The ageing process is a systemic decline from cellular dysfunction to organ degeneration,with more predisposition to deteriorated disorders.Rejuvenation refers to giving aged cells or organisms more youthful characteristics through various techniques,such as cellular reprogramming and epigenetic regulation.The great leaps in cellular rejuvenation prove that ageing is not a one-way street,and many rejuvenative interventions have emerged to delay and even reverse the ageing process.Defining the mechanism by which roadblocks and signaling inputs influence complex ageing programs is essential for understanding and developing rejuvenative strategies.Here,we discuss the intrinsic and extrinsic factors that counteract cell rejuvenation,and the targeted cells and core mechanisms involved in this process.Then,we critically summarize the latest advances in state-of-art strategies of cellular rejuvenation.Various rejuvenation methods also provide insights for treating specific ageing-related diseases,including cellular reprogramming,the removal of senescence cells(SCs)and suppression of senescence-associated secretory phenotype(SASP),metabolic manipulation,stem cells-associated therapy,dietary restriction,immune rejuvenation and heterochronic transplantation,etc.The potential applications of rejuvenation therapy also extend to cancer treatment.Finally,we analyze in detail the therapeutic opportunities and challenges of rejuvenation technology.Deciphering rejuvenation interventions will provide further insights into anti-ageing and ageing-related disease treatment in clinical settings.

Dear Editor,Sebaceous glands(SG)contribute to 90%of the skin surface lipid production and play critical roles in regulating proper skin functions including epidermal barrier maintenance,dermal immune responses,and global body anti-microbial activities.1 Reduced sebaceous lipid has been implicated in diverse skin diseases,including scarring alopecia,inflammation,chronic itch,as well as premature aging of the skin.2 Especially,scar repair after deep burns or large-scale skin defect often leads to irreversible loss of appendages.Due to lacking SG,the regenerated tissues cannot fully recapitulate the structural and functional integrity of normal skin.Therefore,generation of expandable SG cells with developmental and lipid-producing potentials represents a promising approach for treating diseased,damaged,or aged skin with better quality of life.Recently,direct cellular reprogramming has been widely used for obtaining difficult-to-maintain human cells by cell type-specific transcription factors(TFs).

针对我国持续提升风电消纳能力和实现可持续发展的目标,结合电力现货市场建设需求,研究和提出了考虑风电时空特性和相对负荷功率波动对系统运行影响的风电平衡成本分摊方法。借助“等电量-顺负荷”方法构造了零风电平衡成本等效场景,对系统进行日前优化调度,通过对比预测场景和等效场景优化调度的系统总运行成本,求解调度周期风电总平衡成本;在考虑各风电场空间分布及容量的基础上,建立了各风电场间的平衡成本分摊模型;提出了定量描述风电相对负荷波动功率时序演进特征的指标即分时段相对波动率,并给出相应的模型;基于分时段相对波动率获取局部波动影响因子对风电调度周期总平衡成本逐时段进行分摊,得到风电分时段平衡成本。以某含高比例风电地区网架结构为例,对不同风电并网容量、不同风电波动程度系统的分时段平衡成本进行了测算,验证了所提计算方法和模型的有效性。

近废型稠油油藏难以通过常规开发方式有效动用剩余储量,火驱技术为此类油藏进一步提高采收率提供方向。现阶段,火驱技术已趋于成熟,但由于成本投入大、经济性差,火驱技术的推广应用面临困境。通过引入量本利分析法中的盈亏平衡模型和敏感性分析,从多维度的经营视角,揭示不同油价年产油量和成本的平衡关系,明确影响火驱经济性的关键指标,进而推动生产运行优化和决策效能提升。研究结果表明,由于储层前后认识差距大,投入力度大于产出力度,同时管控方向不明确,导致火驱成本有效性低,持续开发经营风险认识不足,影响油藏开发投入决策。研究结果为火驱的经营效益提升指明方向,也为近废型稠油油藏效益开发提供管理新思路。

Myoclonus dystonia syndrome(MDS)is an inherited movement disorder,and most MDS-related mutations have so far been found in theε-sarcoglycan(SGCE)coding gene.By generating SGCE-knockout(KO)and human 237 C>T mutation knock-in(KI)mice,we showed here that both KO and KI mice exerted typical movement defects similar to those of MDS patients.SGCE promoted filopodia development in vitro and inhibited excitatory synapse formation both in vivo and in vitro.Loss of function of SGCE leading to excessive excitatory synapses that may ultimately contribute to MDS pathology.Indeed,using a zebrafish MDS model,we found that among 1700 screened chemical compounds,Vigabatrin was the most potent in readily reversing MDS symptoms of mouse disease models.Our study strengthens the notion that mutations of SGCE lead to MDS and most likely,SGCE functions to brake synaptogenesis in the CNS.

Isolation rearing(IR) enhances aggressive behavior, and the central serotonin(5-hydroxytryptamine,5-HT) system has been linked to IR-induced aggression.However, whether the alteration of central serotonin is the cause or consequence of enhanced aggression is still unknown. In the present study, using mice deficient in central serotonin Tph2-/-and Lmx1 b-/-, we examined the association between central serotonin and aggression with or without social isolation. We demonstrated that central serotonergic neurons are critical for the enhanced aggression after IR. 5-HT depletion in wild-type mice increased aggression. On the other hand, application of 5-HT in Lmx1 b-/-mice inhibited the enhancement of aggression under social isolation conditions. Dopamine was downregulated in Lmx1 b-/-mice. Similar to 5-HT, L-DOPA decreased aggression in Lmx1 b-/-mice. Our results linkthe serotoninergic system directly to aggression and this may have clinical implications for aggression-related human conditions.

目的对一起婚宴引起的副溶血性弧菌食源性疾病暴发事件进行流行病学调查,查明可疑食品、致病因子及污染来源,为防控此类事件提供依据。方法采用现场流行病学、食品卫生学、实验室检测和病例对照分析等方法,调查事件发生的原因,并采取相应的防控措施。结果共调查疑似病例20例,临床表现以腹泻、腹痛、发热、呕吐为主。最短潜伏期为4 h,最长潜伏期为35.5 h,平均潜伏期为16 h。3份病例的肛拭标本副溶血弧菌阳性,脉冲电场凝胶电泳进行图谱比对分析,发现3株菌株同源。病例对照分析表明可疑食品为香酥脆皮咸蹄膀。卫生学调查显示,熟制后的香酥脆皮咸蹄膀改刀时共用了粗加工生海鲜的砧板。结论该事件是一起生熟厨具不分致食品污染副溶血性弧菌引起的食源性疾病暴发事件。建议餐饮单位加强食品卫生安全宣教,生熟用具予以明显区分标识,防止交叉污染,并积极落实其他各项食品安全措施。

目的:采用静息态功能磁共振成像方法,探究精神分裂症患者脑动态活动特征。方法:招募43例符合精神障碍诊断与统计手册第4版(DSM-IV)诊断标准的精神分裂症患者和31例正常对照,每个被试均接受静息态功能磁共振成像扫描,采用阳性与阴性症状量表评估患者症状严重程度,DPABI软件用于计算计算动态局部一致性(dReHo)和动态功能连接(dFC),采用格兰杰因果分析(GCA)计算dReHo显著脑区和全脑其他脑区之间效应连接,采用独立样本t检验比较两组被试dReHo和dFC的组间差异。结果:与正常对照组相比,精神分裂症患者左侧中央后回(P<0.01,团簇水平FWE校正)dReHo下降;患者左侧中央后回和左侧额中回、左内侧额上回、右侧锯状裂、左内侧旁扣带回、右侧补充运动区之间的dFC高于正常对照组(P<0.01);患者从左侧中央后回向右侧豆状壳核的效应连接下降(P<0.02,团簇水平FWE校正)。结论:精神分裂症患者中央后回动态功能连接和效应连接均存在异常。

为探究不同生育期不同程度复合高温干旱胁迫对夏玉米生长发育及产量形成的影响,以夏玉米品种“登海685”为试验材料,于2022年6—9月在衡水农业气象试验站开展夏玉米高温干旱田间胁迫试验。在拔节—抽雄和抽雄—乳熟两个生育期分别进行试验。干旱处理设置为轻旱和重旱两个水平。通过增温箱分别在两个生育期进行高温处理,这样每个生育期有轻旱、轻旱高温、重旱、重旱高温四种胁迫处理,各处理有两个重复。此外设置全生育期增温无旱处理和对照组,共10个处理,对株高、叶面积、干物质重及产量构成要素进行观测。结果显示,夏玉米全生育期增温会增加株高和叶面积,但会降低干物质积累导致产量下降;夏玉米在拔节—抽雄和抽雄—乳熟期发生严重的干旱可显著降低株高,但干旱复合高温后影响程度变弱;两个时期发生高温复合干旱与单一干旱相比对玉米叶面积的降低程度更明显;两个时期发生高温干旱胁迫对玉米植株干物质的降低主要由果实干物质降低带来;这两个阶段高温干旱胁迫均会影响产量形成,但拔节—抽雄期干旱对产量形成的影响更大,而抽雄—乳熟期高温的影响更大。
