Pseudorabies virus VHS protein abrogates interferon responses by blocking NF-κB and IRF3 nuclear translocation

Herpesviruses antagonize host antiviral responses through a myriad of molecular strategies culminating in the death of the host cells. Pseudorabies virus (PRV) is a significant veterinary pathogen in pigs, causing neurological sequalae that ultimately lead to the animal's demise. PRV is known to trigger apoptotic cell death during the late stages of infection. The virion host shutdown protein (VHS) encoded by UL41 plays a crucial role in the PRV infection process. In this study, we demonstrate that UL41 inhibits PRV-induced activation of inflammatory cytokine and negatively regulates the cGAS-STING-mediated antiviral activity by targeting IRF3, thereby inhibiting the translocation and phosphorylation of IRF3. Notably, mutating the conserved amino acid sites (E192, D194, and D195) in the RNase domain of UL41 or knocking down UL41 inhibits the immune evasion of PRV, suggesting that UL41 may play a crucial role in PRV's evasion of the host immune response during infection. These results enhance our understanding of how PRV structural proteins assist the virus in evading the host immune response. © 2024 The Authors

The WRKY Gene Family in Rice （Oryza sativa）

WRKYgenee encode transcription factors that are Involved In the regulation of various biological processes. These zinc-finger proteins, especially those members mediating stress responses, are uniquely expanded In plants. To facilitate the study of the evolutionary history and functions of this eupergene family, we performed an exhaustive search for WRKY genes using HMMER and a Hidden Markov Model that was specifically trained for rice. This work resulted In a comprehensive list of WRKYgene models In Oryza sativa L. eep. indica and L. eep. j.ponica. Mapping of these genes to Individual chromosomes facilitated elimination of the redundant, leading to the Identification of 98 WRKYgenee In j.ponica and 102 In indica rice. These genes were further categorized according to the number and structure of their zinc-finger domains. Based on a phylogenetlc tree of the conserved WRKY domains and the graphic display of WRKY loci on corresponding indica and j.ponica chromosomes, we Identified possible WRKY gene duplications within, and losses between the two closely related rice subspecies. Also reviewed are the roles of WRKY genes In disease resistance and responses to salicylic acid and j.emonlc acid, seed development and germination mediated by glbberelllns, other developmental processes Including senescence, and responses to ablotlc stresses and abeclelc acid In rice and other plants. The signaling pathways mediating WRKY gene expreeelon are also discussed.

In situ digital testing method for quality assessment of soft soil improvement with polyurethane

This study purposes an in situ testing method on quality assessment of soil *** drilling data includes the spatial distribution and in situ strength of untreated and treated soil along three different drillholes measured by on-site drilling monitoring *** factual drilling data can characterize the degree of soil improvement by penetration inj.ction with permeable *** from on-site drilling monitoring shows that the linear zones represent constant drilling speeds shown in the plot of drill bit advancement *** drilling time,which indicates the spatial distributions of soil *** soil profile at the study site is composed of four layers,which includes fill,untreated silty clay,treated silty clay,and mucky *** results of soil profile are verified by the parallel site *** constant drilling speeds profile the coring-resistant strength of drilled *** comparing with the untreated silty clay,the constant drilling speeds of the treated silty clay have been decreased by 13.0-62.8%.Two drilling-speed-based indices of 61.2%and 65.6%are proposed to assess the decreased average drilling speed and the increased in situ strength of treated silty *** tests,*** compressive strength(UCS)test,have been performed with core sample to investigate and characterize in situ strength by comparing that with drilling *** show that the average predicted strengths of treated silty clay are 2.4-6.9 times higher than the average measured strength of untreated silty *** UCS-based indices of 374.5%and 344.2%verified the quality assessment(QA)results by this new in situ *** method provides a cost-effective tool for quality assessment of soil improvement by utilizing the digital drilling data.

Interface structure of α-Mg/14H-LPSO:First-principles prediction and experimental study

The interfacial structure of the α-Mg/14H-LPSO phase in rare earth-including magnesium alloy was investigated via high-angle annular dark-field scanning transmission electron microscopy(HAADFSTEM) imaging and first-principles calculations of density-functional *** possible interfacial models were constructed according to the different terminations of the LPSO phase,and the corresponding interfacial energies were calculated,from which the four most stable structures(Terl-MgY-hollow,Ter2-Zn-hollow,Ter3-MgYII-hollow and Ter4-Mg-bridge) were *** interfacial phase diagrams related to the Y chemical potentials were obtained from the calculations,and the most stable interfacial structure was ***-MgY-hollow and Ter2-Zn-hollow have the lowest interfacial energies in the range of-0.7 eV j.m~2 for the interface model of α-Mg and 14H-LPSO phase structure with Ter2-Zn-hollow termination.

Individual consistency in spatiotemporal characteristics of migratory Whimbrels in the East Asian-Australasian Flyway

Many migratory birds exhibit interannual consistency in migration schedules, routes and stopover sites. Detecting the interannual consistency in spatiotemporal characteristics helps understand the maintenance of migration and enables the implementation of targeted conservation measures. We tracked the migration of Whimbrel (Numenius phaeopus) in the East Asian-Australasian Flyway and collected spatiotemporal data from individuals that were tracked for at least two years. Wilcoxon non-parametric tests were used to compare the interannual variations in the dates of departure from and arrival at breeding/nonbreeding sites, and the interannual variation in the longitudes when the same individual across the same latitudes. Whimbrels exhibited a high degree of consistency in the use of breeding, nonbreeding, and stopover sites between years. The variation of arrival dates at nonbreeding sites was significantly larger than that of the departure dates from nonbreeding and breeding sites. Repeatedly used stopover sites by the same individuals in multiple years were concentrated in the Yellow Sea coast during northward migration, but were more widespread during southward migration. The stopover duration at repeatedly used sites was significantly longer than that at sites used only once. When flying across the Yellow Sea, Whimbrels breeding in Sakha (Yakutia) exhibited the highest consistency in migration routes in both autumn and spring. Moreover, the consistency in migration routes of Yakutia breeding birds was generally higher than that of birds breeding in Chukotka. Our results suggest that the northward migration schedule of the Whimbrels is mainly controlled by endogenous factors, while the southward migration schedule is less affected by endogenous factors. The repeated use of stopover sites in the Yellow Sea coast suggests this region is important for the migration of Whimbrel, and thus has high conservation value.
