Loss of CDX2 expression is associated with poor prognosis in colorectal cancer patients

AIM:To investigate the clinicopathologic characteristics and prognostic implications associated with loss of CDX2 expression in colorectal cancers(CRCs).METHODS:We immunohistochemically evaluated CDX2 expression in 713 CRCs and paired our findings to clinicopathologic and molecular characteristics of each *** included cytokeratin 7 and CK20 expression,microsatellite instability,Cp G island methylator phenotype,and KRAS and BRAF mutation *** and multivariate survival analysis was performed to reveal the prognostic value of CDX2 ***:CDX2 expression was lost in 42(5.9%) ***,loss of CDX2 expression was associated with proximal location,infiltrative growth,advanced T,N,M and overall *** microscopic examination,loss of CDX2 expression was associated with poor differentiation,increased number of tumor-infiltrating lymphocytes,luminal serration and mucin *** of CDX2 expression was also associated with increased CK7 expression,decreased CK20 expression,Cp G island methylator phenotype,microsatellite instability and BRAF *** a univariate survival analysis,patients with loss of CDX2 expression showed worse overall survival(P < 0.001) and progression-free survival(P < 0.001).In a multivariate survival analysis,loss of CDX2 expression was an independent poor prognostic factor of overall survival [hazard ratio(HR) = 1.72,95%CI:1.04-2.85,P = 0.034] and progression-free survival(HR = 1.94,95%CI:1.22-3.07,P = 0.005).CONCLUSION:Loss of CDX2 expression is associated with aggressive clinical behavior and can be used as a prognostic marker in CRCs.

Bile-derived circulating extracellular mi R-30d-5p and mi R-92a-3p as potential biomarkers for cholangiocarcinoma

Background: Cholangiocarcinoma (CCA) is from cholangiocytes, and therefore bile is a potentially rich source of biomarkers for CCA. The aim of the study was to identify and validate microRNAs (miRNAs) in bile samples that are differentially expressed between benign biliary disease (BBD) and CCA. Methods: Bile samples from 106 patients with obstructive biliary disease were allocated consecutively to a discovery set (10 patients with BBD and 11 with CCA) and then a validation set (48 patients with BBD and 37 with CCA). An miRNA microarray platform was used to screen 1209 miRNAs in the discovery set. Quantitative real-time reverse transcription polymerase chain reaction (qRT-PCR) was used to validate the pro ling results in the discovery and validation sets. In addition, the levels of carbohydrate antigen 19-9 (CA19-9) and carcinoembryonic antigen (CEA) were determined from patient serum samples. Results: Microarray pro ling showed that miR-30d-5p and miR-92a-3p were signi cantly upregulated in bile from the CCA group compared with those from the BBD group. qRT-PCR results indicated that the expression levels of miR-30d-5p and of miR-92a-3p were signi cantly upregulated in the CCA group compared to the BBD group, validating the miRNA microarray results. Pathway analysis suggested that putative target genes of miR-30d-5p and of miR-92a-3p were involved in CCA-associated signalling path- ways, such as Hippo, Wnt, p53, MAPK, and EGFR. Receiver operating curve analysis revealed that the areas under the curve for bile miR-30d-5p, miR-92a-3p, serum CA19-9, and CEA were 0.730, 0.652, 0.675, and 0.603, respectively, and bile miR-30d-5p showed the best diagnostic performance with a sensitivity of 81.1% and a speci city of 60.5%. Conclusions: The levels of extracellular miR-30d-5p and miR-92a-3p in bile were signi cantly higher in patients with CCA than those in patients with BBD. Clinical significance of tumor-infiltrating lymphocytes for gastric cancer in the era of immunology

Immunotherapy has begun to revolutionize cancer treatment, by introducing therapies that target the host immune system instead of the tumor, therapies that possess unique adverse event profiles, and therapies that may cure certain types of cancer. The immune microenvironment of tumors is emerging as the most important means of understanding the relationship between a patient' immune system and their cancer, informing prognosis, and guiding immunotherapy, such as an antibody blockade of immune checkpoints. For some solid tumors, simple quantitation of lymphocyte infiltration would seem to have prognostic significance, suggesting that lymphocyte infiltration is not passive but may actively promote or inhibit tumor growth. For gastric cancers, several studies have provided strong evidence that immune cells contribute to determining prognosis. However, the exact role of immune cells in gastric cancer remains unclear. Therefore, this review focuses on the clinical significance of immune cells, especially tumor-infiltrating lymphocytes, in gastric cancer.

Influence of new town development on the urban heat island-The case of the Bundang area

Five new towns have been developed around the Seoul metropolitan area since 1996 However, these new towns generate lots of traffic and related problems in the areas including those new towns and Seoul as a result of increases in population and a lack of ecological-self-sufficiency. Currently, construction of another new town is under deliberation, and what should be a major consider is the notion that the new town be located within a wide, green zone. Many studies have revealed that green space can play an important role in improving urban eco-meteorological capability and air quality. In order to analyze the urban heat island which will be created by the new urban development, and to investigate the local thermal environment and its negative effects caused by a change of land use type and urbanization, Landsat TM images were used for extraction of urban surface temperature according to changes of land use over the last 15 years. These data are analyzed together with digital land use and topographic information. As a study result, it was found the urban heat island of the study area from 1985 to 1999 rapidly developed which showed a difference of mean temperature above +2 0 Before the Bundang new town construction the temperature of the residential area was the same as a forest, but during the new town construction in 1991 analysis revealed the creation of an urban heat island. The temperature of a forest whose size is over 50% of the investigation area was lowest, and thus the presence of a forest is believed to have a direct cooling effect on the urban environment and its surroundings. The mean temperature of the residential and commercial areas in the study was found to be +4 5 higher than the forest, and therefore this part of land use is believed to be the main factor causing the temperature increase of the urban heat island.

Annexin A10 expression in colorectal cancers with emphasis on the serrated neoplasia pathway

Toward Fast and Efficient Mobility in Aquatic Environment： A Robot with Compliant Swimming Appendages Inspired by a Water Beetle

Water beetles are proficient drag-powered swimmers, with oar-like legs. Inspired by this mechanism, here we propose a miniature robot, with mobility provided by a pair of legs with swimming appendages. The robot has optimized linkage structure to maximize the stroke angle, which is actuated by a single DC motor with a series of gears and a spring. A simplified swimming appendage model is proposed to calculate the deflection due to the applied drag force, and is compared with simulated data using COMSOL Multiphysies. Also, the swimming appendages are optimized by considering their locations on the legs using two fitness functions, and six different configurations are selected. We investigate the performance of the robot with various types of appendage using a high-speed camera, and motion capture cameras. The robot with the proposed configuration exhibits fast and efficient movement compared with other robots. In addition, the locomotion of the robot is analyzed by considering its dynamics, and compared with that of a water boatman （Corixidae）.
