Evaluation of the sperm DNA fragmentation index in infertile Japanese men by in-house flow cytometric analysis

Semen analysis has long been used to evaluate male fertility.Recently,several sperm function tests have been developed.Of those,the sperm DNA fragmentation index(DFI),which describes the status of the sperm DNA,is thought to be a suitable parameter for evaluating male fertility.However,there have been no large-scale studies on the sperm DFI of Japanese men.Therefore,we investigated the feasibility of using an in-house flow cytometry-based sperm DFI analysis based on the sperm DNA fragmentation test of sperm chromatin structure assay(SCSA)to assess male fertility in Japan.This study enrolled 743 infertile and 20 fertile Japanese men.To evaluate reproducibility,inter-and intraobserver precision was analyzed.A receiver operating characteristic curve analysis was used to set a cutoff value for the sperm DFI to identify men who could father children by timed intercourse or intrauterine insemination.The variability of the sperm DFI among fertile volunteers was determined.The relationship between semen parameters and the sperm DFI was assessed by Spearman’s rho test.A precision analysis revealed good reproducibility of the sperm DFI.The cutoff value of sperm DNA fragmentation in infertile men was 24.0%.Semen volume had no relationship with the sperm DFI.Sperm concentration,sperm motility,total motile sperm count,and percentage of normal-shaped sperm were significantly and negatively correlated with the sperm DFI.The median sperm DFI was smaller in fertile volunteers(7.7%)than that in infertile men(19.4%).Sperm DNA fragmentation analysis can be used to assess sperm functions that cannot be evaluated by ordinary semen analysis.

The azoospermia factor(AZF)region is important for spermatogenesis,and deletions within these regions are a common cause of oligozoospermia and azoospermia.Although several studies have reported this cause,the present research,to the best of our knowledge,is the first large-scale study assessing this factor in Japan.In this study,1030 male patients with infertility who were examined for Y chromosome microdeletion using the polymerase chain reaction-reverse sequence-specific oligonucleotide(PCR-rSSO)method,a newly developed method for Y chromosome microdeletion screening,were included.The study enrolled 250 patients with severe oligospermia and 717 patients with azoospermia.Among the 1030 patients,4,4,10,and 52 had AZFa,AZFb,AZFb+c,and AZFc deletions,respectively.The sperm recovery rate(SRR)of microdissection testicular sperm extraction in patients with AZFc deletions was significantly higher than that in those without AZF deletions(60.0%vs 28.7%,P=0.04).In patients with gr/gr deletion,SRR was 18.7%,which was lower than that in those without gr/gr deletion,but was not statistically significant.In conclusion,our study showed that the frequency of Y chromosome microdeletion in male patients in Japan was similar to that reported in patients from other countries,and SRR was higher in patients with AZFc deletion.

Non-improvement of atrophic gastritis in cases of gastric cancer after successful Helicobacter pylori eradication therapy

BACKGROUND Helicobacter pylori(H.pylori)infection is closely related to the development of gastric cancer(GC).However,GC can develop even after H.pylori eradication.Therefore,it would be extremely useful if GC could be predicted after eradication.The Kyoto classification score for gastritis(GA)is closely related to cancer risk.However,how the score for GC changes after eradication before onset is not well understood.AIM To investigate the characteristics of the progression of Kyoto classification scores for GC after H.pylori eradication.METHODS Eradication of H.pylori was confirmed in all patients using either the urea breath test or the stool antigen test.The Kyoto classification score of GC patients was evaluated by endoscopy at the time of event onset and three years earlier.In ad-dition,the modified atrophy score was evaluated and compared between the GC group and the control GA group.RESULTS In total,30 cases of early GC and 30 cases of chronic GA were evaluated.The pathology of the cancer cases was differentiated adenocarcinoma,except for one case of undifferentiated adenocarcinoma.The total score of the Kyoto classifi-cation was significantly higher in the GC group both at the time of cancer onset and three years earlier(4.97 vs 3.73,P=0.0034;4.2 vs 3.1,P=0.0035,respectively).The modified atrophy score was significantly higher in the GC group both at the time of cancer onset and three years earlier and was significantly improved only in the GA group(5.3 vs 5.3,P=0.5;3.73 vs 3.1,P=0.0475,respectively).CONCLUSION The course of the modified atrophy score is useful for predicting the onset of GC after eradication.Patients with severe atrophy after H.pylori eradication require careful monitoring.

Effect of Seasonal Variations on the Behavior of Flexible Pavements in Burkina Faso: Towards Alternating and Periodic Loading of Multi-Axle Heavy Goods Vehicles for Road Durability

Bituminous materials are heat-sensitive, and their mechanical properties vary with temperature. This variation in properties is not without consequences on the performance of flexible road structures under the repeated passage of multi-axles. This study determines the influence of seasonal variations on the rate of permanent deformation, the rut depth of flexible pavements and the effect of alternating loading of heavy goods vehicles following the temperature variations on the durability of roads. Thus, an ambient and pavement surface temperature measurement was carried out in 2022. The temperature profile at different layers of the modelled pavement, the evaluation of deformation rates and rutting depth were determined using several models. The results show that the permanent deformation and rutting rates are higher at the level of the bituminous concrete layer than at the level of the asphalt gravel layer because the stresses decrease from the surface to the depth of the pavement. On the other hand, the variations in these rates, permanent deformations and ruts between the hot and so-called cold periods are more pronounced in the bitumen gravel than in bituminous concrete, showing that gravel bitumen is more sensitive to temperature variations than bituminous concrete despite its higher rigidity. Of these results, we suggested a periodic and alternating loading of the different types of heavy goods vehicles. These loads consist of fully applying the WAEMU standards with a tolerance of 15% during periods of high and low temperatures. This regulation has increased 2 to 3 times in the durability of roadways depending on the type of heavy goods vehicle.

Graphical representation of DNA sequences is one of the most popular techniques for alignment-free sequence comparison. Here, we propose a new method for the feature extraction of DNA sequences represented by binary images, by estimating the similarity between DNA sequences using the frequency histograms of local bitmap patterns of images. Our method shows linear time complexity for the length of DNA sequences, which is practical even when long sequences, such as whole genome sequences, are compared. We tested five distance measures for the estimation of sequence similarities, and found that the histogram intersection and Manhattan distance are the most appropriate ones for phylogenetic analyses.

Progress of ulcerative colitis patients during the COVID-19 pandemic

BACKGROUND We have previously demonstrated that the first wave of the coronavirus disease 2019(COVID-19)pandemic caused exacerbations in ulcerative colitis(UC)patients,probably through psychological and physical stress.However,successive waves of the COVID-19 pandemic continuously followed the first.The effects of this chronic stress on the disease condition in UC patients are of interest.AIM To clarify the effect of chronic stress from COVID-19 on disease condition in patients aggravated after the first wave.METHODS Our previous study investigated 289 consecutive UC outpatients treated in one center during March and April 2020,the period of the first wave of the COVID-19 pandemic.In this study,an identical group of 289 UC patients was evaluated using UC-disease activity index(UC-DAI),endoscopic mucosal appearance score,and Matts pathological grade scoring.RESULTS Of the 289 UC patients included in the study in 2020,10 patients dropped out as of 2021 and another 11 patients dropped out as of 2022,making three groups for 2020,2021 and 2022.No significant differences in characteristics were found among the three groups.UC-DAI scores had aggravated during the period of the first wave of the COVID-19 pandemic,but significantly recovered in 2021 and remained stable in 2022.Matts grade scores significantly recovered in 2021 from those in 2020 and remained stable in 2022.CONCLUSION Disease activity of UC patients recovered in 2021 and remained stable in 2022,aggravated by the stress of the first wave of COVID-19 in 2020 despite persistence of the pandemic.

In dentistry,orthodontic root resorption is a long-lasting issue with no effective treatment strategy,and its mechanisms,especially those related to senescent cells,remain largely unknown.Here,we used an orthodontic intrusion tooth movement model with an L-loop in rats to demonstrate that mechanical stress-induced senescent cells aggravate apical root resorption,which was prevented by administering senolytics(a dasatinib and quercetin cocktail).

The recent advancement in lithium-niobite-on-insulator(LNOI)technology is opening up new opportunities in optoelectronics,as devices with better performance,lower power consumption and a smaller footprint can be realised due to the high optical confinement in the structures.The LNOI platform offers both largeχ(2)andχ(3)nonlinearities along with the power of dispersion engineering,enabling brand new nonlinear photonic devices and applications for the next generation of integrated photonic circuits.However,Raman scattering and its interaction with other nonlinear processes have not been extensively studied in dispersion-engineered LNOI nanodevices.In this work,we characterise the Raman radiation spectra in a monolithic lithium niobate(LN)microresonator via selective excitation of Raman-active phonon modes.The dominant mode for the Raman oscillation is observed in the backward direction for a continuous-wave pump threshold power of 20mW with a high differential quantum efficiency of 46%.We explore the effects of Raman scattering on Kerr optical frequency comb generation.We achieve mode-locked states in an X-cut LNOI chip through sufficient suppression of the Raman effect via cavity geometry control.Our analysis of the Raman effect provides guidance for the development of future chip-based photonic devices on the LNOI platform.

Impact of Multi-Axle Vehicles and Road Overloads on the Durability of Asphalt Pavements

The multiplication of heavy vehicle axles and road overloads are phenomena that are becoming increasingly important on the road network of the WAEMU community. These phenomena, although framed by standards, have an impact on the durability of pavements. In this manuscript it is a question of evaluating the life of the road under the effect of traffic of these multi-axle vehicles and the different tolerances of overloads observed on the road network. To achieve this, a modeling of a bituminous pavement was made with the software ALIZE Lcpc Version 231 based on the principle of the French method of sizing. An inventory of multi-axle heavy goods vehicles was also made on a road with a weighing station. This traffic counting made it possible to classify heavy goods vehicles into three categories, namely: 1) trucks, 2) dual-wheeled semi-trailers and 3) single-wheeled semi-trailers. The results obtained show that in terms of aggressiveness, single-wheeled semi-trailers are the most aggressive, followed by heavy goods vehicles in the category of trucks with more than five axles and semi-trailers with dual wheels with more than seven axles. The durability of the road depends on the aggressiveness of heavy goods vehicles, it was found that the tolerance threshold for overloads of 15% of the total permissible rolling weight (TARW) or the total permissible laden weight (TALW) currently granted in the Community area needs to be reviewed. For durable road surfaces, this tolerance may only be allowed for heavy goods vehicles of type P11, P12 and P13. The 5% tolerance can be applied to all vehicles except heavy goods vehicles with single wheels.

CONSPECTUS:“Dynamic exciton”,an umbrella term concept in photochemistry,plays an important role in nature,science,and technology,especially in photoinduced and electrically induced electron transfer(ET)between a donor(D)and an acceptor(A).Typically,an exciton in molecular D−A systems is considered a locally excited(LE)state of a donor(D*)or an acceptor(A*)molecule,but let us extend the terminology of“exciton”to an integrated class of LE,charge-transfer(CT),and charge-separated(CS)states.The degree of CT(0−100%),spin multiplicity,and D−A interaction(i.e.,electronic coupling)are pivotal factors in the“exciton”.Another important aspect of the“exciton”is strongly related to the“dynamic”aspect of the“exciton”by movement of atomic nuclei(i.e.,vibration,rotation,and fluctuation)and their collective motions controlling behaviors of electrons and spins by the passage of time.The concept of“dynamic exciton”should cover a wide variety of the photochemical phenomena that are all essential for the energy conversion devices and processes including various kinds of living systems.In these,a huge amount of the nuclear motional modes may cooperatively be entangled to electronic orbitals.Thus,the idea behind“dynamic exciton”includes usage of the cooperation between nuclear motions and the spin−orbital,as electron−phonon coupling for innovative designs of the energy conversion materials and assemblies.For this,it is particularly important to examine how this wide variety of the motions plays roles on electronic couplings,intermediate geometries and mobilities,exciton energies,CT characters,and so on.We draw researchers’attention to this aspect of the vibronic effect,like the entropy role by collective movement of side chains of the conjugated polymer to modulate the electronic coupling time-dependently at the charge delocalization and dissociation,which is reinforced three-dimensionally at the D/A domain interface of organic photovoltaics(OPVs).In addition,to realize the ultimate high-performance OPVs,the voltage loss must be dimin
