CNKSR2 interactome analysis indicates its association with the centrosome/microtubule system

The protein connector enhancer of kinase suppressor of Ras 2(CNKSR2),present in both the postsynaptic density and cytoplasm of neurons,is a scaffolding protein with several protein-binding *** of the CNKSR2 gene have been implicated in neurodevelopmental disorders,particularly intellectual disability,although the precise mechanism involved has not yet been fully *** has demonstrated that CNKSR2 plays a role in facilitating the localization of postsynaptic density protein complexes to the membrane,thereby influencing synaptic signaling and the morphogenesis of dendritic ***,the function of CNKSR2 in the cytoplasm remains to be *** this study,we used immunoprecipitation and high-resolution liquid chromatography-mass spectrometry to identify the interactors of *** a combination of bioinformatic analysis and cytological experiments,we found that the CNKSR2 interactors were significantly enriched in the proteome of the *** also showed that CNKSR2 interacted with the microtubule protein DYNC1H1 and with the centrosome marker *** colocalization analysis confirmed the centrosomal localization of *** we downregulated CNKSR2 expression in mouse neuroblastoma cells(Neuro 2A),we observed significant changes in the expression of numerous centrosomal *** manipulation also affected centrosome-related functions,including cell size and shape,cell proliferation,and ***,we found that CNKSR2 interactors were highly enriched in de novo variants associated with intellectual disability and autism spectrum *** findings establish a connection between CNKSR2 and the centrosome,and offer new insights into the underlying mechanisms of neurodevelopmental disorders.

Solid-state lithium batteries(SSLBs)are regarded as an essential growth path in energy storage systems due to their excellent safety and high energy *** particular,SSLBs using conversion-type cathode materials have received widespread attention because of their high theoretical energy densities,low cost,and *** the great progress in research and development of SSLBs based on conversiontype cathodes,their practical applications still face challenges such as blocked ionic-electronic migration pathways,huge volume change,interfacial incompatibility,and expensive processing *** review focuses on the advantages and critical issues of coupling conversion-type cathodes with solid-state electrolytes(SSEs),as well as state-of-the-art progress in various promising cathodes(e.g.,FeS_(2),CuS,FeF_(3),FeF_(2),and S)in ***,representative research on conversion-type solid-state full cells is discussed to offer enlightenment for their practical ***,the energy density exhibited by the S cathode stands out impressively,while sulfide SSEs and halide SSEs have demonstrated immense potential for coupling with conversion-type ***,perspectives on conversion-type cathodes are provided at the material,interface,composite electrode,and battery levels,with a view to accelerating the development of conversion-type cathodes for high-energy–density SSLBs.

Using the reference sequences of pgip genes in GenBank,a fragment of 930 bp covering the open reading frame(ORF) of rice Ospgip1(Oryza sativa polygalacturonase-inhibiting protein 1) was *** prokaryotic expression product of the gene inhibited the growth of Rhizoctonia solani,the causal agent of rice sheath blight,and reduced its polygalacturonase *** analysis showed that OsPGIP1 is a hydrophobic protein with a molecular weight of 32.8 kDa and an isoelectric point(pI) of *** protein is mainly located in the cell wall of rice,and its signal peptide cleavage site is located between the 17th and 18th amino *** are four cysteines in both the N-and C-termini of the deduced protein,which can form three disulfide bonds(between the 56th and 63rd,the 278th and 298th,and the 300th and 308th amino acids).The protein has a typical leucine-rich repeat(LRR) domain,and its secondary structure comprises α-helices,β-sheets and irregular *** with polygalacturonase-inhibiting proteins(PGIPs) from other plants,the 7th LRR is absent in *** nine LRRs could form a cleft that might associate with proteins from pathogenic fungi,such as polygalacturonase.

Fracture behavior of sandstone with partial filling flaw under mixed-mode loading: Three-point bending tests and discrete element method

The fracture behavior of natural fracture in the geological reservoir subjected to filling property,affects the crack initiation and propagation under stress *** filling flaws were intermediate between open fractures and filled fractures,the fracture response may be worth *** this work,the effect of the filling property of sandstone with partial filling flaws on the fracture behavior was systematically investigated based on three-point bending tests and the numerical approach of discrete element method(DEM).In the laboratory,semi-circular three-point bending tests were carried out with partial filling flaws of various filling *** on this,numerical simulations were used to further investigate the effect of the filling ratio and the inclination of the partial filling flaw on the mechanical and fracture responses,and the effect of the partial filling flaw under mixed-mode loading on the fracture mechanism was elucidated coupled with acoustic emission(AE)*** obtained results showed that the increase in filling strength and filling ratio of partial filling flaw led to an increase in peak strength,with a decreasing trend in peak strength with the inclination of partial filling *** terms of crack propagation pattern,the increasing filling strength of the partial filling flaw induced the transformation of the fracture mechanism toward deflection,with a tortuosity path,while the filling ratio and inclination of partial filling flaw led to fracture mechanism change from deflection to penetration and attraction,accompanied with a larger AE event source in ***,the b-value based on the Gutenberg-Richter equation fluctuated between 5 and 4 at low filling ratio and inclination and remained around 5 at high filling ratio and inclination of partial filling *** results may provide an application prospective for reservoir stimulation using the natural fracture system.

High-density markers are necessary for map-based cloning of rice genes, but the currently available markers are not satisfactory enough. InDel (insertion-deletion length polymorphism) and SNP (single nucleotide polymorphism) are the new generation of molecular markers and can basically meet the need of fine mapping. InDel and SNP markers can be developed through bioinformatics. These markers are valuable markers with the characters of low cost, high specificity and stability. This article introduced the methods for designing InDel and SNP markers, taking the mapping of a rice rolled leaf gene as an example. In addition, some key factors in improving the design efficiency were also discussed.

Exosomes:the next-generation therapeutic platform for ischemic stroke

Current therapeutic strategies for ischemic stroke fall short of the desired objective of neurological functional ***,there is an urgent need to develop new methods for the treatment of this *** are natural cell-derived vesicles that mediate signal transduction between cells under physiological and pathological *** have low immunogenicity,good stability,high delivery efficiency,and the ability to cross the blood–brain *** physiological properties of exosomes have the potential to lead to new breakthroughs in the treatment of ischemic *** rapid development of nanotechnology has advanced the application of engineered exosomes,which can effectively improve targeting ability,enhance therapeutic efficacy,and minimize the dosages *** in technology have also driven clinical translational research on *** this review,we describe the therapeutic effects of exosomes and their positive roles in current treatment strategies for ischemic stroke,including their antiinflammation,anti-apoptosis,autophagy-regulation,angiogenesis,neurogenesis,and glial scar formation reduction ***,it is worth noting that,despite their significant therapeutic potential,there remains a dearth of standardized characterization methods and efficient isolation techniques capable of producing highly purified *** optimization strategies should prioritize the exploration of suitable isolation techniques and the establishment of unified workflows to effectively harness exosomes for diagnostic or therapeutic applications in ischemic ***,our review aims to summarize our understanding of exosome-based treatment prospects in ischemic stroke and foster innovative ideas for the development of exosome-based therapies.

Sulfur isotopes of S-bearing materials are powerful tools to trace various geological processes and sulfur sources in earth sciences,especially in ore deposits where sulfide-sulfate pair coprecipitates ***,in-situ S isotope determination of barite is challenging without natural matrix-matched reference *** this study,we present two natural barite reference materials(1-YS and 294-YS)for in-situ sulfur isotopic *** LA-MC-ICP-MS laboratories were utilized to test theδ34S micron-scale homogeneity of 1-YS and 294-YS barites that have 2s repeatabilities of better than±0.45‰and±0.41‰,***,the in-situ analysis results are consistent with the results of the bulk analysis by GS-IRMS within *** grand meanδ~(34)S values of 1-YS(13.37‰±0.42‰,2s)and 294-YS(14.38‰±0.44‰,2s)are the final recommended values obtained from four independent *** the results confirm the suitability of 1-YS and 294-YS barite used as calibration materials with respect to in-situ S isotopic ***,the new developed barite reference materials were used as matrix-matched standard to calibrate the barite samples from the Huayangchuan carbonatite-hosted U-polymetallic deposit(Qinling orogenic belt,western China)to obtainδ34S *** the temperaturedependentδ34S fractionation of barite-pyrite pair,we calculate the formation temperature of barite(i.e.,506 to 537°C)and theδ34S value of mineralizing fluid(i.e.,-7.11‰to-7.59‰)in the Huayangchuan *** results indicate an involvement of sedimentary sulfur,presumably acting as a potential uranium source(e.g.,upper crustal materials)for the giant Huayangchuan deposit.

Charge polarity inversion and zincophilicity improvement for chitosan separator towards durable aqueous zinc-ion batteries

Aqueous zinc-ion batteries encounter enormous challenges such as Zn dendrites and parasitic *** modification is a highly effective strategy to address these *** the advantages of low cost,nontoxicity,biodegradability,good film-forming ability,superior hydro phi licity,and rich functional groups,chitosan is an ideal matrix for constructing ***,the presence of positive charges within chitosan in weakly acidic electrolytes is unfavorable for dendrite ***,Schiff base reaction is introduced to modify chitosan matrix,transforming its charge polarity from positive to ***,NbN with excellent zincophilicity is coated onto chitosan matrix,forming a Janus separator with low thickness of 19 μm and considerably improved mechanical *** resultant separator can promote the transport of Zn2+ions while triggering a repulsive shielding effect against anions,therefore dramatically enhancing Zn2+ion transfer number from 0.28 to *** separator can also facilitate desolvation process,improve exchange current density,restrict two-dimensional Zn2+ion diffusion,and enhance electrochemical kinetics,contributing to significantly inhibited dendrite growth,by-product formation,and hydrogen ***,stable and reversible Zn stripping/plating process is enabled for over 2500 h at 2 ma cm-2and 2 mah *** great rate capability and excellent cyclability can be achieved for full batteries even under harsh *** work provides new insights into separator design for Zn-based batteries.
