Regulatory Expression of Peroxisome Proliferator Activated Receptors Genes During Fatty Liver Formation in Geese

In order to investigate the expression pattern of peroxisome proliferator activated receptor （PPAR） genes before and after overfeeding, and estimate the effect of expressed PPAR levels on weights of fatty liver and abdominal fat in geese, the RT-PCR products of PPAR genes in heart, liver, spleen, lung, kidney, stomach, small intestine, brain, breast muscle, leg muscle, and abdominal fat were determined before and after overfeeding. RT-PCR was used to determine the expression levels of PPAR genes. Quantity one software was used to analyze absorbency, and the expression level of GAPDH gene was used as contrast. Expression levels of PPAR-α were relatively high in most of detected tissues but undetectable in abdominal fat tissue before overfeeding, and the level was evidently increased in lung, appeared in abdominal fat tissue, and reduced in the other tissues after overfeeding. Expressed PPAR-γ levels were relatively high in liver, spleen, lung, small intestine, and abdominal fat, and relatively low in the other tissues before overfeeding. Expressed PPAR-γ levels were enhanced in liver, spleen, lung, stomach, and kidney but decreased in abdominal fat and without obvious changes in the other tissues. Expression patterns of PPAR genes show tissue-specific manner. In addition, expression patterns of PPAR-α are different from PPAR-γ after overfeeding. It might suggest that different functions of PPAR subtypes are responsive to overfeeding.

马铃薯StXYLP基因家族的生物信息学分析

[目的]在马铃薯基因组水平上鉴定木质形成素类蛋白(Xylogen-like protein,XYLP)基因家族,分析其理化性质及基因的表达模式。[方法]用生信学方法鉴定马铃薯XYLPs,分析其理化性质、亚细胞定位、进化关系、蛋白质结构、基因结构和染色体定位。利用芯片数据分析马铃薯StXYLPs的时空表达模式及对非生物胁迫的响应。[结果]马铃薯中共有20个StXYLPs;蛋白全长在139~221氨基酸之间,理论等电点在4.36~9.42之间,均锚定在质膜上;蛋白质结构基本相似,均具有保守的nsLTP结构域;StXYLPs在染色体上的定位具有偏好性;StXYLPs具有明显差异的组织表达模式,响应不同的非生物胁迫。[结论]StXYLPs与生长素、细胞分裂素和赤霉素共同调控马铃薯器官发育,19个StXYLPs表达受调控。12个StXYLPs受ABA诱导表达上调,在盐、高温及干旱胁迫下,14个StXYLPs受诱导表达上调,参与对抗非生物胁迫。

Proteinase-activated receptor 2 promotes tumor cell proliferation and metastasis by inducing epithelialmesenchymal transition and predicts poor prognosis in hepatocellular carcinoma

aiM To clarify the role of proteinase-activated receptor 2(PAR2) in hepatocellular carcinoma, especially in the process of *** PAR2 expression levels were assessed by qRT-PCR and immunohistochemistry(IHC) in patient tissues and in hepatocellular carcinoma cell lines SMMC-7721 and Hep G2. Cell proliferation and metastasis were assessed both in vitro and in vitro. Immunoblotting was carried out to monitor the levels of mitogen-activated protein kinase(MAPK) and epithelial-mesenchymal transition *** The prognosis was significantly poorer in patients with high PAR2 levels than in those with low PAR2 levels. Patients with high PAR2 levels had advanced tumor stage(P = 0.001, chi-square test), larger tumor size(P = 0.032, chi-square test), and high microvascular invasion rate(P = 0.037, chi-square test). The proliferation and metastasis ability of SMMC-7721 and Hep G2 cells was increased after PAR2 overexpression, while knockdown of PAR2 decreased the proliferation and metastasis ability of SMMC-7721 and Hep G2 cells. Knockdown of PAR2 also inhibited hepatocellular carcinoma tumor cell growth and liver metastasis in nude mice. Mechanistically, PAR2 increased the proliferation ability of SMMC-7721 and Hep G2 cells via ERK activation. Activated ERK further promoted the epithelial-mesenchymal transition of these cells, which endowed them with enhanced migration and invasion ability. CONCLUSION These data suggest that PAR2 plays an important role in the proliferation and metastasis of hepatocellular carcinoma. Therefore, targeting PAR2 may present a favorable target for treatment of this malignancy.

The visual system continuously adapts to the statistical properties of the environment. Existing evidence shows a close resemblance between deep convolutional neural networks(CNNs) and primate visual stream in neural selectivity to naturalistic textures above the primary visual processing stage. This study delves into the mechanisms of perceptual learning in CNNs,focusing on how they assimilate the high-order statistics of natural textures. Our results show that a CNN model achieves a similar performance improvement as humans, as manifested in the learning pattern across different types of high-order image statistics. While L2 was the first stage exhibiting texture selectivity, we found that stages beyond L2 were critically involved in learning. The significant contribution of L4 to learning was manifested both in the modulations of texture-selective responses and in the consequences of training with frozen connection weights. Our findings highlight learning-dependent plasticity in the mid-to-high-level areas of the visual hierarchy. This research introduces an ai-inspired approach for studying learning-induced cortical plasticity, utilizing DCNNs as an experimental framework to formulate testable predictions for empirical brain studies.

基于丽水国际会展中心项目政务方厅钢结构施工策划实践,介绍双层交错式大跨重型钢梁在受限空间内采用逆序施工技术的具体应用情况。采用先上后下的逆序吊装施工顺序,并结合原有结构支撑反顶、楼板钢筋加密、双机抬吊等施工工艺措施,使得在多种受限工况下的双层交错式重型钢梁仍可以经地面拼装后进行整体吊装,避免因大型机械上楼板造成的楼板开裂以及重型钢梁高空对接导致的精度不足、焊接质量不合格等一系列严重质量问题,最终达到减少安装措施费用、保护成品结构、缩短工期的施工目标,实现工程施工效益最大化。

建立了氢化物发生原子荧光光谱法测定锑精矿中砷、铋、硒、锡的方法。研究了基体及共存元素的干扰情况,实验表明,在酒石酸、硫脲-抗坏血酸存在下,适当的增加酸度可以有效地消除干扰。采用Na2O2熔解样品,用HCl酸化,无需分离基体,实现了锑精矿中砷、铋、硒、锡的连续测定,其回收率为90.6%～103.8%,检出限分别为0.35、0.20、0.65和0.35μg/L。应用该方法分析了锑精矿样品,结果令人满意。

High temperature polyimide nanocomposites containing twodimensional nanofillers for improved thermal stability and capacitive energy storage performance

Future electronic devices toward high integration and miniaturization demand reliable operation of dielectric materials at high electric fields and elevated ***,the electrical deterioration caused by Joule heat generation remains a persistent challenge to ***,the solution-processed polyimide(PI)nanocomposites with unique two-dimensional(2D)alumina nanoplates are *** improvements in the breakdown strength,charge–discharge efficiency and discharged energy density at elevated temperatures have been demonstrated in the composites,owing to simultaneously suppressed conduction loss and increased thermal conductivity upon the incorporation of 2D Al_(2)O_(3) nanofillers possessing excellent dielectric insulation and thermophysical *** predominance of Al_(2)O_(3) nanoplates in enhancing thermal stability and high-temperature capacitive performance over nanoparticles and nanowires is validated experimentally and is further rationalized via finite element ***,the Al_(2)O_(3) nanoplates filled PI nanocomposite exhibits a hightemperature capability up to 200℃ and remarkable efficiency(e.g.≥95% at 200 MV/m)over a wide temperature range,which outperforms commercial dielectric polymers and rivals the state-of-the-art polyimide nanocomposites.
