BACKGrOUnD no guideline recommends antiviral therapy for hepatitis B e antigen(HBeAg)-positive chronic hepatitis B patients with persistently normal alanine amin.t.ansferase levels and a high hepatitis B virus(HBV)DnA viral *** to evaluate the feasibility and safety of a {2. herbal formula as a therapeutic option for chronic HBV *** In.t.tal,395 patients(30–65 years old)with confirmed HBeAg-positive chronic hepatitis B infection and persistently normal alanine amin.t.ansferase were randomized to receive either {2. herbal formula or placebo for 96 *** to evaluate therapeutic efficacy included:(1)HBV DnA levels decreased to less than 4 log10 IU/mL at weeks 48 and 96;and(2.HBeAg clearance and seroconversion rates at weeks 48 and *** HBV DnA levels≤4 log10 IU/mL were 10.05%at week 48 and 18.59%at week 96 in.t.e treatment *** HBeAg clearance and conversion rates were 8.54%and 8.04%at week 48 and 16.08%and 14.57%at week 96,***,HBV DnA levels≤4 log10 IU/mL were 2.55%and 2.55%at weeks 48 and 96,respectively,and the HBeAg clearance rates were 3.06%and 5.61%at weeks 48 and 96,respectively,in.t.e control *** quantitative hepatitis B surface antigen and HBeAg levels at baseline and changes during the treatment period as well as the alanine amin.t.ansferase elevation at weeks 12.and 2. were strong predictors of HBeAg *** High rates of HBV DnA reduction,HBeAg clearance and seroconversion could be achieved with {2. herbal formula treatments,and the treatments were relatively safe for HBeAg-positive chronic hepatitis B-infected patients with persistently normal alanine *** ability of the compound to modulate host immune fun.t.on probably contributed to this effect.

背景:课题组前期研究发现川芎嗪能明显降低骨关节炎关节液中nO、前列腺素e2.平,对减轻骨性关节炎的炎症有一定作用,但具体机制还不清楚。目的:观察川芎嗪治疗膝骨关节炎模型关节软骨中Ⅱ型胶原纤维α1基因、血管内皮生长因子mrnA及mir2.b表达水平,并探讨川芎嗪治疗早期膝骨关节炎大鼠的作用机制。方法:健康雄性SD大鼠随机分为正常组、模型组、川芎嗪高剂量组、川芎嗪低剂量组、阳性对照组,后4组通过木瓜蛋白酶关节腔注射制作早期膝骨关节炎大鼠模型。造模后川芎嗪高、低剂量组分别灌胃川芎嗪100 mg/kg和50 mg/kg,阳性对照组灌服塞来昔布2. mg/kg;正常组、模型组灌服等量生理盐水。干预6周后取材,分别行鼠软骨大体组织学评分、qr t-pCr检测软骨Ⅱ型胶原纤维α1基因、血管内皮生长因子mrnA及mir2.b的表达水平。结果与结论:(1)软骨Mankin评分:川芎嗪高、低剂量组和阳性对照组均较正常组高(pp2.川芎嗪各剂量组和阳性对照组的Ⅱ型胶原纤维α1基因、血管内皮生长因子mrnA表达较正常组上调(ppr2.b表达均较模型组上调(pr2.b从而抑制血管内皮生长因子mrnA的表达,促进软骨的修复。

目的探讨氯离子通道蛋白3(ClC-3)基因过表达对异丙肾上腺素(ISO)诱导的小鼠心肌肥厚的预防作用及机制。方法运用腺相关病毒9(AAV9)感染方法构建ClC-3过表达小鼠模型和原代心肌细胞模型,采用蛋白质印迹法与qrt-pCr检测小鼠心脏组织和原代心肌细胞中ClC-3的表达以确定模型是否建立成功。32.雄性C57BL/6小鼠随机分为4组,每组8只。对照组连续7 d腹腔注射生理盐水,ISO组连续7 d腹腔注射ISO 7.5 mg•kg^-1•d^-1,AAV9-bv+ISO组用空白载体AAV9病毒颗粒感染小鼠4周后连续7 d腹腔注射ISO 7.5 mg•kg^-1•d^-1,AAV9-ClC-3+ISO组用携带ClC-3基因的AAV9病毒颗粒感染小鼠4周后连续7 d腹腔注射ISO 7.5 mg•kg^-1•d^-1。采用心脏超声检查小鼠心功能变化,计算心脏体重指数,采用qrt-pCr检测心肌组织中心房钠尿肽(Anp)、脑钠肽(Bnp)的mrnA表达水平,H-e染色观察左心室形态学变化,苦味酸-天狼星红(pSr)染色观察心脏组织胶原纤维变化。取乳鼠摘出心脏,体外培养原代心肌细胞并分为4组,对照组未予任何干预,ISO组用0.1μmol/L ISO干预48 h,AAV9-ClC-3组用携带ClC-3基因的AAV9病毒颗粒感染细胞48 h,AAV9-ClC-3+ISO组用携带ClC-3基因的AAV9病毒颗粒和0.1μmol/L ISO共同干预48 h。采用芯片膜片钳技术检测小鼠原代心肌细胞的容积激活性氯电流(ICl,vol)。结果蛋白质印迹法和qrt-pCr检测结果均表明ClC-3过表达小鼠和原代心肌细胞模型成功建立。AAV9介导的ClC-3过表达能缓解ISO诱导的小鼠心脏体重指数、收缩末期室间隔厚度、收缩期左室后壁厚度和舒张期左室后壁厚度的增加,改善心脏组织形态学异常和心脏纤维化,减少心脏组织中Anp、Bnp mrnA表达的增加,并能体外抑制ISO导致的心肌细胞ICl,vol降低。结论ClC-3过表达可预防ISO诱导的小鼠心肌肥厚,其机制可能与心肌细胞ICl,vol激活有关。

Granulomatous lobular mastitis(GLM) is a rare and chronic benign inflammatory disease of the breast. Difficulties exist in.t.e management of GLM for many front-line surgeons and medical specialists who care for patients with inflammatory disorders of the breast. this consensus is summarized to establish evidence-based recommendations for the management of GLM. Literature was reviewed using pubMed from January 1, 1971 to July 31, 2.2.. Sixty-six intern.t.onal experienced multidisciplinary experts from 11 countries or regions were invited to review the *** of evidence were determined using the American College of physicians grading system, and recommendations were discussed until consensus. experts discussed and concluded 30 recommendations on historical defin.t.ons,etiology and predisposing factors, diagnosis criteria, treatment, clinical stages, relapse and recurrence of GLM. GLM was recommended as a widely accepted defin.t.on. In addition, this consensus introduced a new clinical stages and management algorithm for GLM to provide individual treatment strategies. In conclusion, diagnosis of GLM depends on a combin.t.on of history, clinical manifestations, imaging examin.t.ons, laboratory examin.t.ons and *** approach to treatment of GLM should be applied according to the different clinical stage of GLM. this evidencebased consensus would be valuable to assist front-line surgeons and medical specialists in.t.e optimal management of GLM.

the Jiangmen Underground neutrino Observatory(JUnO)features a 2. kt multi-purpose underground liquid scintillator sphere as its main *** of JUnO's features make it an excellent location for^8B solar neutrino measurements,such as its low-energy threshold,high energy resolution compared with water Cherenkov detectors,and much larger target mass compared with previous liquid scintillator *** this paper,we present a comprehensive assessment of JUnO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2.MeV threshold for the recoil electron energy is found to be achievable,assuming that the intrinsic radioactive background^(2.8)U and^(2.2.th in.t.e liquid scintillator can be controlled to 10^(-17)g/*** ten years of data acquisition,approximately 60,000 signal and 30,000 background events are *** large sample will enable an examin.t.on of the distortion of the recoil electron spectrum that is domin.t.d by the neutrino flavor transformation in.t.e dense solar matter,which will shed new light on.t.e inconsistency between.t.e measured electron spectra and the predictions of the standard three-flavor neutrino oscillation *** m^(2._(2.)=4.8times10^(-5);(7.5times10^(-5))eV^(2.,JUnO can provide evidence of neutrino oscillation in.t.e earth at approximately the 3sigma(2.igma)level by measuring the non-zero signal rate variation with respect to the solar zen.t. ***,JUnO can simultaneously measureDelta m^2.(2.)using^8B solar neutrinos to a precision of 2.% or better,depending on.t.e central value,and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between.t.e value of Delta m^2.(2.)reported by solar neutrino experiments and the KamLAnD experiment.

JUnO is a multi-purpose neutrino observatory under con.t.uction in.t.e {2. of *** publication presents new sensitivity estimates for the measurement of the △m_(31)^(2.,△m_(2.)^(2.,sin^(2.θ_(12.,and sin^(2.θ_(13) oscillation parameters using reactor antineutrinos,which is one of the primary physics goals of the *** sensitivities are obtained using the best knowledge available to date on.t.e location and overburden of the experimental site,the nuclear reactors in.t.e surrounding area and beyond,the detector response uncertainties,and the reactor antineutrino spectral shape con.t.aints expected from the tAO satellite *** is found that the △m_(2.)^(2. and sin^(2.θ_(12. oscillation parameters will be determined to 0.5%precision or better in six years of data *** the same period,the △m_(31)^(2. parameter will be determined to about 0.2.precision for each mass ordering *** new precision represents approximately an order of magn.t.de improvement over existing con.t.aints for these three parameters.

the Jiangmen Underground neutrino Observatory(JUnO)is a large liquid scintillator detector designed to explore many topics in fundamental *** this study,the potential of searching for proton decay in.t.e p→νK^(+)mode with JUnO is *** kaon and its decay particles feature a clear three-fold coincidence sign.t.re that results in a high efficiency for ***,the excellent energy resolution of JUnO permits suppression of the sizable background caused by other delayed *** on.t.ese advantages,the detection efficiency for the proton decay via p→νK^(+)is 36.9%±4.9%with a background level of 0.2.0.05(syst)±0.2.stat)events after 10 years of data *** estimated sensitivity based on 2.0 kton-years of exposure is 9.6×1033 years,which is competitive with the current best limits on.t.e proton lifetime in.t.is channel and complements the use of different detection.t.chnologies.
