Accurate information on eye position in the orbit is available from visual feedback,efference copy of the oculomotor commands and proprioceptive signals from the extraocular muscles (EOM).Whereas visual feedback and oculomotor commands have been extensively studied,central processing of EOM proprioceptive signals remains to be elucidated.A challenge to the field is to develop an approach to induce passive eye movements without physically contacting the eyes.A novel method was developed to generate passive eye movements in rats.A small rare-earth magnet disk (0.7 mm diameter,0.5 mm thickness) was attached to the surface of a rat's eyeball.A metal rod (5 mm diameter) wrapped with an electromagnetic (EM) coil was placed near the magnet (8-15 mm).By passing currents to the EM coil,electromagnetic force (EMF) was generated and acted upon the magnet and induced passive eye *** EMF induced well-defined passive eye movements,whose directions were dependent on current polarity and amplitudes and peak velocities were dependent on current intensity and *** velocities of the EMF-induced eye movements were linearly related to amplitudes,exhibiting main sequence relationships similar to that of saccades in awake rats and eye movements induced by electrical microstimulation of the abducens nucleus in anesthetized *** examination showed that repetitive EMF stimulations did not appear to result in damages in the EOM *** results validated the EMF approach as a novel tool to investigate EOM proprioceptive signals and their roles in visual localization and gaze control.

Exposure to explosive shockwave often leads to blast-induced traumatic brain injury in military and civilian *** ears are most often damaged following exposure to *** there is an association between tympanic membrane perforation and TBI in blast exposure victims,little is known about how and to what extent blast energy is transmitted to the central nervous system via the external ear *** present study investigated whether exposure to blasts directed through the ear canal causes brain injury in LongEvans *** were exposed to a single blast(0–30 pounds per square inch(psi))through the ear canal,and brain injury was evaluated by histological and behavioral outcomes at multiple *** exposure not only caused tympanic membrane perforation but also produced substantial neuropathological changes in the brain,including increased expression of c-Fos,induction of a profound chronic neuroinflammatory response,and apoptosis of *** blast-induced injury was not limited only to the brainstem most proximal to the source of the blast,but also affected the forebrain including the hippocampus,amygdala and the habenula,which are all involved in cognitive ***,the animals exhibited long-term neurological deficits,including signs of anxiety in open field tests 2 months following blast exposure,and impaired learning and memory in an 8-arm maze 12 months following blast *** results suggest that the unprotected ear canal provides a locus for blast waves to cause *** study was approved by the Institutional Animal Care and Use Committee at the University of Mississippi Medical Center(Animal protocol#0932 E,approval date:September 30,2016 and 0932 F,approval date:September 27,2019).

Genome-edited human induced pluripotent stem cells(iPSCs)hold great promise for therapeutic ***,low editing efficiency has hampered the applications of CRISPR-Cas9 technology in creating knockout and homology-directed repair(HDR)-edited iPSC lines,particularly for silent *** is partially due to chromatin compaction,inevitably limiting Cas9 access to the target *** the six HDAC inhibitors we examined,vorinostat,or suberoylanilide hydroxamic acid(SAHA),led to the highest HDR efficiency at both open and closed loci,with acceptable *** inhibitors equally increased non-homologous end joining(NHEJ)editing efficiencies(~50%)at both open and closed loci,due to the considerable HDAC inhibitor-mediated increase in Cas9 and sgRNA ***,we observed more substantial HDR efficiency improvement at closed loci relative to open chromatin(2.8 vs.1.7-fold change).These studies provide a new strategy for HDRediting of silent genes in iPSCs.

The ears are air-filled structures that are directly impacted during blast *** addition to hearing loss and tinnitus,blast victims often complain of vertigo,dizziness and unsteady posture,suggesting that blast exposure induces damage to the vestibular end organs in the inner ***,the underlying mechanisms remain to be *** this report,single vestibular afferent activity and the vestibuloocular reflex(VOR)were investigated before and after exposure to blast shock waves(~20 PSI)delivered into the left external ear canals of anesthetized *** vestibular afferent activity was recorded from the superior branch of the left vestibular nerves of the blast-treated and control rats one day after blast *** exposure reduced the spontaneous discharge rates of the otolith and canal *** exposure also reduced the sensitivity of irregular canal afferents to sinusoidal head rotation at *** exposure,however,resulted in few changes in the VOR responses to sinusoidal head rotation and *** the best of our knowledge,this is the first study that reports blast exposure-induced damage to vestibular afferents in an animal *** results provide insights that may be helpful in developing biomarkers for early diagnosis of blast-induced vestibular deficits in military and civilian populations.

基于网络药理学的舒心安神汤抗抑郁实验研究

目的:采用网络药理学、分子对接和动物实验探讨舒心安神汤治疗抑郁症的作用及机制。方法:通过TCMSP、Swiss Target Prediction、Uniprot数据库筛选舒心安神汤中的活性成分及作用靶点,采用Gene Cards、OMIM、Drug Bank数据库获得抑郁症疾病靶点;用Cytoscape软件构建可视化网络,利用Venny 2.1软件筛选、收集药物与疾病的共有靶点。将筛选出的药物和疾病的共同靶点输入String数据库,获得蛋白质相互作用(PPI)网络图,进一步筛选出核心靶点。于david数据库中对交集基因进行GO富集和KEGG通路分析,预测其富集的信号通路并进行可视化展示,将核心靶点与相关活性成分在CB-Dock在线平台进行分子对接验证,并采用LC-MS对舒心安神汤主要化学成分进行分析鉴定。构建皮质酮诱导抑郁小鼠模型,利用行为学实验、尼氏染色、Western blotting法进行舒心安神汤药效评估并对脑源性神经营养因子(BDNF)、酪氨酸激酶受体B(TRKB)、环磷腺苷效应元件结合蛋白(CREB)反映突触可塑性相关蛋白的表达进行考察。结果:网络药理学预测得到舒心安神汤治疗抑郁症的潜在活性成分141个,主要为槲皮素、β-谷甾醇、山柰酚、芍药苷、莫诺苷、斯皮诺素及姜辣素等活性成分,关键靶点包括IL6、ESR1、AKT1和EGFR等,GO富集分析结果发现抑郁反应与蛋白磷酸化调控、细胞外基质激酶结合及蛋白激酶活性等相关。KEGG通路富集结果显示,舒心安神汤治疗抑郁可能通过调节PI3K-AKT、HIF-1及MAPK等信号通路发挥作用。分子对接发现芍药苷、莫诺苷、斯皮诺素、姜辣素与ESR、AKT1和BDNF结合能力较强。LC-MS鉴定出舒心安神汤中含有15种高响应化学成分,与网络药理学核心成分预测基本一致。动物实验结果表明,与模型对照组比较,舒心安神汤20.5 g/kg组的小鼠体质量、蔗糖偏好明显增加,强迫游泳时间和新奇抑制摄食试验潜伏期明显减少(P<0.05);尼氏染色显示模型对照组小鼠海马CA3区神经元排列欠规则,细胞周围间隙增宽,胞质内尼氏小体明显减少,而舒心安神汤20.5 g/kg组小鼠海马CA3区神经元排列较为规则,细胞周围间隙缩短,尼氏体较为丰富;舒心安神汤20.5 g/kg组能明显上调小鼠海马组织TRKb、p-AKT、BDNF、CREB的蛋白表达(P<0.05)。结论:舒心安神汤可能通过其含有的槲皮素、β-谷甾醇、芍药苷、姜辣素和斯皮诺素等主要成分作用于ESR1、AKT1等靶基因,并影响BDNF/TRKB信号通路及下游相关蛋白等,发挥改善抑郁症的作用,其机制可能与上调BDNF、TRKB、p-AKT及CREB蛋白表达,改善海马神经元可塑性有关。

Self-reported risk of stroke and factors associated with underestimation of stroke risk among older adults with atrial fibrillation: the SAGE-AF study

Background Though engaging patients with atrial fibrillation(AF) in understanding their stroke risk is encouraged by guidelines, little is known regarding AF patients' perceived stroke risk or its relationship with oral anticoagulation(OAC) use. We aim to identify factors associated with underestimation of stroke risk among older patients with AF and relate this to OAC use. Methods Data are from the ongoing SAGE(Systematic Assessment of Geriatric Elements)-AF study, which included older patients(> 65 years) with non-valvular AF and a CHA2 DS2-VASc score of ≥ 2. Participants reported their perceived risk of having a stroke without OAC. We compared the perceived risk to CHA2 DS2-VASc predicted stroke risk and classified participants as "over" or "under" estimators, and identified factors associated with underestimation of risk using multiple logistic regression. Results The average CHA2 DS2-VASc score of 915 participants(average age: 75 years, 47% female, 86% white) was 4.3 ± 1.6, 43% of participants had discordant predicted and self-reported stroke risks. Among the 376 participants at highest risk(CHA2 DS2-VASc score ≥ 5), 46% of participants underestimated their risk. Older participants(≥ 85 years) were more likely and OAC treated patients less likely to underestimate their risk of developing a future stroke than respective comparison groups. Conclusions A significant proportion of study participants misperceived their stroke risk, mostly by overestimating. Almost half of participants at high risk of stroke underestimated their risk, with older patients more likely to do so. Patients on OAC were less likely to underestimate their risk, suggesting that successful efforts to educate patients about their stroke risk may influence treatment choices.

Detection and Adaptation in Parasitic Angiosperm Host Selection

Developmental transitions in some parasitic angiosperms are tied directly to host-derived chemical cues (xenognosins). The obligate hemi-parasite Striga asiatica, initiates the root apical meristem population (germination), development of the host attachment organ (the haustorium), and shoot apical meristem initiation (seed coat shedding) in response to specific xengonosins. These checkpoints synchronize spatial and temporal tissue development. We have now exploited the external control over these developmental transitions to trace functional expression in haustorial organogenesis. Genes associated with phytohormone regulation, metabolism, vascular tissue development, and reactive oxygen species (ROS) production identified in this study suggest an elaborate and global response closely tied to plant defense and redox chemistry that may also be components of a more general quorum sensing-type mechanism in plants.

目的:通过网络药理学方法、分子对接技术和体外实验,探讨虎杖干预病毒性肺炎的可能作用机制。方法:通过TCMSP平台筛选中药虎杖的活性成分及其作用靶点,从GeneCards数据库筛选病毒性肺炎的相关靶点,取交集得到潜在作用靶点;通过STRING数据库结合Cytoscape 3.7.2软件对潜在作用靶点进行交集靶点蛋白互作(PPI)分析和拓扑学分析,得到虎杖核心成分和病毒性肺炎关键靶点。采用david数据库对交集靶点进行GO功能分析和KEGG通路富集分析,结合Cytoscape 3.7.2软件构建“虎杖-活性成分-靶点-通路”调控网络。采用AutoDock Vina、OpenBabel、PyMOL等软件对虎杖活性成分和病毒性肺炎关键靶点进行分子对接。以脂多糖(LPS)诱导的急性肺损伤人Ⅱ型肺泡上皮细胞A549建立急性肺损伤模型,对网络药理学的研究结果进行验证。结果:共获得虎杖10个活性成分,对应188个作用靶点,病毒性肺炎靶点1216个,取药物与疾病交集靶点93个;槲皮素、木犀草素、毒扁豆次碱、β-谷甾醇等可能是虎杖的核心成分;丝氨酸/苏氨激酶1(Akt1)、血管内皮生长因子A(VEGFA)、肿瘤坏死因子(TNF)、白介素-6(IL6)、白介素-1β(IL1β)、JUN激酶(JUN)、前列腺素G/H合成酶2(PTGS2)、基质金属蛋白酶-9(MMP-9)、半脱氨酸蛋白酶-3(CASP3)等蛋白可能是虎杖干预病毒性肺炎的核心靶点;GO功能分析和KEGG通路富集分析发现,靶点蛋白在细胞外基质、线粒体、核浆、胞质等中通过调控细胞因子、生长因子、蛋白、蛋白激酶以及TNF、PI3K-Akt、Toll样受体等信号通路发挥作用。分子对接发现,虎杖5种核心成分与TNF、Akt1、CASP3、IL1β、JUN、TP53、VEGFA靶点均有强烈的结合能力,结合能<-7.0 kcal/mol,与IL6、PTGS2均有较好的结合能力,结合能<-5.0 kcal/mol,与MMP-9有一定的结合能力,结合能<-4.25 kcal/mol。体外实验结果表明,虎杖可以降低模型组TNF-α、IL-1β、IL-6的表达,与空白组比较,差异有统计学意义(P<0.01,P<0.05),可以抑制Toll样受体4(TLR4)、核因子-κB(NF-κB)表达,差异有统计学意义(P<0.05)。结论:虎杖可能通过调控TNF-α、IL-1β、IL-6、TLR4、NF-κB等靶点干预病毒性肺炎,发挥治疗作用。
