, researchers have successfully modified polymer monoliths to achieve uniform compact pore structures. In particular, microporous materials with pores of 50 nm or less that can provide a large enough surface area are the key to the separation of small molecules. In this review, preparation methods of polymer monoliths for high-performance liquid chromatography, including ultra-high cross-linking technology, post-surface modification, and the addition of nanomaterials, are discussed. Modified monolithic columns have been used successfully to separate small molecules with obvious improvements in column efficiency.

Current design methods for the internal stability of geosynthetic-reinforced soil(GRS)walls postulate seismic forces as inertial forces,leading to pseudo-static analyses based on active earth pressure theory,which yields unconservative reinforcement loads required for seismic stability.Most seismic analyses are limited to the determination of maximum reinforcement strength.This study aimed to calculate the distribution of the reinforcement load and connection strength required for each layer of the seismic GRS wall.Using the top-down procedure involves all of the possible failure surfaces for the seismic analyses of the GRS wall and then obtains the reinforcement load distribution for the limit state.The distributions are used to determine the required connection strength and to approximately assess the facing lateral deformation.For sufficient pullout resistance to be provided by each reinforcement,the maximum required tensile resistance is identical to the results based on the Mononobe-Okabe method.However,short reinforcement results in greater tensile resistances in the mid and lower layers as evinced by compound failure frequently occurring in GRS walls during an earthquake.Parametric studies involving backfill friction angle,reinforcement length,vertical seismic acceleration,and secondary reinforcement are conducted to investigate seismic impacts on the stability and lateral deformation of GRS walls.

Many location-based services need to query objects existing in a specific space,such as location-based tourism resource recommendation.Both a large number of spatial objects and the real-time object access requirements of location-based services pose a big challenge for spatial object storage and query management.In this paper,we propose HGeoHashBase,an improved storage model by integrating GeoHash with key-value structure,to organize spatial objects for efficient range queries.GeoHash is responsible for spatial encoding and key-value structure as underlying data storage.Both the similarity of the encodings for objects in the close geographical locations and the multi-version data mechanism are blended into the proposed model well.Considering the tradeoff between encoding precision and query performance,a theoretical proof is presented.Extensive experiments are designed and conducted,whose results show that the proposed model can gain significant performance improvement.

Organic geochemical characteristics of bark coal in Changguang area: evidence from aromatic hydrocarbons

Seven coal samples were collected from Changguang area,a typical bark coal field in Zhejiang Province,China.The organic geochemical characteristics and depositional environment of bark coal were studied by Rock–Eval pyrolysis,gas chromatography(GC),and gas chromatography–mass spectrometry(GC–MS)analysis.The results show that all samples are type II kerogen.The values of total organic carbon(TOC),potential yield(S1+S2),and hydrogen index of these samples ranged from 65.80 wt%–87.00 wt%,170.43–232.89 mg HC/g,and 234–295 mg HC/g TOC,respectively.In aromatic hydrocarbons compounds of bark coal samples,the phenanthrene series occupy the largest proportion(43.48%–53.74%),followed by the dibenzothiophens(7.33%–16.53%).The distributions of naphthalenes,diphenyls,and chrysene of the samples indicate the source inputs are mainly derived from terrestrial higher plants.Furthermore,the ratios of naphthalene and phenanthrene series compounds reveal the samples are in the mature stage.Some biomarkers(Pr/Ph and C35/C31–35 homohopane),aromatic hydrocarbons(fluorene,dibenzofuran and dibenzothiophene),and high inertinite contents(14.7%–41.8%)suggest that bark coal was formed under relatively oxic conditions and was influenced by marine or brackish lakes environment.

Objective:Although great progress has been made in the field of siRNA gene therapy,safe,efficient,and targeted delivery of siRNA are still major challenges in siRNA therapeutics.Methods:We developed an up-conversion nanoparticle-based nanocage system.This system protected the siRNA from being degraded by nucleases in organisms and selectively delivered the siRNAs to the tumor sites,due to modifications of targeted molecules on the surfaces of nanocages and local inhalation.Results:The siRNAs delivered by the up-conversion nanoparticle nanocages were protected from degradation in transit to the tumor sites,where they accumulated.Compared with the passive target and control groups,the up-conversion nanoparticles based on the nanocage system showed a tumor suppressive effect after approximately 3 weeks of treatment.Conclusions:The up-conversion nanoparticle nanocages efficiently delivered vascular endothelial growth factor siRNAs to tumor sites.Mice with lung tumors treated with tumors targeting up-conversion nanoparticle nanocages showed steady body weight changes,high tumor inhibition ratios,and longer survival times.

Sea cucumber saponins have attracted more attention in recent years due to biological activities.It is a popular practice to soak sea cucumber in Baijiu at home and being applied to industrial manufacturing in China.However,knowledge of the effect of alcohol on the absorption and metabolism of sea cucumber saponins is limited.The effects of alcohol on digestion,absorption and metabolism of sea cucumber saponins in BALB/c mice were investigated after gavage and tail intravenous injection.The results showed that the content of saponins in serum and liver was significantly higher under the influence of alcohol than that in the control group after oral administration.Alcohol promoted the absorption of sea cucumber saponins prototype as well as inhibited the process of saponins being transformed into deglycositic metabolites in the small intestine.Moreover,sea cucumber saponins remained in circulation for a long time and alcohol slowed down the clearance of sea cucumber saponins under the influence of alcohol after intravenous injection.This confirmed the feasibility of marinating sea cucumber in Baijiu to improve the efficacy of saponins and provides an important theoretical basis for the utilization of sea cucumber and the development of sea cucumber liquor.

Background Ginkgo biloba extract(GBE)is evidenced to be effective in the prevention and alleviation of metabolic disorders,including obesity,diabetes and fatty liver disease.However,the role of GBE in alleviating fatty liver hemorrhagic syndrome(FLHS)in laying hens and the underlying mechanisms remain to be elucidated.Here,we investigated the effects of GBE on relieving FLHS with an emphasis on the modulatory role of GBE in chicken gut microbiota.Results The results showed that GBE treatment ameliorated biochemical blood indicators in high-fat diet(HFD)-induced FLHS laying hen model by decreasing the levels of TG,TC,ALT and ALP.The lipid accumulation and pathological score of liver were also relieved after GBE treatment.Moreover,GBE treatment enhanced the antioxidant activity of liver and serum by increasing GSH,SOD,T-AOC,GSH-PX and reducing MDA,and downregulated the expression of genes related to lipid synthesis(FAS,LXRα,GPAT1,PPARγand Ch REBP1)and inflammatory cytokines(TNF-α,IL-6,TLR4 and NF-κB)in the liver.Microbial profiling analysis revealed that GBE treatment reshaped the HFD-perturbed gut microbiota,particularly elevated the abundance of Megasphaera in the cecum.Meanwhile,targeted metabolomic analysis of SCFAs revealed that GBE treatment significantly promoted the production of total SCFAs,acetate and propionate,which were positively correlated with the GBE-enriched gut microbiota.Finally,we confirmed that the GBE-altered gut microbiota was sufficient to alleviate FLHS by fecal microbiota transplantation(FMT).Conclusions We provided evidence that GBE alleviated FLHS in HFD-induced laying hens through reshaping the composition of gut microbiota.Our findings shed light on mechanism underlying the anti-FLHS efficacy of GBE and lay foundations for future use of GBE as additive to prevent and control FLHS in laying hen industry.

Grammar learning has been a bottleneck problem for a long time. In this paper, we propose a method of seman- tic separator learning, a special case of grammar learning. The method is based on the hypothesis that some classes of words, called semantic separators, split a sentence into sev- eral constituents. The semantic separators are represented by words together with their part-of-speech tags and other infor- mation so that rich semantic information can be involved. In the method, we first identify the semantic separators with the help of noun phrase boundaries, called subseparators. Next, the argument classes of the separators are learned from cor- pus by generalizing argument instances in a hypernym space. Finally, in order to evaluate the learned semantic separators, we use them in unsupervised Chinese text parsing. The exper- iments on a manually labeled test set show that the proposed method outperforms previous methods of unsupervised text parsing.

Increasing plant photosynthetic capacity is a promising approach to boost yields,but it is particularly challenging in C3crops,such as soybean(Glycine max(L.)Merr.).Here,we identified GmFtsH25,encoding a member of the filamentation temperature-sensitive protein H protease family,as a major gene involved in soybean photosynthesis,using linkage mapping and a genome-wide association study.Overexpressing GmFtsH25 resulted in more grana thylakoid stacks in chloroplasts and increased photosynthetic efficiency and starch content,while knocking out GmFtsH25 produced the opposite phenotypes.GmFtsH25 interacted with photosystem I light harvesting complex 2(GmLHCa2),and this interaction may contribute to the observed enhanced photosynthesis.GmFtsH25 overexpression lines had superior yield traits,such as yield per plant,compared to the wild type and knockout lines.Additionally,we identified an elite haplotype of GmFtsH25,generated by natural mutations,which appears to have been selected during soybean domestication.Our study sheds light on the molecular mechanism by which GmFtsH25 modulates photosynthesis and provides a promising strategy for improving the yields of soybean and other crops.

温室气体排放导致的全球气候变化问题是社会关注的重要问题,如何较准确地估算各领域内的温室气体排放通量对制定减排策略具有重要意义。为分析研究区水稻—冬小麦轮作系统温室气体(CH_(4)、N_(2)O)排放水平,对比IPCC2006 Tier2及DNDC模型法估算研究区稻麦轮作系统的CH_(4)和N_(2)O排放通量估算差异,定量评估DNDC模型是否适用于轮作系统的温室气体排放估算。于2015—2018年进行稻麦轮作野外试验,利用静态箱-气相色谱分析法定期测定CH_(4)和N_(2)O排放通量,结合田间管理和气象数据开展IPCC2006 Tier2和DNDC模型模拟,利用DNDC模型模拟轮作和非轮作情景的稻麦温室气体排放通量。结果表明,DNDC模型可以较好地模拟出稻田CH_(4)、N_(2)O和麦田N_(2)O排放通量时序变化特征,估算误差分别为-4.8%、-11.6%、-10.8%,精度优于IPCC2006 Tier2;DNDC模型对稻麦轮作的GWP模拟精度高于IPCC2006 Tier2,稻田和麦田GWP模拟相对误差分别为-5.9%、-21.7%;除了冬小麦生育期CH_(4)排放通量,DNDC轮作模式对其他稻麦轮作温室气体排放通量的模拟相对误差均低于非轮作模式。进而为区域农业温室气体估算及制定减排方案提供科学依据。
