Selenium-enriched and ordinary green tea extracts prevent high blood pressure and alter gut microbiota composition of hypertensive rats caused by high-salt diet

High-salt diet is well recognized as a risk factor for hypertension,and dietary intervention plays a critical role in the prevention of hypertension.The current study investigated the effects of selenium-enriched green tea(Se-GT)and ordinary green tea(GT)on prevention of hypertension of rats induced by high-salt diet,as well as their potential regulatory and mechanism.Our results showed that GT and Se-GT supplementations significantly prevented the increase of blood pressure(BP),activated the phosphoinosmde-3-kinase/protein kinase B(PI3K/Akt)signaling pathway,and regulated the gene expression related to BP,as well as improved the tissue damage like heart,liver,and kidneys.Besides,the key parameters associated with oxidative stress,inflammation and endothelial dysfunction were also altered by GT and Se-GT treatments.Importantly,GT or Se-GT administration adjusted the diversity and composition of the intestinal flora.Moreover,GT and Se-GT supplementations increased the abundance of beneficial bacteria and reduced the abundance of harmful or conditional pathogenic bacteria.More specifically,GT intake specifically and significantly enriched the relative abundance of Paraprevotella and Bacteroides,whereas Se-GT was characterized by specific and significant enrichment for Allobaculum and Bifidobacterium.Our results proved that dietary supplement of GT and Se-GT remarkably improved the vascular functions and effectively prevented tissue damage by regulation of intestinal flora,and thus preventing hypertension induced by high-salt diet.

Simulated impacts of 3D urban morphology on urban transportation in megacities: case study in Beijing

Urban morphology and morphology change and their impacts on urban transportation have been studied extensively in planar urban space.The essential feature of urban space,however,is its three-dimensionality(3D),and few studies have been conducted from a 3D perspective,overly limiting the accuracy of studies on the relationships between urban morphology and transportation.The aim of this paper is to simulate the impacts of 3D urban morphologies on urban transportation under the Digital Earth framework.On the basis of the principle that population distribution and movement are largely confined by 3D urban morphologies,which affect transportation,high spatial resolution remote sensing imagery and a thematic vector data-set were used to extract urban morphology and transportation-related variables.With a combination of three research methods-factor analysis,spatial regression analysis and Euclidean allocation-we provide an effective method to construct a simulation model.The paper indicates three general results.First,building capacity in the urban space has the most significant impact on traffic condition.Second,obvious urban space otherness,reflecting both use density characteristics and functional character-istics of urban space,mostly results in heavier traffic flow pressure.Third,no single morphology density indicator or single urban structure indicator can reflect its contribution to the pressure of traffic flow directly,but a combination of these different indicators has the ability to do so.

Protein@PP-Zn nanocomplex assembled by coordination of zinc ions used for intracellular protein delivery

In recent years,intracellular delivery of protein drugs has attracted great attention,and polymer-based systems have been extensively exploited to develop efficient and safe carriers.However,efficient intracellular delivery of protein drugs remains a challenge because of the cell membrane barrier and endosome entrapment.Herein,we report a protein@PP-Zn nanocomplex,which consists of an imidazole-containing block polymer poly(ethylene glycol)-block-poly(β-amino ester)(PEG-b-PAE(Im),PP),zinc ions,and protein drugs,for efficient intracellular protein delivery.PEG-b-PAE(Im)could conjugate proteins via the bridging effect of zinc ions which simultaneously coordinate with imidazole groups on polymer and electron donor groups,such as imidazole and primary amine groups,on protein to improve the loading stability of proteins.Under a slightly acidic environment near cancer cells,the protonation of PAE(Im)backbone increases the positive charge density of the nanocomplex and promotes endocytosis.While under a more acidic environment in endosomes,further protonation of imidazole groups leads to the disintegration of the nanocomplex and the breakdown of endosomes because of the proton sponge effect.Finally,protein is released into the cytoplasm.With the assistance of the nanocomplex,proteins with different sizes and isoelectric points are effectively delivered into cells.This work provides a stable,efficient and universal strategy for intracellular protein delivery.

清入关定都北京建立起全国性政权后,经过百余年时间的不懈努力,最终统一了新疆天山南北,为了巩固统一和加强统治,乾隆二十七年(1762)设立伊犁将军,通过军府制治理新疆。伊犁将军作为新疆的军政首领,其职责十分重要,清廷也很重视伊犁将军的选拔任用,一般都从京城部院及八旗选派满洲或蒙古大员任职。他们在办理公务过程中按规定多以满文书写奏折,向皇帝请示和汇报问题,所以形成了一定数量的满文奏折。这些历任伊犁将军的满文奏折,内容丰富,涉及面广,包括政治、军事、经济、文化等诸多方面,是第一手原始史料,具有其他文献资料无法替代的学术研究价值。

P-band center theory guided activation of MoS_(2) basal S sites for pHuniversal hydrogen evolution

The edge S sites of thermodynamically stable 2H MoS_(2)are active for hydrogen evolution reaction(HER)but the active sites are scarce.Despite the dominance of the basal S sites,they are generally inert to HER because of the low p-band center.Herein,we reported a synergistic combination of phase engineering and NH_(4)^(+) intercalation to promote the HER performance of MoS_(2).The rational combination of 1T and 2H phases raises the p-band center of the basal S sites while the intercalated NH4+ions further optimize and stabilize the electronic band of these sites.The S sites with regulated band structures afford moderate hydrogen adsorption,thus contributing to excellent HER performance over a wide pH range.In an acid medium,this catalyst exhibits a low overpotential of 169 mV at 10 mA·cm^(−2)and Tafel slope of 39 mV·dec^(−1)with robust stability,superior to most of recently reported MoS_(2)-based non-noble catalysts.The combined use of in/ex-situ characterizations ravels that the appearance of more unpaired electrons at the Mo 4d-orbital reduces the d-band center which upshifts the p-band center of the adjacent S for essentially improved HER performance.This work provides guidelines for the future development of layered transition-metal-dichalcogenide catalysts.

Facile generation of highly durable thiol-functionalized polyhedral oligomeric silsesquioxane based superhydrophobic melamine foam

In this study,a durable superhydrophobic/superoleophilic melamine foam was fabricated by a facile and rapid one-step thiol-ene click chemistry and Michael addition reaction,which demonstrated excellent robustness in oil/water separation.First,1H,1H,2H-perfluoro-1-hexene reacted with thiol-functionalized polyhedral oligomeric silsesquioxane via the thiol-ene click chemistry to obtain a fluorinated thiol-functionalized polyhedral oligomeric silsesquioxane solution.Subsequently,the melamine foam was immersed to the solution system to form nanoaggregates on the melamine foam surface by the Michael addition reaction in the presence of ultraviolet light.The micro/nano rough structure and low surface energy of the nanoaggregates layer endowed the pristine melamine foam with superhydrophobicity;the water contact angle was greater than 150°.More importantly,the as-prepared melamine foam could withstand harsh conditions,such as a corrosive solution environment,strong ultraviolet light,mechanical compression,high and low temperature exposure,and ultrasonic washing.Driven by gravity,the as-prepared melamine foam could efficiently separate the oil/water mixtures and maintain 98%separation efficiency at high and low temperatures.In addition,it maintained the desirable absorption capability in different oil/organic solvents even after 15 absorption cycles.Accordingly,this facile,low-cost,and robust onestep method provides important support for the superhydrophobic oil/water separation field.

Effects of miR-200c on the migration and invasion abilities of human prostate cancer Du145 cells and the corresponding mechanism

microRNAs （miRNAs） have played a key role in human tumorigenesis, tumor progression, and metastasis. On the one hand, miRNAs are aberrantly expressed in many types of human cancer; on the other hand, miRNAs can function as tumor suppressors or oncogenes that target many cancer-related genes. This study aimed to investigate the effects of miRNA-200c （miR-200c） on the biological behavior and mechanism of proliferation, migration, and invasion in the prostate cancer cell line Du145. In this study, Du145 cells were transfected with miR-200c mimics or negative control miR-NC by using an X-tremeGENE siRNA transfection reagent. The relative expression of miR-200c was measured by RT-PCR. The proliferation, migration, and invasion abilities of Du145 cells were detected by CCK8 assays, migration assays and invasion assays, respectively. The expressions of ZEB1, E-cadherin, and vimentin were observed by western blot. Results showed that DU145 cells exhibited a high expression of miR-200e compared with immortalized normal prostate epithelial cell RWPE-1. Du145 cells were then transfected with miR-200c mimics and displayed lower abilities of proliferation, migration, and invasion than those transfected with the negative control. The protein levels of ZEB1 and vimentin were expressed at a low extent in Du145 cells, which were transfected with miR-200c mimics; by contrast, E-cadherin was highly expressed. Hence, miR-200c could significantly inhibit the proliferation of the prostate cancer cell line Du145; likewise, miR- 200c could inhibit migration and invasion by epithelial-mesenchymal transition.

New antimicrobial strategies are urgently needed to meet the challenges posed by the emergence of drug-resistant bacteria and bacterial biofilms.This work reports the facile synthesis of antimicrobial dynamic covalent nano-networks(aDCNs)composing antibiotics bearing multiple primary amines,polyphenols,and a cross-linker acylphenylboronic acid.Mechanistically,the iminoboronate bond drives the formation of aDCNs,facilitates their stability,and renders them highly responsive to stimuli,such as low pH and high H2O2 levels.Besides,the representative A1B1C1 networks,composed of polymyxin B1(A1),2-formylphenylboronic acid(B1),and quercetin(C1),inhibit biofilm formation of drug-resistant Escherichia coli,eliminate the mature biofilms,alleviate macrophage inflammation,and minimize the side effects of free polymyxins.Excellent bacterial eradication and inflammation amelioration efficiency of A1B1C1 networks are also observed in a peritoneal infection model.The facile synthesis,excellent antimicrobial performance,and biocompatibility of these aDCNs potentiate them as a much-needed alternative in current antimicrobial pipelines.
