Profiling of N6-methyladenosine methylation in porcine longissimus dorsi muscle and unravelling the hub gene ADIPOQ promotes adipogenesis in an m^(6)A-YTHDF1–dependent manner

Background Intramuscular fat(IMF)content is a critical indicator of pork quality,and abnormal IMF is also relevant to human disease as well as aging.Although N6-methyladenosine(m^(6)A)RNA modification was recently found to regulate adipogenesis in porcine intramuscular fat,however,the underlying molecular mechanisms was still unclear.Results In this work,we collected 20 longissimus dorsi muscle samples with high(average 3.95%)or low IMF content(average 1.22%)from a unique heterogenous swine population for m^(6)A sequencing(m^(6)A-seq).We discovered 70genes show both differential RNA expression and m^(6)A modification from high and low IMF group,including ADIPOQ and SFRP1,two hub genes inferred through gene co-expression analysis.Particularly,we observed ADIPOQ,which contains three m^(6)A modification sites within 3’untranslated and protein coding region,could promote porcine intramuscular preadipocyte differentiation in an m^(6)A-dependent manner.Furthermore,we found the YT521-B homology domain family protein 1(YTHDF1)could target and promote ADIPOQ mRNA translation.Conclusions Our study provided a comprehensive profiling of m^(6)A methylation in porcine longissimus dorsi muscle and characterized the involvement of m^(6)A epigenetic modification in the regulation of ADIPOQ mRNA on IMF deposition through an m^(6)A-YTHDF1-dependent manner.

A kidney-brain neural circuit drives progressive kidney damage and heart failure

Chronic kidney disease(CKD)and heart failure(HF)are highly prevalent,aggravate each other,and account for substantial mortality.However,the mechanisms underlying cardiorenal interaction and the role of kidney afferent nerves and their precise central pathway remain limited.Here,we combined virus tracing techniques with optogenetic techniques to map a polysynaptic central pathway linking kidney afferent nerves to subfornical organ(SFO)and thereby to paraventricular nucleus(PVN)and rostral ventrolateral medulla that modulates sympathetic outflow.This kidney-brain neural circuit was overactivated in mouse models of CKD or HF and subsequently enhanced the sympathetic discharge to both the kidney and the heart in each model.Interruption of the pathway by kidney deafferentation,selective deletion of angiotensin II type 1a receptor(AT1a)in SFO,or optogenetic silence of the kidney-SFO or SFO-PVN projection decreased the sympathetic discharge and lessened structural damage and dysfunction of both kidney and heart in models of CKD and HF.Thus,kidney afferent nerves activate a kidney-brain neural circuit in CKD and HF that drives the sympathetic nervous system to accelerate disease progression in both organs.These results demonstrate the crucial role of kidney afferent nerves and their central connections in engaging cardiorenal interactions under both physiological and disease conditions.This suggests novel therapies for CKD or HF targeting this kidney-brain neural circuit.

Sonic hedgehog signaling in kidney fibrosis: a master communicator

The hedgehog signaling cascade is an evolutionarily conserved pathway that regulates multiple aspects of embryonic development and plays a decisive role in tissue homeostasis. As the best studied member of three hedgehog ligands, sonic hedgehog(Shh) is known to be associated with kidney development and tissue repair after various insults. Recent studies uncover an intrinsic link between dysregulated Shh signaling and renal fibrogenesis. In various types of chronic kidney disease(CKD), Shh is upregulated specifically in renal tubular epithelium but targets interstitial fibroblasts, thereby mediating a dynamic epithelialmesenchymal communication(EMC). Tubule-derived Shh acts as a growth factor for interstitial fibroblasts and controls a hierarchy of fibrosis-related genes, which lead to the excessive deposition of extracellular matrix in renal interstitium. In this review, we recapitulate the principle of Shh signaling, its activation and regulation in a variety of kidney diseases. We also discuss the potential mechanisms by which Shh promotes renal fibrosis and assess the efficacy of blocking this signaling in preclinical settings. Continuing these lines of investigations will provide novel opportunities for designing effective therapies to improve CKD prognosis in patients.

Commercial genetically modified corn and soybean are poised following pilot planting in China

"Who will feed China?"This was a question raised by Lester Brown,the director of the World Watch Institute in the United States,in 1994.He predicted that China's grain production would decrease from 340 million tons in 1990 to 272 million tons by 2030,a 20%decrease,while China's population would reach 1.6 billion(Brown,1995).If the dietary structure was not improved,China would need to import 200-369 million tons of grain,equivalent to the global grain trade at the time.He projected the high possibility of a food crisis in China,which might further trigger a global food crisis.However,with the joint efforts of national agricultural researchers,represented by Yuan Longping,and the Chinese government,China's grain production has achieved 18 consecutive increases from 2004 to 2021.In 2022,grain production in China reached 633 million tons(NBS and Statistics,2022),while the population remained stable at 1.4 billion.As a result,China has achieved complete self-sufficiency in its staple food supply and has demonstrated to the international community through practical actions that the Chinese people can feed themselves.

Geographical patterns and determinants of insect biodiversity in China

Insects play important roles in the maintenance of ecosystem functioning and the provision of livelihoods for millions of people.However,compared with terrestrial vertebrates and angiosperms,such as the giant panda,crested ibis,and the metasequoia,insect conservation has not attracted enough attention,and a basic understanding of the geographical biodiversity patterns for major components of insects in China is lacking.Herein,we investigated the geographical distribution of insect biodiversity across multiple dimensions(taxonomic,genetic,and phylogenetic diversity)based on the spatial distribution and molecular DNA sequencing data of insects.Our analysis included 18 orders,360 families,5,275 genera,and 14,115 species of insects.The results revealed that Southwestern and Southeastern China harbored higher insect biodiversity and numerous older lineages,representing a museum,whereas regions located in Northwestern China harbored lower insect biodiversity and younger lineages,serving as an evolutionary cradle.We also observed that mean annual temperature and precipitation had significantly positive effects,whereas altitude had significantly negative effects on insect biodiversity in most cases.Moreover,cultivated vegetation harbored the highest insect taxonomic and phylogenetic diversity,and needleleaf and broadleaf mixed forests harbored the highest insect genetic diversity.These results indicated that human activities may positively contribute to insect spatial diversity on a regional scale.Our study fills a knowledge gap in insect spatial diversity in China.These findings could help guide national-level conservation plans and the post-2020 biodiversity conservation framework.

Wnt/β-catenin信号是一条进化上高度保守且极度复杂的发育信号通路,可调控胚胎发育、损伤修复以及多种人类疾病的发生、发展。大量证据表明,Wnt/β-catenin信号在肾脏早期发育中起关键作用,而在成年肾脏中则相对沉默。在多种肾脏病动物模型和人类肾脏疾病中它又被重新激活。急性肾损伤后Wnt/β-catenin的激活有助于受损肾小管的修复和再生。然而,该信号的持续激活与慢性肾脏疾病和肾纤维化的发生、发展密切相关。在本文中,我们对Wnt/β-catenin信号通路的组成及其转导机制,以及它在肾脏损伤后修复和纤维化过程中的作用进行较为系统的综述。深入阐明此信号通路的机制可能为将来防治肾脏病提供新的靶点和策略。

Influence of post-grown treatments on CuInS_2 thin films prepared by sulphurization of Cu-In films

Polycrystalline CuInS2 （CIS） films were prepared by sulphurization of Cu-In films. The surface morphology and phase composition of the as-grown film, the KCN-etched film, and the annealed KCN-etched film were investigated. During the sulphurization, the secondary CuxS phase segregated on the surface of the as-grown films. To improve the crystalline quality of CuInS2 films, a series of post-grown treatments, such as KCN-etching and vacuum annealing KCN-etched films, were performed on the as-grown films. Both as-grown and post-treated films were characterized by X-ray diffraction （XRD）, scanning electron microscopy （SEM）, and energy dispersive spectroscopy （EDS）. The results indicated that a CuxS secondary phase segregated on the surface of the as-grown film, which could be removed effectively by KCN etching. After the vacuum annealing treatment, the KCN-etched film had a sphalerite structure with （112） preferred orientation. Meanwhile, the crystalline quality of the CIS film was significantly improved, which provided a novel method to improve the performance of thin film solar cells.

针对目前我国高价值专利评估中存在评估方法科学性不足、定量指标较少及评估体系不完善等问题,以300件中国专利奖获奖专利为研究对象,探讨构建科学合理、实用性强的高价值专利评估指标体系和具体评价方法。首先提出高价值专利是高技术价值、高法律价值、高经济价值、高市场价值的综合体现。其次分别从技术因素、法律因素、经济因素、市场因素等4个层面挖掘高价值专利的典型性特征,构建专利价值评估指标体系和评估模型,并以江苏省获奖专利和普通专利作为测试样本验证模型的有效性,结果表明该模型能够满足对高价值专利评估的要求。最后针对高价值专利评估存在问题,就健全评估体系、增强评估意识与能力、打造评估服务平台等方面提出对策建议。

面对短视频版权侵权乱象,各界呼吁对短视频平台苛以强制性版权过滤义务以治理侵权,其理由是算法推荐的特殊性、平台具备过滤技术能力以及“价值差”的存在。然而,包含算法推荐的平台并不能一概认定为脱离技术中立,其在技术上亦难以准确识别合理使用与侵权使用,设置强制性过滤义务将侵蚀用户的表达自由。从域外发展来看,美国经听证后未设定过滤义务,欧盟虽曾设置但招致持续质疑,且最新立法已显示出矫正态度。当前,我国也不宜对短视频平台设定强制性过滤义务,而应优化现行规则:一是完善“避风港”规则并灵活适用;二是重申“红旗”标准以防“避风港”规则被滥用,从主观和客观方面细化平台构成“应知”的情形;三是探索长短视频版权许可及收益分配机制,实现多方主体的利益平衡。

STUDIES ON THE REGULATION OF MALIGNANT PHENOTYPE AND GENE EXPRESSION IN HUMAN PROMYELOCYTIC LEUKEMIA CELL MUTANT （HL-60-AR）*

The present study has investigated the regulation of malignant phenotype and gene expression in a human promyelocytic leukemia cell mutant （HL-60-AR） by means of cellular engineering technique of cybridization between the fusions of the mutant cells with enucleated mouse reticulocytes. Results indicate that the cybrid cells （HL-R） incorporated with reticulocyte cytoplasts become differentiated, and the malignancy is obviously suppressed or reversed to a certain degree when compared with those of parental tumor cells. They lose the growth ability to form colony soft agar medium, become non-tumorigenic under heterotransplantation to nude mice, and are accompanied by decrease in growth rate, cellular mitotic index and DNA synthesis. No gene transcripts or homologous sequence of mRNA corresponding to c-myc oncogene can be detected in cybrid cells by Northern blot technique with cloned cmyc gene probe, suggesting that the expression of originally active c-myc has been inhibited.
