晋南牛ApoA1基因克隆及组织表达谱分析

为研究晋南牛载脂蛋白A1(ApoA1)基因的结构、生物学功能及其在不同组织中的表达变化规律，本试验以晋南牛肝脏组织总RNA为模板，克隆ApoA1基因序列，并利用多种生物信息学在线分析软件对晋南牛的ApoA1基因进行生物信息学分析及构建系统发育进化树。利用荧光定量PCR方法检测晋南牛心脏、肝脏、脾脏、肺脏和肾脏等组织中ApoA1基因的表达情况。结果表明，晋南牛ApoA1基因的CDS序列全长852 bp，与黄牛ApoA1基因的同源性为98.00%。晋南牛ApoA1蛋白的分子量为30 276.35 Da，由265个氨基酸残基构成，属于亲水性蛋白，有20处磷酸化位点。在ApoA1的二级结构中，α-螺旋占92.08%，属于全α类蛋白。ApoA1蛋白与ApoA2、ApoB、ApoE、ApoC2、ApoC3、SCARB1、LPL、ABCA1等蛋白存在相互作用。基于ApoA1序列同源性构建的系统发育进化树表明，晋南牛与黄牛的亲缘关系最近，与野牛的亲缘关系最远。ApoA1基因在晋南牛肝脏、心脏、脾脏、肺脏和肾脏组织中均有表达，且以肝脏中的相对表达量最高，肾脏中的相对表达量最低。

Chimeric antigen receptor T cell(CAR-T cell) therapy is a novel adoptive immunotherapy where T lymphocytes are engineered with synthetic receptors known as chimeric antigen receptors(CAR).The CAR-T cell is an effector T cell that recognizes and eliminates specific cancer cells, independent of major histocompatibility complex molecules. The whole procedure of CAR-T cell production is not well understood. The CAR-T cell has been used predominantly in the treatment of hematological malignancies,including acute lymphoblastic leukemia, chronic lymphocytic leukemia, lymphoma, and multiple myeloma. Solid tumors including melanoma, breast cancer and sarcoma offer great promise in CAR-T cell research and development. CD19 CAR-T cell is most commonly used, and other targets, including CD20, CD30, CD38 and CD138 are being studied. Although this novel therapy is promising, there are several disadvantages. In this review we discuss the applications of CAR-T cells in different hematological malignancies, and pave a way for future improvement on the effectiveness and persistence of these adoptive cell therapies.

DNA is a biological polymer that encodes and stores genetic information in all living organism. Particularly, the precise nucleobase pairing inside DNA is exploited for the self-assembling of nanostructures with defined size, shape and functionality. These DNA nanostructures are known as framework nucleic acids(FNAs) for their skeleton-like features. Recently, FNAs have been explored in various fields ranging from physics, chemistry to biology. In this review, we mainly focus on the recent progress of FNAs in a pharmaceutical perspective. We summarize the advantages and applications of FNAs for drug discovery, drug delivery and drug analysis. We further discuss the drawbacks of FNAs and provide an outlook on the pharmaceutical research direction of FNAs in the future.

In this paper, we study estimation of a partially specified spatial autoregressive model with heteroskedasticity error term. Under the assumption of exogenous regressors and exogenous spatial weighting matrix, we propose an instrumental variable estimation. Under some sufficient conditions,we show that the proposed estimator for the finite dimensional parameter is root-n consistent and asymptotically normally distributed and the proposed estimator for the unknown function is consistent and also asymptotically distributed though at a rate slower than root-n. Monte Carlo simulations verify our theory and the results suggest that the proposed method has some practical value.

综合考虑流体分子与孔壁间的相互作用、临界性质变化、毛细管力和吸附相的影响,利用修正后的PR(Peng-Robinson)状态方程和气相-液相-吸附相三相相平衡计算方法,分析CO_(2)-页岩油体系在纳米孔隙内的相行为。研究发现:受到纳米孔隙限域效应影响,随着孔径的减小页岩孔隙内流体临界温度和临界压力下降,CO_(2)对体系临界温度的降低起抑制作用、对体系临界压力的降低起促进作用;同时随着CO_(2)物质的量分数的增大,体系临界点左移,相包络线面积减小。大庆古龙A区块页岩储层具有明显的限域效应,当孔径为10 nm时,储层流体逐渐转变为具有凝析气藏流体特征的流体;10 nm孔隙中液相的CO_(2)含量比100 nm孔隙中液相的CO_(2)含量增加了20.0%,而气相中CO_(2)含量减少了10.8%,表明限域效应增强了纳米孔隙中CO_(2)的传质,有利于CO_(2)的埋存和微观驱油。

Cryoablation(CRA)and microwave ablation(MWA)are two main local treatments for hepatocellular carcinoma(HCC).However,which one is more curative and suitable for combining with immunotherapy is still ***,CRA induced higher tumoral PD-L1 expression and more T cells infiltration,but less PD-L1^(high)CD11b^(+)myeloid cells infiltration than MWA in ***,CRA had better curative effect than MWA for anti-PD-L1 combination therapy in mouse ***,anti-PD-L1 antibody facilitated infiltration of CD8^(+)T cells by enhancing the secretion of CXCL9 from cDC1 cells after CRA *** the other hand,anti-PD-L1 antibody promoted the infiltration of NK cells to eliminate PD-L1^(high)CD11b^(+)myeloid cells by antibody-dependent cell-mediated cytotoxicity(ADCC)effect after CRA *** aspects relieved the immunosuppressive microenvironment after CRA ***,the wild-type PD-L1 Avelumab(Bavencio),compared to the mutant PD-L1 atezolizumab(Tecentriq),was better at inducing the ADCC effect to target PD-L1^(high)CD11b^(+)myeloid ***,our study uncovered the novel insights that CRA showed superior curative effect than MWA in combining with anti-PD-L1 antibody by strengthening CTL/NK cell immune responses,which provided a strong rationale for combining CRA and PD-L1 blockade in the clinical treatment for HCC.
