Uhwang Chungsim Won Decreases Blood Oxygen Level-Dependent fMRI Signal Response to A Motor Stimulation Task

Objectives： TO evaluate the effect of Uhwang Chungsim Won [UC, Niuhuang Qingxin Pill （牛黄清心丸] on the blood oxygen level-dependent （BOLD） functional magnetic resonance imaging （fMRI） signal response to a motor stimulation task using a hand grasping paradigm. Methods： Ten healthy right-handed male volunteers were recruited. Their BOLD fMRI signal characteristics during hand grasping were determined before and 1 h after consuming UC. Results： With the administration of UC, BOLD responses to the right hand motor task showed a decrease in both hemispheres including the right cerebellar culmen, right and left medial frontal gyrus, right parahippocampal gyrus, right and left sublobar insula, right and left posterior cingulate, left precuneus, and right precentral gyrus. Decreased BOLD responses to the left hand motor task were shown in the right lingual gyrus, right cerebellar culmen, left superior temporal gyrus, right and left middle frontal gyrus, and right precentral gyrus after UC administration. Increased BOLD responses to the left hand motor task were shown in the left and right posterior cingulate, while there was no increased response to the right hand motor task after UC administration. Conclusions： UC administration significantly decreased fMRI BOLD signal responses to hand grasping in most of the brain areas including the primary motor cortex, but it is not clear whether these decreased responses were caused by a decrease in neuronal activities or by an increase in cerebral blood flow due to the cerebral vasodilatory effects of UC. Further studies using positron emission tomography or single photon emission computed tomography are needed to fully elucidate these findings.

Pharmacological advantages of melatonin in immunosenescence by improving activity of T lymphocytes

Melatonin plays a critical role in regulating photoperiodic signals and has recently been shown to decrease immunosenescence with age. In this study, we examined whether melatonin activates T lymphocytes as major adaptive immune cells in in vitro and in vivo models. Splenocytes, CD4＋, and naive CD4 T lymphocytes were isolated from the spleen of BALB/c mice and the cell population patterns and mRNA profiles associated with T cell activation （CD28 and p21） and the melatonin receptor （MT1A and MT1B） were assessed. The T cell activation- related proteins Ki67 and Bcl2 were also evaluated to confirm the relationship between gene and protein levels. Our data clearly revealed that CD28, p21, MT 1 A, and MT 1B mRNA were highly expressed in the presence of melatonin. Co-culture of CD4＋ T lymphocyte and peritoneal macrophage 7 days after melatonin administration to young and aged mice significantly increased APRIL mRNA, suggesting induction or maintenance of T lymphocyte responses. We also found that the intracellular amount of Ki67 and Bcl2 proteins were significantly upregulated in aged CD4＋ T lymphocytes, suggesting enhancing T cell proliferation and ling-term maintenance of memory T cells. Taken together, we conclude that melatonin supplementation may enhance immunity in aged individuals by upregulating immunosenescence indices in association with T lymphocytes and may be an attractive pharmacological candidate for aged and immunocompromised individuals.

Influence of glass-based dental ceramic type and thickness with identical shade on the light transmittance and the degree of conversion of resin cement

The purpose of this study was to assess the influence of the types and thicknesses of glass ceramic plates on light transmittance and compare the degrees of conversion(DC) of resin cement under the ceramic materials. Three ceramic plates with thicknesses of 0.5, 1.0, 2.0, and 4.0 mm were fabricated from each of five commercial ceramic blocks in shade A2: high-translucency and low-translucency IPS Empress CAD(EmpHT and EmpLT); high-translucency and low-translucency IPS *** CAD(EmxHT and EmxLT); and Vita Mark Ⅱ(Vita). The translucency parameter was obtained using a colorimeter. The light transmittance rate was measured using a photodetector attached to an optical power meter. The DC of a resin cement(Variolink N) underneath the ceramic plates was examined by Fourier transform infrared spectroscopy. The translucency parameter, light transmittance rate, and DC showed significant differences by ceramic type and thickness(P mission and DCs, and the EmxLT showed the least light transmission and the lowest DCs. The high-translucency Empress showed significantly higher DCs than the low-translucency types(P 0.05). Both type and thickness of the glass ceramics significantly influenced the light transmittance and DC of the light-cured resin cement beneath the ceramic of the same shade.

Neuroprotective Effect of Chunghyuldan(Qing Xue Dan)on Hypoxia-Reoxygenation Induced Damage of Neuroblastoma 2a Cell Lines

Objectives： Chunghyuldan （CHD）, a combinatorial drug that has anti-hyperlipidemic and anti inflammatory activities, has been shown to reduce infarct volume in a focal ischemia-reperfusion rat model. To explore the molecular basis of CHD＇s neuroprotective effect, we examined whether CHD shows a cell-protective activity and has a regulatory effect on Bax and/or B-cell leukemia/lymphoma 2 （Bcl-2） expression in mouse neuroblastoma 2a （N2a） cells subjected to hypoxia-reoxygenation （H/R）. Methods： In order to evaluate the effects of CHD on the cytotoxicity induced from hypoxia or H/R condition, lactate dehydrogenase （LDH） assay was performed. To explore whether the suppression of neural damage when pre-treated with CHD is associated with its anti-apoptotic effect, the CHD effect on the expression of Bcl-2 and Bax was analyzed by Western blotting analysis. Results： Cytotoxicity of N2a cell line was slightly increased in 42 h hypoxia condition and dramatically increased under the H/R condition. CHD treatment markedly decreased the cytotoxicity in both conditions （P〈0.01, P〈0.05）. H/R markedly increased the expression of the pro-apoptotic protein, Bax, but slightly increased the expression of the anti-apoptotic protein, Bcl-2, compared with the normoxia or hypoxia group. CHD significantly decreased Bax expression （P〈0.01） and slightly decreased Bcl-2 expression （P〉0.05）, resulted in a reduction of Bax/Bcl-2 ratio in N2a cells subjected to H/R. Conclusion： CHD has neuroprotective effect in N2a cells subjected to H/R, which might be derived at least in part from its ability to decrease the expression of the pro-apoptotic protein, Bax.

Oxygen redox is considered a new paradigm for increasing the practical capacity and energy density of the layered oxide cathodes for Na-ion batteries. However, severe local structural changes and phase transitions during anionic redox reactions lead to poor electrochemical performance with sluggish ***, we propose a synergy of Li-Cu cations in harnessing the full potential of oxygen redox, through Li displacement and suppressed phase transition in P3-type layered oxide cathode. P3-type Na_(0.7)[Li_(0.1)Cu_(0.2)Mn_(0.7)]O_(2) cathode delivers a large specific capacity of ~212 mA h g^(-1)at 15 mA g^(-1). The discharge capacity is maintained up to ~90% of the initial capacity after 100 cycles, with stable occurrence of the oxygen redox in the high-voltage region. Through advanced experimental analyses and first-principles calculations, it is confirmed that a stepwise redox reaction based on Cu and O ions occurs for the charge-compensation mechanism upon charging. Based on a concrete understanding of the reaction mechanism, the Li displacement by the synergy of Li-Cu cations plays a crucial role in suppressing the structural change of the P3-type layered material under the oxygen redox reaction, and it is expected to be an effective strategy for stabilizing the oxygen redox in the layered oxides of Na-ion batteries.

Brain Activity Associated with Memory and Cognitive Function During Jaw-Tapping Movement in Healthy Subjects Using Functional Magnetic Resonance Imaging*

Objective： TO determine whether jaw-tapping movement, a classically described as an indication of personal well-being and mental health, stimulates the memory and the cognitive regions of the brain and is associated with improved brain performance. Methods： Twelve healthy right-handed female subjects completed the study. Each patient performed a jaw-tapping task and an n-back task during functional magnetic resonance imaging （fMRI）. The subjects were trained to carry out the jaw-tapping movement at home twice a day for 4 weeks. The fMRI was repeated when they returned. Results： During the first and second jaw-tapping session, both sides of precentral gyrus and the right middle frontal gyrus （BA 6） were activated. And during the second session of the jaw-tapping task, parts of frontal lobe and temporal lobe related to memory function were more activated. In addition, the total percent task accuracy in n-beck task significantly increased after 4 weeks of jaw- tapping movement. After jaw-tapping training for 4 weeks, brain areas related to memory showed significantly increased blood oxygen level dependent signals. Conclusion： Jaw-tapping movement might be a useful exercise for stimulating the memory and cognitive regions of the brain.

To the Editor： A lumbar epidural block is used to treat patients with a herniated intervertebral disc, spinal stenosis, or lumbosacral radiculopathy. This approach reduces radicular pain by decreasing inflammation of nerve roots through epidural corticosteroid injection. Patients with moderate-to-severe spinal stenosis who taking various anticoagulant agents have more risk for epidural hematoma.

Dear Editor,Here, we report the developmental regulation of proteolytic cleavage of the FLOWERING LOCUS T （FT） protein. FT, a mem- ber of the phosphatidylethanolamine-binding protein （PEBP） family, functions as a florigen or a component of florigen. PEBP binds phosphoethanolamine, and its ligand-binding domain is evolutionarily conserved from bacteria to plants and animals （Schoentgen and Jolles, 1995; Bradley et al., 1996）. The difference between visual and instrumental analyses of a shade selection on natural teeth was investigated. With visual analysis, five prosthodontist examined the middle third of the unrestored maxillary right central incisor of a patient using VITAPEN classical and Vita Toothguide 3D-MASTER tooth shade guide. In instrumental analysis, one prosthodontist examined the same teeth using a spectrophotometer （ShadepilotTM, Degudent, Hanau-Wolfgang, Germany, software 2.40）. Overall, instrumental analysis is more accurate and reproducible than a visual assessment. However, the difference is clinically acceptable.

Traditional herbal medicines,Fuling,Baizhu,and Danggui,promote the proliferation of human Schwann cells

Enhancing Schwann cell proliferation may be beneficial for peripheral nerve repair and nerve regeneration. A traditional herbal formula composed of Fuling （poria cocos）, Baizhu （Atractylodes macrocephala）, and Danggui （Angelica sinensis）（FBD） improves neuronal survival and growth, and FBD may promote the secretion of brain-derived neurotrophic factor. However, the mechanism underlying Schwann cell proliferation remains unclear. We tested whether FBD enhanced the proliferation of human Schwann cells. FBD （20 ug/mL） increased Schwann cell viability and survival and increased the number of cells at G2/M and S phases. FBD also increased nerve growth factor and brain-derived neurotrophic factor expression in Schwann cells, with maximum efficacy at 20 ug/mL.
