Development of Quantitative Real-time Polymerase Chain Reaction for the Detection of Vibrio vulnificus Based on Hemolysin (vvhA) Coding System

Objective To establish a TaqMan real-time fluorescent quantitative PCR to detect Vibrio vulnificus based on the hemolysin gene （vvhA） coding cytolysin. Methods Primers and probes in the conserved region of the vvhA gene sequence were designed for the TaqMan real-time PCR to detect 100 bp amplicon from V. vulnificus DNA. Recombinant plasmid pMD19-vvhA100 was constructed and used as a positive control during the detection. Minimal amplification cycles （Ct value） and fluorescence intensity enhancement （ARn value） were used as observing indexes to optimize the reaction conditions of TaqMan real-time PCR. The TaqMan assay for the detection of Vbirio vulnificus was evaluated in pure culture, mice tissue which artificially contaminated Vibrio vulnificus and clinical samples. Results The established TaqMan real-time PCR showed positive results only for Vibrio vulnificus DNA and pMD19-vvhA100. The standard curve was plotted and the minimum level of the vvhA target from the recombinant plasmid DNA was 103 copies with a Ct value of 37.94±0.19, as the equivalent of 0.01 ng purified genomic DNA of Vibrio vulnificus. The results detected by TaqMan PCR were positive for the 16 clinical samples and all the specimens of peripheral blood and subcutaneous tissue of mice which were infected with Vibrio vulnificus. Conclusion TaqMan real-time PCR is a rapid, effective, and quantitative tool to detect Vibro vulnificus, and can be used in clinical laboratory diagnosis of septicemia and wound infection caused by Vibrio vulnificus.

Background:Quick diagnosis of smear-negative pulmonary tuberculosis(TB)and extra-pulmonary TB are urgently needed in clinical *** research aims to investigate the usefulness of the interferon-γrelease assay(IGRA)for the diagnosis of smear-negative pulmonary and extra-pulmonary ***:We performed TB antibody and TB-IGRA tests on 389 pulmonary TB patients(including 120 smear-positive pulmonary TB patients and 269 smear-negative pulmonary TB patients),113 extra-pulmonary TB patients,81 patients with other pulmonary diseases and 100 healthy *** samples for the TB-Ab test and the TB-IGRA were collected,processed,and interpreted according to the manufacturer’s ***:The detection ratio of smear-positive pulmonary TB patients and smear-negative pulmonary TB patients were 90.8%(109 of 120)and 89.6%(241 of 269),*** was no statistically significant difference of its performance between these two sample sets(P>0.05).The detection ratio of positive TB patients and extra-pulmonary TB patients were 90.0%(350 of 389)and 87.6%(99 of 113),respectively,which was not significantly different(P>0.05).Conclusions:In this work,the total detection ratio using TB-IGRA was 89.4%,therefore TB-IGRA has diagnostic values in smear-negative pulmonary TB and extra-pulmonary TB diagnosis.

30 and 32 kDa outer membrane proteins of Bordetella pertussis as a modulator on promoting degranulation of mast cells

The correlation between the activities of the outer menbrane proteins （OMPs） of Bordetella pertussis and the lgE-mediated asthma was investigated in the present study, in which the OMPs of B. pertussis and their components were prepared by detergent treatment and chromatography, and the molecular weights of the OMPs components were determined by SDS-PAGE. The amounts of total as well as the ovalbumin （OVA）-specific IgE induced by dead B. pertussis whole bacterial vaccine on guinea pigs were detected by ELISA. Meanwhile, the effect of the OMPs and their components to promote the degranulation of guinea pig mast cells was observed by using the mast cell degranulation test, and ELISA assay was used to measure the histatmine levels in the supematants from the mast cell cultures. Histamine sensitive test was used to demonstrate the effects of the OMPs and their components to increase the histamine lethal sensitivity in mice. It was found that four components with molecular weights of 30, 32, 38 and 69 kDa could be obtained from the OMPs of B. pertussts, and the dead whole bacteria vaccine of B. pertussis had the ability to increase the levels of the total as well as the OVA-specific IgE in sera of guinea pigs. The OMPs and their 30 and 32 kDa components demonstrated significantly enhancing effect on the degranulation of guinea pig mast cells, and the histamine levels in the supematants from the mast cell culture treated with OMPs and their 30 and 32 kDa components were also significantly increased. It is evident that the strong adjuvant activity and the enhancing effect to degranulation of mast cells and the release of histamine of certain outer membrane components of B. pertussis could be demonstrated as revealed by the results of the present study, suggesting the possibility of a close relationship between the infection of vaccination with B. pertussis and the IgE-mediated asthma.

Objective:Laboratory diagnosis of neurosyphilis(NS)remains a great *** study was the aimed to identify miRNA candidates as biomarkers to distinguish between NS,non-neurosyphilis,and healthy controls(HCs).Methods:We analyzed miRNA expression profiles in peripheral blood mononuclear cells(PBMCs)from six patients with NS,eight patients with secondary syphilis(SS),and five HCs using microarray *** differentially expressed miRNAs were validated in 33 NS samples,31 SS samples,and 30 HC samples using TaqMan miRNA real-time qPCR(qRT-PCR).Results:Thirty-nine miRNAs were differentially expressed in SS and NS patients compared with *** miRNAs were randomly selected to validate their expression levels in the same samples used in microarray assay by *** miRNAs were upregulated in SS and NS samples compared with ***-PCR analysis of the expression of the 13 miRNAs in a second cohort(76 samples)showed that the average expression levels of nine miRNAs were higher in SS than in NS(SS:0.185,NS:0.136,P=3.8E-10),while the expressions of the other four miRNAs were lower in SS than in NS(SS:0.000757,NS:0.000873,P=0.022).ROC curve analysis of the 13 miRNAs showed the area under the curve value to be 1.00 for distinguishing SS patients from HCs,1.00 for distinguishing NS patients from HCs,1.00 for distinguishing SS and NS patients from HCs,and 0.968 for distinguishing NS from SS ***:The present study is the first one that identified differentially expressed miRNAs in PBMCs from patients with *** results suggest that the 13 candidate miRNAs in PBMCs may be novel noninvasive biomarkers for NS diagnosis.

AIM: To determine the frequencies of HGV and TTV infectionsin serum and saliva samples of non-hepatitis patients withoral diseases in Hangzhou area, and to understand thecorrelation between detected results of HGV RNA and/or TTVDNA in sera and in saliva from the same patientsMETHODS: RT-nested PCR for HGV RNA detection andsemi-nested PCR for nv DNA detection were performed inthe serum and saliva samples from 226 non-hepatitis patientswith oral diseases, and nucleotide sequence ***: Twenty-seven (11.9 %) and 21 (9.3 %) of the226 serum samples were only positive for HGV RNA andlrv DNA, respectively. 10 (4.4 %) and 9 (3.9 %) of the226 saliva samples were only positive for HGV RNA andTTV DNA, respectively. And 7 (3.1%) of the serum samplesand 2 (0.9 %) of the saliva samples showed the positiveamplification results for both HGV RNA and Irv DNA. 12saliva samples from the 34 patients (35.3 %) with HGV orHGV/TTV viremia and 11 saliva samples from the 28 patients(39.3 %) with TTV or HGV/TTV viremia were HGV RNAdetectable, respectively, including two patients positive forboth HGV RNA and TTV DNA in serum and saliva *** saliva samples from the 226 patients were found to beHGV RNA or nv DNA detectable while their serum sampleswere negative for HGV or TTV. Homologies of the nucleotidesequences of HGV and TTV amplification products from theserum and saliva samples of the two patients comparedwith the reported sequences were 88.65-91.49 % and65.32-66.67 %, respectively. 目的:通过测量术眼角膜瓣厚度,观察准分子手术制瓣方式的安全性及可预测性。为临床工作提供帮助。方法:将手术眼按照制瓣方式分为四组:飞秒制瓣组(A组)、Moria-Ⅱ90刀制瓣组(B组)、Moria-Ⅱ110刀制瓣组(C组)、EK制瓣组(D组)。用A超测量角膜及基质床厚度,计算得出实际瓣厚度。所得数据进行统计学分析。结果:制瓣平均厚度A组90.95±10.82μm,B组133.61±11.97μm,C组147.70±16.16μm,D组66.35±11.95μm,各组瓣厚统计学差异明显。制瓣厚度影响因素中,A组与等效球镜低度相关。B组与等效球镜中度相关,与术中档位、术前眼压、平均曲率低度相关。D组与制瓣方法中度相关,与等效球镜低度相关。A组锥镜两次使用;B,C组旋转刀两次使用;D组左右眼制瓣厚度无显著差异。结论:几组制瓣均很稳定,但在制瓣厚度上有明显差异。屈光度数可能是影响制瓣厚度的因素之一。飞秒激光制瓣比常规板层角膜刀在预测性和灵活性方面更有优势。

Role of the globus pallidus in motor and non-motor symptoms of Parkinson's disease

The globus pallidus plays a pivotal role in the basal ganglia circuit. Parkinson's disease is characterized by degeneration of dopamine-producing cells in the substantia nigra, which leads to dopamine deficiency in the brain that subsequently manifests as various motor and non-motor symptoms. This review aims to summarize the involvement of the globus pallidus in both motor and non-motor manifestations of Parkinson's disease. The firing activities of parvalbumin neurons in the medial globus pallidus, including both the firing rate and pattern, exhibit strong correlations with the bradykinesia and rigidity associated with Parkinson's disease. Increased beta oscillations, which are highly correlated with bradykinesia and rigidity, are regulated by the lateral globus pallidus. Furthermore,bradykinesia and rigidity are strongly linked to the loss of dopaminergic projections within the cortical-basal ganglia-thalamocortical loop. Resting tremors are attributed to the transmission of pathological signals from the basal ganglia through the motor cortex to the cerebellum-ventral intermediate nucleus circuit. The cortico–striato–pallidal loop is responsible for mediating pallidi-associated sleep disorders. Medication and deep brain stimulation are the primary therapeutic strategies addressing the globus pallidus in Parkinson's disease. Medication is the primary treatment for motor symptoms in the early stages of Parkinson's disease, while deep brain stimulation has been clinically proven to be effective in alleviating symptoms in patients with advanced Parkinson's disease,particularly for the movement disorders caused by levodopa. Deep brain stimulation targeting the globus pallidus internus can improve motor function in patients with tremordominant and non-tremor-dominant Parkinson's disease, while deep brain stimulation targeting the globus pallidus externus can alter the temporal pattern of neural activity throughout the basal ganglia–thalamus network. 研究重组创伤弧菌溶细胞素融合蛋白(rVvhA)诱导人脐静脉内皮细胞(human umbili-cal vein endothelial cells,human ECV304)凋亡的作用。诱导含pET-28a(+)vvhA重组质粒的BL21大肠杆菌表达创伤弧菌溶细胞素融合蛋白,应用Ni2+亲和层析方法对rVvhA进行纯化;利用分步稀释加透析相结合的方法进行蛋白质复性;绵羊红细胞裂解试验对复性后的rVvhA进行溶血活性初步测定;MTT法检测rVvhA对人ECV304细胞的体外毒性作用;应用Hochest33342/PI活细胞荧光双染及流式细胞术分析rVvhA对人ECV304细胞诱导凋亡的影响。结果显示,用Ni2+-NTAHisBand亲和层析柱纯化rVvhA纯度达88.6%左右;复性后的rVvhA有一定的溶血活性,其溶血活性具有时间-剂量依赖性;MTT结果显示rVvhA具有降低人ECV304细胞的存活率活性;浓度为2.0HU/mlrVvhA作用人ECV30412h后,其诱导凋亡的活性高于对照组和浓度为0.5HU/mlrVvhA处理组,具有剂量依赖性;浓度为2.0HU/mlrVvhA处理组加用40μmol/Lcaspase全酶抑制剂(Z-VAD-FMK)后凋亡率较2.0HU/mlrVvhA处理组有一定程度降低。rVvhA对人ECV304细胞具有诱导凋亡的生物学活性,推测诱导调亡途径可能与caspase家族有关。
