This paper presents an interpolating element-free Galerkin(IEFG) method for solving the two-dimensional(2D) elastic large deformation problems. By using the improved interpolating moving least-squares method to form shape function, and using the Galerkin weak form of 2D elastic large deformation problems to obtain the discrete equations, we obtain the formulae of the IEFG method for 2D elastic large deformation problems. As the displacement boundary conditions can be applied directly, the IEFG method can acquire higher computational efficiency and accuracy than the traditional element-free Galerkin(EFG)method, which is based on the moving least-squares approximation and can not apply the displacement boundary conditions directly. To analyze the influences of node distribution, scale parameter of influence domain and the loading step on the numerical solutions of the IEFG method, three numerical examples are proposed. The IEFG method has almost the same high accuracy as the EFG method, and for some 2D elastic large deformation problems the IEFG method even has higher computational accuracy.

The prevalence of obesity in children has dramatically increased in the last few decades, and obesity has also emerged as an important risk factor for asthma.Innate mechanisms have been shown to be involved in both diseases, particularly through the recently described innate lymphoid cells(ILCs), in which ILC3s have been linked to obesity both in human and in murine models.The aim of this study was to explore whether being overweight in asthmatic children was associated with elevated circulating ILC3 or elevated messenger RNA(mRNA)-levels of RORC, IL-17A and IL-22.Our results showed significantly elevated ILC3 frequencies in overweight asthmatic children compared with non-overweight controls based on the detection of Lin+CD127IL-23R cells by flow cytometry.Moreover, elevated ILC3 frequencies positively correlated with the mRNA expression of RORC which has been identified as a transcription factor of ILC3s.The relative mRNA expression level of IL-17A was also upregulated in overweight compared to non-overweight children, as was the relative mRNA level of IL-22.However, there were no correlations between ILC3s frequencies or the expression of, RORC, IL-17A and IL-22 and asthma severity.These results suggested that childhood obesity is an independent factor that is associated with an elevated frequency of circulating ILC3s and higher expression of RORC, IL-22 and IL-17A.

TANK-binding kinase 1(TBK1)is an essential protein kinase for activation of interferon regulatory factor 3(IRF3)and induction of the type I interferons(IFN-I).Although the biochemical regulation of TBK1 activation has been studied,little is known about how enterovirus 71(EV71)employs the deubiquitinases(DUBs)to regulate TBK1 activation for viral immune evasion.Here,we found that EV71 infection upregulated the expression of ubiquitinspecific protease 24(USP24).Further studies revealed that USP24 physically interacted with TBK1,and can reduce K63-linked polyubiquitination of TBK1.Knockdown of USP24 upregulated TBK1 K63-linked polyubiquitination,promoted the phosphorylation and nuclear translocation of IRF3,and in turn improved IFN-I production during EV71 infection.As a consequence,USP24 knockdown dramatically inhibited EV71 infection.This study revealed USP24 as a novel regulator of TBK1 activation,which promotes the understanding of immune evasion mechanisms of EV71 and could provide a potential strategy for treatment of EV71 infection.

Synthesis of fluorinated CaCO_(3)-based oxygen-supplying nanophotosensitizers to potentiate photodynamic immunotherapy by reversing tumor hypoxia and immunosuppression

Photodynamic therapy is a noninvasive type of phototherapy with a high capacity to boost specific antitumor immunity by causing immunogenic cell death.However,the photodynamic therapeutic potency toward solid tumors is dampened by tumor hypoxia that negatively impairs the generation of cytotoxic singlet oxygen and promotes the formation of tumor immunosuppression.Herein,fluorinated CaCO_(3)(CaF)nanoparticles are prepared with the addition of dopamine-conjugated perfluorosebacic acid and ferric chloride into a calcium chloride ethanol solution via an ammonium bicarbonate-mediated gas-diffusion process.After being coated with commercial lipids and hexadecylamin conjugated chlorin e6(hCe6)via a templated self-assembly process,the yielded PEGylated nanophotosensitizer(hCe6@CaF-PEG)exhibits an effective loading efficiency to perfluoro-15-crown-5-ether(PFCE),a model perfluorocarbon molecule,and thus oxygen molecules.Upon intravenous administration,the obtained PFCE/hCe6@CaF-PEG can alleviate tumor hypoxia by working as an oxygen nanoshuttle.Together with local light emitting diode light exposure,photodynamic treatment with PFCE/hCe6@CaF-PEG can suppress the growth of primary CT26 tumors and unirradiated distant tumors,particularly when synergized with anti-PD-1(aPD-1)immunotherapy to collectively reverse tumor immunosuppression.This work presents an effective strategy to potentiate photodynamic immunotherapy by concurrently reversing tumor hypoxia and immunosuppression.

Fabricating a smart clothing system based on strain-sensing yarn and novel stitching technology for health monitoring

Various yarn-shaped flexible strain sensors have recently been developed.However,research is lacking on additive manufacturing for smart clothing for integrating yarn sensors with commercial garments.Herein,a strain-sensing yarn is sewn into a piece of fabric through a novel stitching technique,and the influence of the stitching method and needle pitch on the sensing performance is investigated using finite element analysis(FEA).The sensing performance could be improved when the sensing yarn is self-locked in the fabric at the needle eyes,and the needle pitch was reduced to 0.5 cm,which is attributed to the enhanced stress and strain concentration.Meanwhile,the composite sensing fabric featured outstanding performance,including a low detection limit(0.1%),rapid response(280 ms),excellent durability(10000 cycles),and high stability(negligible drift and frequency independence).In addition,the remarkable wear resistance,washability,and anti-interference to ambient humidity and perspiration were obtained.Therein,the optimal stitch trace lengths of sensing yarn for detecting elbow motion,breathing,and heartbeats are discussed.Finally,a smart clothing system composed of smart clothing,data acquisition unit,and mobile APP was developed to simultaneously detect human movement and physiological signals.This work provides a reference to produce intelligent garments based on yarn sensors for health monitoring.

Influence of cementite spheroidization on relieving the micro-galvanic effect of ferrite-pearlite steel in acidic chloride environment

The corrosion behavior of the as-received steel and the spheroidized steel in acidic chloride environment was investigated. The results indicate the corrosion mode and corrosion rate of two steels are diverse due to their difference in microstructure. For as-received steel with ferrite-pearlite microstructure, severe localized corrosion happens on the pearlite regions, and plenty of cathodic cementite remains in the pits, further strengthening the micro-galvanic effect and accelerating the corrosion rate. While for spheroidized steel with tempered martensite microstructure, the nanosized cementite particles evenly distributed on the ferrite substrate are easy to fall off, which can significantly reduce the cementite accumulation on the steel surface, relieving the acceleration effect of micro-galvanic corrosion.

Distinct beneficial effect of Sn on the corrosion resistance of Cr-Mo low alloy steel

Retinoid-related orphan receptor α(RORα)is a master regulator of ILC2 s function as a transcript factor.Innate lymphoid cells(ILCs) are shown to mediate the immune pathology of asthma even without adaptive immunity.However,little is known about possible role of RORα alone in asthma.In this context,we evaluated the functions of exogenous RORα in ovalbumin-induced asthma models.RORα-EGFP-expressing adenovirus or EGFP control adenovirus was administered,followed by challenge with aerosolized OVA solution to induce asthma.Th2 cytokine and immune cell profiles of bronchoalveolar lavage fluid(BALF),as well as serum IgE levels were analyzed.Histological analyses were also conducted to demonstrate the effects of RORα expression on airway remodeling and inflammatory responses in the lung.Our data showed that the adenoviral RORα was expressed in lung tissue,and significantly increased numbers of eosinophils,neutrophils,macrophages and lymphocyteswere found in BALF from RORα gene-delivered mice in response to OVA.In addition,RORα expression in the lung increased the levels of Th2-type cytokines and IgE in serum,respectively.Simultaneously,RORα expressionled to an elevation of IL-17 RB and T1/ST2 in lung tissueand BALF in OVA-challenged asthma models.Our results suggest that RORα act as a transcript factor of ILC2 s can promote OVA-induced asthma in mice and present a potential therapeutic target for the treatment of asthma.

In this work,the beneficial effect of Sn addition on the corrosion resistance mechanism of Cr-Mo low alloy steel was studied.Results demonstrated that Sn improves the corrosion resistance of the steel matrix mainly by influencing the microstructural transformation.Sn addition and the synergistic effect of Sn,Cr,and Mo promote the formation of α-FeOOH,SnO_(2),SnO,Cr(OH)_(3),and molybdates,lead to the improved protection and stability of the rust layer.This synergistic effect also endows the inner rust layer with cation selectivity,preventing the further penetration of Cl-and inhibiting the localized corrosion of steel.

Background:The regulatory genes related to lipid metabolism affect the intramuscular fat(IMF)content and improve meat quality traits.Mediator Complex Subunit 4(MED4),a vitamin D receptor protein,affects vitamin D,livestock growth,carcass traits,and triglyceride deposition.However,the physiological function of the MED4 gene on bovine adipocyte differentiation remains unknown.Methods:This study explored the function of the MED4 gene in preadipocyte differentiation in Chinese Red Steppe cattle.The overexpression plasmid and the interference sequences of the MED4 gene were constructed to detect the effects of the MED4 gene on adipogenesis and biomakers using quantitative polymerase chain reaction and western blotting.Results:The MED4 gene had significantly high expression during preadipocyte differentiation(p<0.05).Overexpression of the MED4 gene increased the expression of the PPARγgene,a preadipocyte differentiation biomarker,improved cellular lipid droplets and triglycerides accumulation,and positively accelerated adipocyte maturation(p<0.05).Interference of the MED4 gene can negatively regulate preadipocyte differentiation.Conclusion:This study showed that the MED4 gene may affect the preadipocyte differentiation and adipogenesis in Chinese Red Steppe cattle by regulating the PPARγgene.
