A new outbreak of monkeypox virus(MPV)is currently occurring in several regions of the *** the beginning of May 2022,3040 cases have been reported to the World Health Organization(WHO)*** of its usual“endemic”base in Africa,the virus has so far been detected in 47 countries,including in Europe,America,Oceania,and Asia,where monkeypox is uncommon or previously *** 25 June 2022,the WHO Emergency Committee declared that the outbreak does not currently constitute a Public Health Emergency of International Concern,but unanimously acknowledged the emergency nature of the event and that controlling further spread will require a vigorous ***,new cases of MPV infection have been confirmed in Korea,Singapore,and Taiwan,***,a better understanding of MPV and enhanced surveillance are essential for preventing disease recurrence.

The distribution of the immune system throughout the body complicates in vitro assessments of coronavirus disease 2019(COVID-19)immunobiology,often resulting in a lack of reproducibility when extrapolated to the whole ***,developing animal models is imperative for a comprehensive understanding of the pathology and immunology of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)*** review summarizes current progress related to COVID-19 animal models,including non-human primates(NHPs),mice,and hamsters,with a focus on their roles in exploring the mechanisms of immunopathology,immune protection,and long-term effects of SARS-CoV-2 infection,as well as their application in immunoprevention and immunotherapy of SARS-CoV-2 *** among these animal models and their specific applications are also highlighted,as no single model can fully encapsulate all aspects of *** effectively address the challenges posed by COVID-19,it is essential to select appropriate animal models that can accurately replicate both fatal and non-fatal infections with varying courses and *** animal model libraries and associated research tools is key to resolving the global COVID-19 pandemic,serving as a robust resource for future emerging infectious diseases.

The c-Jun N-terminal kinases （JNKs） are classic stress-activated protein kinases. Many cellular stresses have been shown to stimulate JNK activation. In this review, we focus on ribotoxic stresses based on their multiple biological potencies including anti-HIV-1 activity. Some of the functions of ribotoxins and the signaling transduction pathway that mediated are mentioned. Different from other stimulators, ribotoxic stresses act on special motifs of 28S rRNA in translationally active mammal ribosomes. Binding and damaging on the motif leads to JNK activation and subsequently biological response to the signal initiator, which is named ribotoxic stress response. Laos is the only landlocked country in Southeast Asia and borders Thailand,Myanmar and Cambodia,the three countries in this region that have been hardest hit by human immunodeficiency virus(HIV).Laos has been regarded as a low-HIVprevalence country for *** understand the status of HIV in Laos in recent years,a retrospective study was performed among 2851 patients visiting a hospital in Vientiane,the capital of Laos,from November 2011 to May *** blood samples were obtained from the patients,and DNA was *** status was determined by HIV gag fragment-specific PCR ***-nine samples were detected as HIV proviral DNA positive with a positive rate of 2.4%(69/2851).Sixty-one near full-length genomic sequences were obtained from the positive *** results of phylogenetic analysis showed that the vast majority(91.8%)of the HIV strains belonged to CRF01AE,and the other five(8.2%)strains were identified as a new HIV circulating recombinant form CRF9701B,which had a CRF01AE backbone with an insertion of subtype B in the gag-pol *** analysis revealed that HIV CRF01AE circulating in Laos were multiply introduced from *** results indicated that Laos might be suffering a considerably more serious impact of HIV than previously *** keep this country from undergoing the same increase in HIV prevalence observed in its neighbors,immediate intervention measures and sufficient epidemiological research are urgently needed.

TRIM5α restricts retroviruses in a species-specific manner. Cyclophilin A was independently retrotransposed into the TRIM5 loci in different species, leading to the generation of antiviral TR1M5-cyclophilin A （TRIMCyp） proteins. Previously, we found that assam macaques express a TRIMCyp chimera （amTRIMCyp）, along with a TRIM5α allelic protein （amTRIM5α）. Herein, we investigated the antiviral activity of amTRIMCyp and amTRIM5α individually, as well as their interaction and joint effects. amTRIMCyp showed a divergent restriction pattern from amTRIM5αc Although both proteins potently restricted the replication of HIV-1, only amTRIM5αt inhibited N-MLV. Remarkably, cellular anti-HIV-1 activity increased when amTRIMCyp and amTRIM5αt were coexpressed, indicating a synergistic block of HIV-1 replication. Consistently, PMBCs from heterozygous amTRIM50t/TRIMCyp showed stronger resistance to HIV-1 infection than those from amTRIM5a/TRIM5α homozygotes. The anti-HIV-1 synergistic effect was dependent on the amTRIMCyp-amTRIM5α interaction. In contrast, amTRIMCyp completely abrogated the anti-N-MLV activity mediated by amTRIM5α, showing a dominant-negative effect, indicating that the generation of amTRIMCyp was involved in the trade-off between divergent restriction activities. Our results provide a new paradigm to study functional trade-offs mediated by allelic proteins, a theoretical basis for utilizing animal models with various TRIM5 alleles, as well as novel HIV-1 gene therapy strategies.
