High baseline tumor burden-associated macrophages promote an immunosuppressive microenvironment and reduce the efficacy of immune checkpoint inhibitors through the IGFBP2-STAT3-PD-L1 pathway

Background:Several clinical studies have uncovered a negative correlation between baseline tumor burden and the efficacy of immune checkpoint inhibitor(ICI)treatment.This study aimed to uncover the specific mechanisms underlying the difference in sensitivity to ICI treatment between tumors with high(HTB)and low(LTB)tumor burden.Methods:For in vivo studies,several mouse models of subcutaneous tumors were established,and transcriptome sequencing,immunohistochemistry,and flow cytometry assays were used to detect the immune status in these subcutaneous tumors.For in vitro experiments,co-culture models,cytokine antibody arrays,western blotting,flow cytometry,and enzyme-linked immunosorbent assays were used to explore the underlying molecular mechanisms Results:We found that MC38 or B16 subcutaneous tumors from the HTB group did not show any response to anti-programmed cell death protein-1(PD-1)therapy.Through flow cytometry assays,we found that the infiltration with CD8^(+)T cellswas significantly decreasedwhereasM2-like macrophageswere enriched in subcutaneous tumors of HTB groups compared with those of LTB group.These changes were not affected by the initial number of injected tumor cells or tumor age,nor could they be reversed by surgical tumor reduction.Intraperitoneal colony-stimulating factor 1 receptor(CSF-1R)inhibitor PLX3397 injection at different time points of tumor growth only had an effect when administered in the early tumor stage to maintain the“heat”of the tumor microenvironment during the process of tumor growth,thereby achieving a response to ICI treatment when the tumor grew to a large size.Mechanistically,we found that insulin-like growth factor binding protein 2(IGFBP2)expression levelswere significantly elevated in HTB tumor tissues.IGFBP2 promoted the programmed death-ligand 1(PD-L1)expression in M2-like macrophages by activating signal transducer and activator of transcription 3(STAT3),and PD-L1^(+)M2-likemacrophages exerted an immunosuppressive effect by inhibiting the proliferation and activa

Dear editor,The tumor microenvironment(TME)heavily impacts disease biology and may influence responses to systemic treatments,and thereby,affects patients’prognosis.In our previous study,we found that immune features could predict prognosis and guide the therapy choices for stage I-III colon cancer[1,2].Increasing evidence shows that therapyinduced TME changes can promote tumor progression,metastasis,and the development of resistance[3,4].However,the TME dynamics in colorectal liver metastases(CRLM)under treatment are still incompletely clear.

Depression accelerates gastric cancer invasion and metastasis by inducing a neuroendocrine phenotype via the catecholamine/β_(2)-AR/MACC1 axis

Background:Depression is a common,easily ignored,accompanied disease of gastric cancer(GC)patients and is often observed with elevated plasma catecholamine levels.Depression frequently promotes GC progression and leads to poor clinical outcomes;however,the molecular mechanisms underlying depression-induced GC progression remain poorly understood.We aimed to study the effects of depression on GC progression and explore possible mechanisms mediating the action of depression-associated catecholamines on GC.Methods:Depression states of GC patients were graded using the Patient Health Questionnaire-9,and plasma catecholamine levels were examined by high performance liquid chromatography coupled with tandem mass spectrometry.Migrative and invasive GC cells were examined using transwell assays,and metastatic GC niches were imaged using bioluminescence technology in a depression mouse model established with chronic unpredictable mild stress.Mouse depression-like behaviors were assessed through sucrose preference,forced swimming,and tail suspension tests.Characteristics of the neuroendocrine phenotype were observed via RT-PCR,Western blotting,flow cytometry,and transmission electron microscopy.Results:Fifty-one GC patients(age:53.61±1.79 years;cancer duration:3.71±0.33 months;depression duration:2.37±0.38 months;male-to-female ratio:1.55:1)were enrolled in the study.Depression grade was significantly higher in GC patients showing higher plasma levels of catecholamines(epinephrine:P=0.018;noradrenaline:P=0.009),higher oncogene metastasis-associated in colon cancer-1(MACC1)level(P=0.018),and metastasis(P<0.001).Further,depression-associated catecholamine specifically bound to the beta-2 adrenergic receptor(β_(2)-AR)and upregulated MACC1 expression,and thus promoting neuroendocrine phenotypic transformation through direct binding betweenMACC1 and synaptophysin.Eventually,the neuroendocrine phenotypic transformation accelerated GC invasion in vitro and metastasis in vivo.However,β_(2)-AR antagonist ICI-118,551 or MACC1 silenc

浅埋极软地层超大直径盾构施工存在冒顶漏浆、管片上浮、管片渗漏水、盾构姿态控制困难等重难点问题。依托珠海隧道工程,针对盾构始发一侧隧道埋深浅、周边地层极为软弱的特点,制定并采取了地层预加固处理、盾构机针对性设计、同步双液注浆系统改造及注浆方法优化、改进管片防水体系等系统性措施,浅埋极软地层重难点问题得到有效控制。单一的地层预加固措施对浅埋段管片上浮控制效果有限;厚浆与双液浆组合使用的分区注浆方法可实现缩短顶部浆液初凝时间和早强效果,有效解决了管片上浮及渗漏水;控制管片不均匀上浮可以避免管片渗漏水和裂缝漏水问题。

Dear Editor,Dear Editor,Lung cancer is one of the most common cancers and a leading cause of cancer-related deaths.T cells are known to play a significant role in the destruction of cancer cells.T cells have therefore become the focus of lung cancer immunotherapy.T-cell receptors(TCRs)can recognize antigenic peptides presented by HLA proteins.TCRs are distinct individually and vary with pathophysiological condition,so T cells can respond to a wide range of antigens.TCR repertoire diversity reflects the potential for cellular immunity,and several studies have demonstrated that complementarity determining region 3(CDR3β)diversity is important in cancer therapy and prognosis.1 Liu et al.have reported that the CDR3βdiversity of patients with advanced lung cancer differs significantly from that of healthy individuals.

长期紫云英还田与化肥配施对土壤团聚体中黏粒矿物组成的影响

在湖南南县长期紫云英-双季稻轮作试验的基础上,选择单施紫云英(MV)、单施化肥(F100)和紫云英配施全量化肥(MV+F100)处理,研究不同施肥处理下土壤大团聚体(>250μm)、小团聚体(53~250μm)、粉黏粒(2~53μm)及<2μm团聚体的黏粒组分(100~2000、25~100 nm)中矿物的类型、含量和分布特征。结果表明,不同处理各粒级团聚体黏粒组分中的矿物组成相似,主要以伊利石和高岭石为主,包含少量蛭石和1.4nm过渡矿物。MV和MV+F100处理中,相比于<2μm团聚体,大团聚体、小团聚体及粉黏粒中100~2000 nm组分高岭石含量有所降低,蛭石含量增加。与MV相比,MV+F100处理明显增加了<2μm团聚体颗粒中伊利石的相对含量,以及各粒级团聚体颗粒中蛭石的相对含量。随着各团聚体黏粒组分由100~2000nm减小至25~100nm,黏粒矿物的类型减少,伊利石的相对含量显著提高,高岭石含量降低且结晶度变差。与F100相比,MV+F100处理中各粒级团聚体100~2000 nm颗粒的硅铝率明显降低,出现脱硅富铝化现象。

【目的】双季稻田水旱轮作后能够提高作物产能降低土壤有机质含量，但水旱轮作后化肥氮素配施不同有机物料对作物产量、氮素利用效率及有机质含量影响的报道较少。【方法】以玉米水稻轮作为研究对象，于2015—2021年在洞庭湖区紫潮泥和红黄泥两种双季稻田上设置不施氮肥（CK）、单施化肥（NPK）、NPK+秸秆还田（NPKS）、NPK+有机肥（NPKM）、NPK+生物炭（NPKB）5种处理，探讨在两种土壤上不同施肥处理的作物产量、养分利用效率和土壤碳氮含量及平衡的变化。【结果】紫潮泥和红黄泥6年试验的平均玉米产量、水稻产量和年总产量分别为5.7、7.3和12.9 t·hm-2，三者在紫潮泥中的产量都略高于红黄泥，但差异都不显著。与NPK处理相比，NPKM处理使两种土壤的玉米和水稻平均产量分别显著（P<0.05）提高10.6%、4.20%。在所有处理中，NPKM处理玉米产量最高，为6.0 t·hm-2;NPKB处理水稻产量最高，为7.5 t·hm-2。NPKM和NPKB处理的6年平均年总产量分别显著（P<0.05）高于其他处理，但NPKM和NPKB处理间差异不显著。与其他处理相比，NPKM处理显著（P<0.05）提高了玉米水稻的化肥氮素回收率、农学效率和偏生产力，玉米季6年平均值分别为66.3%、39.5kg·kg-1、56.0 kg·kg-1，水稻季6年平均值分别为53.8%,21.9 kg·kg-1、68.6 kg·kg-1。NPKB处理较NPK、NPKS显著（P<0.05）提高了玉米水稻的化肥氮素农学效率和偏生产力，较NPK处理显著（P<0.05）提高了玉米化肥氮素回收率。两种土壤有机碳、全氮含量从第4年（2019年）开始显著下降。与试验开始前（2015年）相比，2021年紫潮泥和红黄泥施氮处理年平均有机碳含量分别下降了1.8、0.7 g·kg-1，全氮含量分别下降0.4、0.1 g·kg-1；两种土壤NPKM处理的土壤有机碳平均含量下降幅度最小、NPKB处理次之。NPKM处理的碳、氮投入量最高，平均土壤有机碳、全氮损失量最小，分别为0.48 t C·hm-2·a-1、94.7 kg N·hm-2·a-1。【结论】双季稻田水旱轮作后，化肥与有机肥、生物炭配施能提高作物产量和延缓土壤有机质下降，试验期间化肥配施有机肥处理玉米平均产量最高，化肥配施生物炭处理水稻平均产量最高。

Flower-like BiOBr/Bi_(2)WO_(6) Z-scheme heterojunction was prepared by a twostep solvothermal method.BiOBr microspheres were firstly synthesized through solvothermal method.Then the out part of the BiOBr microspheres was designed to react with Na2WO4 forming Bi_(2)WO_(6) nanosheets through ion exchange process.The BiOBr/Bi_(2)WO_(6) heterojunction has a larger BET surface area,smaller energy band gap,faster transfer of charge carriers and a much better visible-light photocatalytic performance than that of BiOBr and Bi_(2)WO_(6).It also has a better cycling stability than that of the BiOBr.Possible photocatalytic mechanism of the BiOBr/Bi_(2)WO_(6) heterojunction is proposed.

Soil bacterial communities interact with silicon fraction transformation and promote rice yield after long-term straw return

Returning crop straw into the soil is an important practice to balance biogenic and bioavailable silicon(Si)pool in paddy,which is crucial for the healthy growth of rice.However,owing to little knowledge about soil microbial communities responsible for straw degradation,how straw return affects Si bioavailability,its uptake,and rice yield remains elusive.Herein,we investigate the change of soil Si fractions and microbial community in a 39-year-old paddy field amended by a long-term straw return.Results show that rice straw return significantly increased soil bioavailable Si and rice yield from 29.9%to 61.6%and from 14.5%to 23.6%,respectively,when compared to NPK fertilization alone.Straw return significantly altered soil microbial community abundance.Acidobacteria was positively and significantly related to amorphous Si,while Rokubacteria at phylum level,Deltaproteobacteria,and Holophagae at class level was negatively and significantly related to organic matter adsorbed and Fe/Mn-oxide-combined Si in soils.Redundancy analysis of their correlations further demonstrated that Si status significantly explained 12%of soil bacterial community variation.These findings suggest that soil bacteria community and diversity interact with Si mobility by altering its transformation,thus resulting in the balance of various nutrient sources to drive biological Si cycle in agroecosystem.

佛手和山金柑果形相关基因CRC的TRV-VIGS体系的构建研究

以芸香科的佛手(Citrus medica L.var.sarcodactylis Swingle)和山金柑(Fortunella hindsii Swingle)花期叶片为材料,以果实发育相关的CRC基因为对象,利用烟草脆裂病毒(tobacco rattle virus,TRV)研究构建病毒诱导的基因沉默(virus-induced gene silencing,VIGS)技术体系,为基因功能研究提供有效简便的方法。待沉默的佛手植株剪枝、抹除花芽后,将CRC基因的沉默载体两次注射于佛手和山金柑的顶部叶片,注射间隔15~30 d。在注射后25~115 d内,佛手新生叶、新生花、果中的CmsCRC表达量均显著下调,且沉默植株的果形聚拢,果实指状结构数增加。CRC基因在佛手中的沉默有效时间是侵染后25~115 d,而在山金柑中沉默有效时间是25~105 d。
