HsfAld, a Protein Identified via FOX Hunting Using Thellungiella salsuginea cDNAs Improves Heat Tolerance by Regulating Heat-Stress-Responsive Gene Expression

Thellungiella salsuginea （formerly T. halophila）, a species closely related to Arabidopsis （Arabidopsis thali-ana）, is tolerant not only to high salt levels, but also to chilling, freezing, and ozone. Here, we report that T. salsuginea also shows greater heat tolerance than Arabidopsis. We identified T. salsuginea HsfAld （TsHsfAld） as a gene that can confer marked heat tolerance on Arabidopsis. TsHsfAld was identified via Full-length cDNA Over-eXpressing gene （FOX） hunt-ing from among a collection of heat-stress-related T. salsuginea cDNAs. Transgenic Arabidopsis overexpressing TsHsfAld showed constitutive up-regulation of many genes in the Arabidopsis AtHsfA1 regulon under normal growth tempera-ture. in Arabidopsis mesophyll protoplasts, TsHsfAld was localized in both the nucleus and the cytoplasm. TsHsfAld also interacted with AtHSP90, which negatively regulates AtHsfAls by forming HsfA1-HSP90 complexes in the cytoplasm. It is likely that the partial nuclear localization of TsHsfAld induced the expression of the AtHsfAld regulon in the transgenic plants at normal temperature. We also discovered that transgenic Arabidopsis plants overexpressing AtHsfAldwere more heat-tolerant than wild-type plants and up-regulated the expression of the HsfAld regulon, as was observed in TsHsfAld-overexpressing plants. We propose that the products of both TsHsfAld and AtHsfAld function as positive regulators of Arabidopsis heat-stress response and would be useful for the improvement of heat-stress tolerance in other plants.

Symptomatic COVID-19 in University Students: A School-Wide Web-Based Questionnaire Survey during the Omicron Variant Outbreak

Aim: To detect risk and preventive factors associated with the Omicron variant infection in university students, a combination of a web-based survey and multivariate logistic regression analysis was introduced as the front-line initiatives by the school health practitioners. Design: Questionnaire survey. Methods: The school-wide web-based questionnaire survey was conducted among our university students as a part of the annual health check-up in April, 2023. The positive outcome was confined to the first symptomatic COVID-19 onset during the Omicron variant outbreak. Results: In this self-administered survey, risk or protective associations were merely estimated statistically in university students (n = 5406). In measured factors, karaoke and club/group activities could maintain the statistical significance in adjusted odds ratios (ORs) as relative risk factors, and science course, measles/ rubella (MR) vaccination, and COVID-19 vaccination remained as relative protective factors in adjusted OR analyses. Club/group activities with member gathering and karaoke sing-along sessions in university students may frequently have WHO’s three Cs. These risk factors are still important topics for the infection control of COVID-19 in university students. Together with some recent reports from other researchers, the significant protective role of MR vaccine in our survey warrants further clinical investigation. If the breakthrough infection continuously constitutes the majority of infection, real data in test-negative case-control or web-based questionnaire design continue to be important for statistical analysis to determine the minimal requirement of our strategies which may be equivalent to or replace COVID-19 vaccines.

In recounting Dr.James M.Tiedje's outstanding research achievements spanning the past 55 years,it is easy to overlook his early and mid-career endeavors.Specifically,his contribution to the aerobic degradation of pesticides and other chemicals,as well as methanogenic degradation of those compounds retains their brilliance.Many researchers in environmental microbiology have gained invaluable knowledge from these studies,which have been applied to the elucidation of previously uncultivated microorganisms.

Characteristics of glucose change in diabetes mellitus generalized through continuous wavelet transform processing:A preliminary study

BACKGROUND The continuous glucose monitoring(CGM)system has become a popular evaluation tool for glucose fluctuation,providing a detailed description of glucose change patterns.We hypothesized that glucose fluctuations may contain specific information on differences in glucose change between type 1 diabetes mellitus(T1DM)and type 2 diabetes mellitus(T2DM),despite similarities in change patterns,because of different etiologies.Unlike Fourier transform,continuous wavelet transform(CWT)is able to simultaneously analyze the time and frequency domains of oscillating data.AIM To investigate whether CWT can detect glucose fluctuations in T1DM.METHODS The 60-d and 296-d glucose fluctuation data of patients with T1DM(n=5)and T2DM(n=25)were evaluated respectively.Glucose data obtained every 15 min for 356 d were analyzed.Data were assessed by CWT with Morlet form(n=7)as the mother wavelet.This methodology was employed to search for limited frequency glucose fluctuation in the daily glucose change.The frequency and enclosed area(0.02625 scalogram value)of 18 emerged signals were compared.The specificity for T1DM was evaluated through multiple regression analysis using items that demonstrated significant differences between them as explanatory variables.RESULTS The high frequency at midnight(median:75 Hz,cycle time:19 min)and middle frequency at noon(median:45.5 Hz,cycle time:32 min)were higher in T1DM vs T2DM(median:73 and 44 Hz;P=0.006 and 0.005,respectively).The area of the>100 Hz zone at midnight to forenoon was more frequent and larger in T1DM vs T2DM.In a day,the lower frequency zone(15-35 Hz)was more frequent and the area was larger in T2DM than in T1DM.The threedimensional scatter diagrams,which consist of the time of day,frequency,and area of each signal after CWT,revealed that high frequency signals belonging to T1DM at midnight had a loose distribution of wave cycles that were 17-24 min.Multivariate analysis revealed that the high frequency signal at midnight could characterize T1DM(odds ratio:1.33,95%confidence i

Saturated hydraulic conductivity of compacted bentonite-sand mixtures before and after gas migration in artificial seawater

To understand the self-healing property of an engineered barrier for radioactive waste disposal,the hydraulic conductivity of compacted bentoniteesand mixtures saturated with artificial seawater(SW)before and after gas migration was examined.Na-and Ca-bentonites were mixed with fine sand at a ratio of 70%bentonite in dry weight.Two aspects were considered during the experiment:the hydraulic conductivity of the specimen that was resaturated after gas migration and the distribution of water content immediately after gas migration to study gas migration pathways.The gas migrated through the entire cross-section of the specimen,and gas breakthrough occurred in the equilibrium swelling pressure range approximately.Subsequently,the gas flow rate reached a sufficient large value when the gas pressure was approximately twice the equilibrium axial pressure(the sum of swelling and confining pressures),which excluded the back pressure.Although the gas migration pathway was not visible when the specimen was observed immediately after gas migration,the water content distribution showed that several parts of the specimen with lower water content were connected in the direction of gas migration.After resaturation,the change in permeability was within a limited rangedtwo to three times larger than that before gas migration for each type of bentonite in SW.This slight change suggests that gas migration creates a pore structure that cannot be sealed via crystalline swelling of montmorillonite in SW,even if highly compacted bentonite is used under a constant-volume condition.

Now and future of mouse mutagenesis for human disease models

One of the major objectives of the Human Genome Project is to understand the biological function of the gene and genome as well as to develop clinical applications for human diseases. For this purpose, the experimental validations and preclinical trails by using animal models are indispensable. The mouse （Mus musculus） is one of the best animal models because genetics is well established in the mouse and embryonic manipulation technologies are also well developed. Large-scale mouse mutagenesis projects have been conducted to develop various mouse models since 1997. Originally, the phenotype-driven mutagenesis with N-ethyl-N-nitrosourea （ENU） has been the major efforts internationally then knockout/conditional mouse projects and gene-driven mutagenesis have been following. At the beginning, simple monogenic traits in the experimental condition have been elucidated. Then, more complex traits with variety of environmental interactions and gene-to-gene interactions （epistasis） have been challenged with mutant mice. In addition, chromosomal substitution strains and collaborative cross strains are also available to elucidate the complex traits in the mouse. Altogether, mouse models with mutagenesis and various laboratory strains will accelerate the studies of functional genomics in the mouse as well as in human.

Plants have the distinctive 3D spatial structure that varies among organs,species and communities,and the spatial structure changes as they interact with their environments.The functions linked to fundamental biological activities such as transpiration,photosynthesis,and growth are also affected by the spatial structure and the environment.In order to promote smart farming using information and communication technology(ICT),it is necessary to measure and utilize information at the cell-organ of plants to the individual and the community levels and the environments in two or even three dimensions.Therefore,this paper introduced the outline of remote sensing of plant functioning and examples of the 3D remote sensing from relatively short distances using drones and ground Lidar.The quality control of rice in the paddy field and chlorophyll fluorescence imaging for photosynthetic diagnosis were also introduced.In addition,a field smart farm and a smart greenhouse,which heavily utilize ICT,built at Takasaki University of Health and Welfare in Gunma,Japan,were also introduced.

Degradation behaviors of nonylphenol ethoxylates by isolated bacteria using improved isolation method

Nonylphenol ethoxylate (NPEO)-degrading bacteria were isolated from activated sludge using an improved isolation method, and the corresponding degradation behaviours were investigated. Eight NPEO-degrading strains distributed in genera Pseudomonas, Sphingomonas, Sphingobium, Cupriavidus, Ralstonia, Achromobacter, and Staphylococcus were acquired. Spectroscopic Determination of C,N,and O Abundances of Solar-analog Stars Based on the Lines of Hydride Molecules

Photospheric C,N,and O abundances of 118 solar-analog stars were determined by applying the synthetic-fitting analysis to their spectra in the blue or near-UV region comprising lines of CH,NH,and OH molecules,with an aim of clarifying the behaviors of these abundances in comparison with[Fe/H].It turned out that,in the range of-0.6■[Fe/H]■+0.3,[C/Fe]shows a marginally increasing tendency with decreasing[Fe/H]with a slight upturn around[Fe/H]~0,[N/Fe]tends to somewhat decrease toward lower[Fe/H],and[O/Fe]systematically increases (and thus[C/O]decreases) with a decrease in[Fe/H].While these results are qualitatively consistent with previous determinations mostly based on atomic lines,the distribution centers of these[C/Fe],[N/Fe],and[O/Fe]at the near-solar metallicity are slightly negative by several hundredths of dex,which is interpreted as due to unusual solar abundances possibly related to the planetary formation of our solar system.However,clear anomalies are not observed in the[C,N,O/Fe]ratios of planet-host stars.Three out of four very Be-deficient stars were found to show anomalous[C/Fe]or[N/Fe]which may be due to mass transfer from the evolved companion,though its relation to the Be depletion mechanism is still unclear.

Measurements of the Ca II infrared triplet lines of young stellar objects

Equivalent widths and line widths of Ca II infrared triplet emission lines were measured in the high-resolution optical spectra of 39 young stellar objects.We found that the equivalent widths of the emission lines decrease with stellar evolution.It has often been claimed that strong chromospheric activity is generated by a dynamo process caused by fast rotation of the photosphere.However,we found no clear correlation between the strength of the Ca II lines and the stellar rotation velocity.Instead,we found that the objects with high mass accretion rates had stronger Ca II emission lines.This correlation supports the turbulent chromosphere model or the magnetic accretion theory for classical T Tauri stars.We also noticed that the equivalent widths of Ca II lines in transitional disk objects are one-tenth of those in classical T Tauri stars,even if the masses of the circumstellar disks are comparable.
