Polymorphisms of Epstein-Barr virus BHRF1 gene, a homologue of bcl-2

Background and Objective: EBV BamHI fragment H rightward open reading frame 1 (BHRF1), the Epstein-Barr virus (EBV) early gene, is structurally and functionally homologous to the oncogene bcl-2 and may play an important role in the development of EBV-associated tumors. To characterize the polymorphisms of BHRF1 in EBV-associated tumors, we analyzed the sequences of BHRF1 in isolates from nasopharyngeal carcinoma (NPC) and EBV-associated gastric carcinoma (EBVaGC) biopsies as well as throat washing (TW) samples from healthy donors. Methods: BHRF1 DNA sequences were analyzed by polymerase chain reaction (PCR) and sequencing for 39 NPC samples, 40 EBVaGC samples, and 53 EBV-positive TW samples from healthy donors. The variants of BHRF1 gene were classified according to the signature changes. The EBV types 1 and 2 at nuclear antigen (EBNA) 3C locus were determined by PCR. Results: Compared with EBV standard cell line B95-8, all isolates carried a silent mutation at amino acid (AA) 80 (nucleotide 54616 T→C), the AA88 L→V mutation was found in most isolates, and the AA79 V→L mutation in a few isolates. Other mutations were sporadically distributed. Based on the mutations at AA88 and AA79, 3 distinct variants of BHRF1 genes, designated as 79V88V, 79L88L, and 79V88L, were identified. The 79V88V was the most common variant. The distribution of the BHRF1 variants among the NPC, EBVaGC, and TW samples was not significant. The corresponding regions of bcl-2 homologues were conserved in all isolates except for 3 samples. The distribution of BHRF1 variants in type 1 and type 2 strains was significant different (P < 0.001, contingency coefficient was 0.554). Conclusions: The 79V88V is the dominant variant in NPC, EBVaGC, and TW samples from healthy donors and preferential linkages between BHRF1 and EBNA3C variants exist. Conserved BHRF1 in Bcl-2 homologous domains is helpful to remain the important role of BHRF1.

The BEL1-like transcription factor GhBLH5-A05 participates in cotton response to drought stress

Drought stress impairs crop growth and development.BEL1-like family transcription factors may be involved in plant response to drought stress,but little is known of the molecular mechanism by which these proteins regulate plant response and defense to drought stress.Here we show that the BEL1-like transcription factor GhBLH5-A05 functions in cotton(Gossypium hirsutum)response and defense to drought stress.Expression of GhBLH5-A05 in cotton was induced by drought stress.Overexpression of GhBLH5-A05 in both Arabidopsis and cotton increased drought tolerance,whereas silencing GhBLH5-A05 in cotton resulted in elevated sensitivity to drought stress.GhBLH5-A05 binds to cis elements in the promoters of GhRD20-A09 and GhDREB2C-D05 to activate the expression of these genes.GhBLH5-A05 interacted with the KNOX transcription factor GhKNAT6-A03.Co-expression of GhBLH5-A05 and GhKNAT6-A03 increased the transcription of GhRD20-A09 and GhDREB2C-D05.We conclude that GhBLH5-A05 acts as a regulatory factor with GhKNAT6-A03 functioning in cotton response to drought stress by activating the expression of the drought-responsive genes GhRD20-A09 and GhDREB2C-D05.

How the enhanced East Asian summer monsoon regulates total gross primary production in eastern China

Recognizing the relationship between gross primary production(GPP)and precipitation in eastern China,the East Asian Summer Monsoon(EASM)plays a crucial role in shaping GPP.Despite confirmation of the strong link between EASM and GPP,there remains a notable research gap in understanding the specific impact of the EASM on GPP in different regions of eastern China.Here we used simulations from Trends in Net Land-Atmosphere Carbon Exchanges(TRENDY)models from 1951 to 2010 and divided eastern China into five subregions for the study.We also used the New East Asian Summer Monsoon Index(NEWI)as a quantitative metric to distinguish between periods of strong and weak EASM.Building on this,this study aims to investigate the response of GPP in different subregions of eastern China.Regionally,under strengthened EASM years(1954,1957,1965,1969,1977,1980,1983,1987,1993 and 1998),East China experienced the most pronounced increase in GPP at 12±21(mean±1 sigma)gC m^(-2) mon^(-1) compared to the weak EASM years(1958,1961,1972,1973,1978,1981,1985,1994,1997 and 2004).In contrast,Southwest China showed a decline in GPP at-4±10 gC m^(-2) mon^(-1).Moreover,GPP also increased in Northeast and North China when EASM strengthened,while South China showed a decline in GPP.This indicated that GPP changed with monsoon intensity.According to the mechanism analysis,during strong EASM,there was intense moisture convergence through alterations in the atmospheric circulation field over East China and abundant precipitation,which further contributed to the increase in GPP.Downward solar radiation in Southwest China decreased with EASM enhancement,which suppressed GPP and hindered vegetation growth.Overall,the results highlight the importance of accurately predicting the impact of different EASM intensities of regional carbon fluxes.

Objective Hypertriglyceridemic waist(HW),hypertriglyceridemic waist-to-height ratio(HWHtR),and waist-to-hip ratio(WHR)have been shown to be indicators of cardiometabolic risk factors.However,it is not clear which indicator is more suitable for children and adolescents.We aimed to investigate the relationship between HW,HWHtR,WHR,and cardiovascular risk factors clustering to determine the best screening tools for cardiometabolic risk in children and adolescents.Methods This was a national cross-sectional study.Anthropometric and biochemical variables were assessed in approximately 70,000 participants aged 6–18 years from seven provinces in China.Demographics,physical activity,dietary intake,and family history of chronic diseases were obtained through questionnaires.ANOVA,χ2 and logistic regression analysis was conducted.Results A significant sex difference was observed for HWHtR and WHR,but not for HW phenotype.The risk of cardiometabolic health risk factor clustering with HW phenotype or the HWHtR phenotype was significantly higher than that with the non-HW or non-HWHtR phenotypes among children and adolescents(HW:OR=12.22,95%CI:9.54-15.67;HWHtR:OR=9.70,95%CI:6.93-13.58).Compared with the HW and HWHtR phenotypes,the association between risk of cardiometabolic health risk factors(CHRF)clustering and high WHR was much weaker and not significant(WHR:OR=1.14,95%CI:0.97-1.34).Conclusion Compared with HWHtR and WHR,the HW phenotype is a more convenient indicator with higher applicability to screen children and adolescents for cardiovascular risk factors.

BACKGROUND Clear cell sarcoma(CCS)is a rare soft-tissue sarcoma.The most common metastatic sites for CCS are the lungs,bones and brain.CCS is highly invasive and mainly metastasizes to the lung,followed by the bone and brain;however,pancreatic metastasis is relatively rare.CASE SUMMARY We report on a rare case of CCS with pancreatic metastasis in a 47-year-old man.The patient had a relevant medical history 3 years ago,with abdominal pain as the main clinical manifestation.No abnormalities were observed on physical examination and the tumor was found on abdominal computed tomography.Based on the medical history and postoperative pathology,the patient was diagnosed with CCS with pancreatic metastasis.The patient was successfully treated with surgical interventions,including distal pancreatectomy and sple-nectomy.CONCLUSION This report summarizes the available treatment modalities for CCS and the importance of regular postoperative follow-up for patients with CCS.

In this study,we reported the discovery and structure-activity relationship analysis of chrysin derivatives as a new class of inhibitors targeting poly(ADP-ribose)polymerase 1(PARP1).Among these derivatives,compound 5d emerged as the most effective chrysin-based inhibitor of PARP1,with an IC50 value of 108 nmol·L^(-1).This compound significantly inhibited the proliferation and migration of breast cancer cell lines HCC-1937 and MDA-MB-436 by inducing DNA damage.Furthermore,5d induced apoptosis and caused an extended G1/S-phase in these cell lines.Molecular docking studies revealed that 5d possesses a strong binding affinity toward PARP1.In vivo,in a xenograft model,5d effectively reduced tumor growth by downregulating PARP1 expression.Overall,compound 5d shows promise as a potential therapeutic agent for the treatment of BRCA wild-type breast cancer.

Source apportionment and suitability evaluation of seasonal VOCs contaminants in the soil around a typical refining-chemical integration park in China

Accurate source apportionment of volatile organic compounds(VOCs)in soil nearby petrochemical industries prevailing globally,is critical for preventing pollution.However,in the process,seasonal effect on contamination pathways and accumulation of soil VOCs is often neglected.Herein,Yanshan Refining-Chemical Integration Park,including a carpet,refining,synthetic rubber,and two synthetic resin zones,was selected for traceability.Season variations resulted in a gradual decrease of 31 VOCs in soil from winter to summer.A method of dry deposition resistance model coupling partitioning coefficient model was created,revealing that dry deposition by gas phase was the primary pathway for VOCs to enter soil in winter and spring,with 100 times higher fux than by particle phase.Source profiles for five zones were built by gas sampling with distinct substance indicators screened,which were used for positive matrix factorization factors determination.Contributions of the five zones were 14.9%,20.8%,13.6%,22.1%,and 28.6%in winter and 33.4%,12.5%,10.7%,24.9%,and 18.5%in spring,respectively.The variation in the soil sorption capacity of VOCs causes inter-seasonal differences in contribution.The better correlation between dry deposition capacity and soil storage of VOCs made root mean square and mean absolute errors decrease averagely by 8.8%and 5.5%in winter compared to spring.This study provides new perspectives and methods for the source apportionment of soil VOCs contamination in industrial sites.

The as-cast amorphous Ti_(48)Zr_(27)Cu_(6)Nb_(5)Be_(14)composites,comprising in situ formedβ-Ti ductile crystalline precipitates,were prepared by water cooled copper mold suction casting.Then,the semi-solid composites were obtained after the as-cast composites were treated by semi-solid isothermal treatment.The microstructure evolution and kinetics of the composites were examined.Results show that the microstructures of both the as-cast and semi-solid composites comprise ofβ-Ti crystal phases and amorphous matrix phases.Before and after treatment,the crystals evolve from fine granular or fine dendritic crystals to coarse crystals.As the treatment temperature increasing or the time prolonging,the average crystal size gradually increases and the surface morphology of the crystals gradually becomes regular.By studying the microstructural evolution and dynamics during the isothermal treatment process,it is found that the final morphology ofβ-Ti crystals is influenced by the isothermal treatment temperature and time(t),and theβ-Ti evolution rate increases with an increase in treatment temperature.In addition,a linear relationship was observed between the size of cubicβ-Ti crystals(D^(3))and t;the growth kinetics factor K is 3.8μm^(3)·s^(-1).As the K value closes to 4μm^(3)·s^(-1),it is inferred the morphology evolution ofβ-Ti crystals is a coarsening behavior controlled by the diffusion of solute elements.

Cytochrome P45026A1 Contributes to the Maintenance of Neuropathic Pain

The cytochrome P450 proteins(CYP450s)have been implicated in catalyzing numerous important biological reactions and contribute to a variety of diseases.CYP26A1,a member of the CYP450 family,carries out the oxidative metabolism of retinoic acid(RA),the active metabolite of vitamin A.Here we report that CYP26A1 was dramatically upregulated in the spinal cord after spinal nerve ligation(SNL).CYP26A1 was mainly expressed in spinal neurons and astrocytes.HPLC analysis displayed that the content of all-trans-RA(at-RA),the substrate of CYP26A1,was reduced in the spinal cord on day 7 after SNL.Inhibition of CYP26A1 by siRNA or inhibition of CYP26A1-mediated at-RA catabolism by talarozole relieved the SNL-induced mechanical allodynia during the maintenance phase of neuropathic pain.Talarozole also reduced SNL-induced glial activation and proinflammatory cytokine production but increased anti-inflammatory cytokine(IL-10)production.The RA receptors RARα,RXRβ,and RXRγwere expressed in spinal neurons and glial cells.The promoter of Il-10 has several binding sites for RA receptors,and at-RA directly increased Il-10 mRNA expression in vitro.Finally,intrathecal IL-10 attenuated SNL-induced neuropathic pain and reduced the activation of astrocytes and microglia.Collectively,the inhibition of CYP26A1-mediated at-RA catabolism alleviates SNL-induced neuropathic pain by promoting the expression of IL-10 and suppressing glial activation.CYP26A1 may be a potential therapeutic target for the treatment of neuropathic pain.

Glioma-derived ANXA1 suppresses the immune response to TLR3 ligands by promoting an anti-inflammatory tumor microenvironment

A highly immunosuppressive tumor microenvironment(TME)and the presence of the blood‒brain barrier are the two major obstacles to eliciting an effective immune response in patients with high-grade glioma(HGG).Here,we tried to enhance the local innate immune response in relapsed HGG by intracranially injecting poly(I:C)to establish a robust antitumor immune response in this registered clinical trial(NCT03392545).During the follow-up,12/27(44.4%)patients who achieved tumor control concomitant with survival benefit were regarded as responders in our study.We found that the T-cell receptor(TCR)repertoire in the TME was reshaped after poly(I:C)treatment.Based on the RNA-seq analysis of tumor samples,the expression of annexin A1(ANXA1)was significantly upregulated in the tumor cells of nonresponders,which was further validated at the protein level.In vitro and in vivo experiments showed that ANXA1 could induce the production of M2-like macrophages and microglia via its surface receptor formyl peptide receptor 1(FPR1)to establish a Treg cell-driven immunosuppressive TME and suppress the antitumor immune response facilitated by poly(I:C).The ANXA1/FPR1 signaling axis can inhibit the innate immune response of glioma patients by promoting an anti-inflammatory and Treg-driven TME.Moreover,ANXA1 could serve as a reliable predictor of response to poly(I:C),with a notable predictive accuracy rate of 92.3%.In light of these notable findings,this study unveils a new perspective of immunotherapy for gliomas.
