Puerariae Radix protects against ulcerative colitis in mice by inhibiting NLRP3 inflammasome activation

Ulcerative colitis(UC)is a common inflammatory disease of the gastrointestinal *** Chinese medicine(TCM)has long been used in Asia as a treatment for UC and Puerariae Radix(PR)is a reliable anti-diarrheal *** aims of this study were to investigate the protective effect of PR using the dextran sulfate sodium salt(DSS)-induced UC model in mice and identify molecular mechanisms of PR *** chemical constituents of PR via ultra-performance liquid chromatography/tandem mass spectrometry and identified potential PR and UC targets using a network pharmacology(NP)approach were obtained to guide mouse experiments.A total of 180 peaks were identified from PR including 48 flavonoids,46 organic acids,14 amino acids,8 phenols,8 carbohydrates,7 alkaloids,6 coumarins and 43 other *** results showed that caspase-1 was the most dysregulated of the core genes associated with UC.A PR dose of 0.136 mg/g administered to DSS treated mice reversed weight loss and decreased colon lengths found in UC *** also alleviated intestinal mucosal shedding,inflammatory cell infiltration and mucin *** treatment suppressed upregulation of NOD-like receptor protein 3(NLRP3),cysteinyl aspartate-specific proteases-1(caspase-1),apoptosis-associated speck-like(ASC)and gasdermin D(GSDMD)at both the protein and m RNA expression *** addition of a small molecule dual-specificity phosphatase inhibitor NSC 95397 inhibited the positive effects of *** results indicated that PR exerts a protective effect on DSS-induced colitis by inhibiting NLRP3 inflammasome activation in mice.

Enhanced editing efficiency in Arabidopsis with a LbCas12a variant harboring D156R and E795L mutations

Cas12a(Cpf1),a Class 2 Type V CRISPR/Cas nuclease,has several unique attributes for genome editing and may provide a valuable alternative to ***,a low editing efficiency due to temperature sensitivity and insufficient cleavage activity of the Cas12a nuclease are major obstacles to its broad *** this report,we generated two variants,ttAsCas12 Ultra and ttLbCas12a Ultra harboring three(E174R,M537R,and F870L)or two(D156R and E795L)mutations,respectively,by combining the mutations from the temperature-tolerant variants ttAsCas12a(E174R)and ttLbCas12a(D156R),and those from the highly active variants AsCas12a Ultra(M537R and F870L)and LbCas12a Ultra(E795L).We compared editing efficiencies of the five resulting Cas12a variants(LbCas12a,ttLbCas12a,ttLbCas12a Ultra,AsCas12a Ultra,and ttAsCas12 Ultra)at six target sites of four genes in Arabidopsis(Arabidopsis thaliana).The variant ttLbCas12a Ultra,harboring the D156R and E795L mutations,exhibited the highest editing efficiency of all variants tested in Arabidopsis and can be used to generate homozygous or biallelic mutants in a single generation in Arabidopsis plants grown at 22 *** addition,optimization of ttLbCas12a Ultra,by varying nuclear localization signal sequences and codon usage,further greatly improved editing ***,our results indicate that ttLbCas12a Ultra is a valuable alternative to Cas9 for editing genes or promoters in Arabidopsis.

The mass balance of the Greenland Ice Sheet(GrIS)plays a crucial role in global sea level *** the 1960s,remote sensing missions have been providing extensive and continuous observation data for change monitoring of the *** this paper,we present our recent research results from remote sensing-based GrIS change ***,historical satellite data are processed and used to fill data gaps and are combined with existing partial maps,completing an ice velocity map of the GrIS from the 1960s to *** map provides valuable data for estimating the historical mass balance of ***,the monthly gravimetry-based mass balance of the GrIS from 2002 to 2020 is estimated by combining Gravity Recovery and Climate Experiment(GRACE)and GRACE Follow On(GRACE-FO)*** is found that the GrIS has lost a total mass of approximately 4443±75 Gt during this ***,based on Global Land Ice Measurements from Space(GLIMS),an updated Greenland glacier inventory is achieved utilizing data collected between 2006 and *** inventory provides more detailed and up-to-data glacier boundaries of ***,these advances provide essential data support for estimating the mass balance of the GrIS,contributing to the advancement of research on global sea level change.

In order to realize a low-frequency and small-size underwater acoustic source,an underwater acoustic transducer has been designed by using Ni Mn ga alloy, which has the advantages of large strain, rapid response and high energy density. Based on the deformation principle of Ni Mn ga alloy, a physical model of a Ni Mn ga longitudinal transducer is established, and the equivalentcircuit of the transducer is derived.A multi-physics coupling model of electromagnetic, mechanical and acoustic fields is established by using the finite element method to predict the underwater acoustic performance of the transducer.A small-scale prototype of the Ni Mn ga longitudinal transducer is fabricated and its sound source level is tested in water within the frequency range from 500 Hz to 800 Hz. Experimental results demonstrate that the Ni Mn ga transducer with an 8 mm diameter of the radiation surface, achieves sound source level of 115.5 d B at 700 Hz.

Traditional garnet solid electrolyte(Li_(7)La_(3)Zr_(2)O_(12))suffers from low room temperature ionic conductivity,poor air stability,high sintering temperature and energy *** the development prospects of high-entropy materials with high structural disorder and strong component controllability in the field of electrochemical energy storage,herein,a novel high-entropy garnet-type oxide solid electrolyte,Li_(5.75)ga_(0.25)La_(3)Zr_(0.5)Ti_(0.5)Sn_(0.5)Nb_(0.5)O_(12)(LGLZTSNO)was constructed by partially replacing the Li and Zr sites in Li_(7)La_(3)Zr_(2)O_(12)with ga and Ti/Sn/Nb elements,*** experimental and density functional theory(DFT)calculation results show that the high-entropy LGLZTSNO electrolyte has preferable room temperature ion conductivity,air stability,interface contact performance with lithium anode,and the ability to suppress lithium *** to the improvement of electrolyte performance,the critical current density of Li/Ag@LGLZTSNO/Li symmetric cell was increased from 0.42 to 1.57 mA cm^(−2),and the interface area specific impedance(IASR)was reduced from 765.2 to 42.3Ωcm^(2).Meanwhile,the Li/Ag@LGLZTSNO/LFP full cell also exhibits excellent rate performance and cycling performance(148 mA h g^(−1)at 0.1 C and 124 mA h g^(−1)at 0.5 C,capacity retention up to 84.8%after 100 cycles at 0.1 C),showing the application prospects of high-entropy LGLZTSNO solid electrolyte in high-performance all solid state lithium batteries.

Identification,characterization,and verification of miR399 target gene in grape

The microRNA miR399 plays an important role in phosphorus signal transduction pathways in ***,miR399 was shown to be closely associated with berry ripening in grape(Vitis vinifera).The objective of the present study was to elucidate the evolutionary characteristics of the miR399 gene family in grape and to verify the cleavage effect on the target *** miR399s were identified by miRNA sequencing and retrieval from the miRBase *** mature sequences and precursor sequences were subjected to phylogenetic analysis to reconstruct evolutionary trees,as well as secondary structure analysis of the precursor sequence,and prediction of target *** cisacting elements in the miR399 promoter were predicted and the cleavage effect of grape miR399b on its target genes was *** grape miR399 family comprised nine precursor sequences and nine mature *** precursor sequences formed a typical and stable stem—loop *** minimum folding free energy ranged from-55.70 kcal·mol^(-1)to-37.40 kcal·mol^(-1).Multiple sequence alignment revealed that the miR399 family was highly *** grape miR399 family was phylogenetically closely related to peach,apple,and citrus *** miR399s were predicted to target inorganic phosphate transporter 1—3,phospholipase D delta-like,and *** cleavage effect of grape miR399b on the target genes was verified by means of a dual-luciferase assay and 5’*** GUS staining showed that the promoter activity of miR399b was promoted by ga3treatment.

Ambient-Condition Strategy for Production of Hollow ga2O3@rGO Crystalline Nanostructures Toward Efficient Lithium Storage

Crystalline γ-ga2O3@rGO core-shell nanostructures are synthesized in gram scale,which are accomplished by a facile sonochemical strategy under ambient *** are composed of uniform γ-ga2O3nanospheres encapsulated by reduced graphene oxide（rGO） nanolayers,and their formation is mainly attributed to the existed opposite zeta potential between the ga2O3and *** as-constructed lithium-ion batteries（LIBs） based on as-fabricated γ-ga2O3@rGO nanostructures deliver an initial discharge capacity of 1000 mAh g-1at 100 mA g-1and reversible capacity of600 mAh g-1under 500 mA g-1after 1000 cycles,respectively,which are remarkably higher than those of pristine γ-ga2O3with a much reduced lifetime of 100 cycles and much lower *** situ XRD and XPS analyses demonstrate that the reversible LIBs storage is dominant by a conversion reaction and alloying mechanism,where the discharged product of liquid metal ga exhibits self-healing ability,thus preventing the destroy of ***,the rGO shell could act robustly as conductive network of the electrode for significantly improved conductivity,endowing the efficient Li storage *** work might provide some insight on mass production of advanced electrode materials under mild condition for energy storage and conversion applications.

Photothermal therapy(PTT)has been widely used in the treatment of tumors,but its efficacy is greatly limited by the inability of precise drug delivery and the increase of heat shock proteins(HSPs)caused by high *** article describes a therapeutic strategy to enhance PTT with starvation therapy in conjunction with ferroptosis mechanism.A nanoparticle platform ZIF-8@ga was constructed by wrapping together glucose oxidase(GOX)and gold nanospheres(AuNPs)loaded with dihydroartemisinin(DHA)with zeolitic imidazolate framework-8(ZIF-8).This platform can take advantage of the micro-environment of osteosarcoma(OS)cells,featuring low pH and high reactive oxygen species(ROS),for precision drug *** can effectively catalyze glucose to produce gluconic acid and H_(2)O_(2),and DHA can also induce ROS production in OS *** produced by GOX and DHA can further generate lipid peroxidation(LPO)and lead to ferroptosis of OS *** the same time,ROS and LPO produced can inhibit the expression of HSPs,thereby increasing the therapeutic effect of *** vitro experiments show that the nanoparticles are pH and ROS responsive.1μg/mL GOX combined with 0.2μg/mL DHA promotes ferroptosis of OS cells,and increases the killing effect of near-infrared(NIR)on OS *** in vivo experiments showed that the nano drug-delivery platform combined with PTT can effectively inhibit the growth of OS ***,this study provides a new idea for the treatment of OS with biomaterials combined with various treatment methods.
