Investigating the mechanism of action of Bu-Yang-Huan-Wu decoction in treating bleomycin-Induced pulmonary fibrosis through the epithelial-mesenchymal transition pathway

Background:To explore the effects and mechanisms of Bu-Yang-Huan-Wu Decoction on pulmonary fibrosis in ***:Forty-five C57BL/6J mice were randomly divided into three groups:Control,Model,and Bu-Yang-Huan-Wu *** fibrosis was elicited in mice through a solitary intratracheal administration of 2.5 mg/kg *** the control group,mice were given a solitary intratracheal administration of a comparable volume of *** began on the first day after the successful model establishment and lasted for 21 *** survival rate and body weight of the mice were recorded daily,and on the 22nd day,bronchoalveolar lavage fluid was collected to determine total cells and total *** wet/dry weight ratio of lung tissue and hydroxyproline were *** tissue pathology was observed using hematoxylin and eosin staining and Masson *** mRNA expression of epithelial-mesenchymal transition-related proteins(E-cadherin and vimentin)was detected by RT-qPCR,and their protein expression was analyzed by western ***:Compared to the model group,the Bu-Yang-Huan-Wu Decoction treatment notably enhanced both the survival rate and body weight in pulmonary fibrosis mice,significantly reduced lung tissue wet/dry weight ratio,total cells,and protein in bronchoalveolar lavage fluid,and hydroxyproline *** pathological morphology of lung tissue was significantly improved,with increased expression of the epithelial cell marker E-cadherin mRNA and protein,and decreased expression of the mesenchymal cell marker vimentin mRNA and ***:Bu-Yang-Huan-Wu Decoction can improve the degree of bleomycin-induced pulmonary fibrosis in mice by inhibiting epithelial-mesenchymal transition.

Myeloid-derived suppressor cells(MDSCs)are a group of heterogeneous immature cells with a strong immunosuppressive function in myeloid cells,which are impeded in the differentiation of myeloid cells under the pathological conditions of hypoxia,inflammation,infection,and *** individuals age,there is a significant increase in myeloid-derived suppressor cells(MDSCs),which subsequently enhance the immunosuppressive functions of Tregs(regulatory T cells)and Bregs(regulatory B cells).Therefore,MDSC may be related to immune system remodeling,thereby preventing excessive lesions caused by *** indicates that MDSC could serve as a potent inducer of immune *** senescence,characterized by immune dysfunction with aging,is closely linked to the onset of diseases like infections,pulmonary fibrosis,and *** achieve the purpose of anti-aging by intervening in immune aging and slow down the occurrence and development of related ***,understanding the biological characteristics of MDSC and its role in immune aging is crucial for immunotherapy targeting *** article reviews the different roles of MDSC in immune aging and its relationship with pulmonary fibrosis,tumor and other related diseases to provide theoretical basis for more comprehensive targeted MDSC immunotherapy.

Background:COVID-19 has had a dramatic impact on human health,economies,societies,and livelihoods around the *** Chinese medicine(TCM)formulae have played an important role in the prevention and treatment of *** evaluated the role of TCM in treating of COVID-19 and encouraged other countries to promote the use of TCM ***,the key is to find the basic core traditional Chinese medicine(BC-TCM)among those ***:For the first time,we mined the data of TCM formulae in CNIPA and analyzed herb characteristics and association *** then determined the BC-TCM and screened main compounds and therapeutic ***,the potential molecular mechanisms were explored by using enrichment analyses and molecular ***:This study screened 123 patented TCM formulae,including 312 *** to frequency statistics and association rules,nine herbs(Gan Cao,Jin Yinhua,Guang Huoxiang,Fu Ling,Huang Qi,Jie Geng,Lian Qiao,Cang Zhu,Ku Xingren)were selected as the *** BC-TCM involved 166 main compounds and 48 therapeutic *** active compounds Hederagenin,Spinasterol,Beta-sitosterol,and Liquiritin had high binding activity to the COVID-19 targets 3CL,ACE2,and core targets RELA,HSP90AA1,STAT3,MAPK3,and TP53 according to molecular docking ***,Hederagenin might be a potential compound for the prevention and treatment of ***:Our research predicted and confirmed the preventive therapeutic effect of BC-TCM on *** has the potential to broaden the scope of TCM,guide people in using clinical formulae,and provide valuable insights for future TCM discovery research.

Study on multi-timescale operation of hydrogen-containing energy coupled with transportation system

With the widespread attention on hydrogen energy, the operation optimization of the coupling system of hydrogen energy and transportation has become a critical issue. Therefore, this study develops a hydrogen-containing energy transportation coupling system for the system collaborative operation framework, operation mode and equipment modelling of the system. Furthermore, a medium- to long-term operation optimization model and a short-term operation optimization model were constructed considering the differences in the operation of coupled systems at different timescales and operating costs as the objective function and power balance and system equipment as constraints. In the medium- and long-term operation optimization model, the planning scenario reduction method is used to reduce the wind power generation scenario. In the short-term operation optimization model, a multivariate uncertainty model is constructed to represent the uncertainty in the coupling system. Subsequently, the solution method of the model is proposed. Finally, a coupled system is simulated to verify the effectiveness of the model. (i) When the initial scene set is 600, the typical scene reduction method using Latin hypercube sampling and the Wasserstein distance can reduce operating costs by 7.60% and 9.49% compared with K-means reduction and K-media reduction methods. (ii) The sensitivity coefficients of hydrogen sales price, electricity sales price and maintenance rate to operating costs are -0.031%, -1.009% and 0.0105%, respectively. (iii) Considering multiple uncertainties can help make optimal decisions based on the overall consideration of disadvantage scenarios, thereby reducing system operating costs. Medium-to-long-term and short-term optimization models are constructed for operation of a hydrogen-containing energy transportation system, considering operation of coupled systems at different timescales. Incremental cost analysis model of distribution network based on economic dispatch of distributed new-energy storage system

With the implementation of China's carbon-peaking and carbon-neutrality strategy, new energy will achieve leapfrog growth. Due to the good economics of distributed new-energy generation, it can not only save users' own investment, but also help to achieve local consumption of new energy. However, it will also bring about a series of incremental costs to the power grid. This paper first enumerates the concept, development status and scheduling mode of a distributed new-energy storage system. Based on the above, it establishes a new-energy power generation model and an energy storage system charging and discharging model, and proposes a global optimization scheduling model for a distributed new-energy storage system, considering the time-of-use electricity price and taking the lowest total operating cost of the distributed new-energy power generation system as the objective function. Finally, it proposes a distribution network incremental cost analysis model based on the penetration of distributed new energy. The calculation results show that the incremental cost of grid-connected distributed new energy is 1.0849, 1.2585 and 1.3473 yuan/kWh, respectively, which indicates that the global dispatching model can optimize the power consumption structure of a distributed power generation system, and has the function of peak shaving and valley filling, but the incremental cost of the distribution network will also increase. A new-energy power generation model and an energy storage system charging and discharging model use a global optimization scheduling method, considering the time-of-use electricity price and taking the lowest total operating cost as the objective function. The results show the incremental cost of grid-connected distributed new energy. Graphical Abstract

Objective:The aim of this study is to discover the possible working mechanisms of Ardisiae Japonicae Herba(AJH)on hepatoma carcinoma(HCC).Methods:In this study,ethanol extract of AJH was prepared and used to treat HCC cell in ***,a genomic wide RNA sequencing(RNA-seq)was performed to screen deregulated genes in HCC cells after the treatment of AJH *** gene and protein expression related to lipid metabolism in HCC cells were also investigated to validate the results obtained from ***:AJH extract could inhibit HCC cell proliferation in ***-seq analysis has identified 1,601 differentially expressed genes(DEGs,fold change≥2.0 or fold change≤0.5,P<0.05)in HCC after AJH extract treatment,which included 225 up-regulated genes and 1,376 down-regulated *** pathway analysis of DEGs demonstrated that lipid metabolism was a potential pathway related to AJH *** agreement with the RNA-seq data,q PCR and Western-blot analysis indicated that expression of genes and proteins related to lipid metabolism(SREBP1,ACC,ACLY and FASN)were significantly downregulated in AJH treatment group as compared with the control ***,AJH extract could also decrease lipid contents and cellular free fatty acid levels in HCC ***:Ethanol extract of AJH could inhibit HCC cell proliferation in vitro,the possible mechanism may be related to the inhibition of lipid metabolism.

Wettability and corrosion behavior between alkaline slag from sodium smelting of vanadium-titanium magnetite and refractory substrates

The sodium smelting of vanadium-titanium magnetite can achieve the comprehensive utilization of Fe,V,and ***,the generation of alkaline slag during this process may cause damage to refractory *** wettability and corrosion behavior of alkaline slag on three types of refractory(MgO-C,SiC,and high alumina refractory)substrates were investigated at temperatures up to 1200℃.The effects of duration on the wettability of molten slag on SiC substrates were also *** showed that the high alumina refractory exhibited better wettability with the molten slag than the others,and thus,it is easier to be *** results of scanning electron microscopy coupled with energy dispersive spectroscopy showed that MgO-C and high alumina refractory substrates were severely *** was a visible and regular interfacial reaction layer between the slag and SiC refractory substrate,which was produced by the redox reaction between the metal oxides in the slag and the SiC refractory *** the increase in holding time,the interface layer expands and silico-ferrite phases are generated at the *** redox reaction between Fe_(2)O_(3) and SiC substrate is the main reason for the *** comparing the differences in wettability and corrosion behavior between the alkaline slag from sodium smelting of vanadium-titanium magnetite and MgO-C,SiC and high alumina refractories,it is concluded that SiC refractory has good corrosion resistance to the *** oxides in the slag accelerate the oxidation rate of SiC refractory.
