Ig SF11 regulates osteoclast differentiation through association with the scaffold protein PSD-95

Osteoclasts are multinucleated, giant cells derived from myeloid progenitors. While receptor activator of NF-κB ligand(RANKL)stimulation is the primary driver of osteoclast differentiation, additional signaling further contributes to osteoclast ***, we demonstrate that immunoglobulin superfamily member 11(Ig SF11), whose expression increases during osteoclast differentiation, regulates osteoclast differentiation through interaction with postsynaptic density protein 95(PSD-95), a scaffold protein with multiple protein interaction domains. Ig SF11 deficiency in vivo results in impaired osteoclast differentiation and bone resorption but no observed defect in bone formation. Consequently, Ig SF11-deficient mice exhibit increased bone *** in vitro osteoclast culture systems, we show that Ig SF11 functions through homophilic interactions. Additionally, we demonstrate that impaired osteoclast differentiation in Ig SF11-deficient cells is rescued by full-length Ig SF11 and that the Ig SF11-PSD-95 interaction requires the 75 C-terminal amino acids of Ig SF11. Our findings reveal a critical role for Ig SF11 during osteoclast differentiation and suggest a role for Ig SF11 in a receptor-and signal transduction molecule-containing protein complex.

To investigate the association between polymorphisms (SNP) in the p53 and murine double minute 2 homolog (MDM2) promoter 309 in cervical carcinogenesis. SNP at p53 codon 72 polymorphisms and MDM2 promoter 309 (T/G) together with human papillomavirus (HPV) types were examined in a total of 187 cervical smear samples using real time PCR. 27 cases with HPV types 16 and/or 18 had significantly higher frequency of the TG + GG genotype and G allele than 56 with other types of high-risk HPV (P = 0.0136). 48 cases with HPV types 52 and/or 58 had significantly higher frequency of the TG + GG genotype and G allele than 56 with other types of high-risk HPV (P = 0.001). Our studies have demonstrated that the frequency of G allele in MDM2 promoter 309 increased from LSIL to HSIL and that there was an increased OR for G allele in HSIL cases with high-risk HPV types including 52 and 58. It is known that geographically different oncogenic HPV types 52 and 58 are more prevalent than 16 and 18 in a Japanese population.

Using a rabbit model, we examined N epsilon-(hexanoyl) lysine (HEL) levels in bone and urine to detect when peroxidative reaction first occurs after steroid administration. Japanese white rabbits weighing about 3.5 kg each were injected with a single intramuscular dose of methylprednisolone 40 mg/kg and divided into groups consisting of 10 rabbits each, which were killed after 1, 3, 5 and 14 days (groups A, B, C and D respectively). As a control, 10 untreated rabbits (group N) were also studied. The proximal femurs were examined histopathologically and immunohistochemically using monoclonal antibody HEL, which is a highly specific antibody against N epsilon-(hexanoyl) lysine, an early peroxidation marker. In addition, urinary levels of HEL were measured by enzyme-linked immunosorbent assay in group N, A, B and C. Osteonecrosis was detected only in group D (90%). Increase of positive reaction of HEL in the bone was observed in group A and D. HEL expression in group D was judged to be a secondary reaction resulting from the development of osteonecrosis. Urinary level of HEL showed a significant increase in only group A (P < 0.001). The present findings suggest that peroxidation in bone occurred within 24 hours after steroid administration in a rabbit model and that it is possible to noninvasively grasp the timing of this peroxidative reaction by measuring the urinary level of HEL.

Leaching Phenomena of Antifouling Agents from Ships＇ Hull Paints

The effects ofpH, dissolved ion content and relative water velocity on the release rate of an antifouling agent, cuprous oxide, from ships＇ hull paint have been investigated by rotating cylinder tests. Additionally, test paint panels were attached to a vessel and recovered after a certain period of voyage for the validation of the laboratory tests. In the initial period, the release rates are influenced by pH, dissolved ion content and water velocity, but once after a certain period of test, those effects become less significant. These phenomena can be explained when the paint film is fresh, the rate is controlled by chemical reaction, the surface and/or diffusion layer in the water phase governs the rate. After the antifouling substance in the paint film leached out from the near-surface region, a diffused layer （leached layer）, that has little antifouling agent remained, is formed at the surface of the coating, and the diffusion in that layer can be a rate-determining process. The development of the leached layer is affected by a balance between the leaching rate of the antifouling ingredient and paint resin determined by the chemical properties and speed of the water. Thus, the leaching rates of antifouling agents are affected by the history of the paint in the water.

This study was undertaken to examine which factors contributed to the correction of crowding in two patients who underwent nonextraction orthodontic treatment. A study model analysis was conducted to determine the effects of the orthodontic treatment for crowding with high canines on crown angulation and dental arch width in two patients. The results showed that the crown angulation was significantly increased, indicating distal tipping in the maxillary dental arch. This tendency was most commonly observed in the premolars among the lateral teeth. With respect to the dental arch width, the largest change was evident in the first molar and first premolar regions in cases 1 and 2, respectively. On the basis of these results, up-righting of mesially tipped lateral teeth and expansion of narrow dental arches could prove to be the keys to the success of space regaining or correction of high canines and mild crowding.

Outcomes of endoscopic endonasal dacryocysto-rhinostomy for intractable lacrimal dacryostenosis and associated factors

AIM：To examine the effects of patient age,canalicular obstruction,mode of anesthesia,and duration of nasolacrimal intubation on the outcomes of endoscopic endonasal dacryocystorhinostomy（DCR）. METHODS： Totally 56 eyes of 46 patients with prolonged epiphora underwent minimally invasive endoscopic endonasal DCR.A successful surgical outcome was defined as a significant improvement in symptoms,adequate water passage from the puncta to the nasal cavity,and patency of the DCR *** outcomes were assessed at least 6mo after ***＇s exact test was used to discuss the factors,and then the logistic regression analysis was made by SAS9.4 ***： The overall success rate was 75.0%,and complete resolution was observed in 27 *** success rate was higher for patients with ≥6mo intubation than for those with 〈6mo ***,there were no significant differences in outcomes between groups stratified by age（〈65 or ≥65y）,presence or absence of canalicular obstruction,mode of anesthesia（local or general）,and use or nonuse of a radiowave *** patient developed subcutaneous emphysema around the eye and nose and one developed subcutaneous hemorrhage after surgery. CONCLUSION： Endoscopic endonasal DCR can be considered safe and minimally invasive with reasonable success rates,particularly when the duration of nasolacrimal intubation is ≥6mo.

In the field of Second Language Acquisition （SLA）, motivation has played a crucial role for language learning and teaching. Just as motivation is important in SLA, demotivation constitutes an important factor for both students and teachers. In our previous study, investigating 97 teachers for demotivational factors, the teachers felt demotivated to see students＇ lack of interest and bad attitudes, as well as dissatisfaction with curriculum, teaching material and poor facilities. We concluded further investigation was necessary as to how teachers regain motivation and hopefully toward directed motivational current （DMC）, which not only provides a direction for action, but it also energizes action （D6myei et al, 2016）. In our present study, compared to our previous study which was analyzed quantitatively, we have collected qualitative data from some 34 college teachers investigating when they felt demotivated, what they did to cope with it, and how they regained their motivation. Data was collected, analyzed qualitatively, and factors which may lead to DMC was extracted. It is hoped that this study will be beneficial for facilitating better SLA learning and teaching, and also for teacher training.

Coronary spasm: It's common, but it's still unsolved

＜正＞FRP are getting more generally known as a viable alternative to conventional steel reinforcement and has been widely accepted as a convenient material for upgrading existing civil infrastructures. Japan has been a leading country in terms of the number of practical applications and the amount of FRP composites （or continuous fiber reinforcement） used for infrastructures since later 1980s. However, Japanese material manufacturers, constructors, and research units are confronting a challenge how to make FRP composites play a leading role for building up a sustainable infrastructure *** paper provides a state-of-the-art review on the use of FRP composites for the infrastructures in Japan. The contents include: development and markets of FRP composites, development of codes, standard applications of FRP, and current barriers for further uptake of FRP. Moreover, this paper outlooks the future potential roles of FRP composites as the sustainability becomes a major concern for our infrastructure system, which calls the need of advanced construction materials having effective cost, long life-span, less maintenance, less environmental impact, innovative functions, and easiness for the purpose of managing social assets. Efforts are suggested to put into verifying the overall advantages of FRP related to all these aspects.

Coronary spasm is caused by a transient coronary nar -rowing due to the constriction of epicardial coronary artery, which leads to myocardial ischemia. More than 50 years have passed since the first recognition of coronary spasm, and many findings on coronary spasm have been reported. Coronary spasm has been considered as having pivotal roles in the cause of not only rest angina but also exertional angina, acute coronary syndrome, and heart failure. In addition, several new findings of the mechanism of coronary spasm have emerged recently. The diagnosis based mainly on coronary angiography and spasm provo-cation test and the mainstream treatment with a focus on a calcium-channel blocker have been established. At a glance, coronary spasm or vasospastic angina(VSA) has become a common disease. On the contrary, there are several uncertain or unsolved problems regarding coronary spasm, including the presence of medically refractory coronary spasm(intractable VSA), or an appropriate use of implantable cardioverter defibrillator in patients with cardiac arrest who have been confirmed as having coronary spasm. This editorial focused on coronary spasm, including recent topics and unsolved problems.
