Comprehensive transcriptome analysis based on RNA sequencing identifies critical genes for lipopolysaccharide-induced epididymitis in a rat model

Epididymitis is a comm only diagnosed disease associated with male ***,little is known about the molecules that are involved in its *** study was to identify critical genes associated with lipopolysaccharide-induced epididymitis and an a lyze the molecular mecha nism of epididymitis through RNA *** ntal epididymitis models were generated by administering male Sprague-Dawley rats'lipopolysaccharide.A total of 1378 differentially expressed genes,including 531 upregulated and 847 down regulated gen es,were identified in the epididymitis model rats compared with those in sham-operated rats by RNA seque *** ctional enrichme nt an a lyses suggested that the upregulated genes were markedly enriched in inflammati *** biological processes,as well as in the tumor necrosis factor(TNF)signaling pathway,cytokine-cytokine receptor interactions,complement and coagulation cascades,and in the chemokine *** downregulated genes(collagen type XXVⅢalpha 1 chain[Col28α1],cyclirndependent kinase-like 1[Cdkl1],phosphoserine phosphatase[Psph],and fatty acid desaturase 2[Fads2])and ten upregulated genes(CCAAT/enhancer-binding protein beta[Cebp0],C-X-C motif chemokine receptor 2[Cxcr2],interleukin 11[ll11],C-C motif chemokine ligand 20[Ccl20],nuclear factor-kappa-B inhibitor alpha[NfKbia],claudin 4[Cldn4],matrix metallopeptidase 9[Mmp9],heat shock 70 kDa protein 8[Hspa8],intercellular cell adhesion molecule-1[Icam1],and Jun)were successfully confirmed by real-time polymerase chai n *** n blot dem on strated that CDKL1 was decreased,while MMP9 and NFKBIA were increased in the experimental model group compared with those in the sham-operated *** study sheds new light on the understand!ng of the early response of the epididymis during bacterial epididymitis.

Change of intestinal mucosa barrier function in the progress of non-alcoholic steatohepatitis in rats

AIM: To explore the change of intestinal mucosa barrier function in the progress of non-alcoholic steatohepatitis (NASH) in rats. METHODS: Thirty-two Sprague-Dawley (SD) rats were randomly divided into control group and model group. Rats in the control group were given normal diet, and rats in the model group were given fat-rich diet. Eight rats in each group were killed at end of the 8th and 12th wk, respectively. The levels of endotoxin, D-xylose, TG, TC, ALT and AST, intestinal tissue SOD and MDA as well as intestinal mucus secretory IgA (sIgA) were measured. The pathology of liver was observed by HE staining. RESULTS: At end of the 8th wk, there was no marked difference in the levels of endotoxin, D-xylose and sIgA between the two groups. At end of the 12th wk, rats in the model group developed steatohepatitis and had a higher serum level of endotoxin (P = 0.01) and D-xylose (P = 0.00) and a lower serum level of sIgA (P = 0.007). CONCLUSION: Intestinal mucosa barrier malfunction may exist in NASH rats and may be an important promoter of NASH in rats.

Ten endo-a-substituted-bicyclo[2.2.2]octenyl(and octanyl)methanols were synthesized,their odors were evaluated. We have previously reported the syntheses and structure-odor relationship of two series of endo-bicyclo[2.2.1]heptenyl(and heptanyl)methanols^(1.2).In order to study the effect of the bicyclic skeleton on odor,ten of the title compounds have been synthesized.

＜正＞The problem of frequency limitation arising in calibration of the test fixtures is investigated in this paper. It is found that at some frequencies periodically,the accuracy of the methods becomes very low,and the denominators of the expressions of the required S-parameters approach *** conclusion can be drawn whetlier the test fixtures are symmetric or not.A good agreement between theory and experiment is obtained.

Prostate cancer(PCa) remains the most commonly diagnosed malignant tumor in men,and is the second highest cause of cancer mortality after lung tumors in the United States. Accumulating research indicates that micro RNAs(mi RNAs) are increasingly being implicated in PCa. mi RNAs are conserved small noncoding RNAs that control gene expression posttranscriptionally. Recent profiling research suggests that mi RNAs are aberrantly expressed in PCa,and these have been implicated in the regulation of apoptosis,cell cycle,epithelial to mesenchymal transition,PCa stem cells,and androgen receptor pathway. All of these might provide the basis for new pproaches for PCa. Here,we review current findings regarding mi RNA research in PCa to provide a strong basis for future study aimed at promising contributions of miRNA in PCa.

Hypoxia-preconditioned bone marrow-derived mesenchymal stem cells protect neurons from cardiac arrest-induced pyroptosis

Cardiac arrest can lead to severe neurological impairment as a result of inflammation,mitochondrial dysfunction,and post-cardiopulmonary resuscitation neurological *** preconditioning has been shown to improve migration and survival of bone marrow–derived mesenchymal stem cells and reduce pyroptosis after cardiac arrest,but the specific mechanisms by which hypoxia-preconditioned bone marrow–derived mesenchymal stem cells protect against brain injury after cardiac arrest are *** this end,we established an in vitro co-culture model of bone marrow–derived mesenchymal stem cells and oxygen–glucose deprived primary neurons and found that hypoxic preconditioning enhanced the protective effect of bone marrow stromal stem cells against neuronal pyroptosis,possibly through inhibition of the MAPK and nuclear factor κB ***,we transplanted hypoxia-preconditioned bone marrow–derived mesenchymal stem cells into the lateral ventricle after the return of spontaneous circulation in an 8-minute cardiac arrest rat model induced by *** results showed that hypoxia-preconditioned bone marrow–derived mesenchymal stem cells significantly reduced cardiac arrest–induced neuronal pyroptosis,oxidative stress,and mitochondrial damage,whereas knockdown of the liver isoform of phosphofructokinase in bone marrow–derived mesenchymal stem cells inhibited these *** conclude,hypoxia-preconditioned bone marrow–derived mesenchymal stem cells offer a promising therapeutic approach for neuronal injury following cardiac arrest,and their beneficial effects are potentially associated with increased expression of the liver isoform of phosphofructokinase following hypoxic preconditioning.

Boosting bidirectional sulfur conversion enabled by introducing boron-doped atoms and phosphorus vacancies in Ni2P for lithium-sulfur batteries

Lithium-sulfur （Li-S） batteries have gained great attention due to the high theoretical energy density and low cost,yet their further commercialization has been obstructed by the notorious shuttle effect and sluggish redox ***,we supply a strategy to optimize the electron structure of Ni2P by concurrently introducing B-doped atoms and P vacancies in Ni2P （Vp-B-Ni2P）,thereby enhancing the bidirectional sulfur *** study indicates that the simultaneous introduction of B-doped atoms and P vacancies in Ni2P causes the redistribution of electron around Ni atoms,bringing about the upward shift of d-band center of Ni atoms and effective d-p orbital hybridization between Ni atoms and sulfur species,thus strengthening the chemical anchoring for lithium polysulfides （LiPSs） as well as expediting the bidirectional conversion kinetics of sulfur ***,theoretical calculations reveal that the incorporation of B-doped atoms and P vacancies in Ni2P selectively promotes Li2S dissolution and nucleation ***,the Li-S batteries with Vp-B-Ni2P-separators present outstanding rate ability of 777 m A h g-1at 5 C and high areal capacity of 8.03 mA h cm-2under E/S of 5μL mg-1and sulfur loading of 7.20 mg *** work elucidates that introducing heteroatom and vacancy in metal phosphide collaboratively regulates the electron structure to accelerate bidirectional sulfur conversion.

Regulated cell death is a form of cell death that is actively controlled by *** studies have shown that regulated cell death plays a key role after spinal cord *** and ferroptosis are newly discovered types of regulated cell deaths that have been shown to exacerbate inflammation and lead to cell death in damaged spinal ***,a complex form of cell death that is interconnected with various regulated cell death mechanisms,has garnered significant attention in the study of spinal cord *** injury triggers not only cell death but also cellular survival *** signaling pathways play pivotal roles in influencing the processes of both deterioration and repair in spinal cord injury by regulating pyroptosis,ferroptosis,and ***,this review aims to comprehensively examine the mechanisms underlying regulated cell deaths,the signaling pathways that modulate these mechanisms,and the potential therapeutic targets for spinal cord *** analysis suggests that targeting the common regulatory signaling pathways of different regulated cell deaths could be a promising strategy to promote cell survival and enhance the repair of spinal cord ***,a holistic approach that incorporates multiple regulated cell deaths and their regulatory pathways presents a promising multi-target therapeutic strategy for the management of spinal cord injury.
